Electron tunneling spectroscopy has been used to characterize the degradation of beta-D-fructose after electron bombardment in a scanning electron microscope. The decrease in intensity of various vibrational bands is correlated with structural changes in the molecule, thereby providing a detailed picture of the degradation process.  相似文献

Purpose

Humic substances, which are integral components of total organic carbon (TOC), influence soil quality. The study aimed to investigate whether humic and non-humic fractions exhibit early, consistent, and measurable changes and affect TOC sensitivity and storage in a tropical sandy loam soils amended with corn cob biochar.

Materials and methods

There were four treatments with four replicates established in a randomized complete block design. Composite soil samples were taken from plots without biochar (CT), from plots incorporated with 15 t biochar ha?1 (BC-15), and 30 t biochar ha?1 without or with phosphate fertilizer (BC-30 and BC-30+P). The TOC, and humin, humic acid (HA), and fulvic acid (HA) fractions of soil organic carbon were determined for each treatment. The optical densities (400–700 nm) were measured on the soil-free extracts by spectrophotometry; the densities measured at 465 and 665 nm were used to calculate the E465/E665 ratios.

Results and discussion

The BC-30 and BC-30+P plots recorded the highest TOC, humin, humic acid (HA), and fulvic acid (FA) contents with respect to the lowest in the CT. The total exchangeable carbon stratification was significantly higher in all the biochar-treated plots relative to the CT. Spectral analysis showed higher values of E465/E665 (5.02 and 5.15) in the CT and BC-15-treated soils, respectively, compared with the BC-30 and BC-30+P-amended soils with E465/E665 ratios of 2.76 and 2.98, respectively.

Conclusions

Corn cob biochar applied to a tropical sandy loam:

? increased the concentrations of HA and FA and led to increased stratification of TOC, with a stronger effect on HA compared with FA;

? significantly lowered E465/E665 at the high biochar application rate of 30 t ha?1, implying the dominance of high molecular weight humic acid-like substances, and increased degree of aromaticity of the TOC.

Phosphate rock (PR) is an alternative fertilizer to increase the P content of P-deficient weathered soils. We evaluated the effects of fertilizer form on indicators of biological cycling of P using an on-farm trial on a Rhodic Kandiudox in western Kenya. Treatment plots were sampled after 13 cropping seasons of P applications as Minjingu phosphate rock (PR) or as triple super phosphate (?TSP) (50 kg P ha?1 season?1), as well as a P-unfertilized control (0 kg P ha?1 season?1). Soils (0–15 and 15–30 cm) were analyzed for microbial biomass P (Pmic), activities of acid phosphomonoesterase, alkaline phosphomonoesterase, and phosphodiesterase, and sequentially extractable P fractions. P additions as Minjingu PR yielded 299% greater Pmic than TSP at 0–15-cm depth despite similar labile P concentrations in the two P fertilization treatments and stimulated activities of acid phosphomonoesterase (+39%). When added in the soluble form of TSP, a greater percentage of total soil P was present in mineral-bound forms (+33% Fe- and Al-associated P). Higher soil pH under Minjingu PR (pH 5.35) versus TSP (pH 5.02) and the P-unfertilized treatment (pH 4.69) at 0–15-cm depth reflected a liming effect of Minjingu PR. The form of P fertilizer can influence biological P cycling in weathered soils, potentially improving P availability under Minjingu PR relative to TSP via enhanced microbial biomass P and enzymatic drivers of P cycling.  相似文献

We quantified fluctuations in the status of individual patches (wetlands) in supporting connectivity within a network of playas, temporary wetlands of the southern Great Plains of North America that are loci for regional biodiversity. We used remote sensing imagery to delineate the location of surface waters in >8,000 playa basins in a ~31,900 km2 portion of Texas and quantified connectivity in this region from 2007 to 2011. We ranked playas as stepping-stones, cutpoints, and hubs at different levels of environmental conditions (regionally wet, dry, and average periods of precipitation) for dispersal distances ranging from 0.5 to 34 km, representing a range of species’ vagilities, to provide baseline dynamics within an area likely to experience disrupted connectivity due to anthropogenic activities. An individual playa’s status as a stepping-stone, cutpoint, or hub was highly variable over time (only a single playa was a top 20 stepping-stone, cutpoint, or hub in >50 % of all of the dates examined). Coalescence of the inundated playa network usually occurred at ≥10 km dispersal distance and depended on wetland density, indicating that critical thresholds in connectivity arose from synergistic effects of dispersal ability (spatial scale) and wet playa occurrence (a function of precipitation). Organisms with dispersal capabilities limited to <10 km routinely experienced effective isolation during our study. Connectivity is thus a dynamic emergent landscape property, so management to maintain connectivity for wildlife within ephemeral habitats like inundated playas will need to move beyond a patch-based focus to a network focus by including connectivity as a dynamic landscape property.  相似文献

The authors have studied microstructure evolution during thermally induced phase separation in a class of binary supported lipid bilayers using a quantitative application of imaging ellipsometry. The bilayers consist of binary mixtures consisting of a higher melting glycosphingolipid, galactosylceramide (GalCer), which resides primarily in the outer leaflet, and a lower melting, unsaturated phospholipid, 1,2-dilauroyl-sn-glycero-3-phosphocholine (DLPC). Three different bilayer compositions of GalCer/DLPC mixtures at 35:65, 20:80, and 10:90 molar ratios were cooled at controlled rates from their high-temperature homogeneous phase to temperatures corresponding to their phase coexistence regime and imaged in real time using imaging ellipsometry. During the thermotropic course of GalCer gelation, we find that two distinct types of morphological features modulate. First, the formation and growth of chain and fractal-like defects ascribed to the net change in molecular areas during the phase transition. The formation of these defects is consistent with the expected contraction in the molecular area during the liquid crystalline to gel-phase transition. Second, the nucleation and growth of irregularly shaped gel-phase domains, which exhibit either line-tension dominated compact shape or dendritic domains with extended interfaces. Quantifying domain morphology within the fractal framework reveals a close correspondence, and the quantization of the transition width confirms previous estimates of reduced phase transition cooperativity in supported bilayers. A comparison of domain properties indicates that thermal history, bilayer composition, and cooling rate all influence microstructure details including shapes, sizes, and distributions of domains and defects: At lower cooling rates and lower GalCer fractions compact domains form and at higher GalCer fractions (or at higher cooling rates) dendritic domains are evident. This transition of domain morphology from compact shapes to dendritic shapes at higher cooling rates and higher relative fractions of GalCer suggests kinetic control of shape equilibration in these phospho- and glycolipid mixtures.  相似文献

Neoplasms produce substances that induce blood vessel formation (angiogenesis). Fractions from ethanol extracts of the Walker 256 carcinoma were isolated by silica column chromatography and C18 reversed-phase high-performance liquid chromatography. Two of the isolated fractions induced neovascularization when tested in the rabbit corneal micropocket assay. One of the fractions was identified as nicotinamide by desorption-electron impact mass spectrometry, nuclear magnetic resonance spectroscopy, and gas chromatography-mass spectrometry. The second active fraction contained nicotinamide as part of a more complex, as yet unidentified, molecular arrangement. Microgram quantities of commercial nicotinamide induced neovascularization in the corneal micropocket assay and in the chick chorioallantoic membrane assay.  相似文献

The accumulation of plastics in the soil ecosystem poses an increasing environmental concern worldwide. However, little is known about the effect of plastic concentrations on soil properties and soil biota. In this study, we investigated the effect of low-density polyethylene (LDPE) microplastics (MPs) on the chemical and microbial properties of agricultural soil using a set of microcosm experiments. The soil was incubated for 100 days with LDPE at concentrations of 0%, 0.1%, 1%, 3%, 5%, and 7% at 25°C with 70% water-holding capacity. Along with soil chemical analysis, we conducted an analysis of soil microbial properties on the first day and again after 100 days of incubation. LDPE concentrations of ≥1% significantly (p < .05) decreased the pH but increased the electrical conductivity of the soil in comparison with the control (0% LDPE at 100 days). Increasing the LDPE concentration did not affect the soil exchangeable cation content or the available Pb concentration. Firmicutes were the most abundant phyla in the soil on the first day, whereas Proteobacteria, Firmicutes and Actinobacteria became dominant in all treatments after 100 days. An increasing LDPE concentration increased the abundance of Actinobacteria and decreased Proteobacteria. Principal component analysis demonstrated that only 7% LDPE was positively correlated with Actinobacteria, indicating that higher concentrations of LDPE contributed to the growth of this phylum. The findings of this study imply that MP contamination could affect soil chemical properties and microbial activity and that these effects primarily depend on MP concentrations in soil.  相似文献
