Antioxidant activity of carotenoids is suggested to be one of the factors for their disease preventing effects. Marine carotenoids fucoxanthin and its two metabolites, fucoxanthinol and halocynthiaxanthin, have been shown to exhibit several biological effects. The antioxidant activities of these three carotenoids were assessed in vitro with respect to radical scavenging and singlet oxygen quenching abilities. The 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity of fucoxanthin and fucoxanthinol was higher than that of halocynthiaxanthin, with the effective concentration for 50% scavenging (EC 50) being 164.60, 153.78, and 826.39 microM, respectively. 2,2'-Azinobis-3-ethylbenzo thizoline-6-sulphonate radical scavenging activity of fucoxanthinol (EC 50, 2.49 microM) was stronger than that of fucoxanthin (EC 50, 8.94 microM). Hydroxyl radical scavenging activity as measured by the chemiluminescence technique showed that the scavenging activity of fucoxanthin was 7.9 times higher than that by fucoxanthinol, 16.3 times higher than that by halocynthiaxanthin, and 13.5 times higher than that by alpha-tocopherol. A similar trend was observed when the hydroxyl radical scavenging was assessed by the electron spin resonance (ESR) technique. ESR analysis of the superoxide radical scavenging activity also showed the superiority of fucoxanthin over the other two carotenoids tested. Singlet oxygen quenching ability of the three carotenoids was lower than that of beta-carotene, with quenching rate constants ( k Q, x10 (10) M (-1) s (-1)) being 1.19, 1.81, 0.80, and 12.78 for fucoxanthin, fucoxanthinol, halocynthiaxanthin, and beta-carotene, respectively. The higher radical scavenging activity of fucoxanthin and fucoxanthinol compared with halocynthiaxanthin is assumed to be due to presence of the allenic bond.  相似文献

Fermentation using native lactic acid bacteria (LAB) was evaluated for its effectiveness in recovering lipids and proteins simultaneously from freshwater fish visceral waste (FVW). Five different LAB isolated from fish processing waste were employed in a fermentation process that involved 10% (w/w) glucose, 2% (w/w) NaCl, and 10% (v/w) LAB. Cultures evaluated included four native isolates (Pediococcus acidilactici NCIM5368, Enterococcus faecalis NCIM5367, Pediococcus acidilactici FM37, and Pediococcus acidilactici MW2) from FVW with E. faecium NCIM5335 as the reference culture. Fermentation with native LAB resulted in recovery of > 90% oil present in the material as against no recovery in case of raw (unfermented) viscera and resulted in > 50% of degree of hydrolysis of proteins. The fatty acid profile of lipids was not affected by the fermentation process. The fermentation liquor, rich in hydrolyzed protein, exhibited antioxidant as well as antagonistic properties against several bacterial pathogens. The results clearly demonstrate the usefulness of fermentation using native isolates for simultaneous recovery of lipids and proteins from fish processing waste. It also asserts the value of fermentation as an eco-friendly method and aids in minimizing disposal/pollution problems associated with these solid wastes.  相似文献

The immunostimulatory role of carotenoid extract from shrimp processing discards was evaluated by feeding common carp fingerlings with a diet containing carotenoid extract for 21 weeks at 100 and 200 mg kg?1 astaxanthin levels. Haemoglobin content was significantly (P < 0.05) higher in the blood of fish fed with carotenoid diet (>8.1 g dL?1) compared with that from fish fed with carotenoid deficient diet (6.86 g dL?1) and also leukocyte counts were higher (P < 0.05). No differences (P > 0.05) were observed in total serum protein, globulin level and albumin‐globulin ratio, but albumin content was higher (P < 0.05). Respiratory burst activity was significantly (P < 0.05) higher, the serum lysozyme activity almost doubled and the serum bactericidal activity was significantly increased when the fish were fed with diet containing 200 mg kg?1 of astaxanthin but no significant differences were observed in serum trypsin inhibitory activity. There was a significant (P < 0.05) increase in leukocyte myeloperoxidase activity due to dietary carotenoids. Challenging fish with Aeromonas hydrophila after the feeding period resulted in 50% mortality in the control group while in the group fed with diet containing 100 mg kg?1 astaxanthin, the mortality rate was 15%. No mortality and even symptoms of infection was not observed in the group fed with diet containing 200 mg kg?1 of astaxanthin. The study indicated that carotenoid extract from shrimp processing discards can effectively be used as immunostimulants in aquaculture of carps and dietary carotenoids were found to enhance various immune defence mechanisms and also provide protection against the infection of pathogen A. hydrophila.  相似文献

Crustacean waste is an important source of natural carotenoids. Carotenoids in shrimp waste occur as a complex with proteins. Investigations were carried out to assess the effect of hydrolyzing shrimp waste with different proteases on the subsequent recovery of carotenoids in vegetable oil. Hydrolyzing the shrimp waste with the bacterial protease alcalase gave higher extraction yields in oil (28.6 μg/g waste) compared to hydrolyzing with the plant protease papain (24.8 μg/g) or the animal protease trypsin (25.3 μg/g). Optimized conditions for alcalase treatment and extraction of carotenoids was determined to be hydrolysis of shrimp waste with 0.75% of alcalase at 37°C for 150 min, adding sunflower oil to hydrolyzed waste in a ratio of 2:1, and heating at 70°C for 90 min. Prior hydrolysis of waste considerably reduced the heating time required for maximum extraction of pigments, which may help in preventing the carotenoid degradation during prolonged heating in oil.  相似文献
