The objective of this study was to determine whether a prophylactic regimen of intramuscular polysulfated glycosaminoglycans (PSGAG), administered to growing Thoroughbred foals, would significantly reduce the incidence or severity of osteochondritis dissecans (OCD) or traumatic joint lesions that had been found in untreated foals (historical controls) from the same farm during the preceding 3 years. The number of PSGAG-treated foals with clinically significant joint lesions was 44.1% lower than the number of foals with OCD/joint injuries that had been found in the untreated historical controls. Surgical intervention of the treated foals declined 87.7% from the incidence on the farm during the preceding 3 years. Although the incidence of OCD lesions of the hock, stifle, and fetlocks in treated foals was not significantly different from that found in the matched controls, the lesions in the PSGAG-treated foals resolved without surgery. These results suggest that PSGAG has the potential to improve the clinical course of osteochondrosis (OC) and OCD. Additional studies are underway to determine whether a shorter or more targeted prophylactic regimen will provide a similar benefit.  相似文献

In a variety of human genetic diseases, replacement of the absent or defective protein provides significant therapeutic benefits. As a model for a somatic cell gene therapy system, cultured murine fibroblasts were transfected with a human growth hormone (hGH) fusion gene and cells from one of the resulting clonal lines were subsequently implanted into various locations in mice. Such implants synthesized and secreted hGH, which was detectable in the serum. The function of the implants depended on their location and size, and on the histocompatibility of the donor cells with their recipients. The expression of hGH could be modified by addition of regulatory effectors, and, with appropriate immunosuppression, the implants survived for more than 3 months. This approach to gene therapy, here termed "transkaryotic implantation," is potentially applicable to many genetic diseases in that the transfected cell line can be extensively characterized prior to implantation, several anatomical sites are suitable for implantation, and regulated expression of the gene of therapeutic interest can be obtained.  相似文献

Abstract.— Fish meal is one of the predominant sources of crude protein used in practical feeds for most cultured species. However, with overfishing and increasing demand for protein feedstuffs, new competitively priced sources of dietary protein are needed. A coextruded soybean meal-red blood cell (SBM-RBC) ingredient was evaluated as a source of crude protein for rainbow trout Oncorhynchus mykiss . The SBM-RBC ingredient was incorporated at either 0, 21, 42 or 63% of the dietary protein, replacing an isoni-trogenous amount of fish meal. All diets were fed for 8 wk to triplicate groups of juvenile rainbow trout (average initial weight 49.5 g) stocked into a recirculating system consisting of 48, 114-L aquaria. There were no significant differences in weight gain, feed conversion ratio, protein efficiency ratio, apparent crude protein digestibility, apparent gross energy digestibility, dress-out percentage, or fillet protein and fat concentrations in fish fed any of the experimental diets. Based on these data, extruded SBM-RBC appears to be a suitable ingredient in diets fed to rainbow trout.  相似文献

Amino acid sequences of encephalitogenic proteins from bovine cord and rabbit brain are reported. The bovine protein contains 45 residues. The rabbit protein is identical except for two isopolar substitutions, a dipeptide and amino acid deletion. Analysis of this protein and a 140-residue myelin basic protein indicates that the smaller protein is a portion of the larger encephalitogen. The larger myelin protein contains at least two encephalitogenic sites.  相似文献

European bat lyssavirus type 2 (EBLV-2) has been isolated once previously from a bat in the UK in June 1996. In September 2002, a Daubenton's bat (Myotis daubentonii) found in Lancashire developed abnormal behaviour, including unprovoked aggression, while it was in captivity. Brain samples from the bat were tested for virus of the Lyssavirus genus, which includes EBLV-2 (genotype 6), and classical rabies virus (genotype 1). A positive fluorescent antibody test confirmed that it was infected with a lyssavirus, and PCR and genomic sequencing identified the virus as an EBLV-2a. Phylogenetic comparisons with all the published sequences from genotype 6 showed that it was closely related to the previous isolate of EBLV-2 in the UK and suggested links to isolates from bats in The Netherlands. The isolation of EBLV-2 from a bat found on the west coast of England provides evidence that this virus may be present within the UK Daubenton's bat population at a low prevalence level.  相似文献

A new assemblage of arthropod cuticles from Upper Silurian rocks in Shropshire, England, includes at least two centipedes and a trigonotarbid arachnid. This unequivocal terrestrial fauna from the Silurian constitutes the earliest direct record of land animals. The presence of predatory arthropods suggests that complex terrestrial ecosystems were in place by the late Silurian (414 x 10(6) years before present) and that the animal invasion of the land occurred earlier than was previously thought.  相似文献

Many marine species are shifting their distributions in response to changing ocean conditions, posing significant challenges and risks for fisheries management. Species distribution models (SDMs) are used to project future species distributions in the face of a changing climate. Information to fit SDMs generally comes from two main sources: fishery-independent (scientific surveys) and fishery-dependent (commercial catch) data. A concern with fishery-dependent data is that fishing locations are not independent of the underlying species abundance, potentially biasing predictions of species distributions. However, resources for fishery-independent surveys are increasingly limited; therefore, it is critical we understand the strengths and limitations of SDMs developed from fishery-dependent data. We used a simulation approach to evaluate the potential for fishery-dependent data to inform SDMs and abundance estimates and quantify the bias resulting from different fishery-dependent sampling scenarios in the California Current System (CCS). We then evaluated the ability of the SDMs to project changes in the spatial distribution of species over time and compare the time scale over which model performance degrades between the different sampling scenarios and as a function of climate bias and novelty. Our results show that data generated from fishery-dependent sampling can still result in SDMs with high predictive skill several decades into the future, given specific forms of preferential sampling which result in low climate bias and novelty. Therefore, fishery-dependent data may be able to supplement information from surveys that are reduced or eliminated for budgetary reasons to project species distributions into the future.  相似文献
