OBJECTIVE: To evaluate the potential of an implant of a GnRH-agonist (deslorelin) to create a progesterone free animal suitable for studying progesterone (P4) metabolism in intact cows by measuring blood P4 and faecal P4 metabolites. METHODS: Experiment 1: Eighteen non-lactating cycling Holstein-Friesian cows, 4 to 7 years old, were allocated to one of three groups to study plasma P4 concentrations preceding an intravaginal insert. These groups comprised: i) a deslorelin group (GnRH-agonist implanted); ii) a PGF group receiving two injections of prostaglandin (PGF2alpha) 12 days apart; and, iii) an ovariectomised (OVX) group. An intravaginal device (CIDR) was inserted into the vagina of each animal and left in place for 11 days. Plasma P4 concentrations were measured during the study period. Experiment 2: Twelve non-lactating cycling Holstein-Friesian cows, 4 to 7 years old, were allocated to two groups: i) a deslorelin group (GnRH-agonist implanted); and ii) an ovariectomised group. Plasma P4 and faecal P4 metabolites (20-oxo-pregnanes, 20alpha-OH and 20beta-OH) were monitored for a period of 5 weeks. RESULTS: Experiment 1: Average plasma P4 concentration did not differ between the three groups (1.28, 1.43 and 1.55 ng/mL for deslorelin, OVX and PGF cows, respectively, P = 0.8) during the period of supplementation. Experiment 2: There was no difference in plasma P4 (mean plasma P4 < 0.02 ng/mL, P = 0.9) and faecal P4 metabolites between deslorelin and OVX cows 2 weeks after the implantation (P = 0.7). CONCLUSIONS: These data showed that a GnRH-agonist (deslorelin) implant may be used as an alternative to ovariectomy to create a progesterone free animal suitable for studying the metabolism of administered P4.  相似文献

Aggression in 108 dog breeds and six crosses was ranked by 185 members of the Companion Animal Society of the New Zealand Veterinary Association. The Rottweiler and German Shepherd were categorised as extremely aggressive and the Akita, Basengi, Chihuahua (long-coated and short-coated), Chow Chow, Cocker Spaniel, Shar pei and Welsh Corgi (Cardigan and Pembroke) were classified as very aggressive. A chi-squared test of data from the twenty most popular breeds showed that significant differences were perceived between the breeds. Cocker Spaniels, Pembroke Welsh Corgis, German Shepherds and Rottweilers were more aggressive than Staffordshire Bull Terriers, Border Collies and Doberman Pinchers, which were more aggressive than Cavalier King Charles Spaniels, Bichon Frise, English Springer Spaniels, German Shorthaired Pointers, Weimaraners, Labrador Retrievers, Golden Retrievers, Newfoundlands, Dalmatians, Boxers, Bulldogs, Bull Mastiffs and Rough Collies.  相似文献

Six 1-year-old farmed red deer were used to compare physiological and behavioural responses to a range of doses of 5% xylazine with or without the addition of 0.4 mg of fentanyl citrate and 3.2 mg of azaperone per ml. Each deer was randomly assigned to one of six treatments: xylazine alone at 0.4 and 0.6 mg/kg, the xylazinelfentanyl citrate/azaperone combination containing 0.2, 0.4 and 0.6 mg/kg of xylazine, or a sterile water control. Injections were given intramuscularly in the anterior neck, operator blind, on each of 6 sampling days between October and January, such that each deer received all treatments with 9-28 days between each treatment. Measurements included heart rate and respiration rate. A 0-3 scoring system (normal to nil response, respectively) was devised to record sedative responses (body stance, head position, degree of eye closure, palpebral reflex, resistance to movement of the head, response to noise) and analgesic responses to touch and pinching of the ear, and response to a needle prick in the gluteal region. Scores were added to produce a sedation score and analgesia score, respectively, for each treatment. Records were taken immediately prior to injection and thereafter at 5, 14, 25, 35, 60, 90, 120, 150, 180, 210, 240 and 300 minutes. All deer given each dose rate of the xylazine and the xylazine/fentanyl citrate/azaperone combination became recumbent. There was a tendency for the time to recumbency and variation of time to recumbency to be shorter at higher dose rates and with the addition of fentanyl citrate and azaperone to xylazine, particularly with xylazine at 0.4 mg/kg. These trends were not statistically significant (p>0.05). The duration of recumbency was shorter with the low dose of the xylazine/fentanyl trateiazaperone combination (0.2 mg/kg of xylazine) than for the higher doses of xylazine alone or the combination of drugs (p<0.05). There were no significant differences in heart rates or respiration rates between treatments, although all treatments significantly reduced both heart and respiration rates (p<0.01). The sedation scores showed similar peak responses and timing to peak responses (14-25 min) to both drug treatments and all dose rates, but the responses were less persistent for lower doses. The analgesia scores showed a similar pattern, with peak responses 14-35 minutes after administration and more persistence at higher dose rates of both xylazine alone and the xylazine/fentanyl citrate/azaperone combination. This study has shown that most physiological and behavioural responses to a range of doses of xylazine or the xylazine/fentanyl citrate/azaperone combination were statistically similar. However, there was a tendency for recumbency to occur more rapidly and with less variation in timing when the mid-range dose of the drug combination was used, supporting the observation by practitioners that the drug combination results in a more rapid and reliable state of recumbency at a lower dose rate of xylazine.  相似文献

Suspected photosensitisation occurred in three groups of lambs grazing birdsfoot trefoil (Lotus corniculatus c.v. Grasslands Goldie). In one group, sucking lambs aged about 10 weeks, grazing birdsfoot trefoil, developed skin lesions while lambs of a similar age and from the same flock grazing lucerne (Medicago sativa) or a mixed sward of both species showed no signs of photosensitisation. Affected lambs had lesions on their backs and ears. In a few animals the tips of the ears were shortened by 2-3 cm. In the affected lambs, serum liver enzymes (gamma-glutamyltransferase, glutamate dehydrogenase), bilirubin and serum Vitamin B12 levels were within the normal range. At necropsy, no significant pathological changes were detected in the liver and histological changes in the skin were consistent with primary photosensitisation. In the second group, three of 80 weaned lambs grazing the same birdsfoot trefoil at a restricted intake were affected in the same manner as the first group. In the third group, 15 animals from 28 sets of sucking twin lambs were also affected. In only two sets of twins were both lambs affected. None of the ewes grazing with the lambs in the first or third groups showed any clinical signs of photosensitisation.  相似文献

Theileria orientalis (also known historically as T. sergenti and T. buffeli) is responsible for benign or non-transforming theileriosis, and exerts its major effect through erythrocyte destruction. The life cycle of T. orientalis is essentially similar to that of other Theileria species, except that the schizonts do not induce transformation and fatal lymphoproliferation. The pathogenesis of anaemia as a result of infection is not clearly established and may be multifaceted. Clinical signs of weakness, reluctance to walk and abortion are early but non-specific indications of disease, particularly if accompanied by a history of cattle being moved. Physical examination may reveal pallor (pale eyes, vaginal mucosa), pyrexia, and elevated heart and respiratory rates. T. orientalis is an economically important parasite of cattle in New Zealand, Australia and Japan, especially where naïve animals are introduced into an endemic area or in animals under stress. Increased awareness of the risks posed by the parasite is required to enable management practices to be implemented to minimise its impact.  相似文献

AIM: To determine the welfare of calves arriving at a slaughter plant by assessing their physical state and plasma biochemical profile. METHODS: Calves (n=7,169) aged 4-10 days, were examined immediately after they were unloaded from trucks (n=20) and categorised on physical grounds as being in an 'unacceptable', 'marginal' or 'acceptable' state. 'Unacceptable' calves were lying down, unable to walk, extremely weak or seriously injured. 'Marginal' calves had a wet umbilicus, were hollow sided, apparently immature, or weak and slow and unsteady on their feet. 'Acceptable' calves were strong, walked freely, round-sided, bright and alert and had a dry umbilicus. After unloading, 27 'unacceptable', 306 'marginal' and 289 'acceptable' calves were weighed, examined clinically and a blood sample was taken from each. A second blood sample was taken from 118 of these calves, 12-15 hours later. The packed cell volume (PCV) and plasma concentrations of glucose, beta-hydroxybutyrate (BHB), total plasma proteins (TPP), gamma-glutamyl transferase (GGT), urea, creatine kinase (CK) and lactate were measured. RESULTS: Only 27 of the 7,169 calves examined (0.4%) were categorised as being in an 'unacceptable' state on arrival and these were euthanased shortly after unloading. Generally, the plasma biochemical profiles of the 'acceptable' calves were similar to those of 'marginal' animals. However the glucose concentration of 'marginal' calves was lower than that of 'acceptable' calves, suggesting that they had lower energy reserves. After 12-15 h in lairage, plasma glucose concentration decreased and the BHB concentration increased in both 'acceptable' and 'marginal' calves compared with concentrations measured on arrival. CONCLUSIONS: The percentage of calves presented for slaughter in an 'unacceptable' state was small (0.4%). However, 4% of calves were classified as 'marginal' and failed to meet all the requirements of the code of recommendations and minimum standards governing the welfare of young calves in New Zealand. The categorisation of calves by a physical assessment was supported by results of plasma biochemistry. As a result, plasma biochemical profiles are considered unnecessary for making decisions regarding the welfare status of calves arriving at a slaughter plant; an assessment of their physical state is sufficient.  相似文献

OBJECTIVE: To test whether injecting lignocaine into the scrotal neck 5 to 10 s before or into both testes immediately after ring castration and docking wound significantly reduce the plasma cortisol response to castration and docking. DESIGN: A physiological study with controls. PROCEDURE: Lambs were given one of six treatments: control handling, injection of lignocaine into scrotal neck, injection of lignocaine into both testes, ring castration and docking, ring castration and docking after lignocaine was injected into the scrotal neck, and ring castration and docking before lignocaine was injected into both testes. Blood samples were taken before and regularly after treatment and analysed for plasma cortisol concentrations. RESULTS: The plasma cortisol concentrations of lambs castrated and docked after lignocaine had been injected into the scrotal neck were significantly lower between 20 and 60 min after treatment than in lambs castrated and docked without local anaesthesia. Injecting lignocaine into the testes after ring application did not significantly reduce the cortisol response to ring castration and docking. CONCLUSIONS: Lignocaine injected into the scrotal neck 5 to 10 s before ring castration will reduce the cortisol response and by inference the pain associated with ring castration.  相似文献
