Five studies were performed to determine factors affecting progesterone concentration in skim milk. Results of the first study indicated that progesterone concentration was higher in skim milk of samples kept 16 hours in an ice bath (0 C) than of those left at room temperature (21 C). In the second study, this temperature effect was found to be reversible, with skim milk progesterone concentration increasing when whole milk samples were cooled prior to centrifugation. In the third study, [3H]-labeled progesterone was used to determine the relationship between fat content of foremilk (the first milk obtained from the teats), midmilk (milk obtained midway through milking), and strippings (milk obtained immediately after milking machines have been removed) samples and temperature (4 C and 21 C) on the percentage of progesterone in the skim milk fraction. The relationship between percentage of butterfat and percentage of progesterone in skim milk was linear when the log of these variables was used for calculations. In the fourth study, assayable progesterone in the skim milk fraction of foremilk, midmilk, and strippings was affected by temperature. In the fifth study, a multiple-regression procedure was used to determine the amount of variation in percentage of radioactive progesterone in the skim milk fraction. The 1998 Report of the American Association of Feline Practitioners and Academy of Feline Medicine Advisory Panel on Feline Vaccines was developed to help veterinary practitioners formulate vaccination protocols for cats. The current panel report updates information, addresses questions, and speaks to concerns raised by the 1998 report. Although there has been increasing research on the adoption of agroforestry technologies over the last decade, few such studies have assessed uptake over a long period and many are based on a single snapshot in time. Furthermore, most of these studies have mainly looked at non-adopters and adopters: only recently have social scientists considered testers. A further category of users neglected in adoption studies has been re-adopters of technologies. Studying this group provides an interesting and more nuanced understanding of adoption and re-adoption. Methodologically, most adoption studies use quantitative methods and fail to link their findings to wider socio-economic, political and institutional settings. This paper presents a study of the dynamics of improved tree fallow use by farmers in Siaya and Vihiga districts of western Kenya over a period of eight years. It uses both qualitative and quantitative data to critically discuss the motivations of adopters, testers/rejecters and re-adopters. The results show that the process of adoption is highly dynamic and variable with farmers planting improved fallows and discontinuing or re-adopting them due to a whole range of factors, of which soil fertility improvement is just one. These factors included incentives from projects, the tying of adoption to credit programmes, prestige, participation in seminars/tours and the availability of a seed market from projects promoting improved fallows. Farmers planting improved fallows for such reasons may be termed ‘pseudo-adopters’. There were significant differences in adoption between the two districts, with more farmers in Siaya planting improved fallows than in Vihiga. A majority of farmers in Vihiga (53%) who were given seed never planted improved fallows, even though they had been exposed to the technology. Some 40% of farmers in Siaya and 38% in Vihiga planted improved fallows but later rejected them. This has some important implications for research and development. For improved fallow technologies to be attractive to farmers, they must provide other tangible economic benefits besides soil fertility improvement. This presents a challenge to researchers who must better attune themselves to the needs and demands of farmers if they wish to see their research findings widely adopted.  相似文献

Our objective was to evaluate the effect of postruminal protein infusion on pancreatic exocrine secretions. One Holstein, two crossbred, and five Angus steers (305 +/- 5 kg) with pancreatic pouch-duodenal reentrant cannulas and abomasal infusion catheters were used in a replicated 4 x 4 Latin square. All steers were abomasally infused with 1,050 g/d of raw cornstarch with treatments of 0, 60, 120, or 180 g/d of sodium casein suspended in water to yield 6,000 g/d of infusate daily. Steers were limit-fed (1.5 x NEm; 12 equal portions daily) a 90% corn silage, 10% supplement diet formulated to contain 12.5% CP. Periods consisted of 3 d of adaptation to infusion, 7 d of full infusion, 1 d of collection, and 7 d of rest. Pancreatic juice was collected in 30-min fractions continuously for 6 h. Total juice secreted and the pH of individual fractions were recorded, a 10% subsample was retained to form a composite sample, and remaining fluid was returned to the duodenum. Juice composite samples were stored (-30 degrees C) until analyzed for total protein and activities of alpha-amylase, trypsin, and chymotrypsin. Casein infusion linearly increased alpha-amylase concentration (182 to 271 units/mL; P < 0.02; 17.5 to 24.6 units/mg of protein; P < 0.03) and secretion rate (26,847 to 41,894 units/h; P < 0.01). Total juice secretion (155 g/h), pH of pancreatic juice (8.13), secretion rate of protein (1,536 mg/h), and concentration of protein (10.2 mg/mL) in pancreatic secretions were not affected (P > 0.05) by casein infusion. Similarly, casein infusion did not change 0.05) trypsin and chymotrypsin concentrations (1,379 and 349 units/L or 0.134 and 0.033 units/mg of protein, respectively) or secretion rates (206 and 52 units/h, respectively). Abomasal infusion of protein with starch stimulated a greater pancreatic secretion of alpha-amylase activity into the intestine than infusion of starch alone.  相似文献

Steers (379 +/- 10 kg) with ruminal, duodenal, and ileal cannulas were used in a 5 x 5 Latin square digestion trial to quantify and evaluate the relationship between intestinal protein supply and intestinal starch disappearance. Treatments were infusions of 0, 50, 100, 150, or 200 g/d of casein along with 1,042 g/d of raw cornstarch. Abomasal infusions were accomplished by passing tubing and a pliable retaining washer through the reticular-omasal orifice into the abomasum. Steers were fed a 93% corn silage, 7% supplement diet that contained 12% crude protein at 1.65% body weight in 12 equal portions/d. Periods lasted 17 d (12 d for adaptation, 2 d of collections, and 3 d of rest). The quantity and percentage of organic matter and protein disappearance from the small intestine increased linearly (P < 0.03) with infused casein. Greater quantities of starch disappeared with increased casein infusion (P < 0.01). The infusion of 200 g/d of casein increased small intestinal starch disappearance by 226 g/d over the control. Casein infusion did not affect the quantity or percent of organic matter, starch, or protein disappearance in the large intestine. Treatments did not change ruminal ammonia N, ruminal pH, or plasma glucose concentrations. Starch disappearance from the small intestine was increased with greater protein flow to the duodenum of steers.  相似文献

On two soils lacking fungi capable of forming mycorrhizas with Pinus, the addition of superphosphate at 40 kgP/ha greatly increased mycorrhiza formation in Pinus radiata D. Don and P. elliottii Little and Dorman. Phosphorus could be partially replaced by increasing inoculum density or avoiding delay in inoculation after sowing. Chlamydospores were less effective than basidiospores as inoculum except at the highest density and in the presence of added P. At low available P concn such as are common in Australian soils, the normal pattern of infection of new short roots is interrupted and re-infection from the soil is of greater importance. Survival of mycorrhizal fungi in the soil in the absence of the host may therefore be a critical factor in inoculation programs.  相似文献

Identification of B-cell epitopes on the betanodavirus capsid protein   总被引:1,自引:0,他引:1  
The pepscan procedure was used to identify betanodavirus B-cell epitopes recognized by neutralizing mouse monoclonal antibodies (MAbs) and serum samples obtained from sea bass, Dicentrarchus labrax, naturally infected with betanodavirus. Pepscan was performed with a panel of thirty-four 12-mer synthetic peptides that mimicked the entire betanodavirus capsid protein. Sea bass serum samples reacted strongly with three regions of the capsid protein comprising amino acid residues 1-32, 91-162 and 181-212. The latter region was also recognized by neutralizing MAbs and coincided with a region of high antigenic propensity identified by an antigen prediction algorithm. These data suggest that a region of the betanodavirus capsid protein spanning amino acid residues 181-212 may represent a neutralization domain that could potentially be used to inform the development of nodavirus vaccines and immunodiagnostic reagents.  相似文献
