In this work we identified specific bovine leukocytes that were bound by the Mannheimia haemolytica adhesin molecule (MhA) and the biological effect on the leukocytes. Histochemical staining and flow cytometry showed that MhA bind neutrophils (90%) and monocytes (5%). MhA induced an oxidative response in purified neutrophils; this effect was 1.5-fold higher than the effect observed with control cells activated with Zymosan. Cellular binding by MhA was inhibited with GlcNAc and its oligomers, as well as by glycoproteins containing tri- and tetra-antennary N-glycosydically linked glycans. MhA-induced oxidative burst was significantly inhibited by GlcNAc, iodoacetamide, superoxide dismutase, and piroxicam (p<0.05). Our findings suggest that among bovine leukocytes, neutrophils are the main target for MhA, inducing production of oxidative radicals by non-opsonic mechanism that seem to play an important role in tissue damage during mannheimiosis.  相似文献

By means of an international project, Argentinian and Italian researchers are carrying out joint research to study biological and ecological aspects of Tuta absoluta biological control. This paper lists indigenous natural enemies reported for T. absoluta, as well as the current results on T. absoluta egg and larval parasitoids in both countries. Parasitoid species that conformed to different guilds are shown to coexist in cropping conditions, and some show positive characteristics as potential biocontrol agents against T. absoluta by means of augmentative releases. Future laboratory and field evaluations of the efficacy of biological control programmes in Argentina and Italy are proposed.  相似文献

The aim of this work was to determine the enzymatic activity of phosphofructokinase (PFK), malate dehydrogenase (MDH) and isocitrate dehydrogenase (IDH) in boar spermatozoa and study their participation in bicarbonate‐induced capacitation and follicular fluid‐induced acrosome reaction. Enzymatic activity of these enzymes was determined spectrophotometrically in extracts of boar spermatozoa. Sperm suspensions were incubated in the presence of bicarbonate (40 mM), a well‐known capacitation inducer, or follicular fluid (30%), as an acrosome reaction inducer, and different concentrations of oxoglutarate, oxalomalate and hydroxymalonate, inhibitors of PFK, IDH and MDH, respectively. Capacitation percentages were determined by the fluorescence technique of chlortetracycline (CTC), and true acrosome reaction was determined by trypan blue and differential–interferential contrast, optical microscopy. The activity of PFK in boar spermatozoa enzymatic extracts was 1.70 ± 0.19 U/1010 spermatozoa, the activity of NAD‐ and NADP‐dependent IDH was 0.111 ± 0.005 U/1010 and 2.22 ± 0.14 U/1010 spermatozoa, respectively, and the activity of MDH was 4.24 ± 0.38 U/1010 spermatozoa. The addition of the specific inhibitors of these enzymes prevented sperm capacitation and decreased sperm motility during capacitation and inhibited the acrosome reaction (AR), without affecting the sperm motility during this process. Our results demonstrate the participation of PFK, IDH and MDH in bicarbonate‐induced capacitation and follicular fluid‐induced acrosome reaction in boar spermatozoa, contributing to elucidate the mechanisms that produce energy necessary for these processes in porcine spermatozoa.  相似文献

The recurrence and persistence of foot and mouth disease (FMD) could be the consequence of cyclic and massive transportation of calves. For this reason, in South America, vaccination strategies related to livestock dynamic are being promoted. In order to aid the evaluation of such strategies, a method for predicting the risk of transportation of nonvaccinated weaned calves was developed; this method combines expert opinion and empirical evidence using Bayesian estimators. It was applied through Monte Carlo simulation to data of Argentina under four hypothetical vaccination schemes: E1, extended vaccination season of 1/6 of the population of calves each month from July to December without second round vaccination (SRV); E2, extended irregular vaccination from July to December with SRV applied to 70% of the calves resembling the scheme applied in Argentina in 2001; E3, vaccination in November and December without SRV; and E4, vaccination concentrated in November. E1 resulted in probability of transporting non vaccinated calves (tnvc) reaching its maximum in the following year in May with mean=0.0250 and percentile 95% (P95)=0.0404; for the same month tnvc estimates for the other schemes were E2: mean=0.0071; P95=0.0162; E3: mean=0.0017; P95=0.0042 and E4: mean=0.0001; P95=0.0004. Bonferroni multiple comparison for simultaneous assertions for May showed that E4 resulted the best scheme, E1 the worst, and E2 and E3 are intermediate with nonsignificant difference observed between overall (p<0.05). Results were consistent with historical records and quantification for future needs for re-vaccination was made possible. While the ratio "total vaccinated"/"total estimated existences" will give a biased vision of vaccination coverage under the situation of extended vaccination campaigns, a model as the one developed here could allow a more accurate assessment and the design of mitigation plans.  相似文献

The lack of efficacy of conventional strategies for the maintenance of healthy udders in domestic cattle has prompted studies on the use of immunomodulators or biological response modifiers (BRM) for this purpose. These compounds are agents that modify the host's response to pathogens leading to beneficial effects on disease outcome. The objective of this study was to evaluate the effects of a single intramammary infusion of Panax ginseng (GS) extract on the amount of pro-inflammatory cytokines and the number of monocytes/macrophages present in bovine mammary tissues at drying off. Eight mammary quarters from six nonpregnant cows in late lactation were infused with 10 mL of GS (3mg/mL), six quarters were treated with 10 mL of placebo (vehicle alone) and six quarters were maintained as uninoculated controls. The analyses of tumor necrosis factor-alpha (TNF-α) by immunohistochemistry revealed that the production of this proinflammatory cytokine significantly increased (P<0.05) in the inoculated mammary glands of cows following BRM inoculation, whereas the interleukin-1 alpha (IL-1α) and IL-6 staining area was not affected by BRM treatment. The number of monocytes/macrophages detected with CD14 antibody was significantly higher (P<0.05) in BRM-treated quarters than in placebo and uninoculated control quarters. These results indicated an immunomodulator potential of the BRM used. The beneficial effect of the extract could be used as alternative therapy in the control of mastitis at drying off, either alone or in conjunction with dry cow antibiotic therapy.  相似文献

Ovine pregnancy toxaemia is a metabolic disorder affecting sheep in their last 6 weeks of pregnancy as a result of their inability to maintain adequate energy homoeostasis. Different alternative treatments are available with variable results. The aim of this research was to evaluate a peroxisome proliferator‐activated receptor alpha (PPARα) stimulant as an alternative to treat clinical pregnancy toxaemia. Thirty‐three adult sheep, with known gestation date and carrying a single foetus, were fasted from day 130 of gestation until animals showed clinical disease. From that moment onwards, sheep were treated during 6 days with three different therapeutic alternatives: 10 mg/kg of 2‐methyl‐2‐phenoxy‐propionic acid; 10 mg/kg of 2‐methyl‐2‐phenoxy‐propionic acid + 100 mL of propylene glycol oral; or 100 mL of propylene glycol oral. Glycaemia and serum β‐hydroxybutyrate (BHOB) were determined daily. Liver biopsies were taken at day 130 of gestation, at the beginning and end of treatments and at 5 days postpartum, evaluating the extent and degree of the steatosis lesion. Even though in sheep treated with 2‐methyl‐2‐phenoxy‐propionic acid, serum concentrations of glucose and BHOB recovered more slowly, we conclude that 2‐methyl‐2‐phenoxy‐propionic acid alone or combined with propylene glycol can be used as an alternative to effectively treat fatty liver, and therefore pregnancy toxaemia.  相似文献

The genetic diversity of several local populations of the fungal pathogen Cochliobolus sativus, collected over 3 years from different regions of the Czech Republic, was examined using amplified fragment length polymorphism (AFLP). A high level of variability was found even among isolates from one lesion. Measures of multilocus linkage disequilibrium suggested that recombination has a minor impact on the genetic structure of populations. Cochliobolus sativus forms genetically divergent populations (FST = 0·33), indicating a low level of geneflow between populations. This was supported by a significant correlation between genetic diversity and geographical distances up to 80–100 km. The most likely explanation for the genetic variability is that the fungus forms conidia with highly variable chromosomal rearrangements. The differentiation observed among local populations implies that genetic drift, including a founder effect, combined with restricted migration generates the structure of C. sativus populations.  相似文献

The sanitary and economic impact of cystic echinococcosis is serious in those countries where it becomes endemic. Ultrasonography is one technique that may be used to diagnose this disease in endemic areas. In parasitized sheep, hydatid cysts appear sonographically as a round hypoechoic structure. Twenty two sheep destined for slaughter were studied sonographically and imaging findings compared to post-mortem findings. Three sheep with hydatid cysts were identified. Eighty additional sheep not destined for slaughter were also studied. Echinococcus granulosus cysts were detected in three animals. Forty sheep from a non-endemic area had no hepatic cysts. The in vivo sonographic study of sheep provides a useful screening tool for echinococcosis.  相似文献

Random S5 inbreds derived from three F2 maize (Zea mays L.) populations (L1934 × LP918, LP915 × LP2541 and L7310 × L7266) were selected for ear rot resistance after inoculation with a low-fumonisin producing isolate belonging to F. proliferatum. The four less susceptible and the four most susceptible inbreds from each population were crossed and F1 seeds were pooled. Resistant and susceptible pools from each population were evaluated for disease severity (percentage of the ear visibly diseased) after inoculation with the isolate used for selection, and high toxigenic isolates belonging to F. verticillioides and F. graminearum. Grain mycotoxin concentration was assessed by ELISA. Differences in disease resistance to each fungus were observed between resistant and susceptible pools in most populations and environments indicating that selection after inoculation with a single species might be effective to develop broad-based resistance to Fusarium. Resistant pools exhibited, after inoculation with F. verticillioides, low grain fumonisin concentrations in most populations and years. Positive genotypic correlations between disease severity and fumonisin concentration (0.89 < rg < 0.98, depending on fungal species and year) indicate that selection for disease severity accounted for most of the variability for field fumonisin accumulation. Selection seemed to be also effective to reduce grain deoxynivalenol and zearalenone concentrations after inoculation with F. graminearum. Ratios between grain deoxynivalenol concentration and disease severity were lower in L7310 × L7266 than those observed in the other populations suggesting that mechanisms affecting mycotoxin accumulation might exist in this population and additional responses should be feasible if including deoxynivalenol concentration as another selection parameter.  相似文献
