Goose fatty liver is a specific type of nonalcoholic fatty liver that is protected from harmful effects associated with severe steatosis. Our previous findings suggest that suppression of the complement C5 may be relevant, but the mechanism is unclear. Therefore, in this study, we first verified the expression pattern of complement genes (including C5) during goose fatty liver formation and then determined the liver fat content and fatty acid composition by high-performance liquid chromatography (HPLC), followed by selecting the differential metabolites to treat HepG2, goose and mouse primary hepatocytes, aiming to explore the mechanism of C5 and inflammation suppression in goose fatty liver. The data confirmed the suppression of complement genes (including C5) in goose fatty livers. Moreover, fat content was significantly higher in fatty liver versus normal ones, with oleic acid and palmitic acid dominantly accounting for the difference. In line with this, high concentration of palmitate led to down regulation of C5 expression in goose primary hepatocytes whereas upregulation in mouse primary hepatocytes and HepG2 cells. In conclusion, regulation on C5 expression by fatty liver related factors including high level of palmitic acid may contribute to the protection of goose liver from severe hepatic steatosis.  相似文献

Twenty-eight Norwegian Red Cattle dairy cows were fed silage ad libitum and restricted amounts of concentrates. Blood samples were collected before morning feeding, once or twice weekly, from 2 weeks before to 12 weeks after calving. Parameters of liver function, carbohydrate status and fertility were recorded in order to assess their interrelationships. Eight cows were treated for clinical ketosis. Four of these had to be treated 2 or 3 times. Aspartate aminotransferase and bilirubin showed the highest within-animal coefficients of correlation with acetoacetate. Analysis of variance revealed a significant effect of carbohydrate status (indicated by plasma acetoacetate levels) on the levels of aspartate aminotransferase, glutamate dehydrogenase and sorbitol dehydrogenase, though only a small part of the total variation was explained by this factor. The estimated volume density of liver fat in the 4th week of lactation averaged 6.0 +/- 6.4% (+/- SD) ranging from 0.1-25.1%. Liver fat content at this stage of lactation was not significantly correlated with other indicators of liver function or carbohydrate status. Cows treated for clinical ketosis had significantly lower plasma progesterone values at the time of first ketosis treatment than untreated multiparous cows. The frequency of high progesterone values (greater than 3 ng/ml) being significantly lower in treated than in untreated cows during the period from 3-5 weeks post partum, though not at later stages. In conclusion, the results revealed a significant relationship between carbohydrate status and liver function, and also between clinical ketosis and luteal function.  相似文献

The effect of separate chilling (5°C for 2 and 4 days) on the shoots and roots of two maize hybrid seedlings on the electric potential, the composition of fatty acids and the ATPase activity in the microsomes of non-chilled leaves and roots (20°C) was investigated. It has been found that the low temperatures induced on the non-chilled organs gave similar changes to organs exposed to a reduced temperature. The changes consisted of a parallel depolarization of leaf and root membranes and similarly a decrease in the ATPase activity as well as changes in the unsaturation of the membranes' fatty acids. The reduction of the acid content 18:2 was particularly significant, which in the leaf microsomes correlated with a drop in the ATPase activity.
The similarity between the reactions of the chilled and non-chilled parts of the plant gives evidence of a communication' between the organs which may facilitate a coordinated reaction of the entire organism to stress. The background for these phenomena could be die action potential of die chilled parts of seedlings which, in the non-chilled parts, might produce changes in the structure of biomembranes and in the activity of enzymes.  相似文献

Abstract— The physiology of essential fatty acid metabolism in the cat is reviewed. Emphasis is placed on those aspects of the n:6 and n:3 fatty acids, their metabolites and interactions, which relate primarily to the skin. The functional roles, if known, of the fatty acids are discussed. Recent clinical research into the use of essential fatty acid supplements in the management of feline dermatoses is presented. 大型Sou的总脂和脂肪酸组成随食物的变化及营养强化的实施而发生显著变化。酵母组总脂为2.5%,明显高于海水小球藻组(1.65%)和有机肥组(1.5%)。经鱼油强化或海水小球藻二次培养后脂类含量明显升高。三组中酵母组的MUFA含量是高为55%。主要是C16:1ω9(23%),而PUFA最低(16.6%),其中EPA(2.7%)、DHA(0.8%)等ω3HUFA含量均很低;与酵母组相比,海水小球藻组和有机肥组的MUFA含量显著下降,PUFA含量显著升高,均为30%-33%左右,三组中海水小球藻组EPA含量最高(14%),其次是有机肥组(5.9%)和酵母组(2.7%);有机肥组DHA含量最高,达11.7%,而酵母组和海水小球藻组DHA含量均低,仅为0.7%-0.8%。经鱼油强化后,酵母组的MUFA含量显著下降,PUFA显著升高,EPA由2.7%提高到10%左右,DHA由0.8%以7%;酵母组经海水小球藻二次培养后MUFA显著降低,PUFA相应升高,EPA由2.7%提高到9.7%,而DHA含量无变化。  相似文献

Ultrasonography of the liver was performed in 200 Holstein-Friesian dairy cattle using a 3.5 MHz transducer with a linear array electronic scanner. Liver samples were taken, processed and examined microscopically and the fat occupying rate was calculated. The hepatic ultrasonograms were evaluated according to the presence of bright pattern, dark pattern, deep attenuation, vascular blurring and blurring of edges. Of the 200 animals, 96 had a normal liver, 63 had hydropic degeneration of the liver, 37 had fatty infiltration of the liver, 3 had liver dystrophy and I had hepatic amyloidosis, diagnosed through histopathological examination. Amyloidosis was characterized by bright pattern and blurring of edges. Liver dystrophy had higher percentages of bright pattern and blurring of edges than normal liver. Hydropic degeneration had higher percentages of dark pattern and blurring of edges than normal liver. Fatty infiltration had higher percentages of bright pattern, deep attenuation, vascular blurring and blurring of edges than normal liver. The present results suggest that different ultrasonographic patterns can be observed in various diffuse hepatocellular disorders in dairy cattle  相似文献

Blood samples were collected every 2 h during a 24 h period from 6 cows of one herd and 10 cows of another herd. In a third herd 9 cows were sampled every 2 h from 6 a.m. to 8 p.m. Concentrations of total bile acids, acetoacetate, glucose and free fatty acids were determined in blood plasma. A marked difference in individual bile acid concentrations and patterns of diurnal variation was found. For most cows the highest bile acid values were observed between 2 and 6 a.m. (overall mean (+/- SD) at 6 a.m.: 104 +/- 84 mumol/l, range: 20-307 mumol/l). Fourteen cows with a bile acid value greater than 90 mumol/l at 6 a.m. ("high BA") were characterized as a group by showing a pronounced decrease in the mean bile acid concentration after morning feeding. In the group of 11 cows with a 6 a.m. bile acid value less than 90 mumol/l ("low BA") the time of day did not contribute significantly to the bile acid variation. For the "high BA" group a nearly synchronous variation between the mean values of the 3 feeding dependent parameters (acetoacetate, glucose and free fatty acids) and the mean values of bile acids was found. The within animal coefficients of correlation between bile acids and the feeding dependent parameters were significantly higher in the "high BA" group than in the "low BA" group. No direct connection was found between bile acid levels and the quantity of concentrates fed or the individual milk yield.  相似文献

Protein kinase C (PKC) activity was evaluated and the phosphorylation of its endogenous substrates was explored in fatty liver induced by administration of ethionine (an analogue of methionine) to cows in order to assess the relevance of PKC-dependent phosphorylation in the development of fatty liver. PKC activity was decreased in both the cytosolic and the total particulate fractions from fatty livers, compared to the corresponding fractions from control liver. The mode of activation by the PKC cofactors (1-oleoyl-2-acetyl-sn-glycerol, 12-O-tetradecanoylphorbol-13-acetate, phosphatidylserine and Ca2+) was similar in both control and fatty livers, suggesting a quantitative but not a qualitative change in PKC in fatty liver. At least three substrate proteins (34 kDa, 26 kDa and 19 kDa) were found in the cytosolic fraction and their phosphorylation was reduced in fatty liver.
