番茄内生细菌种群动态分析及拮抗菌株的筛选   总被引:3,自引:3,他引:3  
摘要:采用研磨组织稀释分离法对3个种植地区的番茄内生细菌的种群分布、数量变化规律进行了初步探讨。共分离、纯化得到内生细菌374株,经形态及理化特征鉴定,分属10个不同的属,13个种。其中假单孢菌属Pseudomonas sp.,黄单孢菌属Xanthomonas sp.;肠杆菌属Entenobacter sp.和芽孢杆菌属Bacillus sp.为优势种群。内生细菌在番茄植株种群数量从0.21x104 到13.82x104 cfu.g -1鲜重之间,但不同品种、组织、生育期及种植地,内生细菌的种群和数量各不相同,各组织中内生细菌的种群及数量根中最多,其次是茎和叶,成株期内生细菌无论种类和数量都明显高于苗期。采用平板对峙法从分离的内生细菌中筛选出对番茄灰霉病菌具有体外拮抗活性的菌株32个,占菌株总数的8.56%; 25个菌株对叶霉病菌有抑制作用,占菌株总数的6.68%;对灰霉病菌和叶霉病菌都具有拮抗作用的菌株20个,占菌株总数的5.35%。对灰霉病菌有强拮抗作用的菌株12个,对叶霉病菌有强拮抗作用的菌株仅1个,其中芽孢杆菌是主要的类群。  相似文献

[目的]研究南五味子果实多糖和脂溶性物质的抗菌抗氧化作用。[方法]采用水提取法、乙醚索氏提取法,分别提取南五味子果实多糖和脂溶性物质,并对其抗菌作用、抑制羟自由基能力进行研究,并初步鉴定果皮脂溶性物质的化学成分。[结果]南五味子果皮、果肉的脂溶性物质对金黄色葡萄球菌有抗菌作用,抑菌圈分别为19.39、18.28 mm;南五味子果皮、果肉、种子的脂溶性物质对枯草芽孢杆菌有抗菌作用,抑菌圈分别为32.87、19.54、7.52 mm。南五味子多糖、果肉和种子的脂溶性物质,抑制羟自由基能力分别为4 011.48、3 079.01、1 082.66 U/m L。[结论]南五味子果实具有较强的抑菌作用及抑制羟自由基能力。南五味子可作为一种野生水果开发。  相似文献

Plant–soil feedbacks are important to productivity and plant community dynamics in both natural and managed ecosystems. Among soil bacteria, the Streptomyces possess particularly strong antagonistic activities and inhibit diverse plant pathogens, offering a clear pathway to involvement in plant–soil feedbacks. We hypothesized that feedback effects and the ability of individual host plant species to foster antagonistic Streptomyces populations may be modified by the richness of the surrounding plant community. To test this, we collected soil associated with four different plant species (two C4 grasses: Andropogon gerardii, Schizachyrium scoparium; and two legumes: Lespedeza capitata, Lupinus perennis), grown in communities that spanned a gradient of plant species richness (1, 4, 8, 16, or 32 species). For each of these soils, we characterized the potential of soil Streptomyces to antagonize plant pathogens, using an in vitro plate assay with indicator strains to reveal inhibition. We cultivated each plant species in each conditioned soil to assess feedback effects on subsequent plant growth performance. Surrounding plant richness modified the impacts of particular plant species on Streptomyces antagonistic activity; A. gerardii supported a higher proportion of antagonistic Streptomyces when grown in monoculture than when grown in 32-spp plant communities, and L. capitata supported more strongly antagonistic Streptomyces when grown in 4- or 32-spp plant communities than in 8-spp plant communities. Similarly, the feedback effects of particular plant species sometimes varied with surrounding plant richness; aboveground biomass production varied with plant species richness for A. gerardii in L. perennis-trained soil, for L. capitata in A. gerardii-trained soil, and for L. perennis in L. capitata-trained soil. Streptomyces antagonist density increased with overall Streptomyces density under low but not under high plant richness, suggesting that plant diversity modifies selection for antagonistic phenotypes among soil Streptomyces. This work highlights the complexity of feedback dynamics among plant species, and of plant–microbiome interactions in soil.  相似文献

Mutants defective in motility or antibiotics production were obtained by Tn5 mutagenesis of a biocontrol agent Pseudomonas fluorescens MelRC2Rif (wt). Tomato or melon seeds were co-inoculated with a Tn5 mutant and wt in a 1:1 ratio and then grown in soil for 10 days. There was no change in ratios of Tn5 mutants defective in antibiosis to wt in the process of rhizoplane colonization, suggesting little contribution of in vitro antibiosis to the rhizoplane competence of P. fluorescens MelRC2Rif. Similar results were also obtained when seeds treated with bacteria were planted in soil artificially infested with fungal pathogens. In contrast, ratios of Tn5 mutants defective in motility to wt significantly decreased, suggesting the contribution of motility to the rhizoplane competence of this bacterium. When a non-motile Tn5 mutant and wt were co-inoculated into soil at a matric potential of pF 2.3 (–20 kPa) and plants were then grown, there was no change in the ratio in rhizoplane colonization, suggesting that motility might have a role in the movement along roots but an insignificant role in the movement from bulk soil towards roots. When they were co-inoculated into 0.2% water agar (WA) instead of soil, a remarkable decline in ratios was detected. Thus it was soil structure that hindered the efficiency of motility. Time course enumeration of rhizoplane colonization of tomatoes grown in WA revealed that motility was an important means of movement towards and/or along roots rather than the multiplication on roots. Verticillium wilt caused by a highly virulent, defoliating (D) pathotype of Verticillium dahliae is threatening olive production in Spain and other Mediterranean countries. This disease must be managed by an integrated strategy, in which biocontrol agents can play an important role. We have investigated the potential of Trichoderma asperellum strains for antagonism against V. dahliae and suppression of Verticillium wilt of olive caused by the D pathotype. First, we tested the antagonistic potential of T. asperellum strains Bt2, Bt3 and T25 against six V. dahliae isolates, four of the D and two of the nondefoliating (ND) pathotypes, in different in vitro assays. All T. asperellum strains overgrew the colonies of all V. dahliae isolates to a similar extent. However, extracellular compounds from strains Bt3 and T25 showed higher anti-V. dahliae activities than those of Bt2 in membrane assays. Also, growth of Bt2 was reduced by ND V. dahliae whereas that of Bt3 and T25 was not affected by V. dahliae-secreted compounds. In planta assays using strains Bt3 and T25, and ’Picual’ olive plants, showed that the two T. asperellum strains significantly reduced the severity of symptoms and the standardized area under the disease progress curve caused by highly virulent D V. dahliae, but not the final disease incidence. Strain T25 significantly increased growth of ‘Picual’ plants and displayed higher ability for colonizing the olive rhizosphere and establishing endophytic infection in olive roots than Bt3.  相似文献

本试验用22种抗菌药物,对70株仔猪腹泻大肠杆菌进行敏感试验,结果表明:抑菌作用最强的是:氟呱酸(高度敏感菌株占100%)、丁胺卡那霉素(高敏、中敏菌占100%)、妥布霉素和先锋霉素_4(高敏、中敏菌均为94.28%),其次为庆大霉素(91.42%)新霉素(90%)和卡那霉素(88.57%)。  相似文献

A method for the eradication of Clavibacter michiganensis subsp. michiganensis was tested for its efficacy in three experiments carried out in the laboratory and greenhouse. In the first experiment, peat moss and sand mix in pots was amended with fresh tomato debris which was either artificially infected with the pathogen, or was not amended. Pots were enclosed in plastic bags or left open. Two temperatures (25 °C and 45 °C) were tested over a 6-week period. The pathogen was not detected in the amended soil after 4 weeks treatment at 45 °C, but was not eradicated after treatments in open pots at 25 °C.In the second experiment, the survival of C.m. michiganensis in either artificially infested soil or in artificially infected tomato plants was studied to determine the behaviour of the pathogen under these conditions. Strains of saprophytic bacteria in the genera Bacillus, Paenibacillus and Brevibacillus were identified under the experimental conditions. In vitro antagonism between Bacillus subtilis and C.m. michiganensis was observed. Finally, the recovery of C.m. michiganensis introduced into disinfected substrate was determined. Survival of C.m. michiganensis in plates artificially inoculated with substrate was greatly reduced after a 4-week treatment at 25 °C, or after 1 week at 45 °C.C.m. michiganensis remained pathogenic on plant tissue after 4 weeks of either thermal treatment.It is important to take these results into account with regard the effect of different soil disinfection techniques or ecological alternatives such as biofumigation, solarization, and the addition of organic matter, as well as for integrated pest management systems.  相似文献

本文对一种经过纳米无机银类抗菌材料和纳米TiO2光催化剂处理的抗菌耐磨实木复合地板的抗菌性能和耐磨性能进行了检测和分析。结果表明,经处理的产品能够抑制室内常见的金黄色葡萄球菌以及大肠杆菌的繁殖,同时对“黑曲霉”有防霉和灭菌的效果,且其表面耐磨性高于实木地板和实木复合地板。  相似文献

 利用经苗期和田间抗性鉴定对白背飞虱(Sogatella furcifera)具有中抗水平以上的国内外品种52个进行拒性和22个品种进行抗生性测定。结果表明,对若虫取食、成虫定居和产卵的拒性以云南品种的毫糯亮、白糯、鬼衣谷、滇瑞337~68等较为明显。抗生性(种群增长、若虫生存率、排泄量)测定结果表明:N22、冷水谷(681)、脚鲁、法炮谷、浙丽1号等5个品种抗生性明显。  相似文献
