抗旋毛虫循环抗原单克隆抗体的制备及初步应用

摘    要:为评价单克隆抗体用于检测旋毛虫循环抗原(CA)的应用价值,作者利用淋巴细胞杂交瘤技术制备了4个抗旋毛虫CA的杂交瘤细胞系,建立了以单克隆抗体双抗体夹心ELISA检测CA的方法,并初步应用于动物血清中CA的检测。应用该法检测旋毛虫病猪血清的阳性率为72.1%(31/43)。60头健康猪、30头感染猪囊虫和30头感染弓形虫的猪均为阴性。对流行地区河南邓县34头和湖北囊樊134头屠宰猪进行流行病学调查,

Abstract:In the present study,a trial was made to evaluate the monoclonal anti-body(McAb ) produced as tool in the detection of circulating antigen(CA)ofTrichinella spiralis. Four hybridomas secreting McAbs against the CA of T. spiraliswere established. The McAbs secreted by F1 , F2 and G5 were IgG1, and that by H8 wasIgM. The titers of F1, F2 and G5 ascites were 1 :160 000,and H8 was 1 :1 000. TheMcAbs secreted by F2and G5were specific to T. spiralis ,while secreted by both F1 andH8 could react with Cysticercus cellulosae. The McAb secreted by F2 was labelled withhorseradish peroxidase(HRP). The labelled McAb was used to detect CA by sandwich ELISA method. Sera of the infected pigs with T. Spiralis(confirmed by musclebiopsy ) were examined for CA of this parasite. The positive rate was 72.1%(31/43). Control serum specimens obtained from 60 healthy swine were all negative. In anothergroup of 60 swine sera, 30 with cysticercosis and 30 with toxoplasmosis, were all nega-tive too. A survey was made on slaughtered pigs naturally infected with T. spiralis inDengzhou and Xiangfan. A total of 168 pigs were examined and the average prevalencerate by CA was 9. 52%(16/168); and 68.8/(11/16) of them was also anti-trichinellaantibody positive. Thirty mice were orally infected with T. spiralis larvae. Their serawere collected at different periods,i.e.4,7,11,18 and 28 days post infection for thedetection of CA which was done by the sandwich ELISA, CA could be detected on day 4post infection, the positive rate was 60%( 18/30);while on day 7 and 11 it was allclearly identified(30/30);on day 18 it could be detected but at a lesser OD reading;however on the 28th day the positive rate dropped to 13.3/(4/30).This study con-firmed that McAb may be a valuable tool in the detection of CA,especially in the earlydiagnosis of trichinosis,and it is convenient for epidemiological survey in field.
