新民猪和皮民猪CAST基因的多态性检测分析

摘    要:为了研究新民猪和皮民猪钙蛋白酶抑制蛋白(CAST)基因的多态性,试验采用PCR-RFLP法对27头新民猪和50头皮民猪F1代CAST基因的2个突变位点进行检测,并与已报道的猪种进行比较。结果表明:新民猪Arg249Lys突变位点的A等位基因频率为66.67%,B等位基因频率为33.33%;Ser638Arg突变位点的A等位基因频率为18.00%,B等位基因频率为82.00%。皮民猪Arg249Lys突变位点的A等位基因频率为94.00%,B等位基因频率为6.00%;Ser638Arg突变位点的A等位基因频率为28.00%,B等位基因频率为72.00%。与其他猪种相比,新民猪和皮民猪的Arg249Lys突变位点的A等位基因频率偏高,Ser638Arg突变位点的A等位基因频率偏低,从而引起有利单倍型249Lys-638Arg频率较低。

Abstract:To study the polymorphisms of the CAST genes from new Min pigs and the F1 generations of Pietrain×new Min pig,the PCR-RFLP method was used to detect the 2QTL in CAST genes from 27 new Min pigs and 50 F(1) generations of Pietrain ×new Min pig,compared with other pig breeds reported.The results showed that in new Min pigs,the A allele frequency of CAST Arg249Lys was 66.67%,and the B allele frequency was 33.33%;the A allele frequency of CAST Ser638Arg was 18.00%,and the B allele frequency was 82.00%.In the F(1) generations of Pietrain ×new Min pig,the A allele frequency of CAST Arg249Lys was 94.00%,B allele frequency was 6.00%;the A allele frequency of CAST Ser638Arg was 28.00%,and the B allele frequency was 72.00%.Compared with the other breeds,in new Min pigs and the F(1) generations of Pietrain ×new Min pig,the A allele frequency of CAST Arg249Lys was much higher than that of the others,the A allele frequency of CAST Ser638Arg was much lower than that of the others,thus causing the lower frequency of favorable haplotype 249Lys-638Arg.
