摘    要:分析比较患病与健康虾夷马粪海胆体腔液菌群的PCR-DGGE指纹图谱.并通过切胶、克隆测序等步骤分析患病虾夷马粪海胆体腔液中的特异菌群.结果显示,健康海胆体腔液中菌群的PCR-DGGE指纹图谱具7个条带,其中有5个与患病海胆相同,2个为不同条带,表明健康海胆体腔液中至少含7种细菌,其中2种以上细菌随机体环境变化而消亡;患...

Abstract:The bacterial community composition in the coelomic fluid of sick and healthy S. intermedius was analyzed using 16S rDNA PCR-DGGE fingerprint, and the special bacterium was analyzed by excising gel cloning and sequencing. The result indicated that 7 bands were observed in the PCR-DGGE fingerprint of coelomic fluid of healthy sea urchin, in which 5 bands were the same as the sick sea urchin, 2 were different bands. This suggested that there were at least 7 different bacteria in coelomic fluid of healthy sea urchin, and more than 2 bacteria disappeared with the internal environment change. 11 bands were found in sick sea urchin, including 6 special bands (Band 1~6) corresponding with each single bacterium, among which one was Sphingomonas sp., 3 were Vibrio sp., and the other 2 bands were not identified. Three vibrio strains had similarity with V. splendidus, V. tasmaniensis and V. cyclitrophicus, and were very likely to be the pathogen bacteria of S. intermedius.
