| 摘 要： | 从黑龙江某猪场疑似猪流行性腹泻猪的粪便中提取总RNA，采用RT—RCR方法扩增，首次获得中国地方流行株PEDVM全基因序列，将其克隆到pMD18-T载体中进行测序，克隆的M基因核苷酸序列由681个核苷酸组成，含有一个完整的开放阅读框架，编码226个氨基酸。与国外已发表的其他毒株进行基因进化分析，GenBank中的6PEDV形成的基因进化树具有3个分支。其中分离株LJB／03形成一个独立的分支，说明LJB／03的M基因序列与其他毒株相比，发生了一定的变异。 |

| Abstract: | A porcine epidemic diarrhea virus (PEDV) LJB/03 was isolated from the faece of piglets infected with PEDV on a pig farm, Heilongjiang province, China. The M gene of LJB/03 was amplified from the RNA extracted directly from the faece samples by RT-PCR and cloned into pMD18-T vector. The complete genomic nucleotide sequence was determined by sequencing, and it encompasses an open reading frame of 681 nucleotides, encoding 226 amino acides. Genetic analysis of the M gene, including genetic distance measurement, phylogenetic tree analysis, and residue substitution analysis, showed that the PED viruses fell into three groups, and the LJB/03 itself was as an independent group. These data revealed that the M gene nucleotide sequence of LJB/03 has some mutations compared with the other PED viruses. |
