黄喉拟水龟松鼠葡萄球菌16S rDNA的序列测定和系统进化分析

摘    要:从患病的黄喉拟水龟肝脏、肾脏分离到1株葡萄球菌(YLD81101),经人工感染实验证实其为该病的病原菌。为了从分子水平上对其进行分类学鉴定,采用扩增细菌16S rDNA的通用引物对该菌的16S rDNA进行PCR扩增、克隆及序列分析。结果扩增出长约1.5kp的目的片段,测序得到1条长度为1515bp的核苷酸序列(GenBank登录号GQ261178)。核苷酸相似性分析表明,所得序列与GenBank公布的松鼠葡萄球菌(Staphylococcus sciud,S.sciuri)CTSP9菌株16S rDNA核苷酸序列相似性最高(99.9%)。同时,用与该序列相关性较高的S.sciuri及常造成动物葡萄球菌病的S.aureus S、saufeuss.bsp.anaerobius、S.gallinarum、S.hyicus代表菌株构建的系统发育进化树显示,菌株YL081101与S.sciuri代表株聚为一簇。上述研究证实,分离菌株YL081101为松鼠葡萄球菌。

Abstract:In the present study, a Staphylococcus strain (YL081101) was isolated from the liver and kidney of the diseased Mauremys mutica Cantor, artificial infection of which proved that Staphylococcus was the pathogen of disease. In order to identify the classification of strain YL081101 at moleculer level, 16S rDNA of strain YL081101 was amplified using the universal primers designed for eubacterial 16S rDNA, and the PCR product was cloned and sequenced. As a result, 1.5kb desired fragment was amplified and nucleotide sequence of 1515 bp was obtained from the bacteria (GenBank accession number GQ261178). The sequence had the highest similarity of 99.9% with 16S rDNA of CTSP strain of Staphylococcus sciuri obtained from GenBank using BLAST. The phylogenetic analysis was conducted based on the 16S rDNA sequence of strain YL081101 and other related organisms, including the representative strains of S. sciuri, S. aureus, S. aureus subsp, anaerobius, S, gallinarum and S. hyicus species in GenBank database. The result showed that the strain YL081101 and all strains of S. sciuri species formed a separate and distinct phylogenetic branch which revealed that the strain YL081101 is S. sciuri species.
