摘    要:为制备克伦特罗(clenbuterol,CL)单克隆抗体,鉴定其特性,采用重氮化法合成克伦特罗-人血清白蛋白(CL-HSA)作为免疫原免疫小鼠,通过杂交瘤技术建立杂交瘤细胞株,利用体内诱生法制备单克隆抗体,用间接ELISA和间接竞争ELISA分别测定抗体效价和特异性.将细胞反复冻存和传代,考察稳定性.获得细胞株C611...

Abstract:To prepare monoclonal antibody against clenbuterol(CL) and identify its characteristics.Balb/c mice were immunized with the conjugate CL-HAS which had been prepared via a diazotization.The splenocytes of immunized mice were fused with Sp2/0 myeloma cells by a routine method.Monoclonal antibody(mAb) was raised in Balb/c mice and was identified by inditect ELISA for the titer,and complete indirect ELISA for its specificity.One stable hybridoma C611,secreting monoclonal antibodies against clenbuterol(CL) was established.It was highly specific for CL with a titer of 1∶64 000 in the ascitic fluid and showed low cross-reactivities less than 7% to other β-agonists.The limit detection of CL was 0.1 μg/L,and the IC50 was about 1 μg/L.The storage condition and cloning would not influence the property of the hybridomas.The mAb against CL were prepared successfully,and possessed high titer and specificity,which is useful for the development of an ELISA kit for screening of CL residue.
