猪德尔塔冠状病毒(PDCoV)是一种新出现的冠状病毒,能够引起猪只发生呕吐和水样腹泻,临床症状与猪流行性腹泻(PED)和猪传染性胃肠炎(TGE)相似。相关研究表明猪氨基肽酶N(pAPN)是PED病毒(PEDV)和TGE病毒(TGEV)入侵宿主细胞的功能性受体。为研究pAPN是否是PDCoV入侵宿主细胞的受体,本研究以TGEV为指示病毒,通过BHK-pAPN细胞感染试验、pAPNRNA干扰试验、ST细胞过表达pAPN试验、可溶性pAPN阻断试验,发现BHK-pAPN细胞是PDCoV非易感细胞,在易感细胞ST上沉默和过表达pAPN对PDCoV的增殖并无影响,可溶性pAPN不能阻止PDCoV感染ST细胞。这些研究结果表明pAPN不是PDCoV入侵宿主细胞的受体,而且对PDCoV的增殖无影响。 相似文献

目的:研究中药复方对人工感染猪流行性腹泻病毒仔猪的治疗效果。方法:本文研究对象为人工感染猪流行性腹泻病毒感染仔猪,一共50头,均采取抽签分组方式分为两组,收取时间在2015年1月2日~2016年2月10日之间,分为观察组一组(25头应用中药复方治疗)、对照组一组(25头应用西药治疗),将两组治疗效果进行对比。结果:观察组人工感染猪流行性腹泻病毒感染仔猪的总有效率(显效20头、有效3头、无效2头)高于对照组(p 0.05);观察组人工感染猪流行性腹泻病毒感染仔猪的不良反应发生率8.00%低于对照组(p 0.05);治疗后观察组仔猪体重(1.81±0.25)kg与对照组具有显著差异(p 0.05)。结论:通过对人工感染猪流行性腹泻病毒感染仔猪实施中药复方后,取得显著效果,其与西药治疗相比,具有多种优势。 相似文献

猪流行性腹泻病毒(PEDV)属于冠状病毒家族成员,是近年来引起新生仔猪水样腹泻致死的主要病原之一,给养殖业带来了巨大威胁。病毒感染后,宿主细胞通过模式识别受体(PRRs)识别病原相关分子模式(PAMPs),促进I型干扰素等细胞因子的产生,进而抑制病毒的增殖。近年来的研究表明,PEDV能够通过其编码蛋白对宿主的抗病毒天然免疫反应进行调节,成功逃避免疫识别或拮抗宿主的天然免疫反应,为自身的快速复制和增殖创造条件。本文结合目前关于PEDV与宿主细胞相互作用的研究进展,综合分析了PEDV编码的各种蛋白在感染过程中拮抗宿主天然免疫系统的分子机制,旨在为进一步认识PEDV乃至其他冠状病毒的致病机制,也为探索新的抗病毒药物靶标提供思路。 相似文献

猪流行性腹泻病是由猪流行性腹泻病毒感染引起的新生仔猪的一种急性传染病,患病仔猪临床表现为急性腹泻、呕吐、脱水,具有较高的发病率和死亡率。我国于20世纪80年代首次发现猪流行性腹泻病毒,随后该病毒在国内迅速传播,并于2010年由猪流行性腹泻病毒变异株引起了该病的大规模发生,给我国生猪养殖业造成了严重的经济损失。本文对猪流行性腹泻病毒的病毒学、流行病学特点及疫苗研究进展进行综述,以期为猪流行性腹泻病的防控和疫苗研发提供理论参考。 相似文献

猪流行性腹泻病毒(PEDV)是冠状病毒科甲型冠状病毒属的一种有囊膜的单股正链RNA病毒,对全世界养猪业造成了巨大的损失。PEDV与宿主之间的致病机制和免疫调节相互作用仍不清楚。有研究表明冠状病毒可以诱导细胞发生自噬,并且与病毒的复制有着密切的联系,病毒与细胞自噬之间的关系十分复杂,虽然病毒感染与细胞自噬之间的关系研究比较广泛,但细胞自噬在PEDV感染中的作用报道较少。研究PEDV与细胞自噬的相互作用可以加速对PEDV感染入侵机制的理解,为未来研究病毒治疗和抗病毒药物提供新的思路。

猪流行性腹泻(Porcine epidemic diarrhea,PED)是由猪流行性腹泻病毒(Porcine epidemic diarrhea virus,PEDV)引起猪的高度接触性肠道传染病,感染PEDV的猪群主要表现为腹泻、呕吐、厌食、脱水等临床症状。PEDV可感染各个阶段的猪群,临床上PEDV主要造成7日龄以内的仔猪高发病率及高死亡率(可达100%)。PEDV的迅速传播给养猪行业造成了巨大的经济损失。本文针对PEDV的病原学、病毒编码蛋白及其生物学功能、感染细胞机制进行综述,以期为深入研究PEDV的致病机制提供参考。 相似文献

近年来,仔猪腹泻呈多发性趋势,因传播速度快、治愈率低、死亡率高,给猪场带来巨大损失[1],研究发现[2],腹泻死亡的仔猪占仔猪死亡总数的38.9%,可见,该病严重制约着仔猪的健康生长。腹泻病毒感染是导致仔猪腹泻最重要的原因之一。许多病毒可致仔猪腹泻,如诺如病毒、博卡病毒、星状病毒等,但最常见、危害最深的是猪传染性胃肠炎病毒(TGEV)、猪流行性腹泻病毒(PEDV)和猪轮状病毒(RV),目前临床上很难区分这3种病毒[3-4],常呈单独或混合感染,严重影响养猪业发展。如何更好地发现、治疗、预防该病,一直是各级兽医从业人员研究的重点。现就其临床症状、剖检病变、诊断、治疗及综合防制等方面进行简要分析。 相似文献

Porcine viral diarrhea is an acute and highly contagious enteric disease in pigs which causes huge economic losses in pig industry worldwide. Transmissible gastroenteritis virus (TGEV) is main pathogens responsible for piglets viral diarrhea. Knockout the host cellular surface receptor for TGEV may be an effective way to accelerate the breeding of resistant pigs. In this study, we applied site‐specific editing pAPN which is effective in swine testis (ST) cells. Site‐specific editing of pAPN reduced TGEV proliferation in ST cells by 96%–99% at different time periods post‐infection. Next, the site‐specific editing of pAPN porcine fetal fibroblasts were produced, and then the cell colonies were used as donor cells to generate the site‐specific editing of pAPN pigs. Our research findings will not only offer a more thorough understanding of the pathogenesis of piglet diarrhea and lay the foundation for breeding TGEV‐resistant piglets, but also understanding the molecular mechanisms involved in coronaviral infections. 相似文献

The inhibitory effects of rhubarb on cell lesion, which was caused by porcine epidemic diarrhea virus (PEDV), and its mechanism had been studied in this research.The maximum safe concentration of rhubarb solution applied to Vero cells was determined through morphological observation, and then the PEDV infected Vero cell model in vitro was established, by which the inhibitory effect of rhubarb on virus was studied.The experimental method had also been established based on this cell model.The result showed that the maximum safe concentration was 3.28 mg/mL.Both morphological observation and MTT assay were indicated that rhubarb could not only block the attachment of the virus to Vero cell and inhibit the synthesis and reproduction of the virus, but also could directly inactivate the virus.This research showed that rhubarb had an inhibitory effect on PEDV, and provided a theoretical basis for the prevention and treatment of clinical porcine epidemic diarrhea. 相似文献

为研究大黄对猪流行性腹泻病毒(PEDV)感染所引起的细胞病变的抑制作用及其机理,本试验采用形态学观察法测定大黄溶液在Vero细胞上的最大安全浓度,建立体外病毒感染Vero细胞模型并利用该模型进行了大黄对病毒抑制作用的研究。结果显示,本试验成功建立了体外病毒感染模型和大黄的抗病毒试验方法;大黄在Vero细胞上的最大安全浓度为3.28 mg/mL;形态学观察法和MTT法均显示大黄不仅能阻断病毒吸附Vero细胞,抑制病毒的合成复制,还可直接灭活病毒。本研究结果表明,大黄具有抗PEDV的作用,为临床上预防和治疗猪流行性腹泻(PED)提供理论基础。 相似文献

猪流行性腹泻病毒在Vero细胞上培养成功后,病毒全基因组的序列和分子结构特征,结构蛋白和非结构蛋白的生物学特性和功能,病原学、血清学和分子生物学的诊断技术,体液免疫、细胞免疫和局部黏膜免疫的机理,疫苗和病毒的细胞受体等方面得到了广泛的研究。但是,与其他冠状病毒相比,猪流行腹泻病毒的研究还比较滞后,许多研究还没有开展或不够深入,为了能加快对猪流行性腹泻病毒研究进程,对其最近的分子生物学研究进展做了综述,为猪流行性腹泻病毒的深入研究提供参考。 相似文献

应用抗猪流行性腹泻病毒(PEDV)单克隆抗体(McAb)杂交瘤细胞株诱生腹水McAb,将3株单抗混合,用于实验室防治试验。结果表明,其对病毒具有中和作用,可使病猪耐过PEDV的侵袭;用于生产场的发病猪,初步证明疗效显著,保护率达91%,而未注抗体的对照发病猪全部死亡。上述结果表明,抗猪流行性腹泻病毒McAb混合制剂确能缓解发病症状,减少新生猪的死亡。 相似文献

Porcine epidemic diarrhea virus (PEDV) was isolated in Vero cell cultures from the small intestine of a piglet experimentally infected with porcine coronavirus 83P-5, that had been isolated during outbreaks of porcine acute diarrhea and passaged in piglets. The isolation of the PEDV was successful only in Vero cells maintained in the maintenance medium (MM) containing trypsin. Infected Vero cell cultures exhibited CPE characterized by cell-fusion and syncytial formation, as well as cytoplasmic fluorescence when examined by the indirect immunofluorescent test using rabbit anti-83P-5 virus serum. The isolate was adapted to serial propagation in Vero cell cultures by adding trypsin to MM. Vero cell-adapted PEDV was successfully propagated in the MA104, CPK and ESK cell lines in the presence of trypsin in MM. Vero cell-adapted PEDV had morphologic and physicochemical characteristics similar to those of other members of the coronaviridae. The isolate differed serologically from porcine transmissible gastroenteritis (TGE) and porcine hemagglutinating encephalomyelitis viruses, and no antigenic relationship between the isolate and TGE virus could be detected by the indirect immunofluorescent test. Attempts to isolate PEDV in 6 types of primary fetal pig cell cultures and 6 of 10 established cell lines resulted in the failure, probably because these cells were damaged by the action of trypsin. 相似文献

Isolation of porcine epidemic diarrhea virus in porcine cell cultures and experimental infection of pigs of different ages 总被引:6,自引:0,他引:6
This paper describes the isolation of porcine epidemic diarrhea (PED) virus in Vero and porcine cell cultures, and the influence of age on disease in experimental infection. PED virus was isolated from the small intestine of piglets inoculated with PED samples and cultured in Vero, porcine bladder and kidney cells propagated in collagen-coated tissue culture plates in maintenance medium (MM) containing trypsin. In porcine bladder and kidney cell cultures inoculated with isolated PED virus, cytopathic effects (CPE) including cell fusion were detected. Specific brilliant fluorescence was observed in the cytoplasm of these cells. Two- and 7-day old, and 2-, 4-, 8- and 12-week old specific pathogen-free (SPF) pigs were orally inoculated with PED virus isolated from an outbreak. All 2- and 7-day old pigs inoculated developed severe watery diarrhea from post-inoculation day (PID) 1 and died between PID 3 and 4. Although three of five 2-week old pigs developed diarrhea on PID 1-4, they eventually recovered. In the 4-week old group, three of five pigs had mild diarrhea for 1-2 days. None of the 8- and 12-week old pigs showed any clinical signs. Antibodies against PED virus were detected in all surviving pigs by virus neutralization (VN) test and immunofluorescence assay (IFA).
