It is well known that after a stressor, levels of plasma cortisol rise, inducing physiological changes within the animal that are directed toward maintaining homeostasis. Less well understood is the role of cortisol in regulating food intake in teleosts. This study investigated the effect of cortisol on food intake and regulation of the neuroendocrine appetite-stimulating hormones, neuropeptide Y (NPY) and ghrelin, in tilapia (Oreochromis mossambicus). Male and female tilapia were randomly assigned to one of the following treatments: unhandled control, vehicle-injected control, or cortisol (2 μg/g BW). Food intake was determined 24 h after injection during a 1-h feeding trial. Cortisol reduced food intake (P < 0.001). An identical study was conducted to measure the effects of 24-h cortisol treatment on the endocrine regulators of food intake. Cortisol reduced stomach expression of ghrelin mRNA (P < 0.05) and plasma concentrations of ghrelin (P < 0.05). In the hypothalamus/optic tectum cortisol reduced levels of GHSR1a-LR (biologically active ghrelin receptor) mRNA. In the telencephalon/preoptic area cortisol significantly reduced levels of NPY and GHSR1b-LR (biologically inactive ghrelin receptor) mRNA. These findings suggest that anorexigenic actions of cortisol may be mediated via two separate pathways: (1) reducing circulating ghrelin levels as well as GHSR1a-LR expression in the hypothalamus/optic tectum and/or (2) suppressing NPY expression in the telencephalon/preoptic area.  相似文献

Ghrelin has been implicated in the control of cell proliferation in reproductive tissue. Here, we provide evidence that both ghrelin mRNA and protein are present in ovarian follicles. Persistent expression of ghrelin was also demonstrated in sheep ovary throughout the estrous cycle and pregnancy. In fact, the relative mRNA levels of ghrelin varied depending on the stage of the cycle, with the highest expression during the development of the corpora lutea (CL) and minimal expression in the regressing CL. A similar pattern was seen during pregnancy. Dynamic changes in the profile of ghrelin expression during the estrous cycle and throughout pregnancy suggest a precise regulation of ovarian expression of ghrelin, which could represent a potential role for ghrelin in the regulation of luteal development. In conclusion, the presence of the ghrelin signaling system within the sheep ovary especially in the oocytes opens up the possibility of a potential regulatory role of this novel molecule in reproductive function.  相似文献

Recent studies have implicated the peripheral actions of ghrelin in reproductive tissues. Here, we provide evidence that both ghrelin and its receptor GHSR-1a (the type 1a growth hormone secretagogue receptor) are expressed at both mRNA and protein levels in sheep oocytes and pre-implantation embryos produced in vitro . Real-time RT-PCR experiments confirmed that ghrelin mRNA levels varied depending on developmental stage, with the highest expression in metaphase II (MII) oocytes, higher expression at the 2-cell stage, and minimal expression in germinal vesicle (GV) oocytes, 4- and 8-cell stages, and in the blastocyst. The levels of GHSR-1a mRNA decreased from GV to MII, increased immediately at the 2-cell stage and then remained stable until the blastocyst stage. Ghrelin protein was detected mainly in the cytoplasm close to the plasma membrane in both inner cell mass and trophectoderm cells, while GHSR-1a protein was most abundant in the plasma membrane. In conclusion, the presence of the ghrelin signalling system within the sheep oocytes and pre-implantation embryos opens up the possibility of a potential regulatory role of this novel molecule in reproductive function.  相似文献

The effect of appetite regulatory hormone cholecystokinin (CCK) on the secretions of oxyntomodulin (OXM) and ghrelin, and the effect of ghrelin on the secretions of CCK and OXM were studied in ruminants. Eight Holstein steers, 7 months old, 243 ± 7 kg body weight (BW), were arranged in an incomplete Latin square design (8 animals × 4 treatments × 4 days of sampling). Steers were intravenously injected with 10 µg of sulfated CCK‐8/kg BW, 20 µg of acyl ghrelin/kg BW, 100 µg of des‐acyl ghrelin/kg BW or vehicle. Blood samples were collected from ?60 min to 120 min relative to time of injection. Plasma concentrations of ghrelin, sulfated CCK and OXM were measured by double‐antibody radioimmunoassay. Plasma acyl ghrelin was increased to peak level (428.3 ± 6 pg/mL) at 60 min after injection of CCK compared with pre‐injected levels (203.3 ± 1 pg/mL). These results showed for the first time, that intravenous bolus injection of CCK increased ghrelin secretion in ruminants. In contrast, injection of ghrelin did not change CCK secretion. Administration of ghrelin or CCK has no effect on plasma OXM concentrations. In conclusion, our results show that administration of CCK increased ghrelin secretion but did not affect OXM release in ruminants. Ghrelin did not affect the secretions of CCK and OXM.  相似文献

In this study, the pharmacokinetic profile of flumequine (FMQ) was investigated in blunt snout bream (Megalobrama amblycephala) after intravascular (3 mg/kg body weight (b.w.)) and oral (50 mg/kg b.w.) administrations. The plasma samples were determinedby ultra‐performance liquid chromatography (UPLC) with fluorescence detection. After intravascular administration, plasma concentration–time curves were best described by a two‐compartment open model. The distribution half‐life (t1/2α), elimination half‐life (t1/2β), and area under the concentration–time curve (AUC) of blunt snout bream were 0.6 h, 25.0 h, and 10612.7 h·μg/L, respectively. After oral administration, a two‐compartment open model with first‐order absorption was also best fit the data of plasma. The t1/2α, t1/2β, peak concentration (Cmax), time‐to‐peak concentration (Tmax), and AUC of blunt snout bream were estimated to be 2.5 h, 19.7 h, 3946.5 μg/L, 1.4 h, and 56618.1 h. μg/L, respectively. The oral bioavailability (F) was 32.0%. The pharmacokinetics of FMQ in blunt snout bream displayed low bioavailability, rapid absorption, and rapid elimination.  相似文献

Ghrelin is a highly conserved peptide hormone secreted by the stomach, which is involved in the regulation of food intake and energy expenditure. Ghrelin stimulates growth hormone (GH) release, and increases appetite in a variety of mammalian and non-mammalian vertebrates, including several fish species. Studies were conducted to investigate the effect of feeding and fasting on plasma and stomach ghrelin, and the growth hormone/insulin-like growth factor I (IGF-I) axis in the Mozambique tilapia, a euryhaline teleost. No postprandial changes in plasma and stomach ghrelin levels or stomach ghrelin mRNA levels were observed. Plasma levels of GH, IGF-I and glucose all increased postprandially which agrees with the anabolic roles of these factors. Fasting for 4 and 8 d did not affect ghrelin levels in plasma or stomach. Plasma GH was elevated significantly after 4 and 8 d of fasting, while plasma IGF-I levels were reduced. Plasma ghrelin levels were elevated significantly after 2 and 4 wk of fasting, but no change was detected in stomach ghrelin mRNA levels. Four weeks of fasting did not affect plasma GH levels, although plasma IGF-I and glucose were reduced significantly, indicating that GH resistance exists during a prolonged nutrient deficit (catabolic state). These results indicate that ghrelin may not be acting as a meal-initiated signal in tilapia, although it may be acting as a long-term indicator of negative energy balance.  相似文献

本试验旨在研究饲料中添加枯草芽孢杆菌对团头鲂幼鱼生长性能、肝脏抗氧化指标、肠道菌群结构和抗病力的影响。选取初始体重为(1.81±0.01)g的团头鲂幼鱼360尾,随机分为4组(每组3个重复,每个重复30尾),分别饲喂添加0(T0组,作为对照组)、2×10~7(T1组)、2×10~8(T2组)、2×10~9CFU/g枯草芽孢杆菌(T3组)的等氮等脂饲料。试验期为8周。结果表明:1)与对照组相比,T1组和T2组团头鲂幼鱼的增重率、特定生长率显著升高(P0.05),且T1组的饲料系数显著降低(P0.05)。2)与对照组相比,T1组团头鲂幼鱼的肝脏抗氧化酶(超氧化歧化酶、过氧化氢酶和谷胱甘肽过氧化物酶)活性显著升高(P0.05),同时该组肝脏丙二醛含量显著降低(P0.05)。3)各组全鱼水分、粗蛋白质、粗脂肪和粗灰分含量均无显著差异(P0.05)。4)变性梯度凝胶电泳(DGGE)图谱显示摄食不同添加量枯草芽孢杆菌饲料的团头鲂幼鱼的肠道菌群结构发生了改变。5)采用嗜水气单胞菌对鱼体进行攻毒,各组96 h后的累积死亡率未产生显著差异(P0.05),但T1组和T2组的累积死亡率低于对照组和T3组。由此可见,饲料中添加2×10~7CFU/g的枯草芽孢杆菌能够提高团头鲂幼鱼的生长性能、抗氧化能力,并改变肠道菌群结构,但对其体成分和抗病力没有显著影响。  相似文献

The larval development of the southern African endemic sparid Spondyliosoma emarginatum is described and illustrated from specimens collected in Algoa Bay and the Swartkops, Sundays and Swartvlei estuaries of the Cape Province. S. emarginatum has demersal eggs. Preflexion larvae are moderately elongate (BD = 18–23% BL) with body depth increasing to 31 % BL in postflexion stages. The head is rounded and the snout blunt but becomes more elongate in postflexion stages. There are short spines on the preoperculum. Flexion occurs at about 6 mm BL and postflexion larvae are characterized by distinct medio-lateral pigmentation. Osteological development is described from a series of cleared and stained specimens. All fins have the adult complement of spines and rays by 12 mm BL. The larvae of S. emarginatum are compared with those of the European S. canthurus and other southern African sparids.  相似文献

Ghrelin, an orexigenic hormone, is the endogenous ligand for the growth hormone secretagogue receptor (GHSR). Obestatin is produced from the same precursor peptide as ghrelin, and although obestatin was initially thought to promote actions opposite to those of ghrelin, many studies have failed to confirm this hypothesis. In the current study, multiparous cows were continuously infused with ghrelin (n = 10) or obestatin (n = 10) for 8 wk and compared to an untreated group (n = 10) to examine the effects of these hormones on somatotropic and liporegulatory gene expression. The expression of key genes was measured by quantitative real-time polymerase chain reaction. Growth hormone secretagogue receptor mRNA expression was altered in ghrelin- and obestatin-infused cows in a similar manner, as expression was increased at 4 wk, however it had decreased by 8 wk. Obestatin-infused cows presented with a significant decrease in the expression of ATP-binding cassette A1 (ABCA1) in adipose tissue, suggesting changes in cholesterol transport. Liver insulin-like growth factor (IGF) binding protein-3 mRNA displayed a week-by-treatment interaction, as expression was increased in control and obestatin-infused cows; however, expression decreased in ghrelin-infused cows. Adipose expression of hormone sensitive lipase (LIPE) mRNA was not altered by treatment or time, suggesting hormone infusion is not initiating lipolysis. The expression of lipogenic genes in adipose tissue increased with time in all groups, consistent with the general lactational profile of lipogenesis in dairy cows. These data indicate that continuous infusion of ghrelin or obestatin does not alter the expression of key somatotropic or liporegulatory genes in the lactating dairy cow, although obestatin infusion may alter cholesterol transport.  相似文献

Ghrelin, a novel motilin-related endogenous ligand for growth hormone secretagouge receptor, is implicated in various biological functions, including regulation of female reproduction. But the presence of ghrelin and its role in reproductive functions in buffalo, a species with poor reproductive efficiency, is not known. In the present study full-length ghrelin cDNA was isolated from bubaline abomasum, which encodes the entire prepropeptide of 116 amino acids. The deduced amino acid sequence of ghrelin of buffalo showed >95% and 31% identity with that of ruminants (cattle, sheep, and goat) and humans, respectively. Analysis of synonymous and nonsynonymous nucleotide substitutions in the coding region of ghrelin indicated that these sequences of different species have been under purifying selection. The 3995-bp amplicon of ghrelin gene consisting of 4 exons and 3 introns was cloned with genomic DNA from buffalo. Further, ghrelin expression was determined by quantitative real-time PCR, in situ hybridization, and immunohistochemistry in bubaline endometrial tissues at different stages of the estrous cycle and early pregnancy. Our results indicated the persistent expression of ghrelin mRNA and protein in the endometrium during stage I (day 3–5), stage II (day 6–15), and stage III (day 16–21) of the estrous cycle and also during early (∼day 30–40) pregnancy. Immunohistochemistry and quantitative real-time PCR experiments indicated the relatively higher expression of ghrelin in the endometrium during stage II (day 6–15) of the estrous cycle and early pregnancy than during stage I (day 3–5) and stage III (day 16–21) of the estrous cycle, but no statistically significant difference in ghrelin expression was observed among stages. To conclude, the results of the present study indicate the persistent expression of ghrelin in the uterine endometrium throughout the estrous cycle and in early pregnancy which might be helpful in determining its role in buffalo reproduction.  相似文献

家蚕幼虫血清蛋白(BmLSP)是由脂肪体细胞合成的一种贮藏蛋白,是研究家蚕幼虫期发育调控的理想靶标。基于家蚕基因组数据,克隆了BmLSP基因5'端侧翼区~1.6 kb的调控序列,构建成以红色荧光蛋白基因DsRed为报告基因的pig-gyBac表达载体并进行转基因注射,在个体水平上验证该启动子的特性。结果表明:BmLSP-DsRed表达框以单拷贝形式整合至家蚕基因组;DsRed在转基因家蚕脂肪体中特异转录表达,其时期表达特征与BmLSP基因基本一致,随发育阶段的不同呈现有规律的变化,即幼虫期表达,但1龄、2龄眠期不表达,3龄、4龄眠期有微弱表达,上蔟后表达量逐渐降低直至化蛾阶段消失。提示BmLSP可能通过其启动子区的特异元件受激素的精确调控而行使功能。  相似文献

Ghrelin is involved in many biological processes, ranging from appetite regulation and the release of growth hormone to the regulation of gastrointestinal motility and secretion processes. Ghrelin expression is not homogenously distributed throughout the gastrointestinal tract; expression is species-specific and can also depend on the animal age. This study was performed to investigate ghrelin immunolocalization in the gastrointestinal tract of pigs at different ages: 1 day (birth), 28 days (weaning), 2 months, 4 months, and 7 months (pre-puberty). Tissue samples were collected along the entire gastrointestinal tract and were examined by immunohistochemistry and double-immunofluorescence. Histometry was performed by counting the number of endocrine ghrelin immunopositive cells in the gastrointestinal mucosa. Ghrelin was found to be present along the swine alimentary canal from the stomach to the caecum. In all regions of the alimentary canal of the animals studied, ghrelin-immunoreactive (IR) cells co-localized with chromogranin-A and were therefore identified as endocrine cells. In the gastric fundus, ghrelin-immunoreactivity was partially detected in co-localization with H-K-adenosine triphosphatase and pepsinogen. Ghrelin-IR endocrine cells were abundant in the oxyntic mucosa but less present in the small intestine and rare in the large intestine. The cell density of the ghrelin-IR endocrine cells was lowest in the oxyntic mucosa of 1-day-old pigs. We can conclude that gastric ghrelin expression is not related merely to age but could also potentially be influenced by food intake.  相似文献

为了探讨Ghrelin在绵羊卵母细胞体外成熟过程中与BCL-2、BAX的相关性及Ghrelin在绵羊卵母细胞成熟过程中可能的作用机理,试验在绵羊卵母细胞成熟液中添加500 ng/mL Ghrelin,采用实时荧光定量PCR分别在0,8,16,24小时时检测卵母细胞BCL-2、BAX的表达量。结果表明:试验组添加Ghrelin后8,16小时时卵母细胞BCL-2 mRNA相对表达量显著升高(P<0.05),但成熟后16,24小时时较8小时时有下降趋势;下降到24小时时,其表达量与0小时时没有显著变化(P>0.05);8,16小时时试验组与对照组比较差异显著(P<0.05),0,24小时时差异不显著(P>0.05)。试验组添加Ghrelin后8,16,24小时卵母细胞BAX mRNA相对表达量较0小时时下降,且差异显著(P<0.05),但成熟后16,24小时时与8小时时比较有上升趋势;在8,16小时时试验组较对照组BAX mRNA相对表达量均有所下降,且差异有显著性(P<0.05),而在0,24小时时差异不显著(P>0.05)。说明Ghrelin在绵羊卵母细胞成熟过程中与BCL-2和BAX的表达存在相关性,推测Ghrelin在卵母细胞成熟过程中存在积极调控作用。  相似文献

Ghrelin is a peptide hormone that has been implicated in the regulation of feed intake, but little is known about its secretion in pigs. Hence, the effect of feeding pattern on the regulation of ghrelin secretion was tested. In experiment 1, barrows were allotted randomly into 1 of 2 groups, (1) ad libitum fed (CONT) and (2) limited access to feed (once per day, MEAL). Blood samples were taken through jugular catheters every 15 min for 6 h after 7 d on the experimental feeding regimen. Plasma concentrations of ghrelin and insulin were determined by radioimmunoassay. Ghrelin concentrations in the MEAL pigs were elevated before feeding and declined after feeding (P < 0.01). No pattern in plasma ghrelin concentrations was observed in the CONT pigs, but ghrelin concentrations were lower than in the MEAL group. Insulin concentrations were greater in CONT pigs (P < 0.01) during most of the sampling and increased after feeding in the MEAL pigs (P < 0.01). In experiment 2, the treatments were the same as in experiment 1; however, the amount of feed was increased in the MEAL group so that their daily intake was similar to the CONT pigs. Ghrelin concentrations in the MEAL group were again elevated before the meal and declined afterward (P < 0.01). Insulin but not glucose concentrations were negatively correlated with ghrelin. Once-per-day feeding resulted in increased plasma concentrations of ghrelin, which decreased after feeding. Ghrelin may be involved in the regulation of feed intake in pigs.  相似文献

昆虫蜕皮激素受体(ecdysone receptor,EcR)是一种核内受体,为蜕皮激素的分子靶标。用0.002 g/L蜕皮激素溶液浸泡过的桑叶饲喂家蚕5龄幼虫,采用双跟踪标定定量PCR(dual-spike-in qPCR)方法,检测家蚕蜕皮激素受体基因BmEcR-A、BmEcR-B1和超气门蛋白(ultraspiracle,USP)基因BmUSP在家蚕幼虫马氏管、脂肪体、中肠组织的转录水平变化,分析蚕体组织以及BmEcR和BmUSP基因在蜕皮激素代谢过程中的作用与表达调控特征。结果表明:家蚕EcR的2种同工型基因BmEcR-A和BmEcR-B1的mRNA转录水平均变化显著,在脂肪体中的转录水平最高,其它组织相对较低;BmUSP的mRNA转录水平同样变化显著,且在脂肪体中的转录水平最高,马氏管次之,中肠最低。另检测正常情况下4龄眠期和5龄末期BmEcR-A、BmEcR-B1和BmUSP的mRNA转录水平相对于其它发育时期较高,且脂肪体中的转录水平要明显高于其它组织。BmEcR-A、BmEcR-B1和BmUSP均在家蚕幼虫脂肪体中高水平表达,表明了脂肪体在蜕皮激素代谢过程中的重要性。  相似文献

This study was designed to investigate the effect of ghrelin on gastric acid secretion in weaning piglets both in vivo and in vitro. Thirty newborn piglets were selected from six litters and on 28, 35 (weaning), 38, 42 and 45d of age, respectively, one piglet from each litter was killed and the mucosal tissue from gastric fundus was collected for detecting ghrelin mRNA as well as H(+)-K(+)-ATPase mRNA expression and activity. Primary cultures of gastric mucosal cells from 5-week-old weaning piglets were challenged with 3x10(-5), 3x10(-4), 3x10(-3), 3x10(-2) and 3x10(-1)nmol/ml h-ghrelin, respectively, for 4h in order to further clarify the effect of ghrelin on gastric H(+)-K(+)-ATPase mRNA expression and activity. Ghrelin mRNA expression in gastric fundus kept stable from 28d to 42d, followed by a sudden increase on 45d, exhibiting a peak that was significantly higher than any other age groups investigated. H(+)-K(+)-ATPase activity and mRNA expression showed similar trends of increase with slightly different timing. H(+)-K(+)-ATPase mRNA expression tended to increase on 42d, while H(+)-K(+)-ATPase activity started to rise from 35d, but neither of them reached significantly higher levels until 45d. In vitro, ghrelin significantly increased H(+)-K(+)-ATPase activity of gastric mucosal cells at 3x10(-4), 3x10(-3), and 3x10(-2)nmol/ml, but augmented H(+)-K(+)-ATPase mRNA expression only at 3x10(-4)nmol/ml. The results indicate that ghrelin mRNA expression is up-regulated 10 days after weaning in the gastric fundus of piglets, coinciding with the increase of H(+)-K(+)-ATPase mRNA expression and activity. Ghrelin acts on gastric mucosal cells to stimulate both mRNA expression and activity of H(+)-K(+)-ATPase in vitro.  相似文献

本试验旨在探讨高温季节饥饿后补偿摄食对团头鲂幼鱼生长性能、血清生化指标、肠道消化酶活性及肝脏抗氧化酶活性的影响。试验设计5个组,分别为饥饿0、5、10、15、20 d后再补偿摄食3周,依次标记为S0(对照)、S5、S10、S15、S20组。选择初始均重为(22.03±0.04)g的团头鲂幼鱼270尾,随机分入这5组,每组3个重复,每个重复18尾。结果显示:1)S10组试验鱼末体均重、增重率、特定生长率和饲料效率均达到最高,且补偿摄食期间其日摄食量和饲料效率显著高于对照组(P0.05),说明团头鲂幼鱼补偿生长是通过提高摄食率和饲料效率共同实现的。2)S5、S10组血清甘油三酯水平显著低于其他组(P0.05),而S20组血清谷草转氨酶活性显著高于其他组(P0.05),说明饥饿时间过长鱼体肝脏受到一定程度损伤;血清中其他生化指标如葡萄糖、总蛋白、白蛋白、总胆固醇水平和谷丙转氨酶活性在各组间无显著差异(P0.05)。3)补偿摄食后,S15组肠道脂肪酶活性显著高于其他组(P0.05);S20组肠道蛋白酶活性显著高于其他组(除S5组外)(P0.05),说明饥饿时间过长在补偿摄食后鱼体内蛋白质消化能力增强。4)各饥饿后补偿摄食组肝脏超氧化物歧化酶(除S5、S20组外)和过氧化氢酶活性均显著高于对照组(P0.05),而S20组肝脏谷胱甘肽过氧化物酶活性显著低于其他饥饿后补偿摄食组(P0.05),说明饥饿时间过长对机体抗氧化能力的恢复有所影响。由此可见,在本试验中,高温季节团头鲂幼鱼饥饿10 d后补偿摄食3周,补偿生长效果达到最佳,鱼体免疫与抗氧化能力也得到一定增强。  相似文献

丙酮酸脱氢酶激酶(PDK)在生物体的新陈代谢中具有重要的生理功能。为了探讨家蚕(Bombyx mori)PDK基因的表达特性,用蛾区半分法将二化性家蚕品种的活化越年卵(丙2期)分别以常温(25℃)光照和低温(15℃)黑暗催青,通过实时荧光定量PCR技术检测分析2种温度催青处理后家蚕不同发育阶段和不同组织的BmPDK表达水平。常温光照催青区BmP-DK的表达水平表现出发育时期和组织差异:胚胎发育阶段BmPDK在己4期的表达水平最高,其次是戊2、己3和己5期;幼虫发育阶段BmPDK的表达水平在蚁蚕期极显著高于其它龄期,5龄期在卵巢和精巢的表达水平高于其它组织;蛹期BmPDK的表达水平比其它发育时期低;成虫期的表达水平与胚胎发育末期相当,其中雌蛾的脂肪体和卵巢中BmPDK的表达水平较高。推测BmPDK在家蚕胚胎发育末期及产卵过程中有重要作用。催青温度影响BmPDK在家蚕各发育时期及卵巢组织中的表达水平:胚胎发育阶段戊2期BmPDK的表达水平为常温催青区显著高于低温催青区,己3~己5期常温催青区BmPDK的表达呈低-高-低的趋势,而低温催青区则呈上升趋势;蚁蚕期BmPDK表达水平为低温催青区极显著低于常温催青区;蛹期和成虫期卵巢中,常温催青区BmPDK的表达水平极显著高于低温催青区。催青温度对二化性家蚕BmPDK的表达的影响主要出现在胚胎戊2期、胚胎己3期~蚁蚕期、蛹期、成虫期。  相似文献
