以单猪屎豆碱(MO)、槲皮素(QU)和环磷酰胺(CY)为参照,观察了狗舌草600mL/L乙醇提取物(EX)对淋巴细胞性白血病L1210细胞体外试验的形态变化;利用流式细胞术,从DNA分子水平上检查了EX对L1210细胞各周期相的影响,探讨EX对L1210细胞的分化机理。结果发现,EX能够使L1210细胞向淋巴细胞方向发展;经EX作用24h后,L1210细胞G0+G1期的百分比较对照组明显升高。提示EX对L1210细胞增殖的抑制作用可能是由于G1期的阻滞所致。  相似文献

狗舌草抗淋巴性白血病有效部位的急性毒性   总被引:2,自引:1,他引:1  
利用体重 19~ 2 2 g的 BAL B/ c-C雌性小鼠 ,通过腹腔注射 ,从急性半数致死量 L D50 和供试鼠的临床表现等方面 ,检查了狗舌草 60 %乙醇提取物冻干粉的急性毒性。结果发现 ,狗舌草 60 %乙醇提取物对雌性 BAL B/ c-C小鼠腹腔注射的 LD50 为 ( 791.2 2± 170 .17) mg/ kg,95 %可信限为 63 9.15~ 979.49m g/ kg。急性死亡小鼠 (剂量达到 3 3 6.11mg/ kg以上 )在死前表现四肢抽搐 ,盲目运动 ,呼吸急促 ,翘尾 ;未死亡的小鼠表现食欲减退 ,反应迟钝 ,在随后的长期饲养中 ,临床表现逐步恢复正常  相似文献

将从早花期和盛花期狗舌草中提取得到的双稠比咯啶生物碱(PAs),按100mg/kg分别给两组SD大鼠腹腔注射。结果表明,两个时期狗舌草中PAs对SD大鼠均具有一定的肝毒性、肺毒性和中枢神经毒性,但盛花期狗舌草中PAs的毒性较弱;新发现狗舌草对心、肾也具有一定的毒性作用。  相似文献

将人参总皂甙经化学转化成转化型人参皂甙后分离得8个组分,在体外细胞上进行了转化型人参皂甙8个组分对CEF细胞毒性、生长活性、诱导肿瘤细胞凋亡、诱导单克隆抗体生成以及抗IBDV、AIV(H9)、NDV、MDV病毒的活性研究。结果发现,转化型人参皂甙各组分的细胞毒性作用在23.26~39.53mg/L之间;对鸡胚成纤维细胞(CEF)的生长和活性呈促进作用的工作浓度在10~20mg/L之间;转化型人参皂甙8号组分12.5mg/L诱导48h后,肝癌细胞7402株有60%的细胞凋亡,大鼠脑胶质瘤细胞C6株有35%的细胞凋亡;3、4、7号组分12.5mg/L和6.25mg/L对单克隆抗体6E6株杂交瘤细胞和单克隆抗体10B11株杂交瘤细胞的单抗生成具有明显促进作用,其HI效价为2^6~7与对照组的2^5相比有明显差异,IFA效价为10^4~5与对照组的10^2相比有极显著差异;4、7、8号组分15mg/L阻斯IBDV、AIV(H9)、NDV感染CEF细胞的作用都达到了51.1%以上,抑制IBDV、AIV(H9)、NDV在CEF上增殖的作用都在30%以上;6、7号组分12.5mg/L在阻断感染、抑制增殖、直接杀灭3种方式的抗MDV—RB1B强毒株在SPF鸡CEF细胞上的空斑减数率都达了到ACV抗MDV空斑减数率的80%,其中4、6号组分对MDV—RBIB株的直接杀灭作用与ACV的杀灭作用相等。表明人参总皂甙经化学处理转变成转化型人参皂甙,可明显改变其生物学活性。  相似文献

以体外培养的猪卵巢颗粒细胞为模板,研究了玉米赤霉烯酮(ZEA)对该细胞的毒性作用.结果表明,在所测浓度范围内,ZEA对卵巢颗粒细胞活性具有明显的抑制作用,且抑制作用随着浓度的增强而加大,并呈现剂量效应关系;60、90和120 μmol/L的ZEA使细胞中的MDA含量均升高,呈现剂量效应关系;ZEA作用后,细胞中GSH-Px酶活力下降;TUNE检测发现凋亡细胞呈棕褐色,凋亡细胞间间隙明显加大,随着ZEA剂量的加大,棕褐色细胞逐渐增多,尤其是高剂量ZEA组,细胞核大部分被染成棕褐色,细胞皱缩,胞间几乎无连接;当Annexin标记FIFC、流式细胞仪检测发现,高剂量组(120 μmol/L) ZEA处理后其凋亡率可达60.83%,坏死率很低,显示ZEA抑制猪卵巢颗粒细胞增殖主要是通过细胞凋亡,并非坏死途径.  相似文献

塞内卡病毒A(SVA)作为一种新发病病原,致病机制仍不清楚。通过显微镜观察发现,SVA感染PK-15细胞后能使细胞产生明显的细胞病变(CPE),同时伴随着严重的细胞凋亡。为了深入研究SVA诱导凋亡的情况,在验证SVA各蛋白真核质粒正常表达后,通过Annexin V-FITC/PI双染流式方法检测SVA各蛋白诱导凋亡的情况,Annexin V-FITC/PI和Hoechst染色后显微镜观察磷脂酰丝氨酸和核凝聚程度,通过Western blotting和RT-PCR技术分析SVA-VP1对凋亡通路中主要调控分子Caspase3、Caspase8和Bax蛋白质和mRNA水平的影响,发现SVA-VP1能够显著诱导细胞发生早期和晚期凋亡,并且能够促进凋亡蛋白Caspase3、Caspase8及Bax蛋白和mRNA水平的上调。本研究结果为深入研究SVA调控宿主细胞凋亡的分子机制和致病机制奠定了理论基础。  相似文献

利用细胞病变抑制法测定了蚯蚓寡肽的体外抗伪狂犬病毒(PRV)作用,分别用AO/EB染色法和DNA琼脂糖凝胶电泳法测定了蚯蚓寡肽对HeLa细胞凋亡的影响。结果显示,蚯蚓寡肽可以抑制PRV感染细胞,同时存在剂量效应和时间效应。蚯蚓寡肽浓度越大细胞病变程度越轻,各处理组在72h的细胞病变程度最轻。蚯蚓寡肽可以诱导HeLa细胞凋亡,且具有浓度效应,1.5mg/mL组、0.75mg/mL组的细胞凋亡率分别为60.20%和48.12%。  相似文献

在28℃、60%~80%RH、16L∶8D的实验条件下研究了葎草乙醇和水提取物对朱砂叶螨的生物活性。结果表明,葎草乙醇提取物对朱砂叶螨有较强的触杀作用,80g/L浓度的提取物平均死亡率高达91.11%,80g/L浓度的水提物下平均死亡率51.11%;乙醇提取物对朱砂叶螨有较强的驱避作用,在浓度为10g/L的醇提物下驱避率达60%,醇提效果优于水提;乙醇提取物对朱砂叶螨成螨产卵量有较强的抑制作用,在浓度为10g/L的醇提物和水提物下,减少率分别为62.39%与47.93%。研究结果有助于朱砂叶螨的综合治理。  相似文献

用60%乙醇作溶剂制备千里先抗菌有效部位冻干粉(即千里光60%乙醇提取物).分析其化学成分.并用改良寇氏法测定其对小鼠腹腔注射的急性半数致死量(LD50)。结果发现,千里光60%乙醇提取物合有生物碱、黄酮类化合物,但不含双稠吡咯啶生物碱;测得千里光60%乙醇提取物对小鼠腹腔注射的LD50为2206mg/kg,95%可信限为1867—2607mg/kg。表明千里光60%乙醇提取物属低毒性物质,并可保留千里光的有效成分生物碱和黄酮类化合物而排除其中的双稠吡略啶生物碱.进一步提高了千里光的安全性。  相似文献

以匍匐翦股颖克罗米和狗牙根新农1号为材料,通过愈伤诱导、继代和悬浮培养,进而细胞融合来探讨草坪草品种改良的新方法。结果表明:(1)匍匐翦股颖和狗牙根原生质体融合时酶解液的最佳组合为:1.5%纤维素酶、0.1%果胶酶、0.7M甘露醇,10h酶解时间,且酶解前应用高渗溶液预处理效果更佳;(2)细胞融合时,原生质体密度2×10^5个/mL,融合剂由30%的PEG4000、15mmol/LCaCl2·2H20、0.5mmol/L KH2PO4·H2O、0.5mol/L甘露醇组成,并调pH至7.5效果较好;(3)匍匐翦股颖和狗牙根细胞PEG融合率约为5%,以30min融合时间效果较好,产生的多核融合体少,杂种细胞多呈条形,具有较强的活力。  相似文献

1-(2-Chloroethyl)3-cyclohexyl-1-nitrosourea (CCNU) is an alkylating agent in the nitrosourea subclass. A prospective evaluation of CCNU was done to determine the maximally tolerated dosage of CCNU in tumor-bearing cats. Response data were obtained when available. Twenty-five cats were treated with CCNU at a dosage of 50-60 mg/m3 body surface area. Complete hematologic data were available for 13 cats. Neutropenia was the acute dose-limiting toxicity. The median neutrophil count at the nadir was 1,000 cells/microL (mean, 2,433 cells/microL; range, 0-9,694 cells/microL). The time of neutrophil nadir was variable, occurring 7-28 days after treatment, and counts sometimes did not return to normal for up to 14 days after the nadir. Based on these findings, a 6-week dosing interval and weekly hematologic monitoring after the 1st treatment with CCNU are recommended. The nadir of the platelet count may occur 14-21 days after treatment. The median platelet count at the nadir was 43,500 cells/microL. No gastrointestinal, renal, or hepatic toxicities were observed after a single CCNU treatment, and additional studies to evaluate the potential for cumulative toxicity should be performed. Five cats with lymphoma and 1 cat with mast cell tumor had measurable responses to CCNU. Phase II studies to evaluate antitumor activity should be completed with a dosing regimen of 50-60 mg/m3 every 6 weeks.  相似文献

Human and equine athletes are reported to have a high prevalence of gastric disease, and anecdotal evidence suggests a similar phenomenon applies to racing sled dogs. To investigate the prevalence of gastric disease in racing sled dogs, we conducted 2 gastroscopy studies on dogs competing in the annual Iditarod Sled Dog Race. A pilot study of dogs that were either dropped from the 2000 Iditarod Sled Dog Race because of illness or that finished the race indicated that, approximately 5 days after competing, 10 of 28 dogs (35%) had endoscopic evidence of gastric ulceration, erosion, or hemorrhage. The next year, an endoscopic study of 73 dogs participating in the 2001 Iditarod race was performed in order to evaluate a larger population of dogs. Data from 70 of these dogs could be used; 34 (48.5%) had ulceration, erosion, gastric hemorrhage, or some combination of these findings. When this group of 70 dogs was compared retrospectively to a control group of 87 dogs presented to the Texas A&M University (TAMU) Veterinary Medical Teaching Hospital, the Iditarod sled dogs had a significantly higher prevalence (P = .049) of gastric lesions. These findings suggest that, similar to athletes of other species, elite canine athletes have an increased prevalence of gastric disease compared to the canine population at large.  相似文献

Continuous rate infusion (CRI) of furosemide in humans is considered superior to intermittent administration (IA). This study examined whether furosemide CRI, compared with IA, would increase diuretic efficacy with decreased fluid and electrolyte fluctuations and activation of the renin-angiotensin-aldosterone system (RAAS) in the horse. Five mares were used in a crossover-design study. During a 24-hour period, each horse received a total of 3 mg/kg furosemide by either CRI (0.12 mg/kg/h preceded by a loading dose of 0.12 mg/kg IV) or IA (1 mg/kg IV q8h). There was not a statistically significant difference in urine volume over 24 hours between methods; however, urine volume was significantly greater after CRI compared with IA during the first 8 hours ([median 25th percentile, 75th percentile]: 9.6 L [8.9, 14.4] for CRI versus 5.9 L [5.3, 6.0] for IA). CRI produced a more uniform urine flow, decreased fluctuations in plasma volume, and suppressed renal concentrating ability throughout the infusion period. Potassium, Ca, and Cl excretion was greater during CRI than IA (1,133 mmol [1.110, 1,229] versus 764 mmol [709, 904], 102.7 mmol [96.0, 117.2] versus 73.3 mmol [65.0, 73.5], and 1,776 mmol [1,657, 2.378] versus 1,596 mmol [1,457, 1,767], respectively). Elimination half-lives of furosemide were 1.35 and 0.47 hours for CRI and IA, respectively. The area under the excretion rate curve was 1,285.7 and 184.2 mL x mg/mL for CRI and IA, respectively. Furosemide CRI (0.12 mg/kg/h) for 8 hours, preceded by a loading dose (0.12 mg/kg), is recommended when profound diuresis is needed acutely in horses.  相似文献

The epidemiology of H. placei and of other gastrointestinal nematodes in yearling dairy cattle was examined on two farms in Kiambu District, central Kenya during each of 13 one-month periods from April 1993 to April 1994. On each farm, 32 newly weaned dairy calves were given a single dose of albendazole and then placed on experimental pastures. Twelve of the animals were designated for bi-monthly slaughter (n = 2) and analysis of worm population characteristics and 20 were designated for blood and faecal collection and for weighing. Two parasite-free tracer calves were grazed alongside the weaner calves each month throughout the study period and were also slaughtered for analysis of worm populations. Faecal egg counts, haematological and serum pepsinogen determinations, herbage larval counts, and animal live weight changes were recorded monthly. The study revealed that Haemonchus placei, Trichostrongylus axei, Cooperia spp. and Oesophagostomum radiatum were responsible for parasitic gastroenteritis and that H. placei was the predominant nematode present in the young cattle on both farms. Faecal egg counts from resident cattle and necropsy worm counts revealed that pasture larval levels were directly related to the amount of rainfall. The total worm burdens in the animals were highest during the rainy season (March–June and October–December) and lowest during the dry seasons (July–September and January–February). The very low recovery of immature larvae of H. placei from the tracer calves indicated that arrested development is not a feature of the life cycle of this parasite in central Kenya. The maintenance of the parasite population depended on continuous cycling of infection between the host and the pasture. The agroclimatic conditions of the study area were such that, in general, favourable weather conditions for the development and survival of the free-living stages of gastrointestinal nematodes existed all year round.  相似文献

The occurrence of ectoparasites in sheep flocks is frequently reported but seldom quantified. Sheep production used to be a predominantly family activity in the state of Sa~o Paulo (Brazil), but it began to become a commercial activity in the past decade. Thus, information about the ectoparasites existing in sheep flocks has become necessary. The present data were obtained by means of questionnaires sent to all sheep breeders belonging to the `Associaça~o Paulista de Criadores de Ovinos' (ASPACO; Sa~o Paulo State Association of Sheep Breeders). Response reliability was tested by means of random visits paid to 10.6% of the respondents. Most of the properties (89.5%) reported the presence of one or more ectoparasites. Screw-worm (Cochliomyia hominivorax) was the most frequent ectoparasite (72.5%), followed by bot fly larvae (Dermatobia hominis, 45.0%), ticks (Amblyomma cajennense) and Boophilus microplus, 31.3%) and finally lice (Damalinia ovis, 13.8%). Combined infestations also occurred, the most common one being screw-worm with bot fly larvae (36.0%) followed by bot fly larvae with ticks (13.9%), screw-worm with ticks (9.3%), bot fly larvae with lice (6.9%), and ticks with lice (5.0%). The most common triple combination was screw-worm, bot fly larvae and ticks (12.8%). Breeds raised for meat or wool were attacked by bot fly larvae and ticks more often than other breeds. Lice were only absent from animals of indigenous breeds. The relationships among these ectoparasites are discussed in terms of sheep breeds, flock size, seasonality and the ectoparasitic combinations on the host.  相似文献

An experiment was conducted to study disappearance of C14 to C18 fatty acids, lag times and biohydrogenation (BH) rates of C18 fatty acids of ground soya bean and canola seeds in situ. Three ruminally fistulated Dallagh sheep were used to determine ruminal BH of unsaturated fatty acids (UFAs). Differences in the disappearance of fatty acids through the bags and lag times were observed between the oilseeds. We saw that the longer the incubation time of the oilseeds in the rumen, the lower the content of C18:2 and C18:3. Significantly higher lag times for both C18:2 and C18:3 were observed in ground canola compared to ground soya bean. BH rates of C18:2 and C18:3 fatty acids in soya bean were three times higher than those of canola. These results suggest that the fatty acid profile of fat source can affect the BH of UFAs by rumen micro‐organisms. So that UFAs of canola had higher ability to escape from ruminal BH. It seems that fatty acid profile of ruminant products is more affected by canola seed compared to soya bean seed.  相似文献

Serum biochemical values and the wet weight to dry weight ratios of tissues were determined in yaks in Shandan county of Gansu province. The liver, kidney, heart and muscle contents of seven elements in yaks were also analysed. Most serum biochemical values were similar to those of cattle, camels and sheep, but the calcium concentration was considerably above the normal range for other ruminants.
