口蹄疫(foot-and-mouth disease,FMD)是由口蹄疫病毒(foot-and-mouth disease virus,FMDV)感染偶蹄兽所引起的一种急性、热性、高度接触性传染病,FMDV有7个血清型,加之病毒传播迅速,严重影响畜牧业的发展。FMDV为小RNA病毒科、口蹄疫病毒属的唯一成员,其基因组编码4种结构蛋白和10种非结构蛋白。FMDV感染宿主后利用自身蛋白通过多种途径和方式来影响宿主天然免疫应答,从而有利于FMDV复制的微环境。这些策略包括FMDV参与细胞自噬、内质网应激和应激颗粒形成的细胞过程,破坏多种宿主蛋白的功能,如劫持、裂解宿主蛋白或干扰宿主蛋白的表达、去除宿主蛋白的泛素化以及抑制宿主蛋白的磷酸化。这些逃避天然免疫的策略也是目前研究的热点。基于现有的研究结果,作者总结了近几年FMDV蛋白在抑制宿主天然免疫方面的研究进展,以期为FMDV的研究与防控提供参考。  相似文献

口蹄疫(foot-and-mouth disease, FMD)是由口蹄疫病毒(Foot-and-mouth disease virus, FMDV)感染引起的急性、高传染性的牲畜疾病,爆发期间给养殖业造成重大经济损失。当FMDV进入宿主体内时会被免疫系统识别,Ⅰ型和Ⅲ型干扰素会介导增强免疫反应。FMDV通过多种途径抑制抗病毒应答反应,其中包括在转录和翻译水平上抑制干扰素的产生;劫持、裂解宿主蛋白或干扰宿主蛋白的表达;阻断天然免疫过程中蛋白质的翻译后修饰;调节自噬;抑制应激颗粒(stress granules, SGs)的形成。本文综述了FMDV蛋白颉颃抗宿主免疫的相关机制,以期对FMDV致病机理有一个全面的认识,为FMDV的防治提供理论基础。  相似文献

Since then, FMDV has become a model system in animal virology and a considerable amount of information on its structure, biology and vaccinology has been obtained. However, the disease that this virus produces (FMD) still constitutes one of the main animal health concerns. 口蹄疫病毒能引起牛、羊等偶蹄动物发生高度接触性的传染病口蹄疲,该病常常影响着全球畜牧业的发展.FMDV是小RNA病毒科口蹄疫病毒属的成员,口蹄疫病毒为单股正链RNA病毒,病毒基因组全长约8.5 kb,基因组分为5'非编码区、3'非编码区和一个开放阅读框(ORF).基因组的中部是一大的开放阅读框,编码一多聚蛋白,多聚蛋白在翻译的同时,经二级裂解后,形成3种病毒结构蛋白(VP0,VP3和VP1)和8种非结构蛋白(L,2A,2B,2C,3A,3B,3C和3D).其中3C全长639 bp,编码213个氨基酸.3C蛋白酶是小RNA病毒的共同裂解酶,在多聚蛋白成熟过程中起着极为重要的作用,且在抗病毒药物打靶方面具有一定研究价值.因此,对3C蛋白酶的结构及功能研究进展进行综述很有必要.  相似文献

Foot-and-mouth disease (FMD) is an acute, hot, and highly contagious infectious disease caused by foot-and-mouth disease virus (FMDV) infection of cloven-hoofed animals. FMDV has seven serotypes and it spreads rapidly, which seriously affects the development of animal husbandry. FMDV is the prototype member of the Aphthovirus genus within the Picornaviridae family, and its genome encodes 4 structural proteins and 10 non-structural proteins. After infecting the host, FMDV uses its proteins to affect the host innate immune response through a variety of pathways and methods, which is beneficial to the microenvironment of FMDV replication. These strategies include FMDV involvement in autophagy, endoplasmic reticulum stress and stress granule formation of cellular processes, subverting the functions of various host proteins, such as hijacking, cleaving host proteins or interfering with the expression of host proteins, removing ubiquitin from host proteins, and inhibiting the phosphorylation of host proteins, which are also the focus of current research. Based on the existing research results, we summarized the research progress of FMDV protein in suppressing the innate immunity of host in recent years, intending to provide a reference for the research and prevention of FMDV.  相似文献

2014~2015年,美国、巴西等国爆发了一种症状类似于口蹄疫的水泡性疾病,病原被确定为塞尼卡病毒A(Senecavirus A,SVA)。为了鉴别和诊断塞尼卡病毒A和口蹄疫病毒(Foot-and-mouth disease virus,FMDV),本研究针对塞尼卡病毒的VP1基因和口蹄疫的5’UTR基因,合成特异性引物,优化了反应体系和扩增条件,建立了一种可同时检测塞尼卡病毒A和口蹄疫病毒的双重RT-PCR方法。特异性和敏感性试验结果表明,建立的双重RT-PCR检测方法特异性好,敏感性强,对塞尼卡病毒A和口蹄疫病毒最低检出量分别为104 copies/μL、103 copies/μL。本研究建立的方法对临床快速鉴别塞尼卡病毒A和口蹄疫病毒感染具有重要意义。  相似文献

Foot-and-mouth disease (FMD) is a highly contagious disease of cloven-hoofed animals. In Uganda, FMD outbreaks are mainly controlled by ring vaccination and restriction of animal movements. Vaccination stimulates immunity and prevents animals from developing clinical signs which include lameness, inappetence, and decreased production. Ring vaccination and restriction of animal movements have, however, not successfully controlled FMD in Uganda and outbreaks reoccur annually. The objective of this study was to review the use of FMD virus (FMDV) vaccines and assess the effectiveness of vaccination programs for controlling FMD in Uganda (2001–2010), using retrospective data. FMD vaccine distribution patterns in Uganda (2001–2010) matched occurrence of outbreaks with districts reporting the highest number of outbreaks also receiving the largest quantity of vaccines. This was possibly due to “fire brigade” response of vaccinating animals after outbreaks have been reported. On average, only 10.3 % of cattle within districts that reported outbreaks during the study period were vaccinated. The average minimum time between onset of outbreaks and vaccination was 7.5 weeks, while the annual cost of FMDV vaccines used ranged from US $58,000 to 1,088,820. Between 2001 and 2010, serotyping of FMD virus was done in only 9/121 FMD outbreaks, and there is no evidence that vaccine matching or vaccine potency tests have been done in Uganda. The probability of FMDV vaccine and outbreak mismatch, the delayed response to outbreaks through vaccination, and the high costs associated with importation of FMDV vaccines could be reduced if virus serotyping and subtyping as well as vaccine matching were regularly done, and the results were considered for vaccine manufacture.  相似文献
