为延长生物能源作物柳枝稷(Panicum virgatum)的叶片功能期,以柳枝稷‘西稷1号’品系的成熟胚诱导的愈伤组织为受体,采用农杆菌介导法转化叶片衰老延缓基因PSAG12-IPT.结果表明:经过农杆菌转化,共培养,抗性愈伤组织筛选、再生和生根培养,在农杆菌介导处理的8500块愈伤组织中共获得了320株转化植株;经PCR检测分析,转化植株中共有19株植株扩增出标记基因潮霉素B磷酸转移酶基因的目标片段,初步表明PSAG12-IPT基因已整合到柳枝稷基因组中,转化率为0.223%.对柳枝稷转基因植株的叶片叶绿素含量、净光合速率、胞间CO2浓度和气孔导度进行测定,表明PSAG12-IPT基因在部分转基因植株中已经表达.  相似文献

以美国从南到北11个不同生态型柳枝稷(Panicum virgatum L.)品种的成熟种子为外植体,进行愈伤组织诱导和分化,对其愈伤组织各种形态及比例进行统计分析,以评价不同生态型柳枝稷的组织培养反应。结果表明:低地型品种Alamo和Performer的种子发芽率和愈伤诱导率均较低,但组织培养中结构松脆、生长旺盛、色泽鲜亮、增殖较快、容易分化成苗的Ⅱ型愈伤组织比例高,容易再生成苗;虽然高地型品种Dacotach,Blackwell和Nebraska28等的愈伤诱导率和种子发芽率较高,但较难挑选出Ⅱ型愈伤组织,不易获得组培再生苗。本研究结果为柳枝稷的遗传改良和生物技术育种奠定基础。  相似文献

高效再生体系是建立遗传转化体系的基础和前提,本研究以柳枝稷(Panicum virgatum)成熟种子为外植体,对柳枝稷再生体系的建立进行探索。使用70%乙醇及0.1% HgCl2采用两步法灭菌,以MS培养基为基本培养基,研究不同质量浓度2,4 D与6 BA组合对愈伤组织诱导的影响,不同质量浓度GA3对愈伤组织分化的影响。结果表明,70%乙醇处理20 s,然后用0.1% HgCl2处理5 min,种子活力保存最好,愈伤组织诱导率最高;愈伤组织诱导培养基添加5 mg·L-1 2,4 D与1.2 mg·L-1 6 BA为最佳组合,其诱导率最高,质量最好;添加0.5 mg·L-1GA3分化成苗的效果最好。最终得到柳枝稷最优的再生体系:MS培养基为基本培养基,添加5 mg·L-1 2,4 D及1.2 mg·L-1 6 BA进行愈伤组织诱导;添加0.5 mg·L-1 GA3分化、再生。  相似文献

试验旨在研究日粮中添加肠膜蛋白粉和发酵豆粕替代鱼粉对断奶仔猪生长性能和养分消化率的影响。试验选用180头28日龄(杜×长×大)断奶仔猪,公母各半,体质量(8.0±0.5)kg,随机分为3个处理,每处理6个重复,每重复10头猪。试验期14 d,试验期间常规饲养管理。对照组日粮为17.0%豆粕+6.0%鱼粉,进口肠膜组日粮为16.6%豆粕+4.0%发酵豆粕+4.0%进口肠膜蛋白粉,国产肠膜组日粮为16.6%豆粕+4.0%发酵豆粕+4.0%国产肠膜蛋白粉。结果表明:在采食量和增质量方面,进口肠膜组高于对照组,国产肠膜组低于对照组,差异均不显著(P≥0.05);在料重比方面,进口肠膜组最低,国产肠膜组高于对照组,差异均不显著(P≥0.05);进口肠膜组和国产肠膜组腹泻率、腹泻指数和死淘率均高于对照组,但差异均不显著(P≥0.05);进口肠膜组钙的消化率比对照组下降13.21%(P0.05);干物质、能量、粗蛋白质、粗脂肪、磷和必需氨基酸的消化率处理间无显著差异(P≥0.05)。因此,断奶仔猪日粮中使用进口或国产肠膜蛋白粉和发酵豆粕等替代鱼粉,不影响生长性能、腹泻和养分消化率。  相似文献

柳枝稷(Panicum virgatum)可广泛应用于盐渍化地区的土壤改良。本试验以柳枝稷品种Alamo(低地型)和Blackwell-1(高地型)为材料,采用浓度为60 mmol/L的Na2CO3溶液(盐胁迫)和3种不同碱度NaOH溶液即pH值分别为7.0(CK,无胁迫)、8.0、9.0组合处理,探究盐碱不同胁迫时间对柳枝稷抗氧化及渗透调节的影响。结果表明:(1)盐碱胁迫下柳枝稷的光合色素含量均随胁迫时间的增加而下降,并在最后一次取样时显著下降(P<0.05),柳枝稷光合色素的合成受到了影响。(2)盐碱胁迫下柳枝稷的相对水分含量、电导率和丙二醛(MDA)含量随胁迫时间的增加呈先上升后下降的趋势。(3)盐碱胁迫下柳枝稷的抗坏血酸过氧化物酶(APX)、过氧化氢酶(CAT)和过氧化物酶(POD)含量均有显著上升(P<0.05),对柳枝稷维持正常生长起重要作用。(4)盐碱胁迫下柳枝稷可溶性蛋白含量上升但未见明显差异(P>0.05),可溶性糖含量出现显著差异(P<0.05)。Alamo的可溶性蛋白含量低于Blackwell-1...  相似文献

试验用黄孢原毛平革菌(Phanerochaete chrysosporium)、乳白耙菌(Irpex lacteus)、虫拟蜡菌(Ceriporiopsis subvermispora)3种白腐真菌及其混合菌对柳枝稷进行预处理,研究白腐真菌种类、时间、温度、水分、菌液量对柳枝稷降解的影响,为柳枝稷的生物预处理及利用提供理论依据。结果表明:黄孢原毛平革菌、乳白耙菌和虫拟蜡菌两两之间均无拮抗反应,可以相互混合生长;当温度为28°C,加水5mL,菌液量为1mL时,混合真菌对木质素的降解与单一白腐真菌相比并没有优势,在3种白腐真菌中,乳白耙菌效果最好,几乎不降解纤维素,并且在第20天时木质素降解率达到最高为36.63%;当改变条件时,发现温度为28°C,水分为5mL,菌液量为3mL是乳白耙菌对柳枝稷进行预处理20d效果最好的条件,此时木质素降解率最高为63.45%,半纤维素降解率为24.82%,而纤维素和空白组差异不显著。因此,在三种白腐真菌中,乳白耙菌对柳枝稷的预处理效果最好,木质素降解率最高,半纤维素略有降解,而且几乎不降解纤维素。  相似文献

以柳枝稷品系‘西稷1号’、‘西稷2号’和‘西稷3号’为主区,以农杆菌菌液浓度为副区,对农杆菌浸种处理后柳枝稷种子萌发及幼苗生长发育相关性状进行了研究。结果表明,品系与农杆菌菌液浓度的互作效应不显著;不同基因型品系对农杆菌的敏感程度不同,其中西稷3号最不敏感;随着农杆菌菌液浓度的升高,柳枝稷种子发芽率、幼苗成苗率及叶绿素含量呈下降趋势,幼苗POD活性、CAT活性、MDA含量呈上升趋势,SOD活性呈先升后降的趋势。综合农杆菌对柳枝稷种子萌发及幼苗生理生化特性影响,在柳枝稷农杆菌浸种转化法中菌液浓度控制在1.2 OD600较为适宜。  相似文献

植物修复技术是一种清除土壤中重金属有效、经济的生态技术。为在重金属污染的土壤环境下建植柳枝稷进行植物修复提供基础理论依据,本试验采用三因素五水平正交旋转试验设计,研究镉、pH和浸种及其耦合作用分别对高地型Blackwell(B品种)和低地型Kanlow(K品种)柳枝稷种子萌发及幼苗发育的影响。结果表明,不同镉处理下两个品种柳枝稷种子发芽率与对照差异均不显著,但是浸种可显著提高柳枝稷种子发芽指数和种子活力指数。随着镉浓度的增加,其幼苗根长、苗长、及种子活力均呈缓慢下降趋势,且镉对幼根生长的影响大于其对幼苗生长的影响。B品种比K品种更加耐受镉胁迫;播种前浸种可以缓解镉胁迫,促进柳枝稷幼苗和根的发育。镉和pH对柳枝稷种子萌发和幼苗发育存在耦合作用,同时pH为4.8时柳枝稷幼苗根长苗长及种子活力有最大值。在较低浓度的镉胁迫环境下(150μmol/L),高地型B品种比低地型K品种种子萌发和幼苗生长综合表现更好,同时适当浸种和偏酸性条件均有利于柳枝稷幼苗在镉污染土壤环境中的生长。本试验表明柳枝稷对镉胁迫有较强的耐受性,具有一定修复镉污染土壤的潜能,在实际应用中可通过推广种植柳枝稷来改善镉污染的土壤环境。  相似文献

为明确柳枝稷对水胁迫和酸碱胁迫的耐受力,论文以盆栽的柳枝稷(Panicum virgatum)Alamo为试验材料,测定在5个土壤含水量水平(8%,16%,20%,24%和32%)和5个pH水平(4.9,6.3,7.0,7.7和9.1)柳枝稷的产量(干鲜重、分蘖和株高)、生理反应(可溶性糖、脯氨酸、丙二醛、叶绿素含量和相对电导率)和品质(纤维素、半纤维素、酸性洗涤纤维、中性洗涤纤维和酸性洗涤木质素)。结果表明:干旱胁迫和酸碱胁迫对柳枝稷的多项指标影响不显著,柳枝稷对干旱和pH胁迫有一定的耐受力;土壤含水量和pH对柳枝稷的鲜重和脯氨酸含量产生了显著的耦合作用,为柳枝稷在边际土地上的引种种植提供了理论依据;纤维素与干鲜重、酸性洗涤纤维、酸性洗涤木质素、中性洗涤纤维含量、株高显著正相关,与可溶性糖、脯氨酸、丙二醛含量和相对电导率显著负相关。  相似文献

为了提高柳枝稷(Panicum virgatum L.)种子萌发率,探究不同植物生长物质处理对柳枝稷种子萌发特性的影响,本研究以柳枝稷(Panicum virgatum‘Forestburg’)种子为材料,采用不同浓度(0,10,50,200,400,1 000μmol·L~(-1))的赤霉素(GA3,GA4+7)、脱落酸(ABA)、6-苄氨基腺嘌呤(6-BA)、吲哚乙酸(IAA)、氟啶酮(FL)和多效唑(PA)等7种外源植物生长物质处理种子,分析柳枝稷种子萌发特性对不同浓度外源植物生长调节剂的响应。结果表明:10~200μmol·L~(-1) IAA和GA4+7,10~50μmol·L~(-1) GA3,50μmol·L~(-1) 6-BA及10μmol·L~(-1) FL均可对柳枝稷种子萌发产生显著促进作用(P0.05),但是当其浓度过高时则会抑制柳枝稷种子萌发;ABA会抑制柳枝稷种子的萌发,PA对柳枝稷的萌发无显著作用;GA4+7对柳枝稷种子萌发影响效果最好,其中经200μmol·L~(-1) GA4+7处理后种子萌发率最高,可达71%,较对照提高30%。  相似文献

以紫羊茅的成熟种子为外植体材料,研究了2,4-D、有机物及光照对愈伤组织诱导发生、生长状态及绿苗分化能力的影响.结果表明,3~5 mg/L 2,4-D为愈伤组织诱导和继代培养基的适宜浓度,诱导率为46.6%~57.9%,并使愈伤组织保持胚性状态.在继代培养基中添加脯氨酸和天门冬酰氨,并且每培养1~2个月给予15 d左右的散射光培养,使培养了2年多的愈伤组织仍保持99%以上的绿苗分化率.利用基因枪将GUS基因转入继代2年多的愈伤组织,通过组织染色法检测到了高频率的GUS基因瞬时表达.  相似文献

对双纸片协同试验不同指示底物及纸片间放置距离进行探讨。选择临床分离菌10株、实验室诱导菌5株及肺炎克雷伯菌700603(SHV-18)共16株细菌,先做双纸片增效实验,然后每株细菌对CTX、CAZ、AZT、FEP、CRO及CF六种药敏纸片逐个分别在15 mm、20 mm、25 mm、和30 mm处做双纸片协同实验。试验结果表明:FEP、CTX、AZT、CRO在15 mm处检出率均为100%;在20 mm处FEP、CTX、AZT检出率为88%,CRO检出率为75%。双纸片协同实验中检出率高的底物为FEP、CTX、AZT、CRO,其最佳协同距离为15 mm。  相似文献

OBJECTIVE: To quantify the association between intervertebral disk calcification and disk herniation in Dachshunds. DESIGN: Longitudinal study. ANIMALS: 61 Dachshunds that had been radiographically screened for calcification of intervertebral disks at 2 years of age in other studies. Thirty-seven of the dogs had survived to the time of the present study and were > or = 8 years of age; 24 others had not survived. PROCEDURES: Radiographic examination of 36 surviving dogs was performed, and information on occurrence of disk calcification at 2 years of age were obtained from records of all 61 Dachshunds. Information on occurrence of disk herniation between 2 and 8 years of age was obtained from owners via questionnaire. Associations between numbers of calcified disks and disk herniation were analyzed via maximum likelihood logistic regression. RESULTS: Disk calcification at 2 years of age was a significant predictor of clinical disk herniation (odds ratio per calcified disk, 1.42; 95% confidence interval, 1.19 to 1.81). Number of calcified disks in the full vertebral column was a better predictor than number of calcified disks between vertebrae T10 and L3. Numbers of calcified disks at > or = 8 years of age and at 2 years of age were significantly correlated. CONCLUSIONS AND CLINICAL RELEVANCE: Number of calcified disks at 2 years of age was a good predictor of clinical disk herniation in Dachshunds. Because of the high heritability of disk calcification, it is possible that an effective reduction in occurrence of severe disk herniation in Dachshunds could be obtained by selective breeding against high numbers of calcified disks at 2 years of age.  相似文献

On 80 bovine hindclaws from slaughtered cows a correct functional claw trimming (sole horn thickness 5 mm; group 1) and on further 80 hindclaws a standardised, incorrect claw trimming (sole horn thickness 2 mm; group 2) was performed using 4 different disks (one cutting, three grinding disks). Two thermocouples were inserted into the solar corium and the heat production on the corium was measured during claw trimming. During correct functional claw trimming with each of the 4 disks a temperature increase in the corium with a median of < or = 0.3 degrees C was calculated. During incorrect claw trimming (2 mm sole horn thickness) a temperature increase with a median of < or = 0.3 degrees C for the first three disks was determined. During claw trimming using the abrasive semiflexible plastic disk a median of 1.3 degrees C temperature increase was calculated. Comparison of the temperatures measured in the claws from group 1 and 2 showed a statistically significant difference between all four disks. Comparison of the temperature increase within each group (group 1 and 2) revealed a statistically significant difference between the cutting disk with steel blades and the abrasive, semiflexible plastic disk in both groups. The highest temperatures measured were 41.5 degrees C in group 1 using the disk with 70% amount of fitted hard metal granulate, and 42.2 degrees C in group 2 (temperature increase of 10.6 degrees C) using the abrasive semiflexible plastic disk. Under the experimental conditions of this study, only a very slight thermogenesis was found when a functional and correct claw trimming using the 4 tested disks was performed. This slight thermogenesis should not be able to cause thermic injury of the corium. By trimming the sole horn incorrectly (2 mm) the measured heat generation in the corium was minor, without danger of thermic insult, excepted for using the disk with a 70% amount of fitted hard metal granulate and the abrasive semiflexible plastic disk. Even, it is unlikely that a short term temperature increase of 10.6 degrees C can cause thermic insults of the solar corium.  相似文献

紫花苜蓿(Medicago sativa L.)愈伤组织再生率受基因型限制,是影响苜蓿遗传转化效率的主要因素之一。研究发现,GRF转录因子家族基因在提高多种植物愈伤再生中起着重要促进作用。本试验以拟南芥(Arabidopsis thaliana)AtGRF序列为模板,在紫花苜蓿基因组序列中查找,获得了含有miR396靶点的9个MsGRFs基因。除MsGRF3aMsGRF4a外,其余7个MsGRFs在紫花苜蓿胚性愈伤组织中的表达量均显著高于非胚性愈伤。为了进一步研究MsGRFs对紫花苜蓿愈伤再生效率的影响,我们在紫花苜蓿中转入MIM396基因,降低miR396的表达。结果显示:MIM396植株叶片愈伤组织再生时间显著缩短,再生率显著提高;与野生型愈伤组织相比,在MIM396植株叶片诱导的愈伤组织中,除MsGRF3a外,其余MsGRFs显著高表达。以上研究表明,miR396-MsGRF通路对苜蓿愈伤组织再生起着重要调控作用,这为提高苜蓿再生效率,并打破基因型对不同苜蓿品种再生的限制提供候选基因。  相似文献

收集国内外主要特殊医学用途婴儿配方食品营养数据,分别对国内外4 类特殊医学用途婴儿配方食品中三大产能营养素、维生素、矿物质以及在标准中要求强制添加或选择性添加的其他营养元素含量进行比较分析,并对国内外4 类特殊医学用途婴儿配方食品进行主成分分析。结果表明:氨基酸配方或乳蛋白深度水解配方中,国内品牌脂肪、VK、泛酸、牛磺酸含量显著大于国外品牌(P<0.05),但国内品牌叶酸含量显著低于国外品牌(P<0.05);乳蛋白部分水解配方中,国内品牌脂肪、锰、牛磺酸、肉碱含量显著高于国外品牌(P<0.05);  相似文献

假俭草抗寒性差是限制其广泛应用的主要因素之一。本研究目标是以优良假俭草选系E-126为材料,经低温诱导和筛选获得体细胞抗寒突变体。结果表明,假俭草种子诱导的愈伤组织在继代培养的过程中,经0℃的低温条件下培养26 d,获得了2块存活的愈伤组织,该愈伤组织经过继代增殖后,进行分化、生根和移栽,获得株系1和株系2。苗期外部形态观察结果表明,假俭草低温诱导的株系1、株系2和对照在叶色、叶毛、叶长和叶宽上均没有显著性差异。半致死温度分析结果表明,株系1、株系2以及对照叶片半致死温度(LT50)分别为-6.646,-6.546和-5.351℃,处理与对照之间差异显著,且都低于对照,但株系1和株系2之间无显著性差异。SRAP的结果表明,假俭草低温诱导的株系1和株系2在110,230以及240 bp处均存在相同特征带,表明假俭草体细胞突变体植株的变异稳定且在分子水平与对照存在差异,但株系1和株系2无差异。因此,株系1和株系2可作为同一体细胞抗寒突变体株系加以利用。  相似文献

 为探索适合草原放牧条件下西门塔尔牛的超数排卵技术,分别采用国产及进口促卵泡素(FSH)和三种方案(A:FSH+PG;B:FSH+PG+GnRH;C:FSH+PG+CIDR +GnRH)对放牧条件下西门塔尔牛进行超数排卵试验。结果表明,采用国产FSH超排时,方案A、方案B和方案C所获头均可用胚数分别为(3.43±2.41)枚、(5.93±3.54)枚和(7.21±3.19)枚,采用进口FSH超排时,三种超排方案所获头均可用胚数分别为(4.34±2.71)枚、(6.76±3.79)枚和(7.79±4.23)枚,采用两种FSH超排,方案B、方案C所获头均可用胚数明显高于方案A(P<0.05);3种方案中,国产与进口FSH所获头均可用胚数差异均不显著(P>0.05)。试验表明,在放牧条件下,采用三种超排处理方案,以FSH+PG+CIDR+GnRH法最好,FSH+PG+GnRH法次之,FSH+PG法效果最差;国产FSH与进口FSH超排效果差异不显著。  相似文献

OBJECTIVE: To assess risk factors for recurrence of clinical signs associated with thoracolumbar intervertebral disk disease (IVDD) in dogs that had decompressive laminectomy without attempted prophylactic treatment of other disk spaces. DESIGN: Retrospective study. ANIMALS: 229 dogs. PROCEDURE: Medical records of dogs that had decompressive laminectomy without prophylactic fenestration for a first episode of IVDD and were available for follow-up were reviewed. Information on 7 clinical and 8 radiographic potential risk factors were recorded. RESULTS: Clinical signs associated with recurrence of IVDD developed in 44 (19.2%) dogs. Ninety-six percent of recurrences developed within 3 years after surgery. Recurrence developed in 25% of Dachshunds and 15% of dogs of other breeds combined. Number of opacified disks was a significant risk factor for recurrence. Risk increased with number of opacified disks in an almost linear manner; each opacified disk increased risk by 1.4 times. Dogs with 5 or 6 opacified disks at the time of first surgery had a recurrence rate of 50%. CONCLUSIONS AND CLINICAL RELEVANCE: When all likely episodes of recurrence are considered and a long follow-up period is achieved, true rate of recurrence of IVDD appears to be higher than in many previous reports. Dogs with multiple opacified disks at the time of first surgery should be considered a high-risk subpopulation.  相似文献

本试验以3个结缕草(Zoysia spp.)(品)种(兰引3号,马尼拉,青岛)的茎基作为外植体材料,诱导愈伤组织,观察出愈率,然后用不同剂量(5~40 Gy)的60Co γ射线辐照处理愈伤组织,观察其胚性愈伤组织诱导情况。结果表明,较低剂量(15~20 Gy)的辐照处理可以促进结缕草愈伤组织向胚性愈伤组织转化,在这一剂量范围内,3个结缕草(品)种不但可以获得很高的出愈率,同时也可获得较高的胚性愈伤组织形成率。胚性愈伤组织诱导的临界剂量是20 Gy, 此时,兰引3号、马尼拉和青岛愈伤组织的胚性转化率均达到了峰值,分别为76.47%、63.64%和37.5%。3个结缕草品种愈伤组织辐照的半致死剂量为30 Gy。  相似文献
