Analysis of the partial bovine leukaemia virus (BLV) gp51 gene sequences obtained from five BLV strains isolated in different regions of Iran and BLV-FLK strain was carried out. The Iranian BLV gp51 sequences were compared with seven other corresponding sequences of BLV strains isolated in different countries. Nucleotide sequence analysis showed a variability of 0.003–5.1% and the phylogenetic tree constructed revealed three clusters. The first cluster included French, German and FLK-BLV samples; the second cluster included four Iranian samples; and the third cluster included Australian, Korean, Japanese, Brazilian and Belgian reference strains and one Iranian sample. Iranian samples were had significantly similarity to European and Australian samples.  相似文献

Data on the worldwide distribution of bovine immunodeficiency virus (BIV) and bovine leukemia virus (BLV) is limited. A prevalence study of antibodies to BIV and BLV was conducted in six different cattle herds in Brazil. Out of a total of 238 sera analyzed, 11.7% were found positive for anti-BIV p26 antibodies as determined by Western blot analysis, 2.1% were positive for anti-BLV gp51 antibodies as detected by immunodiffusion test. Peripheral blood mononuclear cells from BIV seropositive cattle were found to have BIV-provirus DNA, as detected by nested polymerase chain reaction. A nucleotide sequence corresponding to a 298 bp fragment of the BIV pol gene was also analyzed. Amino acid sequences of these Brazilian pol gene products showed 98.0 to 100% homology to the American strain BIV R29, 97.0 to 99.0% to Japanese BIV isolates, and divergence ranged from 0 to 4.0% among Brazilian BIV isolates. This evidence of the presence of BIV and BLV infections in Brazil should be considered a health risk to Brazilian cattle populations and a potential causative agent of chronic disease in cattle.  相似文献

With the aim of achieve a better understanding of the epidemiology and distribution of bovine leukaemia virus (BLV) infection in Chile, we assessed the suitability of using DNA isolated from the leukocyte fraction of bulk milk samples to carry out PCR-RFLP and DNA sequence analysis. The env fragment of BLV was successfully amplified from 33 serologically positive bulk milk samples collected from different geographical areas in the south of Chile. Restriction analysis allowed to classify 17 isolates within the Australian subgroup and 16 within the Belgium subgroup. DNA sequence and multiple alignment analysis of eight Chilean isolates showed a significantly higher frequency of single and double nucleotide substitutions. Most of these mutations were non-silent, resulting in changes at the protein level in several important epitopes of gp51. The Chilean sequences and 59 BLV env sequences available at GenBank, were subjected to a phylogenetic analysis, resulting in four different clusters. The groups identified were not related to those previously defined by restriction analysis. Chilean isolates were included in two different clusters and were genetically not related to isolates collected from neighbouring countries. Considering our results we can conclude: (i) bulk milk samples are suitable to identify the presence of BLV allowing epidemiological and genetic studies to be conducted on large geographical areas; (ii) at least four different genetic groups of BLV were identified by phylogenetic analysis, with Chilean isolates included in two different sub clusters.  相似文献

Bovine leukemia virus (BLV) is the etiological agent of enzootic bovine leukosis, the most common neoplastic disease of cattle worldwide and a serious problem for the cattle industry. Previous studies have shown the molecular prevalence of BLV and the coexistence of BLV genotype-1 and -4 in Egyptian dairy cattle; however, the molecular characteristics of BLV in Egyptian beef cattle are unknown. Therefore, we collected blood samples of 168 beef cattle from slaughterhouses in three governorates in Egypt. Based on BLV-CoCoMo-qPCR-2 targeting long terminal repeats and nested PCR targeting the env-gp51 gene, the BLV provirus infection rates were found to be 47/168 (28.0%) and 42/168 (25.0%), respectively. Phylogenetic analysis based on 501 bp of the BLV env-gp51 gene from 42 BLV isolates revealed that at least six distinctive strains (b, e, f, g, x, and z) were prevalent in cattle across the examined regions. Furthermore, phylogenetic analysis of the 420 bp sequence of the BLV env-gp51 region of the six strains against 11 known genotypes showed that the strains b, e, f, and g were clustered into genotype-1, and strains x and z were clustered into genotype-4. Our results also indicated that strains b and x exist in both dairy and beef cattle in Egypt. The present study is the first to detect and genotype BLV among beef cattle in Egypt.  相似文献

A first attempt for the investigation of molecular epidemiology of BLV was carried out. PCR amplicons of a part of the env gene of BLV isolated from 309 cattle of different geographical origin were compared with known BLV env sequences. Using RFLPA most of the PCR products can be assigned to the Australian, the Japanese or the Belgian subgroup. A phylogenetic tree resulting from the comparison of the sequences of these env fragments demonstrates the relations and differences between and within the subgroups.  相似文献

Three natural recombinant avian leukosis viruses (ALV; PDRC-1039, PDRC-3246, and PDRC-3249) expressing a subgroup A gp85 envelope protein and containing long terminal repeats (LTR) of endogenous ALV-E viruses were isolated from contaminated commercial Marek's disease vaccines, cloned, and completely sequenced. Their full genomes were analyzed and compared with representative strains of ALV. The proviral DNA of all three isolates displayed 99.3% identity to each other, suggesting a possible common ancestor, even though the contaminating viruses were obtained from three separate vaccine serials produced by two different vaccine manufacturing companies. The contaminating viruses have a genetic organization typical of replication-competent alpharetroviruses. The proviral genomes of PDRC-1039 and PDRC-3246 are 7497 bp long, and the PDRC-3249 is three base pairs shorter because of a deletion of a threonine residue within the gp85 coding region. The LTR, gag, pol, and the transmembrane (TM) region (gp37) of the env gene of all three viruses displayed high identity to endogenous counterpart sequences (>98%). Only the surface (SU) region (gp85) of the env gene displayed high identity with exogenous ALV-A (98.7%). Locus-specific polymerase chain reaction (PCR) analysis for ALV endogenous sequences (ev loci) in the chicken embryo fibroblasts used to produce the original vaccine vials identified the presence of ev-1, ev-2, ev-3, ev-4, and ev-6 in all three vaccines. Homologous recombination most likely took place to involve the SU region of the env gene because the recombinant viruses only differ in this particular region from the consensus ALV-E. These results suggest that the contaminating ALV isolates probably emerged by recombination of ALV-A with endogenous virus sequences before vaccine preparation.  相似文献

A serologic subgroup of bovine leukemia virus (BLV) has not been identified, whereas genetic diversity among BLVs has been reported by polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP). To investigate the distribution of BLV provirus variants, 42 isolates from Argentina and Japan were examined by nested PCR for a segment of the env gene, followed by DNA sequencing. The nucleotide sequences were compared with other previously characterized BLV variants from different geographical areas (Belgium, France, Italy, North America, Australia, Japan and Argentina). The majority of analyzed segments had a tendency for nucleotide substitution without changing the amino acid. The constructed phylogenetic tree showed the relations and differences between proviruses and within each one. Most of the samples in Argentina formed one cluster. The samples in Japan, except one, also formed one cluster and some of them showed high homology with the isolates from Australia and the USA. Considering the sequence analysis of env PCR products of all Japanese and Argentine samples and comparing them with the other previously isolated sequences, the variation was up to 3.5% and was characterized geographically in each area.  相似文献

为了解广东某七彩山鸡种禽场禽白血病流行情况,通过接种DF-1细胞、ELISA抗原检测、聚合酶链式反应(PCR)、囊膜基因克隆测序等方法分离并鉴定出两株ALV-F,分别命名为FGD1801和FGD1802。为进一步了解分离株遗传进化特点,利用PCR方法扩增出env基因并测序,同时与各亚群参考毒株的gp85核苷酸序列进行对比。结果显示:这两个分离株env基因gp85片段长度都为1080 bp,预计编码360个氨基酸,FGD1801和FGD1802分离株gp85基因核苷酸序列的相似性为92.8%,与A、B、E、J和K亚群共26株ALV参考毒株相似性在47.0%~64.5%之间,而与F亚群参考株的相似性在92.0%~92.5%,显著高于其他亚群,且位于同一进化分支上。研究表明,从七彩山鸡分离的FGD1801和FGD1802属于ALV-F,是我国华南地区新发现的ALV亚群。  相似文献

为了解我国东北地区部分养鸡场禽白血病病毒(ALV)的基因组序列特征及其变异情况,本研究从具有典型血管瘤病变的禽白血病发病鸡中分离到一株J亚群ALV(ALV-J)命名为JL0901,并进行了全基因测序.将该序列与已发表的ALV-J毒株序列进行比较研究,结果表明JL0901基因组的gag和pol基因相对保守,而env基因和3'端非编码区(3'UTR)的变异较大.对JL0901的env基因核苷酸序列进一步分析发现,在其gp85基因和gp37基因交界位置发生J亚群和E亚群ALV重组现象.本研究证实国内鸡群中存在J亚群和其他亚群ALV的自然重组现象,并表明国内ALV已出现新的变异趋势.  相似文献

本试验通过PCR技术获得了安徽省地方品种五华鸡禽白血病病毒J亚群(avian leukosis virus subgroup J,ALV-J)env部分基因序列ALV-J-env1,并将该序列与GenBank数据库中登录的8条env基因相应序列进行了比对分析。结果表明,五华鸡已经感染了J亚群禽白血病,且部分鸡个体已经发病。通过分析可见,ALV-J-env1与所比较的基因序列同源性介于94.2%~96.6%之间,说明不同毒株之间的env基因有一定变异。但与ALV-J-env1同源性最高的是来自于中国ALV-J毒株的HQ425636和HM235665基因序列,且在进化树中聚集为一组,暗示它们之间亲缘关系较近,由共同的毒株进化而来。与ALV-J-env1同源性最低的是来源于马来西亚毒株的AY312965基因序列,遗传进化分析也进一步证实,两者之间亲缘关系较远。  相似文献

Three kinds of recombinant vaccinia virus (RVV)--mO-HA/ATI, LO1-HA/ATI and mO-HA/7.5kD--expressing bovine leukosis virus (BLV) envelope glycoprotein (gp60) were constructed. The BLV envelope gene of RVV mO-HA/ATI and LO1-HA/ATI or of RVV mO-HA/7.5kD was expressed under control of the promoter of A-type inclusion body (ATI) protein gene of cow-pox virus or vaccinia virus 7.5-kD protein gene, respectively. The vaccinia virus strain, LC16mO, was used as vector for RVV mO-HA/ATI and mO-HA/7.5kD, and strain LO-1 was used for RVV LO1-HA/ATI. Strains LC16mO and LO-1 are attenuated vaccine virus strains originating from the Lister original vaccinia virus. All 3 kinds of constructed RVV expressed gp60 in cultured rabbit kidney cells after infection; mO-HA/ATI expressed more antigen than did mO-HA/7.5kD. Rabbits vaccinated with RVV produced considerable antibody capable of inhibiting syncytium formation, as well as antibody with virion-binding ability. The RVV that used ATI promoter induced higher antibody titer than did the RVV that used 7.5-kD promoter. Results indicate that BLV gp60 is responsible for induction of neutralizing antibodies that suppress in vitro formation of syncytia among BLV-infected cells. Applicability of RVV, especially those using ATI promoter, was evaluated in a vaccine against bovine leukosis.  相似文献

Anaplasma marginale is an obligate intraerythrocytic rickettsial pathogen (order, Rickettsiales: family, Anaplasmataceae) that causes bovine anaplasmosis. This disease is widely distributed in tropical and sub-tropical regions of the world and causes important economic losses to cattle production. Major surface protein (MSP)1a (msp1alpha gene) is one of the six MSPs identified on A. marginale from cattle, whose sequence and size vary according to the number of tandem 28- to 29-amino acid repeats. This study characterized the msp1alpha and msp4 genes obtained from three distinct Brazilian herds from the State of Paraná. Three strains of the msp1alpha and one strain of the msp4 gene were sequenced. The strains evaluated revealed PCR products of different size, representing three, five and six internal repeats. Sequence analyses confirmed the number of tandem sequence copies and revealed a high degree of sequence identity with strains from other Brazilian States, as well as strains from the USA, Europe and Israel. The msp1alpha DNA and amino acid sequences from A. marginale and DNA sequences of msp4 strains did not reveal distinct phylogeographical segregation. However, the amino acid sequences of msp4 demonstrated definite phylogeographical relationship. These results suggest that the amino acid sequences of msp4 should be used for phylogenetic identification of A. marginale strains and may be an important tool for the epidemiology and control of anaplasmosis. Additionally, the close similarity of the Paraná strains of A. marginale with strains from USA, Europe and Asia may reflect the introduction of these genes during the development of the Brazilian bovine herd.  相似文献

本试验通过病理剖检、接种DF-1细胞、ELISA及RT-PCR检测,从北京某种鸡场的疑似禽白血病的父母代蛋鸡中分离到1株A亚群禽白血病病毒,命名为QC6281.并根据GenBank中已发表序列设计引物,扩增env基因片段并将其克隆到pMD19-T-simple 载体中,扩增的env基因片段大小为1 239 bp,其中包括完整的gp 85基因,将gp 85基因亚克隆到pMD19-T-simple载体中并测序,gp 85基因大小为1 032 bp,编码344个氨基酸.将env基因片段和推导的氨基酸序列与GenBank中9株ALV的env基因相应序列做同源性分析并制作进化树图谱.发现QC6281与已发表的A亚群禽白血病病毒株该区域核苷酸同源性在83.8%~98%,氨基酸同源性在86.9%~98.5%.其中与Myeloblastosis-associated virus type 1(L10922.1)同源性最高,核苷酸同源性为98%,氨基酸同源性为98.5%;且与Myeloblastosis-associated virus type 1(L10922.1)亲缘关系最近.本研究为该毒株的生物学特性以及致病性研究打下基础.  相似文献

应用RT-PCR对2株禽白血病病毒(ALV)和1株劳斯肉瘤病毒(RSV)进行了检测试验.试验提取了病毒RNA,并使用3对ALVgp85基因引物对其进行了反转录、扩增,从而建立了ALV-RSV RT-PCR.使用无相关性的禽RNA和DNA病毒进行特异性试验和使用ALV进行的敏感性试验表明,该方法是一种快速、特异、敏感的体外试验,可用于禽源病毒种毒和疫苗中ALV和RSV的污染检测.  相似文献

Enzootic bovine leukaemia (EBL) which is caused by the bovine leukaemia virus (BLV) still plays a remarkable role despite a significant success in sanitation programmes. In the Federal Republic of Germany it was not possible to eradicate the disease until today. Sporadically during slaughter or necropsy of cattle neoplastic lesions of the lymphatic tissues are observed that need to be clarified with regard to BLV as etiological agent. Due to the fact that in most instances no serological data are available from the respective animals and blood drawings from the original holdings are not easy to obtain the polymerase chain reaction (PCR) opens new avenues as supplementary diagnostic tool to test unfixed lymphatic tissues for the presence of BLV proviral DNA. Lymph node tissues from 10 naturally or experimentally BLV-infected cattle, which have been monitored virologically and serologically, and tissues from 4 negative animals were processed, DNA was extracted and subjected to PCR to amplify BLV env gene specific sequences. The results show that in cattle with BLV-induced leukosis as well as in cattle, which were clinically healthy and unsuspicious at slaughter or at post-mortem, either with persistent lymphocytosis (PL) or without, BLV proviral DNA could be detected easily in samples of lymphatic tissues and in high concordance with serological data. In this article data from the National and OIE reference laboratory for EBL at the Friedrich-Loeffler-Institut (FLI, Germany) are presented. Elaborated laboratory protocols for processing of tissue samples and performing of BLV-PCR are recommended.  相似文献

为了建立一种快速诊断牛白血病(BLV)的检测方法,本研究从BLV中扩增出其囊膜糖蛋白gp51基因,经测序后克隆于表达载体pET32a(+)中,构建了重组表达质粒pET32a-gp51。将其转化受体菌BL21,用IPTG诱导后表达出约42ku的目的蛋白,经western blot检测表明目的蛋白具良好的反应原性。以胶体金标记的羊抗牛IgG(Fc)抗体作为标记抗体,将纯化的His-gp51重组蛋白和羊抗牛IgG分别标记于硝酸纤维素膜上作为检测线和质控线,各部件按序装配形成快速诊断试纸条。对22份样本分别用试纸条和ELISA试验进行检测,2种方法的阳性符合率为88.9%(8/9),表明本研究建立的胶体金免疫层析方法简便、快捷,具有较好的特异性和一定的敏感性,适宜基层初筛诊断和现场应用。  相似文献

为阐明我国马传染性贫血病毒(Equine infectious anemia virus,EIAV)弱毒疫苗致弱及免疫保护的分子机制,作者分析了疫苗株及其亲本强毒株共34个囊膜基因序列。结果发现疫苗株在膜基因gp90V3区的潜在N-连接糖基化位点出现稳定的碱基替换,使该位点消失。为研究囊膜糖基化的作用,以疫苗株全长感染性克隆pLG—FD3—8为亲本,利用反向遗传技术对该突变位点进行糖基化序列回复操作,构建全基因感染性克隆pLGFDg5。将pLGFDg5转染驴胎皮肤细胞(FDD),通过逆转录酶活性和RT-PCR方法评价其感染性。结果表明,将pLGFDg5在FDD细胞中盲传3代后,可在细胞培养上清中检测到逆转录酶活性,用RT—PCR检测到EIAV保守基因片段,在电镜下可见典型的EIAV粒子。在FDD细胞上的病毒复制动力学分析显示,与其亲本克隆pLGFD3—8的衍生病毒pLGFD3-V相比,回复突变克隆衍生的pLGFDg5-V复制速度较慢,获得较低的病毒载量。体外抗体中和试验表明,与其亲本pLGFD3-V相比,引入潜在的糖基化位点g5降低了衍生病毒对中和抗体的敏感性。这一结果为N-连接糖基化在我国马传贫弱毒疫苗致弱机理的作用研究提供了重要依据。  相似文献

A total of 59 monoclonal antibodies (mAbs) specific against the bovine leukaemia virus (BLV) using different antigen preparations was produced. The five antigen preparations for immunizing BALB/c mice were: live cells (CEL), sonicated and ultracentrifuged cells (SOC), cell lysates (LYS), semi-purified BLV (PV), and formalin-treated cells (FOR) from two cell lines permanently infected with BLV (FLK-BLV and BLV-bat2). These viral component presentations were selected to obtain mAbs against specific BLV proteins: located on the cell surface (FOR and CEL), in free virus particles (PV) and intracellular viral proteins (SOC and LYS). Two antigen preparations (SOC and LYS) were lethal to the mice following the intravenous and intrasplenic routes. Six fusions were performed in this study that rendered specific antibodies against BLV. The highest number of hybridomas was produced with SOC; however, the majority of the hybridomas produced (> 90%) were against cellular proteins. Even though immunization with PV gave the lowest number of hybridomas, the majority of them were specific against BLV. Based on the reactivity of the mAbs in Western blot (WB), we classified the mAbs into five groups, namely anti-gp51SU (39 mAbs), anti-gp30TM (six mAbs), anti-Pr72env (nine mAbs), anti-Pr66gag-pro (one mAb) and anti-Prgag (four mAbs). A very high percentage of the mAbs produced (48 of 59) reacted with gp51SU, suggesting that this is the most immunogenic and accessible BLV protein presented in the different antigen preparations. The majority of our mAbs recognized more than one band in WB, suggesting that, aside from reacting with mature proteins, the mAbs also recognized viral precursors.  相似文献

Expression of bovine leukemia virus (BLV) antigens in vivo has not been shown. After BLV infection, however, production of antibodies directed towards BLV proteins (e.g. gp51) can be easily demonstrated. Thus, production of BLV proteins has to take place somewhere in infected cattle. Tissues and organs of experimentally infected cattle were fixed in acetone and embedded in paraffin. Monoclonal antibodies directed to gp51 were used to demonstrate BLV expression immunohistologically by the peroxidase-antiperoxidase (PAP) method. The same samples were also used to demonstrate a tumor associated antigen (TAA) employing a monoclonal antibody. Our results indicate that very few cells, found in the intestinal mucosa, produce gp51 in vivo. The expression of TAA, however, increases significantly shortly after infection with BLV and remains high throughout life.  相似文献
