养殖鳗鲡肠道益生菌的筛选   总被引:9,自引:0,他引:9

从正常养殖的鳗鲡肠道中分离得到477株细菌,通过对常见病原菌的拮抗试验和产酶能力测试筛选出47株细菌。通过对鳗鲡饲料培养基的利用能力从中筛选出19株细菌,鉴定分类后选择7株可能的益生菌进行鱼粉分解试验,筛选得到2株细菌,即A40209CDC4和A31009NA。最后测定这2株细菌对欧洲鳗鲡的毒力和对鳗鲡饲料的表观消化率。结果显示,在急性毒性试验中,试验剂量的细菌对欧洲鳗鲡无毒力,且能有效地提高对饲料的利用率,表明筛选出的2株肠道益生菌A40209CDC4和A31009NA具有良好的开发和应用前景。  相似文献

为探讨壳聚糖和益生菌是否通过甲状腺激素、皮质醇对异育银鲫的非特异免疫功能的进行调控,试验以基础饲料为对照,在基础饲料中分别添加0.50%壳聚糖A、0.50%壳聚糖B、0.20%益生菌以及0.50%壳聚糖A与0.05%益生菌的混合物制备试验饲料,饲养异育银鲫64d后,测定各组鱼外周血白细胞数量及其分类、白细胞吞噬活性、脾脏溶菌酶活性、脾脏淋巴细胞转化能力;同时采用放射免疫分析法测定异育银鲫血清中三碘甲腺原氨酸(T3)、甲状腺素(T4)及皮质醇含量。结果表明,饲料中添加壳聚糖、益生菌或其混合物对异育银鲫白细胞总数无显著影响,但是可显著提高异育银鲫外周血淋巴细胞数量、白细胞吞噬活性和脾脏溶菌酶活性(P<0.05),显著降低外周血多形核细胞数量(P<0.05)。除了壳聚糖A能够显著提高异育银鲫脾脏B淋巴细胞转化能力之外,其余试验组的脾脏T、B淋巴细胞转化能力均与对照组无显著差异(P>0.05)。同时发现饲料中添加壳聚糖、益生菌或其混合物可以显著提高异育银鲫血清中T3的含量(P<0.05)、显著降低异育银鲫血清中皮质醇的含量(P<0.05),除了壳聚糖B对异育银鲫血清中T4的含量无显著影响(P>0.05)之外,其余均可...  相似文献

为建立鳗鲡外周血白细胞体外吞噬嗜水气单胞菌的数量模型,研究了感染时间一定、感染复数变化的情况下鳗鲡外周血白细胞吞噬的细菌数量和白细胞的存活率,同时还研究了感染复数一定、感染时间变化的情况下鳗鲡外周血白细胞吞噬的细菌数量和白细胞的存活率。结果发现,鳗鲡外周血白细胞对嗜水气单胞茵有较强的吞噬作用,在感染复数为100:1、感染时间为30min时细胞吞噬的细菌量达到饱和,而细胞本身仍可维持较高活性。表明100:1的感染复数及30rain的感染时间,是研究鳗鲡外周血白细体外吞噬嗜水气单胞菌数量模型的最佳参数。本研究还通过检测吞噬速率和杀菌速率评估建立起来的数量模型发现,鳗鲡外周血白细胞吞噬嗜水气单胞菌的速率较吞噬大肠杆菌的速率快,但杀灭该茵的速率却均显著低于杀灭大肠杆菌和金黄色葡萄球菌的速率,进一步证明建立的数量模型具有良好的可行性。  相似文献

实验组光合细菌质量浓度分别为2.5、5.0、10.0g/m3,在第1、7、14、21、28天检测鲤鱼血液白细胞吞噬活性和血清溶菌酶活性等指标.结果表明,该光合细菌制剂可以提高鲤鱼白细胞的吞噬活性,增强血清溶菌酶活力.鲤鱼的非特异性免疫功能随着光合细菌制剂浓度的增加和泼洒时间的延长呈现增强趋势.  相似文献

研究了苏云金芽孢杆菌对黄河鲤血液免疫应答的影响.将苏云金芽孢杆菌按1.0×10~(11)、3.0×10~(11)、5.0×10~(11) cfu/kg 3个浓度添加在鱼用全价颗粒饲料中,投喂经嗜水气单胞菌疫苗免疫组(A_0~A_3)和未免疫组(B_0~B_3)黄河鲤,分别于0、15、30、40 d时检测各组黄河鲤白细胞吞噬活性、血清溶菌酶活性以及血清凝集抗体效价,并进行攻毒试验.结果表明:添加苏云金芽孢杆菌可使黄河鲤血液中白细胞吞噬活性和血清溶菌酶活性显著提高.添加组与对照组差异显著(P<0.05),添加浓度越高,血液白细胞活性和溶菌酶活性越强.免疫组与未免疫组白细胞吞噬活性差异不显著(P>0.05),而溶菌酶活性差异显著(P<0.05).在停止投喂后10 d,白细胞活性、血清溶菌酶活性逐渐下降,但差异不显著(P>0.05).添加苏云金芽孢杆菌还可提高黄河鲤的凝集抗体效价及攻毒后的存活率,其中,当菌株添加浓度为5.0×10~(11) cfu/kg时,受免疫的黄河鲤免疫保护率最高.即投喂苏云金芽孢杆菌对黄河鲤血液免疫应答有明显促进作用.  相似文献

在饲料中添加不同剂量的Vc、酵母多糖和壳聚糖,连续投喂42d,并在试验的第14、28、35和42天取样检测,研究其对草鱼血清溶菌酶活性、血清超氧化物歧化酶活性(SOD)活性、血清碱性磷酸酶(AKP)活性、补体C3、含量丙二醛(MDA)含量和血液白细胞吞噬活性共6种指标的影响。结果显示:实验组与对照组相比,饲料中添加Vc可以提高血清溶菌酶活性、SOD活性、AKP活性、补体C3含量和血液白细胞吞噬活性高,并降低MDA含量。添加酵母多糖可以提高血清溶菌酶活性、AKP活性、补体C3含量和血液白细胞吞噬活性,并降低MDA含量,但不能提高SOD活性。添加壳聚糖可以提高血清溶菌酶活性、SOD活性、AKP活性、补体C3含量和血液白细胞吞噬活性,但不能降低MDA含量。饲料中添加Vc、酵母多糖和壳聚糖均对草鱼的非特异性免疫功能具有一定的促进作用。  相似文献

蝇蛆既是优质饲料蛋白质又是新型免疫增强剂。为探明蝇蛆蛋白混合物对美洲鳗鲡非特异性免疫功能的影响,水温为25~28℃时,将750尾初始体质量(95.0±5.0) g的美洲鳗鲡随机分为5组,每组3个平行,每个平行50尾鱼,分别投喂在基础饲料中添加质量分数为0%(对照组)、5%、10%、15%和20%蝇蛆蛋白(由蝇蛆蛋白混合物折算而来)的饲料,投喂养殖60 d后,测定美洲鳗鲡血清中溶菌酶、补体3、补体4、酸性磷酸酶、碱性磷酸酶、超氧化物歧化酶、谷丙转氨酶和谷草转氨酶等免疫指标的变动情况及嗜水气单胞菌攻毒后美洲鳗鲡的死亡率。试验结果显示:美洲鳗鲡血清中补体3、补体4含量随蝇蛆蛋白混合物添加比例增加而显著提高(P<0.05),其中补体3以15%添加组含量最高,补体4以20%添加组含量最高;血清中溶菌酶、酸性磷酸酶、碱性磷酸酶和超氧化物歧化酶活性均随蝇蛆蛋白混合物添加量的增加先升后降,除5%添加组碱性磷酸酶活性与对照组差异不显著外,其余均显著高于对照组(P<0.05),其中溶菌酶、酸性磷酸酶和超氧化物歧化酶均以15%添加组活性最高,碱性磷酸酶以10%添加组最高;谷丙转氨酶活性也随蝇蛆蛋...  相似文献

将阿维拉霉素按照一定的浓度添加到饲料中,投喂经注射接种福尔马林灭活的嗜水气单胞菌(Aerom onashydrophila)菌苗的鲤,于免疫后的第7 d、14 d、21 d、28 d和35 d检测供试鲤血液中的白细胞吞噬活性、血清中溶菌酶活性、抗体效价、补体C3含量及免疫保护率(RPS),探讨了阿维拉霉素对受免鲤免疫应答的影响。结果表明,饲料中添加阿维拉霉素(4 mg/kg和8 mg/kg),投喂较长时间(≥28 d)会降低受免鲤的血液白细胞吞噬活性、血清中的溶菌酶活性和抗体效价,减少受免鲤血清中补体C3的含量,减弱鲤抵抗嗜水气单胞菌人工感染的能力,每公斤饲料中添加8 mg的阿维拉霉素其免疫抑制作用更强。  相似文献

为了探讨 A_(3α)肽聚糖对灭活嗜水气单胞菌疫苗免疫效果的影响,用添加肽聚糖的饲料投喂注射接 种经甲醛灭活的嗜水气单胞菌菌苗的彭泽鲫,测定鲫鱼白细胞吞噬活性、血清和体表粘液溶菌酶活性、抗体 效价以及活菌攻毒后的免疫保护率。结果表明,肽聚糖与灭活菌苗联合使用,鱼体表粘液和血清溶菌酶活性、 白细胞吞噬活性、抗体效价以及活菌攻毒后的免疫保护率显著提高,表明 A_(3α)肽聚糖可增强灭活菌苗的免疫效 果。  相似文献

中草药添加剂对日本鳗鲡生长和非特异性免疫效应的研究   总被引:4,自引:2,他引:2  
在基础饲料中分别添加0.5 g/kg、1.0 g/kg和1.5 g/kg的自制中草药免疫添加剂,配制成3种试验饲料,以基础饲料为对照组饲料,每处理设3个平行样,对体重为30~40 g的日本鳗鲡(Anguilla japonica)进行为期60 d的饲养试验,每20 d取样一次,以相对增重率、成活率和血清中的溶菌酶(LZM)、一氧化氮合酶(NOS)、碱性磷酸酶(AKP)和超氧化物歧化酶(SOD)活性为指标,探讨了中草药免疫添加剂对日本鳗鲡生长和非特异性免疫效应的影响。结果显示:饲料中添加中草药免疫添加剂可以显著提高日本鳗鲡的增重率,降低饲料系数(P<0.05)。试验组日本鳗鲡血清中的LZM、NOS、AKP和SOD等活性均高于对照组。因此,中草药免疫添加剂具有显著促生长和增强机体免疫能力的作用,建议在实际生产中的添加量为1.0 g/kg较为合适。  相似文献

The environmental processes associated with variability in the catch rates of bigeye tuna in the Atlantic Ocean are largely unexplored. This study used generalized additive models (GAMs) fitted to Taiwanese longline fishery data from 1990 to 2009 and investigated the association between environmental variables and catch rates to identify the processes influencing bigeye tuna distribution in the Atlantic Ocean. The present findings reveal that the year (temporal factor), latitude and longitude (spatial factors), and major regular longline target species of albacore catches are significant for the standardization of bigeye tuna catch rates in the Atlantic Ocean. The standardized catch rates and distribution of bigeye tuna were found to be related to environmental and climatic variation. The model selection processes showed that the selected GAMs explained 70% of the cumulative deviance in the entire Atlantic Ocean. Regarding environmental factors, the depth of the 20 degree isotherm (D20) substantially contributed to the explained deviance; other important factors were sea surface temperature (SST) and sea surface height deviation (SSHD). The potential fishing grounds were observed with SSTs of 22–28°C, a D20 shallower than 150 m and negative SSHDs in the Atlantic Ocean. The higher predicted catch rates were increased in the positive northern tropical Atlantic and negative North Atlantic Oscillation events with a higher SST and shallow D20, suggesting that climatic oscillations affect the population abundance and distribution of bigeye tuna.  相似文献

In this experiment, a feeding trial was performed to determine the effects of fructooligosaccharide (FOS) on growth performance, digestive enzyme activity and immune response of Japanese sea bass, Lateolabrax japonicus juveniles (initial weight 38.3 ± 0.5 g), and the fish were examined following feeding with six levels of FOS (0, 0.5, 1, 2, 4 and 6 g/kg) for 28 days. Significant enhancement of weight gain (WG) and specific growth rate (SGR) was found in fish fed 1 g/kg FOS incorporated diets (p < .05), while the feed conversion ratio (FCR) in the 1, 2 g/kg FOS groups reduced significantly compared with the control (p < .05). Besides, the crude lipid in the 4, 6 g/kg FOS groups increased significantly compared with the control (p < .05). On the other hand, the erepsin and lipase activities significantly elevated in intestine of fish fed 2 g/kg FOS (p < .05) and the lysozyme activity in serum of fish fed 2 g/kg FOS were significantly higher than that in the control (p < .05). Moreover, the alkaline phosphatase activities in serum of fish fed 0.5, 1, 2 g/kg FOS were significantly higher than in control (p < .05). Regression analysis showed that the relationships between dietary FOS levels and either SGR, FCR, erepsin or lysozyme activities were best expressed by regression equations, and the optimal inclusion levels are 1.37, 1.80, 3.06, 3.11, 1.93 and 1.80 g/kg for SGR, FCR, erepsin, lipase, lysozyme and total superoxide dismutase activities, respectively. Overall, this study revealed that FOS incorporated diets could beneficial for L. japonicus culture in terms of increasing the growth, digestion and immune activities. Under the present experimental condition, the optimal supplementary level of FOS in the diet of L. japonicus is 1–3 g/kg.  相似文献

Previously, we reported 10 PEGE types of 85 tilapia Streptococcus agalactiae(GBS), which shifted from Streptococcus iniae in China, by using PEGE method. Presently, larger and more representative tilapia GBS were isolated, for the ?rst time in China, to characterize their serotypes and genetic diversities more precisely than had done before. 168 GBS strains were distributed in ?ve provinces of China, in which Guangdong, Guangxi and Hainan were the major ones, holding36.9%(62/168), 37.5%(63/168) and 19.6%(33/168), respectively. Serotypes, Ia, Ib and III, were observed in these strains and the most predominant one was Ia(95.2%), which mainly distributed in Guangdong, Guangxi and Hainan. Ia initially occurred in 2009, it shoot up to 32.1% in 2010,but decreased to 16.1% in 2011 before went up to 45.2% in 2012. Ib sporadically occurred during2007–2011, III onlyoccurred in 2012. 14 different PFGE types, including 4 new types(N, O,P and Q), were observed, in which B, D, F and G were the predominant types, holding 83.9%(141/168) of the total GBS strains. Ia corresponded to 11 PFGE types(A–H, N–P), in which type D predominated(51%). Ib represented 3 genotypes(I, J and Q) and III harbored only 2genotypes(N and F). Type N and Fsynchronously presented in Ia and III. In summary, the genetic diversity of tilapia GBS varied by serotypes and changed with geographical locations and years.Although Iastillpredominated, new rareserotypeIII alreadyoccurred in China.  相似文献

The purpose of this work was to evaluate the suitability of a method based on the presence of two restriction sites (for Hae III and Hindf I) in the mitochondrial NADH dehydrogenase subunit 5 (mt ND5) gene to identify Scomber species. The evaluation was performed on 144 reference and market samples by sequencing of the entire 505-bp fragment of the mt ND5 gene and of a 464-bp fragment of the Kocher fragment of the cytochrome b gene (mt Cytb). Sequence analysis of any of the two fragments allows the identification of each of the four Scomber species, but S. japonicus and S. colias had the same restriction sites at the ND5 amplicon and would not have been differentiated by this analysis. Similarly, loss of the Hae III site in some S. scombrus individuals would have misidentified them as not being Scomber. All the market products were correctly labeled except one acquired in Spain labeled as originating in the Atlantic and containing S. japonicus.  相似文献

Interaction between growth and reproduction occurs in many vertebrates and is particularly obvious at certain stages of the life cycle in fish. Endocrine interactions between the gonadotropic axis and the somatotropic axis are described, the potential role of GH being emphasised. A comparative analysis of these phenomena in mammals, amphibians and fish, suggests a specific role of GH in the physiology of puberty, gametogenesis and fertility. It also shows the original contribution made by studies on the fish model in this field of investigations.  相似文献

Two gonadotropins (GtH; Qa and Qb) were purified by gel filtration and ion exchange chromatography from the pituitaries of Indian walking catfish (Clarias batrachus). The presence of GtH during purification was assessed by in vitro oocyte maturation and in vivo steroidogenic activity, and their identities were determined by elution profiles, molecular weight, biological activities and yield. The molecular weights of Qa and Qb were 37 and 42 kDa, respectively, and composed of distinct subunits (Qa: 20 and 14 kDa and Qb: 26 and 18 kDa). Polyclonal antibodies raised against Qa immunostained Qa, Qb and pituitary GtH cells. A competitive Qa‐ELISA was developed whose sensitivity was 6.25 ng mL?1 (1.25 ng well?1) with intra‐ (3.5%) and inter‐ (12.4%) assay coefficients of variation. Displacement curves parallel to the standard were obtained with plasma and pituitary extracts of catfish, Qb and carp GtHII. The assay was validated by measuring the plasma Qa levels after LHRH treatment and in relation to ovarian growth in the female catfish during different reproductive phases. Based on the results, Qa and Qb corresponded to fish LH and FSH respectively. The findings will increase the knowledge of the mechanisms controlling fish reproduction and identification of sensitive phases in fish in captivity for hormonal manipulation.  相似文献

Tetrahymena is a protozoan parasite, which infects guppy, Poecilia reticulata Peters, and causes substantial economical losses in commercial farms worldwide. Studies of guppy infected by Tetrahymena require standardized infection protocols. The LD50 for Tetrahymena infection of guppies by intraperitoneal (IP) injection was calibrated, and the level obtained was 946 parasites per fish. Guppy infection with Tetrahymena by immersion, imitating the natural route of infection via the integument, was studied under normal or stress conditions. Exposure to cold and netting (CNI) and to cold only (CI) followed by immersion exposure to 10 000 Tetrahymena per mL resulted in 22.5% and 19.2% mortality, respectively, as compared to 14.2% and 10% in groups that were netted only (NI) or non‐stressed (I). Histopathology revealed that immersion infection resulted in a systemic infection. Lysozyme levels, measured 3 weeks after infection, were significantly higher in the CNI group (288 μg per mg protein) compared with CI‐, NI‐ and I‐treated groups (94.5, 64 and 62.3 μg mg?1, respectively). There was no evident parasite immobilization activity in body homogenates, suggesting no development of acquired immunity. Re‐infection by IP injection revealed no increase in protection in any of the treatment groups, mortality range of 56.3–75%, higher than in the non‐exposed control (40.6% mortality).  相似文献
