利用蜱传播试验确定小亚璃眼蜱对中国新分离的牛的巴贝斯虫未定种和环形泰勒虫的传播能力与传播方式,进而明确其在中国牛梨形虫病传播中的流行病学意义。试验结果表明:小亚璃眼蜱可在雌虫阶段受到巴贝斯虫未定种的感染,并可在次代若虫(2/2)和成虫(3/3)阶段将病原传播给试验牛;小亚璃眼蜱幼虫和若虫吸入环形泰勒虫,饱血脱落的幼虫和若虫所蜕化发育的饥饿若虫(2/2)和成虫(2/2)均可将病原传递给敏感动物;感染巴贝斯虫未定种的小亚璃眼蜱饱血雌虫所孵育出的次代幼虫和若虫仍可被环形泰勒虫感染,所发育出的若虫(2/2)和成虫(2/2)可在一次传播试验中将环形泰勒虫和巴贝斯虫未定种同时传播给试验动物。  相似文献

An unidentified Babesia sp. which causes a mild disease in cattle was isolated in a splenectomized ox that received pooled blood from field cattle. That this organism is pleomorphic and resembles Babesia occultans makes it difficult to differentiate between these organisms microscopically. Initially, it was suspected that this Babesia could be B. occultans. Several attempts to transmit this parasite transovarially with Hyalomma marginatum rufipes, the vector of B. occultans, failed. Continued efforts to identify possible vectors, using Boophilus microplus, Rhipicephalus evertsi evertsi and Rhipicephalus appendiculatus, all failed. The only tick thus far identified that could have transmitted the infection transovarially in the adult stage was the two-host tick Hyalomma truncatum.  相似文献

Antigens derived from partially engorged nymphs of Hyalomma anatolicum anatolicum were used in immunizing crossbred (Bos indicus×Bos taurus) cattle against larval, nymphal and adult H. a. anatolicum and H. dromedarii. The cattle were either infected with Theileria annulata at low parasitaemia or were uninfected. Whole nymphal extract (WNE), nymphal membrane antigens (NMA) and nymphal soluble antigens (NSA) were used for immunization. The group immunized with WNE showed significant and better rejection of H. a. anatolicum ticks as compared to calves immunized with either NMA or NSA. The moulting rates of both engorged larvae and nymphs remained unaffected. Nymphs which engorged on the immunized calves were fully susceptible to infection by T. annulata as indicated by the intensity and abundance of Theileria infections in the resulting adult ticks from immunized and unimmunized Theileria infected cattle. These ticks also transmitted fatal theileriosis to susceptible calves.  相似文献

In this comparative study unfed nymphs of four Hyalomma tick species (Hyalomma anatolicum anatolicum, Hyalomma anatolicum excavatum, Hyalomma detritum and Hyalomma marginatum marginatum) were allowed to engorge on calves experimentally infected with Theileria annulata. The infection prevalence in the salivary glands of the adult female and male ticks of each Hyalomma species used in the study were assessed. The infection prevalence with T. annulata was high and did not vary markedly in the four Hyalomma tick species. The mean number of infected acini per tick in female and male ticks was different with female ticks having higher numbers of infected acini than the male ticks. The sex difference was more significant between H.a. anatolicum and H.a. excavatum than between H. detritum and H.m. marginatum. This study clarifies the roles of four Hyalomma tick species, and their sex, in the development of T. annulata.  相似文献

Experiments on the transmission of an unidentified Theileria sp. infective for small ruminants by Haemaphysalis qinghaiensis and Hyalomma anatolicum anatolicum were carried out. Three Theileria-free batches of adult, larvae, and nymphs of laboratory reared H. qinghaiensis and Hy. a. anatolicum ticks were infected by feeding them on sheep infected with Theileria sp. The Theileria sp. was originally isolated from adult ticks of H. qinghaiensis, by inoculation of blood stabilates or tick transmission. H. qinghaiensis has been shown to be capable of transmitting the Theileria sp. infective for small ruminants transstadially to sheep and goats. The nymphs developed from the larvae engorged on the sheep infected with the parasite transmitted the pathogen to splenectomized sheep with prepatent periods of 30, 31 days, respectively; but the subsequent adult ticks of H. qinghaiensis derived from the nymphs did not transmit the pathogen to sheep. However, adults developed from the nymphs engorged on the sheep infected with the parasite transmitted the pathogen to sheep with prepatent periods of 24-27 days. The larvae, nymphs and adult ticks derived from female H. qinghaiensis ticks engorged on infected sheep were not able to transmit the parasite transovarially. The same experiments were done with Hy. a. anatolicum, but examination for presence of piroplasma of Theileria sp. from all animals were negative, demonstrating that Hy. a. anatolicum could not transmit the organism to sheep or goats.  相似文献

A total of 2388 cattle and 442 shelters, from two provinces (Elazig and Malatya) endemic for tropical theileriosis in the east of Turkey, were studied for Hyalomma tick populations from July 1993 to July 1995 in Elazig and from May 1998 to January 1999 in Malatya. Four thousand five hundred and eighty one of 7455 Hyalomma ticks were collected from cattle, the other ticks (2874) were collected from shelters. All of the ticks collected from shelters were Hyalomma anatolicum anatolicum. Two thousand eight hundred and ninety five (63.1%) of 4581 Hyalomma ticks collected from cattle were H.a. anatolicum. 23.8% (1047/4581), 11.7% (536/4581) and 0.6% (3/4581) of Hyalomma ticks were Hyalomma anatolicum excavatum, Hyalomma detritum and Hyalomma marginatum marginatum, respectively. A total of 5909 Hyalomma adult ticks collected from cattle (3362/5909) and shelters (2447/5909) were dissected and salivary glands were stained with Methylgreen/Pyronin method. Thousand one hundred and fifty (46.9%) of 2447 H.a. anatolicum collected from shelters and 412 (19.1%) of 2147 H.a. anatolicum collected from cattle were positive for Theileria infection. Twenty (2.4%) of 820 H.a. excavatum and 23 (4.6%) of 495 H. detritum collected from cattle were positive. The mean number of infected acini per infected male and female ticks collected from cattle were 11.3 and 22.4 in H.a. anatolicum, 4 and 6.8 in H.a. excavatum, 17.9 and 18.3 in H. detritum, respectively. In H.a. anatolicum collected from shelters, the above rates were 11.8 and 17.6 in male and female ticks, respectively. The prevalence and intensity of Theileria infection was greater in female ticks than in males.  相似文献

OBJECTIVE: To assess the efficacy of ivermectin and moxidectin to prevent transmission of Babesia bovis and Babesia bigemina by Boophilus microplus to cattle under conditions of relatively intense experimental challenge. DESIGN: Naive Bos taurus calves were treated with either pour-on or injectable formulations of either ivermectin or moxidectin and then exposed to larvae of B microplus infected with B bovis or larvae or adults of B microplus infected with B bigemina. One calf was used for each combination of haemoparasite, B microplus life stage, drug and application route. PROCEDURE: Groups of calves were treated with the test drugs in either pour-on or injectable formulation and then infested with B microplus larvae infected with B bovis or B bigemina. B bigemina infected adult male ticks grown on an untreated calf were later transferred to a fourth group of animals. Infections were monitored via peripheral blood smears to determine haemoparasite transmission. RESULTS: Cattle treated with either pour-on or injectable formulations of ivermectin and moxidectin became infected with B bovis after infestation with infected larvae. Similarly, larvae infected with B bigemina survived to the nymphal stage to transmit the haemoparasite to animals treated with each drug preparation. Cattle treated with pour-on formulations of ivermectin and moxidectin then infested with adult male ticks infected with B bigemina did not become infected with B bigemina whereas those treated with the injectable formulations of ivermectin and moxidectin did show a parasitaemia. CONCLUSIONS: Injectable or pour-on formulations of ivermectin and moxidectin do not prevent transmission of Babesia to cattle by B microplus. Use of these drugs can therefore not be recommended as a primary means of protecting susceptible cattle from the risk of Babesia infection.  相似文献

The 18S small subunit ribosomal RNA (18S rRNA) gene of an unnamed Babesia species (designated B. U sp.) was sequenced and analyzed in an attempt to distinguish it from other Babesia species in China. The target DNA segment was amplified by polymerase chain reaction (PCR). The PCR product was ligated to the pGEM-T Easy vector for sequencing. It was found that the length of the 18S rRNA gene of all B. U sp. Kashi 1 and B. U sp. Kashi 2 was 1699 bp and 1689 bp. Two phylogenetic trees were, respectively, inferred based on 18S rRNA sequence of the Chinese bovine Babesia isolates and all of Babesia species available in GenBank. The first tree showed that B. U sp. was situated in the branch between B. major Yili and B. bovis Shannxian, and the second tree revealed that B. U sp. was confined to the same group as B. caballi. The percent identity of B. U sp. with other Chinese Babesia species was between 74.2 and 91.8, while the percent identity between two B. U sp. isolates was 99.7. These results demonstrated that this B. U sp. is different from other Babesia species, but that two B. U sp. isolates obtained with nymphal and adultal Hyalomma anatolicum anatolicum tick belong to the same species.  相似文献

Theileria annulata was experimentally transmitted to cattle on two occasions by the two-host tick Hyalomma marginatum rufipes. Transmission was transstadial; engorged nymphs fed on Theileria annulata-infected calves transmitted the disease as adults. Salivary glands of all partially fed and incubated adult ticks were heavily infected with Theileria parasites. Immatures attached rapidly and fed successfully on cattle. However, since the immature stages of this species normally feed on birds, this tick is unlikely to be an important vector in the field.  相似文献

本研究旨在建立小亚璃眼蜱、亚洲璃眼蜱和残缘璃眼蜱的分子生物学鉴定方法,并探讨它们的系统发生关系。在新疆、内蒙古从动物体表采集寄生蜱,形态学鉴定后,PCR扩增测序获得3种璃眼蜱的16S rRNA及线粒体色素氧化酶亚基Ⅰ基因(cytochrome oxidaseⅠ,COⅠ)序列后进行同源性分析。用Mega 5.0和Mrbayes 3.2软件分别构建系统进化树,亚洲璃眼蜱、小亚璃眼蜱样本16S rRNA序列与GenBank中已知相应蜱虫16S rRNA序列聚类,而残缘璃眼蜱COⅠ序列与GenBank中已知残缘璃眼蜱COⅠ序列聚类,与形态学鉴定结果一致。在传统形态学分类的基础上,结合分子生物学鉴定方法能简易、准确鉴定小亚璃眼蜱、亚洲璃眼蜱和残缘璃眼蜱。  相似文献

Studies were undertaken to determine the effect of repeated pure infestations with Boophilus microplus on susceptibility to subsequent pure infestations with Hyalomma anatolicum anatolicum, and the effects of pure infestations with both species of tick on susceptibility to a series of mixed infestations. Crossbred (Bos indicus X Bos taurus) calves were infested with Boophilus microplus (17 times), H. a. anatolicum (four times), followed by five mixed infestations of B. microplus and H. a. anatolicum. The decline in B. microplus engorgement from a mean yield of 274.4 +/- 60.3 ticks per host after the first exposure, to a mean yield of 9 +/- 4.6 per animal after the seventeenth exposure, was observed in animals exposed to only B. microplus. This might be due to acquired resistance. However, these animals were found to be as susceptible to H. a. anatolicum as animals which had never been exposed to ticks of either species. A decline in the yield of H. a. anatolicum from a mean yield of 92.1 +/- 10.7 after the first exposure to 54.7 +/- 11.3 after the fourth exposure, indicated that the cattle could also acquire resistance to repeated pure infestations with this species. After repeated pure infestations with both tick species, cattle reacted to five mixed infestations showing a high degree of resistance to B. microplus and low resistance to H. a. anatolicum (mean yield for B. microplus was only 10 +/- 8.1 ticks per host after the first mixed exposure and declined to 1.3 +/- 1.7 after the fifth, whereas the mean yield for H. a. anatolicum was 71.4 +/- 11.3 ticks per host following the first exposure and declined to 37.3 +/- 7.8 after the fifth). Host responses elicited to one species do not provide cross-resistance to the second species used in this study.  相似文献

将实验室培育的“洁净”长角血蜱的幼虫、若虫、成虫及微小牛蜱的幼虫,先后分别释放到人工感染卵形巴贝斯虫单一种牛体上不同部位事先粘贴好的布袋中,使其自行叮咬吸血。待饱血脱落后亦分别收集,置28℃、相对湿度约90%的条件下蜕皮或产卵孵化。而后用不同世代和各变态期的蜱,分别叮咬感染除脾和非除脾健康易感牛。结果表明,长角血蜱的当代若虫和成虫对卵形巴贝斯虫没有传播能力,幼虫和若虫吸入的病原亦不能经卵传递;饱血雌虫吸入的病原可经卵传递,次代幼虫、若虫和成虫三个阶段都具有传播能力。次代感染幼虫经兔体后的若虫和成虫也具有感染力。 微小牛蜱不能传播该种病原。  相似文献

Theileria sp occurs in cattle in Australia where Boophilus microplus is not present. Two species of ixodid ticks namely Haemaphysalis longicornis and H. bancrofti have been implicated as natural vectors. Experimental transmission was obtained with these two species but attempts to transmit the infection with B. microplus, Ixodes holocyclus and Amblyomma triguttatum were not successful. Although calves born in the enzootic area may become infected before one-month-old most show evidence of infection by 5 months of age. Calves born in the winter also become infected earlier than those born at other times of the year.  相似文献

Hyalomma anatolicum anatolicum nymphs were collected from two localities in the Sudan: Eddamer in Northern Sudan and Wad-Medani in Central Sudan. They were allowed to moult to adult ticks, which were assessed for Theileria infection in their salivary glands using Feulgen stain. At Eddamer, 49.6% of 123 ticks examined were infected with Theileria and the mean intensity of infection was 1.3 (i.e. the number of infected acini/number of infected ticks). At Wad-Medani, 8.6% of 162 ticks were infected and the mean intensity of infection was 7.9. The prevalence of infection was higher in female than in male ticks at both localities. When adult H. a. anatolicum were applied onto two susceptible calves, both animals developed the severe form of theileriosis.  相似文献

Hyalomma anatolicum anatolicum ticks were collected around cattle pens at two locations close to Khartoum. They were assessed for theileria infection by four methods. Salivary glands were stained whole with methyl green pyronin and examined for parasite masses. Adult ticks were partially fed on rabbits, ground up in medium, and the suspensions were examined in Giemsa stained smears and by inoculation into bovine lymphocyte cultures. Ticks were fully fed on calves which were monitored for developing theileriosis. H a anatolicum were found infected with parasite masses similar to those seen in experimental infections with T annulata. At one site 38 per cent of 102 ticks were infected and the mean number of parasite masses per tick for the whole sample was 37. At the other site 86 per cent of 156 ticks were similarly infected and the mean parasite masses per tick was 19.5. Suspensions of sporozoites contained sporozoites typical of those found in experimental preparations of T annulata. Sporozoites harvested from ticks from both locations infected and transformed normal bovine lymphocyte cultures. H a anatolicum ticks from both locations produced fatal theileria infection in susceptible calves.  相似文献

A total of 13 ixodid tick species, Boophilus microplus, Haemaphysalis bispinosa, Haemaphysalis cornupunctata, Haemaphysalis himalaya, Heamaphysalis montgomeryi, Hyalomma anatolicum anatolicum, Hyalomma dromedarii, Hyalomma marginatum isaaci, Hyalomma (Hyalommina) brevipunctata, Hyalomma (Hyalommina) hussaini, Nosomma monstrosum, Rhipicephalus haemaphysaloides, and R. turanicus were collected off 424 buffaloes from the northwestern states of India. Ten tick species, Amblyomma testudinarium, B. microplus, Haemaphysalis anomala, Haemaphysalis arborensis, Haemaphysalis bispinosa, Haemaphysalis intermedia, Haemaphysalis nepalensis, Haemaphysalis neumanni, R. haemaphysaloides, and R. turanicus parasitising 194 buffaloes were collected from the northeastern states of India. In addition to tick-buffalo relationships, the incidence of haemoparasites in buffalo from these two regions was studied and the possible role of these ticks in disease transmission was discussed. Examination of blood films and lymph smears revealed Anaplasma marginale and Babesia bigemina in 6.2 and 2.6%, respectively, of buffaloes tested in the northeastern states, and 14.9 and 4.7%, respectively, in the northwestern states.  相似文献

Babesia caballi, isolated from a horse that originated from South West Africa/Namibia, was transmitted transovarially by adult Hyalomma truncatum. B. caballi proved to be highly infective for adult H. truncatum. Forty-five per cent of ticks feeding on a reacting animal with an extremely low parasitaemia became infected. In spite of a low parasitaemia, the ticks were severely affected by the parasite. Seventy per cent of the infected ticks either died during oviposition or after laying only a few eggs. The features of the infection in horses were: a prepatent period of 10 days, very low parasitaemias with low pathogenicity and spontaneous recovery of the infected animals.  相似文献

Hyalomma anatolicum anatolicum engorged nymphs and flat adults were collected from two areas in northern Sudan. Various developing stages of Trypanosoma theileri-like flagellates were observed in the engorged nymphs, freshly moulted adults and mature adults partially engorged on rabbits. When these ticks were applied to two calves, one calf became infected with the trypanosome. The parasites were observed for one day in the enlarged lymph node nearest to the tick-feeding site 5 days after the tick application. Subsequently the trypanosomes were re-isolated in vitro from the infected calf. Inoculation of a ground-up tick supernatant suspension from the infected batch of ticks containing 10(4) trypanosomes into a calf did not produce a patent infection.  相似文献

Bovine babesiosis is responsible for serious economic losses in Uruguay. Haemovaccines play an important role in disease prevention, but concern has been raised about their use. It is feared that the attenuated Babesia bovis and Babesia bigemina vaccine strains may be transmitted by the local tick vector Boophilus microplus, and that reversion to virulence could occur. We therefore investigated the possibility that these strains could be transmitted via the transovarial route in ticks using a Babesia species-specific polymerase chain reaction (PCR) assay. DNA was extracted from the developmental stages of the tick vector that had fed on calves immunized with the haemovaccine. It was possible to detect Babesia DNA not only in adult ticks, but also in their eggs and larvae. In addition, it was shown that calves infested with larvae derived from eggs laid by ticks fed on acutely infected calves, were positive for Babesia using PCR. Caution should therefore be shown with the distribution of the haemovaccine in marginal areas. It is still advisable that suitable tick control measures be used to prevent transovarial transmission and the potential risk of attenuated Babesia reverting to virulence.  相似文献
