Lipids and essential fatty acids, particularly the highly unsaturated fatty acids, 20:5n‐3 (eicosapentaenoic acid; EPA), 22:6n‐3 (docosahexaenoic acid; DHA) and 20:4n‐6 (arachidonic acid, AA) have been shown to be crucial determinants of marine fish reproduction directly affecting fecundity, egg quality, hatching success, larval malformation and pigmentation. In Atlantic cod (Gadus morhua L.) culture, eggs from farmed broodstock can have much lower fertilization and hatching rates than eggs from wild broodstock. The present study aimed to test the hypothesis that potential quality and performance differences between eggs from different cod broodstock would be reflected in differences in lipid and fatty acid composition. Thus eggs were obtained from three broodstock, farmed, wild/fed and wild/unfed, and lipid content, lipid class composition, fatty acid composition and pigment content were determined and related to performance parameters including fertilization rate, symmetry of cell division and survival to hatching. Eggs from farmed broodstock showed significantly lower fertilization rates, cell symmetry and survival to hatching rates than eggs from wild broodstock. There were no differences in total lipid content or the proportions of the major lipid classes between eggs from the different broodstock. However, eggs from farmed broodstock were characterized by having significantly lower levels of some quantitatively minor phospholipid classes, particularly phosphatidylinositol. There were no differences between eggs from farmed and wild broodstock in the proportions of saturated, monounsaturated and total polyunsaturated fatty acids. The DHA content was also similar. However, eggs from farmed broodstock had significantly lower levels of AA, and consequently significantly higher EPA/AA ratios than eggs from wild broodstock. Total pigment and astaxanthin levels were significantly higher in eggs from wild broodstock. Therefore, the levels of AA and phosphatidylinositol, the predominant AA‐containing lipid class, and egg pigment content were positively related to egg quality or performance parameters such as fertilization and hatching success rates, and cell symmetry.  相似文献

The present study investigated the biochemical composition and quality of turbot (Scophthalmus maximus) eggs throughout the reproductive season. Results showed that the fertilization, hatching and egg floating rates were variable throughout the reproductive season, with the highest values recorded during the mid-season. Meanwhile, positive correlations were found between fertilization, hatching rate and floating rate. The composition of turbot eggs, including total lipid, protein, carbohydrate, moisture and dry weight showed no significant differences during the reproductive season. Furthermore, no correlations were found between egg compositions and viability parameters (VPs), including fertilization and hatching rates as well as larval deformity rate. However, egg diameter varied and correlated with fertilization, hatching and egg floating rates. The fatty acid in eggs at mid-season had significantly higher levels of C14:0, C16:0, C16:1n-7, C18:0, C18:1, C20:4n-6, C20:5n-3 and C22:6n-3. Moreover, significant relationships were found between fatty acids and VPs. Eggs of the middle season had significantly higher concentration of isoleucine, leucine, lysine, methionine, valine, alanine, aspartate, glutamate and serine, whereas no significant relationships were found between amino acids and VPs. These observations suggest that the biochemical profile of eggs may be useful in evaluating egg quality and improving broodstock management for turbot.  相似文献

Problems concerning egg viability or egg quality have received increasing attention, in relation to cultivation as well as to the assessment of reproduction of wild fishes. One important aspect of this paper is to discuss the use of cell morphological characteristics as indicators of egg quality in cod. Early cellular malformation rates in egg batches show a positive correlation with their later egg and larval survival, and can be used as a rough prediction of normal development and mortality rates.
The natural mortality rate offish eggs is not well understood, and observed egg viability of captive cod is are discussed in relation to egg viability in the wild. Results from recent work indicated that approximately 10% of naturally spawned cod eggs from both wild fish (planktonic samples) and from a broodstock had abnormal cleavages at the 2–128 cell stage. Eggs with observed early abnormal cell-cleavage showed very poor survival and hatching rates. For broodstock cod, egg viability was highest during the peak of the spawning season.
The North Atlantic cod may provide a good model species for studies of egg viability variations in wild and captive individuals, as its reproductive biology is well studied. It is now documented that the egg production in cod is dependent on fish age, nutritional status, batch number, and stress factors.  相似文献

In Eurasian perch (Perca fluviatilis), the variability in spawning quality is a major limiting factor for successful production, especially when breeders are fed with an artificial diet. The influence of the dietary DHA/EPA/AA ratio on the egg and larval quality and on the fatty acid and lipid class composition of eggs has been investigated in perch broodstock. Two experimental diets (16% lipids) with two different DHA/EPA/AA ratios, D1 (3/2/2) and D2 (23/9/1), were compared with a natural diet consisting of cultured carp juveniles, CC (10/10/1) and with a commercial diet for salmonids, CDS (14/16/1). Percentages of fertilization and hatching were comparable between fish fed D1, D2 and CC, with the highest hatching rate observed for D1 (63.5 ± 3.8%). These diets supported better values than the CDS. Larval survival and TL50 observed after osmotic stress were higher for the D1 group, followed by larvae produced by fish fed D2 and CC. Larvae from fish fed D1, D2 and CC were significantly more robust than larvae from the CDS group. Differences were observed regarding the fatty acid (FA) profile in the eggs, which was related to the dietary FA composition. The results indicate that a ratio of 3/2/2 seemed to be effective for obtaining eggs and larvae of good quality.  相似文献

The present work reports a characterization of mean wet weight and moisture, the lipid class and fatty acid (FA) composition from the total lipids (TL), of both culture and wild eggs of the cuttlefish, Sepia officinalis , throughout the embryonic development. Additionally, reproductive data, such as the number of spawnings, number and mean weight of eggs and duration of spawning period of cultured cuttlefish is provided. Both types of eggs were similar in mean wet weight, moisture content, TL content and lipid composition throughout embryonic development. Females from the cultured group spawned 13 times and laid 8654 eggs in 64 days, with a mean weight of 0.607 ± 0.179 g. A sex ratio of 1.57 (11♀ for 7♂) promoted an individual fecundity of 787 eggs/♀ (the biggest until now on our culture facilities), which might be related to increased bottom areas. The TL increased with day/stage of embryonic development ( P  < 0.05) only in the cultured egg group. However, no differences were found on TL between culture and wild eggs at the same day/stage ( P  > 0.05). Eggs displayed predominant levels of phosphatidylcholine, phosphatidylethanolamine (PE), cholesterol and triacylglycerol at the end of embryonic development. Polar and neutral lipids of both eggs groups remained consistently proportional (∼50% for each lipid fraction) and a significant increase ( P  < 0.05) was observed in phosphatidylserine, PE and free FA throughout the embryonic development. In either egg type and day, 16:0, 18:0, 20:5n-3 and 22:6n-3 accounted for approximately 70 g Kg−1 of all FA and saturated and n-3 totals seemed to have the same proportion in the cuttlefish eggs. The present results suggest that lipids are not used as energetic substrate but as structural components in cuttlefish egg.  相似文献

Mahi‐mahi Coryphaena hippurus is a promising species for aquaculture development and has been used as a model species for oil toxicology and physiology studies. This species has one of the fastest growth rates of any marine teleost and a unique reproductive biology due to its high spawning frequency and reproductive energy allocation. These characteristics lend the species to being an excellent model for understanding broodstock nutrition for other high energetic pelagic species. In this study, egg morphometrics and larval survival were tracked over a 10‐week period from the initial capture of wild mahi‐mahi broodstock. Larval quality from subsequent spawns collected over time was quantified using larval survival activity indices (SAIs) as a metric to assess egg quality. Larval SAIs were maintained and did not significantly decrease (< .05) over the time course of this study. A multiple linear regression based on the elapsed time in captivity of the broodstock, egg diameter and larval SAI at 1 dph provided the most accurate prediction of larval SAI at 3 dph (R2 = 0.996 < .05). There were strong positive correlations with larval SAIs at 1 and 3 dph and the key nutrients: eicosapentaenoic acid (C20: 5n‐3, EPA) and related fatty acid ratios, vitamin E and nearly all amino acids under investigation. This study demonstrated that larval survival was maintained over time due to the supply of these key nutrients in the broodstock diet.  相似文献

Two Swedish landlocked (nonanadromous) salmon (Salmo salar) stocks were investigated with the aim of characterising their egg fatty acid (FA) and carotenoid profiles. Fish from one of the stocks were also cultured over the past few decades as part of the Swedish program for genetic preservation, allowing a comparison between the eggs from females on a diet based on lipids of limnic (natural food chain) origin and eggs from females fed an artificial diet of marine origin. No significant differences in the FA profile of the phospholipid (PL) or triacylglycerol (TAG) fraction were found between the two wild stocks. The content of EPA (20:5n-3, eicosapentaenoic acid) in PL fraction was significantly higher in eggs from cultured females (13.0%) compared with eggs from both wild stocks (5.7 and 6.4%). Further, in PL fraction, AA (20:4n-6, arachidonic acid) levels in these eggs were significantly lower (2.4% versus 6.7 and 6.2%). The AA content of the TAG fraction differed greatly between wild (4.4 and 4.9%) and cultured (1.2%) eggs, whereas this fraction showed almost no corresponding difference in EPA content. The level of DHA (22:6n-3, docosahexaenoic acid) did not differ between the two wild stocks or between wild and cultured fish. This was in spite of widely different levels of DHA in the diet. The composition of carotenoids was altered in the cultured eggs which had a higher proportion and higher content (1.16 g egg–1) of astaxanthin than the wild eggs (0.56 and 0.62 g egg–1, respectively). Hatching success varied markedly between wild (>95%) and cultured fish (40–75%). We conclude that changes in the lipid source in the diet of female salmon during gonadal maturation will alter the egg fatty acid composition with an increased risk of disturbances in embryonic development as a consequence. Further, the lack of any difference between wild and cultured females in terms of their egg DHA content indicates that there is a strong genetic influence on levels of this fatty acid in salmon eggs.  相似文献

Morphometric characteristics, proximal composition and fatty acids profiles of eggs and larvae at hatching and mouth opening were studied as indicators for assessing the potential differences between spawns obtained from wild or captive females of pikeperch (Sander lucioperca). Captive females were acclimated to hatchery conditions and fed to satiation with chopped marine fish for more than a year, guaranteeing that captivity covered the full cycle of gonad maturation. The number of larvae obtained from the spawnings did not differ between captive and wild females, but both mean diameter and mean weight of the eggs collected from the captive females were significantly larger. Proximal compositions were similar in eggs and larvae from both groups, but fatty acids (FA) profiles showed striking differences: polyunsaturated FA consisted mainly of linoleic acid in eggs and larvae from wild females, while they consisted mainly of long chains of n-3 series in eggs and larvae from captive females. The diet of captive females largely modified the FA profiles of eggs and of hatching or mouth opening larvae, but did not affect the growth potential of early larvae, demonstrating that they were able to satisfy their early physiological needs regardless of the striking differences in their FA.  相似文献

Five diets that contained fresh squid meat as the basic constituent and were supplemented with different amounts of highly unsaturated fatty acids (HUFA) and astaxanthin were fed to pond‐reared Penaeus monodon broodstock. Diet A was sole squid meat. Diets B and C were supplemented with astaxanthin 50 and 100 mg kg?1 respectively. Diets D and E were supplemented with HUFA 5 and 10 g kg?1 and astaxanthin 50 mg kg?1 respectively. The result showed that the group fed diet E had the best reproductive performance in all experimental groups. It had a higher proportion of spawns (71.5%), spawning rate (0.047), a shorter latency period (7.7±0.3 d), higher absolute fecundity (× 103) (361.6±5.5) and egg production/female (× 103) (597.0±18.0) than all the other experimental groups. The fatty acid composition in broodstock diets strongly affected the tissue and fecundity of broodstock. Good correlations between the content of 20:4n‐6 in eggs and the fecundity (r2=0.6109) and egg production (r2=0.9876) of broodstock were found. On the other hand, 22:6n‐3 and DHA/EPA ratio was negatively correlated with the fecundity of broodstock (r2=0.5362, 0.8702 respectively). The result also showed that the balance between n‐3 and n‐6 fatty acid families, total polyunsaturated fatty acids and total saturated fatty acid and 20:5n‐3 (EPA) and 22:6n‐3 (DHA) may play vital roles in maturation and reproductive performance of P. monodon broodstock.  相似文献

Despite carotenoids and essential fatty acids seem to play important roles in fish reproduction, no studies have yet been conducted to determine the effect of dietary carotenoids on gilthead seabream broodstock performance and their relation as antioxidants with dietary n‐3 highly unsaturated fatty acid (HUFA) levels. In addition, the high cost of synthetic sources of carotenoids is leading to the search for new natural carotenoid sources such as paprika oleoresin. Four experimental diets containing two combined levels of carotenoids from paprika oleoresin (40 and 60 mg kg?1) and n‐3 HUFA (25 and 40 g kg?1) were respectively fed to triplicate groups of gilthead seabream (Sparus aurata) broodfish. Elevation of n‐3 HUFA dietary levels from 25 to 40 g kg?1 significantly improved gilthead seabream broodstock performance in terms of egg viability, hatching rates and fecundity. Besides, it markedly increased egg contents in HUFAs which play important energetic and structural roles and improve embryo development. Both arachidonic acid and eicosapentaenoic acid (20:5n‐3) egg contents were more readily affected by dietary n‐3 HUFA than docosahexaenoic acid. HUFA levels did not caused any negative effect suggesting an optimized content of antioxidants in broodstock diets. Increase in dietary carotenoids from 40 to 60 mg kg?1 increased carotenoid contents in eggs and significantly improved egg fertilization rates suggesting an important sperm cell’s protective role by reducing the risk of lipid peroxidation which is detrimental for sperm motility. The increased inclusion of dietary paprika oleoresin enhanced egg carotenoid deposition and improved fish reproductive performance, denoting the high nutritional value of this product as a source of carotenoids for broodstock of this species.  相似文献

We investigated the effect of high levels of n−3 highly unsaturated fatty acids (n−3 HUFA) in broodstock diet on egg quality and chemical composition of eggs of Japanese flounder. The broodstock were fed diets containing three levels of n−3 HUFA (2.1%, 4.8% or 6.2%) 2 months before and during the spawning period. No significant difference was found for weight gain of broodstock among the treatments. Egg production was highest in fish fed the highest level of n−3 HUFA. However, egg quality parameters, such as percentage of buoyant eggs, hatching rate and percentage of normal larvae, were significantly higher in the group fed the lowest n−3 HUFA diet. The fatty acid composition of eggs was influenced more markedly in the neutral lipid fraction than in the polar lipid fraction by dietary n−3 HUFA levels. Arachidonic acid (AA; 20:4n−6) and egg quality parameters both decreased with increasing dietary n−3 HUFA levels. The results suggest that a high level of n−3 HUFA in broodstock diet negatively affects egg quality of Japanese flounder.  相似文献

In addition to the strong influence of the broodstock diet on the development and survival of offspring, domestication may also interfere with the larval life success. We obtained eggs from wild and domesticated Salminus hilarii females and domesticated males. Wild females were caught in the Tiete River and tributaries, and the domesticated females were born three years before the beginning of the experiment in the Ponte Nova Fish Farm. Animals from both groups were fed with the same feed to exclude feed variables. The eggs and larvae were sampled at 0, 8, 16, and 28?h after spawning (HAS), with the last sampling (28 HAS) coinciding with hatching time. After hatching, samplings proceeded at 32, 48, 66, and 96 HAS, with the last sampling (96 HAS) corresponding to the end of yolk sac consumption. Finally, the last experimental period was during the larvae exogenous feeding phase, at 102, 118, 166, and 214 HAS. Our data revealed that domestication of S. hilarii females influenced fatty acid (FA) metabolism during embryo and larva development. However, the structure of membrane phospholipid FA remained mostly stable, with changes principally in the neutral fraction. When the external conditions, mainly water and feed quality, remained constant, domestication of S. hilarii females did not significantly affect the structural FA composition but influenced the selectivity of consumption and/or storage of specific FA.  相似文献

At Doi Inthanon Fisheries Research Unit (DIFRU), Thailand (13°N), rainbow trout were exposed to natural (13°N) and artificial (51°N) photoperiods, and natural water (NW) temperatures and cooled water (CW) 8 months before first spawning. In group I (51°N, CW), water temperatures of 18°C were never reached. In group II (51°N, NW) and group III (13°N, NW), the mean water temperatures in May exceeded 20°C, and 19°C in June and July. Eggs from 94% of all females in group I were obtained before January. This percentage diminished to 84% and 68% in groups II and III. The weight of the spawners and the size of the eggs were significantly lower in group III than in the other groups. No significant differences were observed for egg number per kg body weight of spawners between the groups. The mean fertilization rate of eggs was the highest, with 71%, in group I, and the lowest, with 50%, in group II. For hatching rates, on average 27%, 24% and 30% in groups I, II and III, respectively, differences were not significant. In group III, 37% of all batches reached fertilization rates above 80% and 16% of egg batches showed hatching rates of more than 60%.  相似文献

Brood red snapper, Lutjanus campechanus, were captured from the wild and induced to ovulate by human chorionic gonadotropin (hCG) injection; eggs and sperm were manually stripped; and the eggs were artificially fertilized. Eggs were evaluated by the following egg quality parameters: buoyancy, fertilization, egg size, and oil globule size and number. The relationship of these egg quality parameters and brood characteristics (female size, fecundity, time of year, and response time to ovulation) to 36 h posthatch larval survival was considered. Injection of wild‐caught red snapper females with 1100 IU/kg of hCG resulted in 75% of the females ovulating. The average fecundity was 343,377 ± 30,805 eggs/kg, with a mean percent fertilization of 79.0 ± 1.74%. The mean percentage of floating eggs per spawn was 91.8 ± 1.75%. Mean egg diameter for floating eggs was 778.3 ± 2.09 μm, with a mean oil globule diameter of 117.5 ± 1.53 μm. Brood‐related characteristics were a better predictor of larval survival than postovulation egg characteristics. The percentage of floating eggs in a spawn was not correlated to larval survival. Spawns with eggs having a single oil globule had a similar larval survival as those eggs where multiple globules were common. No clear relationships were found for any one factor and larval survival, but rather a combination of factors was more predictive of survival, most notably spawn date, fecundity, and response time following hCG injection.  相似文献

For the first time, a spontaneous spawning of hake was recorded in Spain in April 2009. The spawn was obtained from broodstock kept in captivity for two years at the facilities of the Spanish Institute of Oceanography in Vigo (NW Spain). Eggs were transparent, spherical and had an average diameter of 1.067 ± 0.024 mm; yolk occupied the majority of egg volume. The oil droplet had a diameter of 0.27 ± 0.03 mm. The incubation period of the eggs lasted for 4 days at 14°C and the duration from hatching to the total absorption of the yolk sac was between 5–7 days after hatching, at the same temperature. Newly hatched larvae had an average total length of 3.20 ± 0.13 mm and began feeding 6 days after hatching; a daily growth rate of 0.158 mm day‐1 was observed from hatching to yolk sac consumption. This paper describes the daily evolution of biometric and morphological characteristics of the different stages of embryos and larvae of European hake up to the age of 19 days.  相似文献

Cultivated Atlantic cod (Gadus morhua) entering their first year of gamete maturation were fed diets with different levels of arachidonic acid (ARA) and eicosapentaenoic acid (EPA) for 6.5 months prior to commencement of spawning. Gravid females were stripped three times: at the beginning, peak and end of spawning. Lipid composition and egg and larval quality of 34 family crosses were investigated. Results indicated that ARA uptake into eggs from broodstock diet was highly efficient achieving proportions of ARA up to 84% higher in eggs than in the diet. EPA was 42–76% higher, and DHA was 155–173% higher in eggs than in diets. Cod fed the diet with the lowest EPA/ARA ratio had the greatest egg production. Eggs from fish on a diet with high ARA level had significantly higher fertilization and hatching success than those fed low levels of ARA. This diet produced on average 71 viable eggs g?1 female compared with 32.5 and 4 eggs in diet B and C, respectively. Furthermore, larval survival until 8 days posthatch was higher in diets with lower ARA levels. The combined results showed that ARA dietary supplementation and low EPA/ARA ratio yielded a greater number of viable larvae kg?1 female.  相似文献

The collection of fertilized eggs for mass culture of Pacific bluefin tuna (PBT) relies on the spontaneous spawning of broodstock in captivity, and the spawning season of the broodstock is generally from mid‐May to September. The diameter of fertilized eggs from the broodstock decreases during the spawning season. To investigate the influence of three potential factors, the egg diameter (larger or smaller), the rearing water temperature (25 or 28 C), and the aeration rate during the night (strong or weak), on early survival and on growth of PBT larvae, replicate trials were conducted to assess these three factors until 7 d after hatching (d.a.h.). At 7 d.a.h., survival rates of larvae reared with strong nighttime aeration were found by a three‐way ANOVA to be significantly higher than for larvae reared with weak aeration. Furthermore, growth rates of PBT larvae hatched from larger eggs were significantly faster than those from smaller eggs. However, a significant difference in the survival rate was not detected for the factors, egg diameter and rearing water temperature. The results indicate that rearing with strong nighttime aeration significantly improved the early survival of PBT larvae and the egg diameter (a proxy for egg quality) and influenced growth.  相似文献

A three‐month feeding trial was conducted to investigate the effects of dietary krill meal on the reproductive performance of tongue sole broodstock. Three diets were formulated to contain different levels of krill meal, 0 (Diet K‐0), 10 (K‐100) and 200 (K‐200) g kg–1 dry matter, replacing the corresponding contents of protein and lipid from fishmeal and fish oil. Each diet was assigned to triplicate tanks. Compared with K‐0, both K‐100 and K‐200 increased the egg properties such as buoyant eggs rate, egg diameter, oil droplet diameter and SOD activity of the fertilized egg. However, the maturation rate, serum estradiol content, the relative fecundity and hatching rate were improved only by K‐100. The krill meal supplementation increased the concentrations of astaxanthin and n‐3 fatty acids in eggs, but decreased the n‐6 fatty acid concentrations. Both K‐100 and K‐200 increased the survival activity index, but the larval deformity rate was decreased only by K‐100. At 15 DPH, the larval length and the digestive enzyme activities were significantly higher in group K‐200 compared with groups K‐0 and K‐100. In conclusion, dietary supplementation of krill meal, at the level of 100 or 200 g kg–1 dry matter in exchange of fishmeal and fish oil had the considerable positive effects on C. semilaevis reproduction.  相似文献

采捕山东威海外海黄海海域的太平洋鳕(Gadus macrocephalus)亲鱼进行驯化和培育,在人工条件下成功驯化存活野生亲鱼49尾,经短期促熟培育后,通过人工授精方式获得了多批次受精卵.对胚胎和早期仔鱼发育过程进行了观察,详细描述了从受精卵到早期仔鱼各发育时期的形态特征.结果显示,太平洋鳕成熟卵子为沉性卵,圆球形,卵径为0.9-1.1 mm,无油球.胚胎发育分为5个时期,分别为卵裂期、囊胚期、原肠胚期、神经胚期和器官形成期.在水温为9-10℃、盐度为27-29的海水中孵化,受精卵历时312 h 30 min孵化出膜.初孵仔鱼全长为(3.85±0.12) mm,6日龄仔鱼开口,肛门与外界相通,进入混合营养期.8日龄仔鱼卵黄囊消耗殆尽,开始进入外源性营养阶段.仔鱼开口饵料为轮虫(Rotifer),12日龄开始摄食卤虫(Artemia saline)无节幼体.6日龄仔鱼鳔原基形成,16日龄鳔充气成为亮泡状.12日龄仔鱼形成肠道第1个生理弯曲,22日龄仔鱼第2个肠道生理弯曲形成.研究结果可为太平洋鳕亲鱼驯化培育和苗种培育提供基础资料.  相似文献
