Scanning electron microscopical (SEM) studies on the tegument of the bovid schistosomes, Schistosoma margrebowiei and Schistosoma mattheei have yielded conflicting results; certain authors observed the tubercles on the tegument of these species to be spined, while others reported that they are spineless. The present study indicates that the protrusion of tubercular spines is subject to phenotypic plasticity regulated by external factors such as the identity of the host species and whether or not the schistosome is paired.  相似文献

Fasciola hepatica, the liver fluke, is a highly evolved endo-parasite that uses various mechanisms to evade the host immune system. The immunosuppressive capabilities of the parasite's excretory/secretory products have been well demonstrated by previous independent studies. However, the role of the parasite's tegument in the immune responses remains to be investigated. In this study, the effect of the tegument and other fractions of adult F. hepatica (excretory/secretory, liver fluke homogenate and liver fluke homogenate without tegument) in the activation of naïve macrophages in vitro was investigated using an ovine model. In addition, an immunoproteomic approach was used to investigate the characteristics of humoral antibody responses developed in sheep against the tegument fraction. The results indicated significantly increased arginase expression in macrophages incubated with the tegument and excretory/secretory fractions. Two dimensional gel electrophoresis of the tegument demonstrated approximately 100 protein spots, with only four of these spots were highly reactive with the positive serum as determined by 2-DE immunoblotting. These results give a preliminary indication that the liver fluke tegument may play role in avoiding hosts’ protective immune responses against itself.  相似文献

The teguments of males from 5 populations of S. mattheei, of which 3 were sympatric and 2 allopatric with S. haematobium, were studied by means of scanning electron microscopy (SEM). A certain percentage of the males of each sympatric population bore tubercle spines while the allopatric populations were spineless. 为观察表皮生长因子 ( epidermalgrowth factor,EGF)对成年水牛晶状体上皮细胞增殖作用的影响。将不同浓度 EGF作用于体外培养的成年水牛晶状体上皮细胞 ,采用 MTT法测定细胞的增殖能力。结果 :EGF在浓度为 1ng/ m L和 1 0 ng/ m L作用的前 3 d促增殖作用逐渐加强 ,呈现时间依赖性 ,且在第 3 d达到最大促增殖效果。不同浓度的 EGF对晶状体上皮细胞的增殖作用不同 ,浓度在 1 0 ng/ m L作用 2 4h后即有明显的促增殖作用 ( P <0 .0 5) ,而作用72 h后最低的有效浓度为 1 ng/ m L,而且 2 50ng/ m L EGF有最大促增殖作用。结论 :EGF是诱导成年水牛晶状体上皮细胞增殖的重要因素。  相似文献

To determine whether Schistosoma mattheei is present in the south-western Transvaal, sixty habitats were searched for the intermediate host snail, Bulinus africanus. Ten populations of this snail were located, 2 of which were infected with S. mattheei. Eggs of one of these isolates, originating from a spring in the Mooi River, were examined with an optical microscope. Scanning electron micrographs of the teguments of adult male worms and the terebratorial membranes of miracidia are described. These parasites are morphologically similar to some previously described from another habitat in the same geographical area and both populations can be regarded as typical S. mattheei.  相似文献

The phylogenetic relationships of five isolates of Pasteurella multocida serotype B:2 belonging to buffalo, cattle, pig, sheep and goat were investigated by comparative sequence analysis of 16S rRNA gene. The 1468bp fragment of 16S rRNA gene sequence comparison showed that the isolates of cattle (PM75), pig (PM49) and sheep (PM82) shared 99.9% homology with the buffalo isolate (vaccine strain P52) whereas, the goat isolate (PM86) shared 99.8% homology with the vaccine strain. The 16S rRNA gene sequences of these isolates were also found monophyletic with type B reference strain NCTC 10323 of P. multocida subsp. multocida. The present study indicated the close relationships of haemorrhagic septicaemia causing P. multocida serotype B:2 isolates of buffalo and cattle with other uncommon hosts (pig, sheep and goat).  相似文献

Each year, hundreds of aquatic migratory birds migrate from northern hemisphere to the Mazandaran Province, northern Iran. Little information is available on prevalence and density of schistosomes in water birds in Iran and around the world. The objectives of this study were to determine definitive and intermediate hosts of avian schistosomes as well as to assess human cercarial dermatitis (HCD) in a wildlife refuge in Mazandaran Province. Of 1106 examined people, 589 (53.2%) had maculopapular rashes mainly on feet but also on hand. The majority of cases were adults and local residents. Of 260 ducks, 41 (15.8%) were found to be infected with Trichobilharzia spp. eggs or adult worms. Prevalence was highest in Anas clypeata and Anas platyrhynchos, 79% and 18.9%, respectively. A total of 1.2% snails, examined by both shedding and crushing methods, were infected with furcocercariae belonging to avian schistosomes. The most frequently infected snail was Lymnaea gedrosiana (5.9%). Our results showed that cercarial dermatitis and avian schistosomiasis is a common and yet neglected disease in this area. Anas clypeata played the most important role in exposing snails to miracidia in ponds and paddy fields. Moreover, because of the high prevalence in ducks and high prevalence of HCD in the region, it is considered as a new endemic focus in Iran.  相似文献

Serine proteinase inhibitors (serpins) represent an important superfamily of endogenous inhibitors that regulate proteolytic events active in a variety of physiological functions. Immunological screening of a Schistosoma japonicum adult worm cDNA expression library with sera of Microtus fortis, a naturally resistant vertebrate host, has identified one clone that encoded for a sequence homologous to those of the serpin superfamily. The full-length sequence encoding S. japonicum serpin (Sj serpin) was amplified from adult worm cDNA by using 5'-RACE-PCR and subsequently cloned into the prokaryotic expression vector pET28c. The full-length Sj serpin fusion-protein with his-tag was expressed in E. coli, purified by affinity chromatography and used to immunize New Zealand white rabbits. Sj serpin is located on the tegument in S. japonicum adult worms. C57BL/6 mice immunized with Sj serpin induced the production of high levels of specific IgE and IgG1 subclass antibodies as well as a marked IL-4 response. Lymphocyte surface marker analysis revealed proliferation of CD19 expressing B cells, indicating a predominant Th2-type response to Sj serpin. Immunized mice developed moderate protection against infection of S. japonicum as demonstrated by a 36 and 39% reduction in the recovery of adult worms and eggs, respectively. These data suggested a role for Sj serpin as a vaccine candidate or as a novel target for anti-schistosome drugs.  相似文献

Development of a vaccine against Schistosoma japonicum which can protect both man and the domestic animal zoonotic reservoirs of infection would be an invaluable tool in attempts to control this infection in those areas in which conventional control methods have failed to break transmission. The pig is a natural host of S. japonicum and because of its anatomical and immunological similarities to humans, it is a potentially valuable host for studies on S. japonicum in particular and schistosomes in general. Radiation-attenuated cercariae are highly effective in inducing immunity in experimental schistosomosis and there are promising reports of partial protection against schistosomes with recombinant-derived individual antigens. In the present study we have set out to establish a protocol for inducing protection with gamma-irradiated cercariae in pigs and to assess the protective capacity of recombinant and naked DNA formulations of Sj62, a 62kDa region of S. japonicum myosin. The corresponding S. mansoni version or Sj62, recombinant IrV-5, has previously been implicated in irradiated vaccine immunity in S. mansoni infections and has been shown to induce high levels of immunity in a variety of hosts.Groups of pigs were immunised three times at 2-week intervals with 2000 cercariae irradiated at 20krad, with Sj62 as a recombinant (rSj62) incorporated in Freund's adjuvant, a micellar preparation, or as a naked DNA construct. Vaccination with irradiated cercariae did not induce significant anti-Sj62 antibody but following intramuscular challenge with 2000 cercariae, the vaccinated pigs showed >95% resistance as assessed by reduced faecal egg output, worm tissue egg burdens and also reduced septal fibrosis. Immunisation with each of the Sj62 formulations induced significant anti-Sj62 antibody responses, the highest titre (>12,800) being with the Freund's preparation but none of the Sj62-immunised groups showed significant resistance to challenge. The data suggest that Sj62 shows little promise as a vaccine candidate for schistosomosis.  相似文献

In order to measure different lymphocyte populations in buffalo (Syncerus caffer) and waterbuck (Kobus defassa), we analysed the monoclonal antibodies from the 1st International Workshop on Leukocyte Antigens in Cattle, Sheep and Goats for suitable cross-reactive reagents. Peripheral blood mononuclear cells from three buffalo and three waterbuck were tested with the whole panel of monoclonal antibodies (mAbs) together with some additional antibodies against MHC and Ig. In some clusters almost all antibodies cross-reacted (CD2, CD8), in others almost none cross-reacted (CD4, CD5) and in cluster CD6, mAbs only reacted with buffalo but not waterbuck. Double staining experiments were performed on buffalo PBM with the cross-reacting antibodies, to confirm that they detected similar cell populations as in bovine PBM. This was shown with reagents against CD2, CD4, CD6, CD8, CD11, WC1, WC3 and Ig. The molecular weights of the buffalo antigens correlated well with those of the homologous cattle antigens. In the CD5 cluster, only one mAb reacted with the two wild species, and defined an unusual CD2+ CD5- cell population in buffalo. Also mAbs cross-reacting with buffalo MHC class II detected unusual expression on resting T cells. From the results presented, it is clear that the workshop panel contains mAbs against the most important T and B cell antigens of buffalo and probably waterbuck, which will allow us to compare functional lymphocyte populations in cattle and wild ruminants.  相似文献

对江苏省盱眙县183头水牛外形特征及体尺指标进行了调查和测量,同时随机抽取48头母牛,用垂直平板聚丙烯酰胺凝胶电泳(PAGE)和水平淀粉凝胶电泳(SGE)对编码血液蛋白18个基因座的基因频率进行检测,同时引用相同方法所得东南亚21个水牛群体5个座位的研究资料,进行聚类分析,结果表明:成年公牛体高、体长、胸围和管围分别为129.16,147.69,198.66和23.12 cm,成年母牛分别为128.43,143.81,193.11和21.62 cm,估计体重公母牛分别为516.39和479.83 Kg。在所检测18个基因座中,有5个存在多型,多态位点百分比为27.78%。群体内遗传变异水平较低,平均杂合度和平均有效等位基因数分别为0.0793和1.1355。聚类分析表明:盱眙水牛与台湾和泰国水牛亲缘关系较近,孟加拉国为沼泽型水牛和河流型水牛的分界线,不同水牛群体的聚类结果基本与其地理分布吻合。  相似文献

Schistosoma hippopotami were collected from the right heart chambers and pulmonary arteries of Hippopotamus amphibius culled in the Kruger National Park. The schistosomes were subjected to scanning electron microscopy as well as optical microscopy. The results indicate that S. hippopotami is not conspecific to S. mansoni as suggested in the literature. On account of the morphology of certain tegumental structures of both male and female parasites, it is suggested that S. hippopotami is adapted to the pulmonary arterial circulation of its host.  相似文献

This study was aimed to set up a simple, economic and pure method to culture and identify buffalo mammary epithelial cell in vitro, which were collected from the fresh milk. The milk was collected in the late lactation period of the high yield milk buffalo, then it was mixed with the PBS in the ratio of 2:1 and centrifugated. A few suspernatants and the pellets were resuspended with the PBS when the milk fat were decanted, then recntrifuged. The final pellets and 1 mL of suspernatants were seeded in petri dish without fat, respectively. The isolated cells were mammary epithelial cells which were identified by cell morphology, immunofluorescence, growth curve, cell viability and RT-PCR. The buffalo mammary epithelial cells were successfully isolated from both the pellets and supernatants. The cells and impurities were less in the supernatants than that in the pellets. The cells were cobblestone-like without fibroblasts and most in cell division. In the post-confluent culture, mammary epithelial cells formed dome structures and layer-separated growth. The cells expressed cytokeratin 18 was identified by immunofluorescence which was the marker of mammary cells. The cell growth curve and cell survival rate were measured. The results showed that the buffalo mammary epithelial cells were activity and easy to attach. The mammary epithelial cells were expressed of milk protein and milk fat related genes detected by RT-PCR. The buffalo mammary epithelial cell line were successfully isolated and identified from fresh milk, which estabilished foundation for the study of the mechanism of lactation, the amelioration of the quality of milk and the improvement of milk yield of buffalo.  相似文献

Monoclonal antibody-based competitive ELISA (C-ELISA) has been used for the specific measurement of antibodies to peste des petits ruminants (PPR) viruses in sheep, goats, cattle and Buffalo. Serum samples from sheep (n = 232), goats (n = 428), cattle (n = 43), buffalo (n = 89) were tested. The animals had not been vaccinated against rinderpest or PPR. Findings suggested that the sero-positive cases were significantly higher in sheep (51.29%) than in goats (39.02%) (P = 0.002). The overall sero-prevalence of PPRV in small ruminants was 43.33%. The PPR antibodies seroprevalence was 67.42% in buffalo and 41.86% in cattle which was significantly higher in buffalo (P = 0.005). The overall sero-prevalence of PPRV in large ruminants was 59.09%. Cattle and buffalo sera showed a high prevalence of antibody against PPR virus which may explain the difficulty experienced in achieving high post-vaccination immunity levels against rinderpest. Because antibodies against PPR virus are both cross-neutralizing and cross-protective against rinderpest virus, further vaccination in the presence of antibodies against PPR virus may be a waste of national resources. It was also suggested that antibodies to PPR virus could prevent an immune response to the rinderpest vaccine. This paper presents serological evidence for the transmission of PPR virus from sheep and goats to cattle and buffalo and highlights the need to include PPR serology in the sero-monitoring programme to give a better indication of national herd immunity of sheep and goats against PPR.  相似文献

试验旨在从水牛(Bubalus bubalis)乳汁中分离培养水牛乳腺上皮细胞并对其进行鉴定,建立经济、便捷的水牛乳腺上皮细胞分离方法。通过无菌收集高产奶水牛泌乳后期乳汁,将其与PBS按2:1(V/V)混匀离心;去除上层乳脂,取带少量上清的沉淀,与PBS按上述比例混匀再次离心;将脂肪层去除干净,取沉淀上方1 mL上清及沉淀分别接种于细胞培养皿。通过细胞形态、免疫荧光、生长曲线测定、细胞接种活力、RT-PCR等对分离的细胞进行鉴定。结果表明,试验成功从水牛乳汁的上清和沉淀中分离获得了水牛乳腺上皮细胞,上清部分细胞和杂质较少;而沉淀部分则细胞和杂质较多。细胞呈典型鹅卵石样,无成纤维细胞,细胞活力好,多处于分裂期。当细胞增殖高度融合时,出现乳头状结构和分层生长现象。经免疫荧光鉴定,分离获得的水牛乳腺上皮细胞表达上皮细胞标志蛋白--细胞角蛋白18。水牛乳腺上皮细胞的生长曲线呈S型,符合一般生物学规律。细胞接种存活率的测定结果显示,细胞活力好、贴壁能力强。经RT-PCR检测,水牛乳腺上皮细胞表达乳蛋白及乳脂合成相关基因。从乳汁中分离水牛乳腺上皮细胞并对其进行鉴定,成功建立了水牛乳汁分离乳腺上皮细胞的方法,为研究水牛泌乳机制、改善乳品质、提高产奶量奠定基础。  相似文献

Components of three cytoskeletal elements, namely, microtubule, intermediate and actin filaments have been localised in the tegument of the 3-week-old juvenile and adult Fasciola gigantica by means of immunofluorescence and immunoperoxidase techniques, using mouse monoclonal anti-alpha-tubulin, anti-cytokeratin antibodies and biotinylated-phalloidin, respectively. The immunostaining with the above probes were also performed in adult Schistosoma mansoni for comparison. The presence of tubulin, indicative of microtubules, was demonstrated in the tegumental cell bodies, their cytoplasmic processes, and the basal layer of the tegumental syncytium of F. gigantica. While in S. mansoni, tubulin appeared as vertical lines stretching across the whole thickness of the syncytium. Cytokeratin, representing one type of intermediate filaments, was detected in the tegumental cell bodies, their cytoplasmic processes, tegumental syncytium and spines of F. gigantica. In contrast, cytokeratin was evident only in the syncytium of S. mansoni, but not in the spines. Phalloidin, which could bind to actin, a subunit of microfilament, was detected in the tegumental cell bodies, their processes, and the microtrabecular network which form the scaffold of the tegumental syncytium of F. gigantica. In S. mansoni, actin was localized in similar tissues except the syncytium was not stained while spines exhibited intense staining. In F. gigantica, the presence of microtubules and actin filaments in the tegumental cells, their processes and in the syncytium could mediate the movement of secretory granules from the cell bodies towards the basal as well as the apical layer of the tegument. Cytokeratin filaments may serve to reinforce the integrity of the tegumental syncytium as well as the spines.  相似文献

[目的]探索摩拉水牛改良滇东南水牛的杂交效果。[方法]采用直肠把握输精法,用摩拉水牛冻精冷配滇东南水牛。[结果]表明,摩本杂F1代成年公牛的各项体尺体重指标与滇东南水牛的差别不大,而摩本杂F1代成年母牛的体重、体高、体斜长、十字部高、髋宽、胸围、管围等比滇东南水牛有不同程度的提高(P0.05或P0.01),而其他体尺二者间差别不大(P0.05)。至一岁前的摩本杂F1代生长速度较快,1岁公母牛的体重分别为274.08±36.40kg和278.25±55.05kg,较国内其他地方水牛品种的杂交一代高。摩本杂F1代成年公母牛的体重分别为474.54±63.00kg和459.39±68.12kg,摩本杂F1代母牛的体格得到明显的改善,体重增大。[结论]用摩拉水牛与滇东南水牛杂交,可以提高滇东南水牛摩本杂F1代的体格、早期生长速度、生长性能及泌乳性能。  相似文献

Humoral immune response of water buffalo naturally infected with Toxocara vitulorum was monitored using three different antigens of this parasite in serum and colostrum of buffalo cows and calves. Soluble extract (Ex) and excretory/secretory (ES) larval antigens and perienteric fluid antigen (Pe) of adult T. vitulorum were used to measure the antibody levels by an indirect ELISA. Serum of 7-12 buffalo cows for the first 365 days and colostrum of the same number of buffalo cows for the first 60 days of parturition, and serum of 8-10 buffalo calves for the first 365 days after birth were assayed. The ELISA detected antibodies against all three T. vitulorum antigens in the colostrum and serum of 100% of buffalo cows and calves examined. The highest antibody levels against Ex, ES and Pe antigens were detected in the buffalo cow sera during the perinatal period and were maintained at high levels through 300 days after parturition. On the other hand, colostrum antibody concentrations of all three antigens were highest on the first day post-parturition, but decreased sharply during the first 15 days. Concomitantly to the monitoring of immune response, the parasitic status of the calves was also evaluated. In calves, antibodies passively acquired were at the highest concentrations 24 h after birth and remained at high levels until 45 days coincidentally with the peak of T. vitulorum infection. The rejection of the worms by the calves occurred simultaneously with the decline of antibody levels, which reached their lowest levels between 76 and 150 days. Thereafter, probably because of the presence of adults/larvae stimulation, the calves acquired active immunity and the antibodies started to increase slightly in the serum and plateaued between the days 211 and 365. All three antigens were detected by the serum antibodies of buffalo calves; however, the concentration of anti-Pe antibody was higher than anti-EX and anti-ES, particularly after 90 days of age. By conclusion, the buffalo cows develop immunity and keep high levels of antibodies against T. vitulorum-Ex, ES and Pe antigens and these antibodies are transferred to their calves through the colostrum. This passively acquired immunity does not protect the calves against the acquisition of the infection, but these antibodies, passively or actively acquired, may have an important role during worm rejection by the calves and prevention of intestinal reinfection.  相似文献
