猪圆环病毒2型ORF2重组腺病毒的构建与鉴定及其免疫效果研究 总被引:5,自引:2,他引:3
为了研究含有猪圆环病毒2型(porcine circovirus type 2,PCV2)的重组腺病毒作为基因工程疫苗的潜在应用价值,本试验根据GenBank中猪圆环病毒2型基因序列,设计了1对引物,用于猪圆环病毒2型ORF2基因的扩增。将目的基因T/A克隆后,亚克隆至腺病毒转移载体pShuttle-CMV,构建重组穿梭质粒pShuttle-CMV-ORF2。经PCR方法和限制性内切酶酶切法及测序证明该基因已成功连接后,重组腺病毒转移载体经PmeⅠ酶切线性化,在BJ5183细菌中与腺病毒骨架载体pAdEasy-1同源重组获得重组腺病毒质粒pAd-CMV-ORF2。经PCR方法和PacⅠ酶切方法及测序鉴定表明该重组腺病毒载体已构建成功。PacⅠ酶切线性化pAd-CMV-ORF2,脂质体法转染AD293细胞进行病毒的包装和扩增。PCR及RT-PCR、IFA、Western blotting检测目的基因及其表达。结果表明,本试验成功构建了重组腺病毒pAd-CMV-ORF2。3次噬斑试验纯化重组腺病毒,测得其TCID50为10-8.75/0.1 mL。将重组腺病毒接种SPF级雌性BALB/c小鼠,用ELISA抗体检测试剂盒检测特异性抗体水平。小鼠免疫试验测得其特异性抗体水平较高,为PCV2重组腺病毒基因工程疫苗的进一步研究打下基础。 相似文献

Rabies pre-immunization has been recommended for high risk professions, including veterinarians. Cell-cultured rabies vaccines have considerably reduced the risk of post-vaccination neurological reactions found in earlier vaccines. However, some adverse reactions have been reported with Human Diploid Cell Vaccines. 329 French veterinary students were surveyed about their rabies and brucellosis vaccination status, the occurrence of adverse reactions to rabies vaccine, and their antibody titer monitoring practices. Questions also were asked to determine if mandatory rabies pre-exposure immunization upon entry to veterinary school motivated students to maintain their rabies pre-exposure vaccination. The overall vaccination rate was 98.5% for rabies and 17% for brucellosis. 19% of the rabies vaccinated students reported some form of adverse reaction, whatever the vaccine brand used, but not experienced systemic allergic reaction. Adverse reactions were twice more frequent in female than male students and were more frequent after primary series than revaccination series (Relative Risk = 1.76). Despite the mild reactions encountered, rabies pre-exposure vaccination has been well-accepted by French veterinary students. Rabies is a neglected disease with an estimated annual mortality of 55,000 human deaths, affecting mainly low-income countries. Over 95% of these cases result from virus transmission through the bite of infected dogs and for this reason there is a real need for a cheap and effective rabies veterinary vaccine to be used in mass vaccination campaigns. In this work, we describe the establishment of a simple platform for the production of a virus-like particles based rabies vaccine using mammalian cells and roller bottles as culture system. Adherent cells were cultured during more than 15 days and VLPs were continuously produced and secreted to the culture supernatant. Immunogenicity and protective efficacy of VLPs were tested through rabies virus neutralizing antibody test and NIH potency test. These viral particles induced high titer of long lasting neutralizing antibodies and protected mice against active virus challenge. Therefore, this development represents a promising platform for the production of a new generation and virus-free rabies vaccine candidate for veterinary applications. 相似文献

Rabies is an incurable zoonotic disease caused by rabies virus, a member of the rhabdovirus family. It is transmitted through the bite of an infected animal. Control methods, including oral rabies vaccination (ORV) programs, have led to a reduction in the spread and prevalence of the disease in wildlife. This study evaluated the stability of RABORAL, a recombinant vaccinia virus vaccine that is used in oral rabies vaccination programs. The vaccine was studied in various field microenvironments in order to describe its viability and facilitate effective baiting strategies. Field microenvironments influenced the stability of this vaccine in this study. This study emphasizes the importance of understanding how vaccines perform under varying field conditions in order to plan effective baiting strategies. 相似文献

为快速检测犬狂犬病毒IgG抗体,应用狂犬病毒G蛋白作为捕获抗原,鼠抗狂犬病毒G蛋白单克隆抗体和羊抗犬IgG分别作为胶体金标记物和诊断抗体,羊抗鼠IgG作为质控线,制备胶体金免疫层析抗体检测试纸。该试纸卡具有良好的特异性、敏感性、重复性和稳定性,与快速荧光灶抑制实验检测值对比总符合率为82.1%。研究显示,本试纸卡能够快速、准确检测犬狂犬病毒IgG抗体,可为临床诊断和宠物犬狂犬疫苗免疫效果的评价提供参考。 相似文献

Rabies virus and canine distemper virus were grown simultaneously, and possibly symbiotically, in the same chick embryos. There seemed to be no adverse effect on either virus when cultured in such manner.
Bivalent vaccines for rabies and canine distemper were produced. The potencies and the virus titers of such vaccines were comparable to those of rabies vaccine and canine distemper vaccine produced separately.

狂犬病是由狂犬病病毒引起的以中枢神经系统感染为特征的一种人兽共患病。狂犬病病毒具有嗜神经性,能在人和多种哺乳动物中引起致死性感染,病死率几乎为100%。除常规的临床诊断外,狂犬病的确诊依赖于实验室诊断方法,方法主要有病原学诊断、血清学诊断和分子生物学诊断方法等。狂犬病作为一种人兽共患烈性传染病,呈全球分布的态势,中国每年因狂犬病病毒感染而导致死亡的人数居全球第2位。因此,加强对该病的病原学、流行病学特征的认识、加快实验室诊断方法的研究对其综合防控工作具有十分重要的意义。作者主要介绍了目前国内外狂犬病流行病学特征和诊断方法的研究进展,并比较了各种方法的优点和缺点,为狂犬病的临床快速诊断和深入研究提供科学依据。 相似文献
