本研究着重对公兔精液冷冻保存所用的防冻剂的选择及其用量、精液的稀释方法、平衡时间、降温速度及解冻温度等方面进行了一系列试验研究,最后进行输精试验,以验证试验效果。公兔采精用假阴道法,采精后立即检查精液一般性状,精子活率达0.7以上始供试验用。稀释液用 Tris 缓冲液为基础液,其成份为:Tris2.52克,葡萄糖1.05克,柠檬酸1.41克,蒸馏水84毫升,卵黄16毫升。稀释液中的防冻剂以 DMSO 和甘油并用效果较好。当 DMSO 最终浓度从0增至9%时,解冻后精子活率亦随之而提高;当甘油最终浓度为1.25、2.5和3.75%时,解冻后精子活率较好,三者间无明显差异(P>0.05),最终浓度为0和5%时,冻后精子活率则比以上三者有明显的下降(P<0.05)。解冻后精子顶体完好率则随着 DMSO 或甘油浓度的增加而下降。以冻后精子活率和顶体完好率这两个因素综合衡量,DMSO 和甘油的最终浓度分别以4.5%和1.25%效果较好。稀释精液时,采用两次稀释法的冷冻效果比一次稀释法的好。但两者差异不显著(P>0.05)。平衡时间以0.5—1小时效果较好,而2小时和3小时的效果较差。冷冻精液时,在-25—-50℃间的降温速度以每分钟下降3—8℃效果较好,如超过11℃,解冻后精子活率则有下降的趋势。颗粒冻精在40—50℃进行干解冻的效果比30℃和60℃的好。输精时,母兔先静注 HCG50—100单位以诱发排卵,输精量为0.5—0.8毫升。输精母兔230头,情期受胎率73.9%,平均每窝产仔数4.4头。  相似文献

本研究开始于1978年11月,结束于1980年12月。在精液制冻方面着重研究颗粒精液剂量的大小及平衡时间,以求制冻操作简便,使用方便;在解冻方面着重研究解冻液的配方、解冻温度及方法,以求精液解冻后在室温中能保存较久时间,便于在生产中应用。试验取公猪浓份精液用葡萄糖4克、蔗糖1.2克、新鲜牛奶30毫升、新鲜蛋黄10毫升、甘油3毫升、蒸馏水57毫升等配成的稀释液按1:1比例稀释,在8—10℃平衡,采用“浮盆法”制冻,在液氮中保存。试验表明:颗粒精液每颗剂量1.0毫升,质量并不受影响,这在实际应用时,就较小剂量方便得多。平衡时间以2小时的效果最好,解冻后精子活力0.34±0.049,精子复苏率45.2±6.236%。用葡萄糖30克、柠檬酸钠(二水)1.5克、蒸馏水100毫升、安钠加注射液2.0毫升等配成的解冻液解冻效果最好,解冻后精子活力0.37±0.061,精子复苏率47.2±4.578%,精子顶体完好率73.70%,精子顶体评分0.498;解冻后的精液在20—21℃保存4小时精子活力仍达0.34±0.067,精子顶体完好率51.92%,精子顶体评分0.957。解冻温度50℃较37℃好,解冻方法以先干后湿的效果较好。母猪输精试验先后进行8批,共输精母猪132头,情期受胎率71.21%。平均每窝产仔数10.19头。  相似文献

本研究的目的是比较采用由流式细胞仪分离的牛X和Y精子进行胞质内精子注射(ICS I)后,两种不同激活方法对ICS I卵母细胞的激活效果。ICS I后,卵母细胞用5μm o l/L的Ionom yc in(离子霉素)处理5 m in后,先在化学成分明确培养液(CDM-1)中培养3 h,然后再在含有1.9 mm o l/L 6-DM AP(6二-甲氨基嘌呤)的培养液中培养3 h(Ionom yc in CDM-1 DM AP),或用5μm o l/L的Ionom yc in处理5 m in后,不经过CDM培养直接转入1.9 mm o l/L DM AP的培养液中培养3 h(Ionom yc in DAM P)。结果发现,两种激活处理的效果差异不显著。  相似文献

栉江珧(Atrina pectinata)和近江牡蛎(Crassostrea rivularis)是我国重要的海产经济贝类,建立其精子冷冻保存技术及精子冷冻库,对种质资源的保护和遗传育种具有重要意义.本研究对其精子冷冻保存过程中冷冻剂种类和浓度的选择、冷冻程序、平衡时间、保存体积、精液稀释方式、保存时间等多个影响因素进行了比较筛选,并利用显微镜直接检测和曙红Y(eosin Y)染色检测结合的方式检验精子存活率.结果表明,二甲亚砜(dimethyl sulfoxide,DMSO)作为抗冻剂的保存效果要明显优于甘油(glycerol,Gly),终浓度为8%~10%的二甲亚砜保存效果最好,其栉江珧和近江牡蛎精子活率均可达60%以上,而甘油保护下精子活率小于40%.精液的稀释方式采用将抗冻液等分为3份,依次与精液混合,每次间隔8 min,整个过程约30 min;此种精液稀释方式与一次性混合方式相比,精子活率可提高约30%.稀释后精液需要在4℃下平衡15~25 min,平衡样品与直接冷冻样品相比,精子活率提高约50%.精液降温程序为在样品投入液氮前,于液氮面以上15 cm处停留5 min,5 cm处停留10 min,精子活率可达60%以上;而未经此降温程序直接投入液氮中的样品精子活率接近0.与此降温程序相对应的样品冷冻体积在1.0~1.4 mL范围内为宜.冷冻180 d内复苏的精子活率由90%左右(鲜精)下降至60%左右,180 d后精子活率下降至56%左右.研究结果为栉江珧和近江牡蛎精子超低温冷冻保存、双壳类种质保护和遗传育种提供了理论和技术依据.  相似文献

研究旨在建立优化的猪精液冷冻保存方法.实验表明,采用ZORLESCO(ZO)液对猪精液进行预稀释,室温平衡1 h.加入冷冻Ⅰ液后于5℃水浴平衡1.5 h,再加入冷冻Ⅱ液平衡2 h后装入0.25 mL细管,液氮面上3cm处平衡10 min投入液氮,采用37℃水浴解冻30 s,可获得最佳冷冻效果.解冻后的精子活力平均为0.58±0.03,质膜完整率为(63.2±1.2)%,顶体完整率为(51.4±2.6)%;精子畸形率最低,仅为(14.0±3.0)%.  相似文献

为了探讨孕酮对绵羊精子体外获能的影响,将绵羊精子分别加到含不同浓度(1、5、10μm o l/L)孕酮的输卵管合成液(SOF)中,作用不同时间后分别取出部分精子样本进行金霉素荧光染色(Ch lortetracycline,CTC),通过精子与CTC结合染色的不同类型来评定孕酮对绵羊精子的影响。结果显示,在240 m in和360 m in时,各实验组中获能精子的百分数均明显高于对照组的(P<0.05),说明一定浓度的孕酮有利于绵羊精子的体外获能。  相似文献

针对星斑川鲽(Platichthys stellatus)养殖群体中雄性个体较雌性小,产精液量低,限制了人工繁殖育苗和杂交育种开展等问题,本研究以性成熟的星斑川鲽为实验材料,对精子稀释液成分、稀释液渗透压、激活海水适宜温度和冷冻精液的适宜稀释比例等进行筛选,并用冷冻精液和鲜精分别与星斑川鲽卵进行受精和孵化实验,实验结果利用SPSS软件中单因素方差分析法和Student-Newman-Keuls进行比较分析。结果表明,利用Ts-2(37.65 g/L蔗糖+10.01 g/L KHCO3+1.21 g/L Tris碱)和SFs-4(60 g/L葡萄糖+10.01 g/L KHCO3+1.21 g/L Tris碱)冷冻保存的精子活力最高,分别为(56.67±5.78)%和(66.67±5.77)%。利用计算机辅助精子分析(computer assisted sperm analysis,CASA)系统对星斑川鲽精子进行分析,测定了用Ts-2和SFs-4保存的冷冻精液中精子运动方式的各项参数。结果显示,测定的各项参数(曲线运动速度(curvilinear velocity,VCL)、直线运动速度(straight line velocity,VSL)、平均鞭打频率(beat cross frequency,BCF)、运动的直线性(linearity,LIN)和运动的前向性(straightness,STR)表明,利用SFs-4保存的精子各项运动参数明显高于Ts-2。利用蔗糖、KHCO3和Tris碱配制不同渗透压稀释液中,在渗透压为319.94 Pa时,精子活力最高,为(61.67±5.00)%,与鲜精相比无显著差异(P0.05)。利用3~18℃海水激活精子,结果显示,当激活海水温度在12℃时精子活力最高,与鲜精相比无显著性差异(P0.05),而其他温度下显著低于鲜精活力(P0.05)。将解冻后的精液稀释10~40倍,与星斑川鲽卵受精,结果显示,在稀释20倍后冷冻精液的受精率最高。本研究利用Ts-2+20%二甲基亚砜(dimethyl sulfoxide,DMSO)和SFs-4+20%DMSO对星斑川鲽精子成功进行冷冻保存,为星斑川鲽精子冷冻保存、种质库的建立和应用提供理论和技术依据。  相似文献

为研制更为简易、高效的保存稀释液配方及操作程序,探讨4°C条件下稀释保存猪精液的可行性,本实验首次采用带精浆直接稀释法对猪精液的低温保存技术进行了研究。结果表明,在4°C条件下,用Ⅱ液(葡萄糖-卵黄-蔗糖)和Ⅳ液(葡萄糖-卵黄-乳糖)保存猪精液,精子的存活时间和活率分别为144 h、0.517和132 h、0.510,而Ⅰ液(葡萄糖-卵黄-蔗糖-柠檬酸钠)和Ⅲ液(葡萄糖-卵黄-乳糖-柠檬酸钠)精子活率保持0.5以上不到24 h。随着保存时间的延长,四种稀释液中保存的精子活率和顶体完整率均逐步下降,其中Ⅱ液和Ⅳ液均分别在48 h和24 h内下降较快,以后则呈缓慢下降趋势。综上所述,在低温条件下,采用Ⅱ液和Ⅳ液稀释保存猪精液,其精子活率能分别在144 h和132 h内保持0.5以上;所研制配方(Ⅱ液和Ⅳ液)组分简单、配制方便;采用带精浆直接稀释法低温保存猪精液可行,且操作程序简易。  相似文献

以药理学的方法研究了铝对根系分泌有机酸的诱导作用及钙离子的调控作用。50μm o l/L A l处理24 h后的黑麦根系分泌物的高效液相色谱(HPLC)分析结果表明,铝诱导黑麦根系分泌柠檬酸(C it)与苹果酸(M a l)。在铝溶液中加入阴离子通道抑制剂尼氟灭酸(NA,1~2μm o l/L)、苯甲酰甲醛(PG,10μm o l/L),有机酸的分泌受到极显著的抑制。C a2 通道抑制剂L a(NO3)3(25μm o l/L)、异搏定(VP,25、50、100μm o l/L)显著或者极显著抑制铝诱导的有机酸分泌,而且在50μm o l/L A l溶液中加入的C a2 专一性螯合剂乙二醇-双-(2-氨基乙基)四乙酸(EGTA,250、500μm o l/L)能显著抑制柠檬酸的分泌。这些结果表明,阴离子通道是铝胁迫下黑麦根系分泌有机酸的重要通道,C a2 可能介导此分泌过程。  相似文献

【目的】以富士(Fuji)、 秦冠(Qinguan)嫁接在平邑甜茶(Malus hupehensis Rehd.)上的当年生盆栽苗为试验材料,采用砂培方法,研究了缺氮胁迫和干旱对富士和秦冠生长情况、 光合参数、 植株各部位氮磷钾含量及氮素利用效率的影响,分析比较了低氮干旱条件下富士和秦冠生长及氮素利用的差异,以期为果树生产高效肥水利用提供理论指导。【方法】试验共设四个处理: 正常氮正常水(ZZ)、 低氮正常水(DZ)、 正常氮干旱(ZG)、 低氮干旱(DG)。氮素和水分均设置两个水平,分别为正常氮(6 mmol/L NO-3-N)、 低氮(0.3 mmol/LNO-3-N)、 正常供水(保持盆中砂子相对含水量为饱和含水量的80%~85%)、 干旱处理(保持盆中砂子相对含水量为饱和含水量的60%~65%)。【结果】富士和秦冠的生物量(茎和叶)、 株高茎粗等生长指标以及光合速率、 气孔导度、 蒸腾速率均为正常氮正常水(ZZ)>低氮正常水(DZ)>正常氮干旱(ZG)>低氮干旱(DG),并且相对应处理下秦冠的以上指标均高于富士;正常供水下,缺氮处理使富士、 秦冠的根冠比比正常氮处理均有所增加,富士提高了2.05%,秦冠提高了22.40%。富士和秦冠的氮、 磷、 钾含量均表现出正常氮正常水(ZZ)>低氮正常水(DZ)>正常氮干旱(ZG)>低氮干旱(DG); 氮、 钾元素含量在植株各部位的分布顺序依次是叶>根>茎,磷元素则是根>叶>茎;光合氮素利用效率(PNUE)和氮素利用效率表现为秦冠处理之间差异极显著,富士处理之间差异不显著;秦冠的PNUE和NUE明显高于富士,在低氮正常水(DZ)处理下,秦冠氮肥利用率比富士高42.07%,在低氮干旱(DG)处理下高64.14%;低氮胁迫下富士和秦冠的NUE显著提高,并且秦冠提高的幅度高于富士。【结论】施用氮肥能够显著提高富士与秦冠的干物质量,同等水肥条件下,秦冠生长优于富士;水分亏缺会减少叶片对氮的吸收,干旱条件下适度增施氮肥,可提高果树的抗旱能力;低氮干旱胁迫下秦冠的生长指标、 光合指标及氮素利用效率指标均优于富士,表现出较强的抗低氮干旱胁迫的能力。  相似文献

Staff members of the Department of Botany of Palacký University in Olomouc and Gene Bank Department – Workplace Olomouc, Research Institute of Crop Production in Prague, Czech Republic, conducted an expedition in seven European countries (Austria, Czech Republic, France, Germany, Italy, the Netherlands, Switzerland) in August/September 1999 to collect wild Lactuca spp. germplasm and study its geographic distribution, ecology and biodiversity. During the mission, more than 600 locations were visited resulting in the collection of 602 seed samples (accessions) of wild Lactuca species and 13 seed samples of related genera (Chondrilla and Mycelis). Lactuca serriola f. serriola, L. serriola f. integrifolia, L. saligna and L. viminea subsp. chondrilliflora were prevalent in southern Europe (Italy, France), however, only L. serriola was common in central and western Europe (Austria, Czech Republic, Germany, Netherlands, Switzerland). The greatest diversity of Lactuca species was found in France, where also the most seed samples (165) were collected. The most characteristic habitats with a high density of Lactuca spp. populations were observed along roads and highways, grassy ditches, ruderal communities, and dust-heaps. Natural infections by powdery mildew (Erysiphe cichoracearum) and downy mildew (Bremia lactucae) on some wild Lactuca spp. were observed. Recent observations concerning the geographic distribution, population structure, habitats, and natural occurrence of diseases of Lactuca spp. are discussed. This assemblage of genetic resources of Lactuca spp. can serve as the basis of future studies of species diversification, spatial population structure, plant microevolution, domestication processes, and genetic variability of host-parasite interactions.  相似文献

Laser-induced breakdown spectroscopy (LIBS) is a new technique for the analysis of plant material. This study investigates the application of LIBS to pasture-based plant samples. The LIBS measurements were obtained from pelletized pasture samples (100 samples) that had been also analyzed by inductively coupled plasma–optical emission spectroscopy (ICP-OES) following microwave digestion for calibration and comparison purposes. Comparisons for elements sodium (Na), potassium (K), magnesium (Mg), calcium (Ca), manganese (Mn), iron (Fe), copper (Cu), zinc (Zn), boron (B), phosphorus (P), and sulfur (S) showed that LIBS could be used for almost all the standard profile total elements with concentrations down to low mg/kg levels (observed error of Na: 0.024 percent, K: 0.18 percent, Mg: 0.016 percent, Ca: 0.073 percent, P: 0.017 percent, Mn: 31 mg/kg, Fe: 150 mg/kg, Zn: 6.6 mg/kg, and B: 1.1 mg/kg). Elemental analysis at less than mg/kg levels was not possible using LIBS. The elements S and Cu were particularly difficult to analyze with reliability using LIBS at the concentration levels found in the plant samples. Replacing microwave digestion and subsequent ICP analysis with a direct analysis of dried plant samples using LIBS has the potential to improve the productivity and reduce the cost of testing.  相似文献

We have studied the effect of anaerobically digested (ADMSW) and composted municipal solid waste (CMSW) on mineralization and foodweb dynamics to verify the hypothesis that ADMSW would immobilize N right after addition to soil in contrast to addition with CMSW. Another hypothesis was that the mesofauna (enchytraeids and microarthropods) would stimulate N release from the decomposer community. We measured excretion of -N and urea-N from the mesofauna and hypothesized that enchytraeids would release urea. ADMSW and CMSW were amended to pots with sandy loam and barley. The pots were divided into treatments with or without mesofauna. Mesofauna, plant N and biomass, soil N and ergosterol (fungal biomass) were measured over a 113-day period of four equidistant samplings. Soil respiration, N mineralization and N release by the mesofauna were modelled from concurrent studies. ADMSW- and CMSW-treated soils initially (<20 days) immobilized N. The amendments did not increase plant growth substantially, and this was probably due to N-limitation in the early stages of plant growth. Enchytraeid abundance was about three times higher in ADMSW- than CMSW-treated soils, indicating that ADMSW contained more labile compounds, bacteria, and microfauna. The mesofauna did not affect N-content, but the cumulated -N excreted by the mesofauna was estimated to be substantial and about one-fifth of total plant N in ADMSW. An explanation for this discrepancy might be that in the absence of the mesofauna, other members of the detrivore and microbivore community performed the mesofauna’s function. Neither enchytraeids nor microarthropods excreted urea.  相似文献

“Wicked” problems are those that are complex and that change when solutions are applied. Many conflicts in conservation fall into this category. The study approached the problem of how to constrain the apparent wickedness of a problem in the conservation management of a species by using simple empirical indicators to carry out iterative assessment of the risk to a population and to document how this risk evolves in relation to the addition of new data and the implementation of management actions. Effects of high levels of uncertainty within data and also concerning population structure were examined through stochastic simulation and by exploration of scenarios. Historical trends in the example used, the Steller sea lion, showed rapid declines in abundance in some regions during the 1980s. The current total population is 130,000-150,000 Steller sea lions through Alaska and British Columbia and this number has been stable since about 1990 in spite of regional differences in population dynamics. Regional differences in the sequence of changes in the number of pups and non-pups, suggested that an internal re-distribution of juveniles could have happened between 1980 and 1990. Current productivity also appears close to the long-term mean. Stochastic population projection using various scenarios showed that, based upon this history, the risk of extinction for the population has declined and is below reasonable thresholds for considering the population to be endangered. The trends in risk suggest that management actions taken since 1990 have probably been effective. Consequently, the conservation management objectives for the Steller sea lion are probably being met. The approach provides a mechanism, based upon experience and scenario analysis, for exploring future policy options and may help to constrain the debate amongst stakeholders about the cost-benefit trade-offs associated with different options.  相似文献

Nutrient distributions under no tillage (NT) compared with conventional disk-and-bed tillage (CT) management in the warm, humid region of the southeastern USA need to be assessed so that future placement, quantity, and type of fertilizers can be altered, if necessary, to efficiently match crop demands. We determined soil-profile distributions of pH, N, P, S, K, Ca, Mg, Na, Zn, Fe, Mn, and Cu to a depth of 0.9 m at the end of 8.5 years of continuous CT and NT management on a Weswood silty clay loam (fine, mixed, thermic Fluventic Ustochrept) in southcentral Texas. Most dramatic changes occurred within the 0–0.05 m depth, where soil under NT had lower pH, Fe, and Cu than under CT, but greater P, K, Zn, and Mn. Greater P and K under NT than under CT also occurred below the till-zone (0.15–0.3 m). At a depth of 0–0.3 m, soil under NT contained greater amounts of extractable P, K, Zn, Fe, Mn, and Cu than under CT. Nitrogen fertilization had little effect on nutrient distributions, except resulting in greater extractable K at 0–0.05 m and greater nitrate at 0–0.15 m. Few changes in soil-profile distributions were observed for extractable S, Ca, Mg, and Na. Long-term continuous use of NT on this fine-textured, high-fertility (except for N) soil had no apparent adverse effects on nutrient distributions relative to CT, but enhanced conservation and availability of P, K, Zn, Fe, Mn, and Cu near the soil surface where crop roots proliferate.  相似文献

The responses of tallgrass prairie plant communities and ecosystem processes to fire and grazing are well characterized. However, responses of invertebrate consumer groups, and particularly soil-dwelling organisms, to these disturbances are not well known. At Konza Prairie Biological Station, we sampled soil macroinvertebrates in 1994 and 1999 as part of a long-term experiment designed to examine the effects and interactions of annual fire, mowing, and fertilization (N and P) on prairie soil communities and processes. For nearly all taxa, in both years, responses were characterized by significant treatment interactions, but some general patterns were evident. Introduced European earthworms (Aporrectodea spp. and Octolasion spp.) were most abundant in plots where fire was excluded, and the proportion of the total earthworm community consisting of introduced earthworms was greater in unburned, unmowed, and fertilized plots. Nymphs of two Cicada genera were collected (Cicadetta spp. and Tibicen spp.). Cicadetta nymphs were more abundant in burned plots, but mowing reduced their abundance. Tibicen nymphs were collected almost exclusively from unburned plots. Treatment effects on herbivorous beetle larvae (Scarabaeidae, Elateridae, and Curculionidae) were variable, but nutrient additions (N or P) usually resulted in greater densities, whereas mowing usually resulted in lower densities. Our results suggest that departures from historical disturbance regimes (i.e. frequent fire and grazing) may render soils more susceptible to increased numbers of European earthworms, and that interactions between fire, aboveground biomass removal, and vegetation responses affect the structure and composition of invertebrate communities in tallgrass prairie soils.  相似文献
