The innervation pattern of vasoactive intestinal polypeptide-immunoreactive (VIP-IR) nerves in the quail internal carotid artery (ICA) and cerebral arterial tree was investigated and compared with that of acetylcholinesterase-positive (AChE-P) nerves. The supply of VIP-IR nerves to the two arterial systems was distinctly richer than that of AChE-P nerves. It was focused mainly on the walls from the distal ICA to the caudal half of the anterior ramus (AR) through the cerebral carotid artery (CCA). Indeed, double staining clearly showed that numerous VIP+/AChE-axons were distributed over these arterial regions where VIP+/AChE+ or AChE+/VIP- axons were sporadic or often lacking. The finding that nerve bundles accompanying the ICA within the carotid canal contained abundant VIP+/AChE- nerve cells suggests that cerebrovascular VIP-IR nerves in the quail have their major source at these neurons and enter the cranial cavity through the CCA. Another significant finding was that a small number of nerve cells, which were mostly stained for AChE alone and occasionally for VIP alone or both, occurred in the major arteries located more rostral than the middle AR. Thus, the quail cerebral arterial tree, at least the rostral segment of the anterior circulation, is multiply innervated by these three distinct categories of the extracranial and intracranial VIP-IR and AChE-P neurons.  相似文献

In the present study, the hemagglutinating activity of 9 reference strains (serovars A-I) of Ornithobacterium rhinotracheale was investigated by using fresh erythrocytes from 15 different species: chicken (broiler, rooster, hen), turkey, pigeon, quail, duck, Harris hawk (Parabuteo unicinctus), house finch (Carpodacus mexicanus), cow, sheep, horse, dog, rabbit, pig, human (groups A, B, AB, and O), and rainbow trout (Oncorhynchus mykiss). All 9 strains agglutinated rabbit erythrocytes. None of the strains was able to agglutinate hen, cow, horse, or rainbow trout erythrocytes. The number of positive reactions among the remaining species varied. Results indicate that the use of rabbit erythrocytes is better suited for testing the hemagglutinating activity of O. rhinotracheale.  相似文献

The pattern of cerebrovascular substance P (SP)- and calcitonin gene-related peptide (CGRP)-immunoreactive (IR) innervation was investigated in the quail. SP- and CGRP-IR nerves were relatively a few in the rostral part of the anterior circulation, and very scanty or lacking in its caudal part and the whole of the posterior circulation. A significant finding was that the anterior circulation in the majority of individuals is furnished with a varying proportion of SP-IR nerves with or without CGRP immunoreactivity. There was a good correlation in the expression of CGRP immunoreactivity between SP-IR cells in the ophthalmic division of the trigeminal ganglion and SP-IR nerves supplying the major cerebral arteries. In the quail, SP- and CGRP-IR fiber bundles are usually present in the internal ethmoidal artery (IEA). From these and other findings, it is most probable that cerebral perivascular SP- and CGRP-IR nerves are mainly derived from the same categories of neurons in the primary sensory ganglion via the IEA. The close association of varicose SP-IR axons to the nerve cells in the pial arteries suggests that these intrinsic neurons may play some vasocontrolling roles through the modulatory effect of their pericellular SP-IR axons.  相似文献

Fertility after insemination of cryopreserved boar semen is currently below that of fresh semen. In an attempt to improve the post-thaw motility and acrosome integrity of boar sperm, semen was frozen using an adapted Westendorf method in which the chicken egg yolk was replaced by either duck or quail egg yolk. The different composition of the yolk types, particularly the amount of cholesterol, fatty acids and phospholipids, were thought to potentially afford a greater level of protection to sperm against damage during freezing and thawing. Sperm frozen in medium containing chicken egg yolk displayed higher motility immediately after thawing, but there was no difference in the motility of sperm frozen with different types of egg yolk 3 or 6 h after thawing and maintenance at 37 degrees C. Sperm frozen in media containing chicken or duck egg yolk had a higher proportion of intact acrosomes immediately after thawing than sperm frozen in medium containing quail egg yolk, but 6 h after thawing and maintenance at 37 degrees C the sperm that had been frozen in medium containing chicken egg yolk had a higher proportion of intact acrosomes than the sperm frozen in media containing duck or quail egg yolk. Analysis of the composition of the different yolk types showed that the basic components of the yolks were similar, but the ratios of fatty acids and phospholipid classes differed. Duck egg yolk had more monounsaturated fatty acids (MUFA) than chicken egg yolk, which had more MUFA than quail egg yolk. Duck egg yolk contained more phosphotidylinositol (PI) than chicken or quail egg yolks and quail egg yolk contained more phosphotidylserine than either chicken or duck egg yolks. The differences in post-thaw motility and acrosome integrity of boar sperm when frozen in media containing the different types of egg yolk may be due to the variation in composition.  相似文献

Development of cerebrovascular noradrenergic (NA) innervation was investigated in the Japanese quail, using histofluorescence technique and quantitative analysis. Cerebral perivascular NA nerves arose from the cerebral carotid (CCA), internal ethmoidal (IEA) and vertebral (VA) arteries before hatching, with the first appearance on the CCA and IEA at embryonic day (E) 8 and of the VA at E14. Nerves arising from the IEA were greater in number and spread more rapidly than those from the CCA and VA. On E16, fluorescent fiber bundles lying on the anterior circulation subdivided rapidly into thin fibers. Consequently, the number of NA nerves over the arterial system increased greatly at hatching, particularly along the distal portion of the anterior ramus. At the same developmental stage, all the major arteries of the anterior and posterior circulation were almost entirely covered by NA nerves. The abrupt reorganization of cerebrovascular NA innervation in newborn quails may be related to some specific trophic and vasomotor roles for structural and functional improvement of the cerebral circulation that is required for its brain differentiation at this critical period of development. The supply of NA nerves to the anterior and posterior circulations sequentially increased during development from posthatching day (P) 1–15. Plexuses of NA nerves in each of the major cerebral arteries at P22 and P42–50 were similar in meshwork construction to each other, and to those seen at P15. Likewise, there was no clear statistical difference in the nerve density of the corresponding arteries among the three posthatching ages, except for the caudal basilar artery.  相似文献

为了探明禽类蓝斑与蓝斑下核至脊髓传导通路的起源的细胞结构,采用HRP微量注射法和微电泳法对北京鸭、麻鸭、鸡、鸽、鹌鹑和鹅6种主要家禽进行了逆行追踪的综合性研究。实验结果发现,在6种动物一侧脊髓的外侧索或背侧索的外侧,于颈中部、颈膨大部或腰膨大部分别引入HRP后,各种动物的同侧蓝斑核内出现了大量的标记细胞;而双侧的蓝斑下核中出现了一些散在性的标记细胞。两核团内的标记细胞胞体呈梭形或多角形。蓝斑核内的  相似文献

从以产蛋下降为主的樱桃谷种鸭以及出现神经症状的雏鸭各分离出1株病毒,分别命名为BZ株和LC株.该2株病毒对SPF鸡胚和健康鸭胚均能产生相同的病变,分离病毒不能凝集鸡、鸭、鹅、鸽等的红细胞,在鸭胚成纤维细胞(DEF)能够产生典型的细胞病变(CPE),电镜下观察到约50 nm的病毒粒子.病理组织学研究表明,二者在临床上均可导致脑组织危害,表现为脑膜水肿、血管充血和皮质层神经胶质细胞增生等.血清学检测表明,分离病毒与禽流感病毒(AIV)、鸭瘟病毒(DEV)、新城疫病毒(NDV)等病原无交叉.生物学特性鉴定该病原为有囊膜单股RNA病毒.利用不同禽病的特异性引物分别进行PCR或RT-PCR,均未扩增出特异条带.设计随机引物进行RT-PCR,扩增出基因片段,利用GenBank进行Blast同源比较,结果发现,分离病毒与以色列火鸡脑膜脑炎病毒(Israelturkey meningo-encephlitis virus,TMEV)和在马来西亚发现的Tembumu病毒至少在2段基因上具有较高的同源性,属于黄病毒属.测序表明,分离病毒与Tembumu病毒的非结构蛋白(NS5基因)和囊膜蛋白(E基因)的核苷酸同源性为86.7%~90.2%和87.0%~91.8%,与TMEV的NS5基因和E基因的同源性为72.4%~73.2%和72.7%~72.8%.2分离株之间E基因和NS5基因的核苷酸同源性均为99.5%.血清中和试验表明,BZ株阳性血清可以中和LC病毒,因此证实二者可能是同一种病毒.综合以上研究,建议将该病命名为"鸭病毒性脑炎"(Duck viral encephalitis disease).  相似文献

Interleukin (IL)-1beta-encoding regions of chicken, duck, goose, turkey and pigeon were cloned and sequenced. Each IL-1beta-encoding region of chicken, duck, goose and turkey is 804 nucleotides long and encodes IL-1beta protein that is 268 amino acids. Pigeon IL-1beta-encoding region is 810 nucleotides long and encodes IL-1beta protein that is 270 amino acids. Two one-nucleotide and one four-nucleotide insertions of pigeon IL-1beta-encoding region sequence were found, resulting in two amino acid insertions in pigeon IL-1beta. Pairwise sequence analysis showed that the sequence identities of IL-1beta-encoding genes ranged from 77% to 99%, which were also found for IL-1beta protein sequence identities, with an average level of both sequence identities of 89%. Phylogenetic analysis indicated that IL-1beta-encoding regions and the encoded proteins of chicken, duck, goose and turkey clustered together and evolved into a distinct phylogenetic lineage from that of pigeon which evolved into a second lineage. The results from the binding reaction of antiserum against each recombinant IL-1beta (r IL-1beta) protein to homologous or heterologous rIL-1beta, the enhancement levels of K60 mRNA expression in rIL-1beta-treated DF-1 cells or the reduction levels of K60 mRNA expression in DF-1 cells treated with rIL-1beta that was preincubated with homologous or heterologous antiserum showed that all five rIL-1beta were functional active and shared significantly structural and functional homology.  相似文献

The objective of the study was to demonstrate and characterize IgG Fc receptors of Mycoplasma synoviae. Two IgG Fc receptors were recognized with molecular weights (MW) of 80,000 and 90,000 and isoelectric focusing points (pI) of 5.3 and 4.3, respectively. The activity of the IgG Fc receptors was eliminated by exposure to 0.1 unit of protease for 10 minutes. Mild reduction in activity was observed with trypsin between 100 to 1000 units for 10 minutes. The IgG Fc receptors were resistant to exposure to 60 C for 60 minutes and to 100 C for 20 minutes. The M. synoviae IgG Fc receptors were strongly reactive to affinity-purified Fc Fragment of chicken IgG; affinity-purified chicken IgG; and serum IgG of chicken, quail, pigeon, and turkey. A moderate reaction was detected to human affinity-purified IgG, and weak reactions were detected to affinity-purified IgG of cat, cow, dog, goat, guinea pig, horse, and rabbit. No reaction occurred with IgG of duck, goose, mouse, pig or rat.  相似文献

用家兔A型魏氏梭菌培养液处理的6株鸡传染性支气管炎病毒,以方阵试验分别与人O型红细胞及羊、猪、兔、鸡、鸭、鹌鹑、麻雀、小鼠等8种动物的红细胞作血凝试验,结果证明,鸡传染性支气管炎病毒H_(120)株和M_(41)株均能凝集人O型以及羊、猪、兔、鸡、鸭、鹌鹑、小鼠的红细胞,而不能凝集麻雀的红细胞;GIBV株和Connecticut株能凝集人O型以及免、鸡、鹌鹑、麻雀的红细胞,而不能凝集猪、羊、鸭、小鼠的红细胞;Gray株能凝集兔、鸡、鹤鹑的红细胞,不能凝集人O型以及猪、羊、鸭、麻雀、小鼠的红细胞;而经家兔A型魏氏梭菌培养液处理的T株和未经处理的6株病毒对人O型以及8种动物的红细胞都没有凝集性。试验还证明,M_(41)株和H_(120)株对人O型及8种动物的血凝活性水平有差异。  相似文献

An isolate of egg drop syndrome-76 virus replicated best in primary chicken embryo liver cells and less well in duck embryo liver cells, duck embryo fibroblast cells and chicken embryo kidney cells. The cytopathic effect in chicken embryo liver cells was marked by the presence of round and refractile cells and detachment of cells from the glass surface. The intranuclear eosinophilic inclusion bodies were observed by 24 to 48 hours after infection. No virus multiplication was observed in primary quail embryo fibroblast cells, chicken embryo fibroblast cells or mammalian cells like Vero, BHK-21 and MDBK. Duck embryos supported the maximum growth of the virus, with allantoic fluid having the highest haemagglutinin titre, followed in order by chorioallantoic membrane, skin and internal organs. Chicken and quail embryos did not support the growth of the virus.  相似文献

根据GenBank发表的鸡刺鼠信号蛋白(ASIP)基因序列的同源保守区域,设计了1对特异性引物。以番鸭、高邮鸭和英系北京鸭(即樱桃谷鸭)毛囊RNA为模板,经扩增、测序及拼接得到的部分mRNA序列均为编码序列,共308bp,对应编码102个氨基酸。与已发表的原鸡和鹌鹑的ASIP对应的mRNA核苷酸序列进行比对,番鸭、高邮鸭和英系北京鸭的同源性均在92.53%以上,相应编码的氨基酸序列同源性均高于92.16%。高邮鸭与英系北京鸭核苷酸,氨基酸序列一致,同源性为100%。半定量PCR结果显示,番鸭ASIP基因表达具有较高的普遍性,在皮肤组织(毛囊和皮肤)和中枢神经系统(下丘脑)及其他非皮肤中均有表达。皮肤和毛囊的表达差异不显著(P>0.05),下丘脑中的ASIP基因相对表达量最高,极显著高于皮肤中ASIP基因的表达量(P<0.01),而与毛囊中的差异不显著(P>0.05)。  相似文献

采用PCR方法扩增了云南矮马和北京油鸡的PrP^C基因,并进行序列测定,结合已发表种属朊病毒基因序列,运用分子生物学软件进行同源性分析。结果表明,云南矮马PrP^C基因与已报道的马PrP^C基因比较,其同源性达99%,氨基酸同源性达100%。与牛PrP^C基因比较,同源性达89%,氨基酸同源性达91%。北京油鸡PrP^C基因与鸡的已知序列比较,同源性达99%;与鸭、鸽、鹌鹑的已知序列比较,同源性94%~97%。其翻译的氨基酸序列与鸡的已知序列比较,同源性达99%,与鸭、鸽、鹌鹑比较,氨基酸同源性88%~99%。从本试验结果来看,云南矮马PrP^C基因与牛的PrP^C基因同源性较远,因此感染牛源朊病毒的风险较小,禽类与哺乳动物PrP^C氨基酸属于2个不同的进化分支,因此哺乳动物海绵状脑病不易传染给禽类或引起朊蛋白构型上的改变。这些结果可为海绵状脑病种间屏障补充详细的数据,也为制定相关预防控制措施提供了一些理论依据。  相似文献

Development of cerebrovascular nitrergic nerves was investigated in the rat, using immunohistochemistry for nitric oxide synthase (NOS) and quantitative analysis. Cerebral perivascular NOS nerves usually appeared on the walls of both the intracranial part of the internal carotid artery (ICA) and the internal ethmoidal arteries (IEA) at birth. NOS nerves via the IEA grew more rapidly than those via the ICA. They extended over all the major arteries located more rostral than the middle part of the basilar arteries during the third postnatal week, while those from the ICA remained limited to the caudal segment of the anterior circulation and to the rostral segment of the posterior circulation throughout development. The appearance of NOS nerves on the vertebrate artery (VA) was not demonstrated before the third postnatal week, being apparently far late in development as compared to that of the same nerve type on the ICA and IEA.  相似文献

Normal tables of chicken embryo development are used to define specific stages of morphogenetic progression from the first cleavage divisions through hatching. Although established for the turkey and Pekin duck, the application of the normal tables of chicken embryo development to other birds of commercial and research importance needs be examined. Chicken, turkey, Japanese quail, and Pekin duck blastoderms from oviductal eggs showed differences in the rate of development that were inversely correlated with egg size. Oviposited eggs from these and additional species (goose, Muscovy and mule ducks, and Guinea fowl) were examined after 24 to 72 h of storage and at 6-h intervals up to 72 h of incubation. There was variation in the developmental stages of the blastoderm at the time of oviposition between and within the species and strains examined. Although it is recognized that the temporal rate of development will differ between different species and strains, the external features of any embryo in any given stage will be nearly identical.  相似文献

The aim of the present study was to examine whether chicken semen can be substituted for quail semen to conduct in vitro experiments on fertilization. Chicken spermatozoa was incubated with the inner perivitelline layer (IPL) isolated from the largest follicle in the quail ovary under in vitro conditions. The perforation of chicken and quail spermatozoa were assessed by counting the number of all visible holes in the pieces of IPL. No difference was found in the number of holes formed by the chicken sperm and quail IPL interaction compared with that between intraspecies. In addition, the number of holes in the IPL was significantly increased with the increase in sperm concentration from 1 × 106 to 8 × 106 sperm/mL (P < 0.05). Interestingly, after treatment of chicken spermatozoa with 2.5 mg/mL of the solubilized quail IPL followed by incubation with the intact chicken IPL, the number of holes in the intact chicken IPL was significantly decreased as compared with that of spermatozoa treated without the solubilized IPL (P < 0.05). This indicates that sperm receptors in the solubilized quail IPL and binding ligands on the chicken spermatozoa would find each other and bind, forming complexes and these complexes blocked the interaction between chicken spermatozoa and intact chicken IPL. These results show that: (i) chicken spermatozoa possess the penetrability into quail IPL; and (ii) a high degree of affinity via the receptor interactions exists between chicken spermatozoa and quail IPL. Therefore, it appears that the substitution of chicken semen for quail semen is possible to use as an in vitro technique to examine the sperm‐IPL interaction during fertilization in quail.  相似文献

从临床表现为产蛋下降、头颈歪斜、软脚为主要特征的发病蛋鸭卵巢、脑浆、肝、脾等组织中分离到1株病毒。分离毒能致死番鸭胚和鸡胚,人工感染产蛋期的蛋鸭能导致体温升高、产蛋急速下降,卵泡出血和萎缩;分离毒不能凝集鸡和鸽红细胞,能被鸡黄病毒高免血清特异中和;用禽黄病毒特异引物扩增为阳性。上述结果证实分离毒是黄病毒科黄病毒属的鸭黄病毒,是导致蛋鸭产蛋下降的病原。  相似文献

From eight falcons and one pigeon which died from NDV over a period of 15 months in Dubai, United Arab Emirates, PMV-1 viruses were isolated on quail embryo cell cultures. The identification of all 9 strains were achieved with the haemagglutination inhibition test against polyclonal chicken PMV-1 antiserum, against mouse monoclonal antibodies as well as with the immunoperoxidase test. Intracerebral pathogenicity index and intravenous pathogenicity index tests were also carried out. Although the virus isolates in this study fell into two distinct groups, the overall clinical symptoms displayed by the falcons tailed to demonstrate any trends or specificity unique to a group. The isolate obtained from a pigeon was similar to the isolates from one group of the falcons and showed no identity with the pigeon variant virus.  相似文献
