Spermiation in 3‐year‐old sterlet, Acipenser ruthenus (L.), males maintained under warm water conditions was induced by intramuscular injection of either (i) (D‐Ala6)‐GnRH‐ProNHEt (Kobarelin); (ii) mammalian GnRH analogue+metoclopramide (Ovopel); or (iii) human chorionic gonadotropin (Biogonadyl). The volume of milt, sperm concentration and motility were measured. A higher percentage of spermiating males was obtained after Kobarelin or Ovopel treatment (81.8% and 77.7% respectively) in comparison with fish treated with Biogonadyl (40.0%). However, only stimulation with Ovopel guaranteed motile spermatozoa in all spermiating males. Moreover, treatment with Ovopel resulted in the highest average milt volume, sperm concentration and motility. The average total number of motile spermatozoa (milt volume×sperm concentration×sperm motility) per individual was 0.99×109, 5.31×109 and 0.02×109 after stimulation with Kobarelin, Ovopel or Biogonadyl respectively. Our data indicate that Ovopel is a good stimulator of spermation in sturgeons. Moreover, the time of sexual maturation could be significantly reduced in sturgeons maintained in cages under warm water conditions.  相似文献

The effectiveness of applying Ovaprim [(D-Arg6, Pro9NEt)-sGnRH + domperidone] and Ovopel [(D-Ala6, Pro9NEt)-mGnRH + metoclopramide] to male barbel Barbus barbus (L.) 6, 12 and 24 h after hormonal stimulation was analyzed. The control group (Control) during each time interval was stimulated with 0.9 % NaCl. Milt was collected from seven fish only once (n = 7) for Ovopel, Ovaprim and Control group in order to determine total volume of milt, volume of milt per kg of body weight, sperm concentration, total sperm production, seminal plasma osmotic pressure, pH of milt and pH of seminal plasma. Woynarovich’s solution (68 mM NaCl + 50 mM urea) with the addition of 0.5 % BSA (pH 7.7; 181 mOsm kg?1) was used as the activating liquid. Selected parameters of sperm motility (MOT %) and progressively motile sperm (%), curvilinear velocity (VCL, μm s?1), straight-line velocity (μm s?1), movement linearity (%), wobbling index (%), amplitude of lateral head displacement (μm) and beat cross frequency (Hz) were determined using the Computer-assisted sperm analysis system. A time of 6 h proved to be too short to obtain milt from barbel following hormonal stimulation with Ovaprim and Ovopel. Extending the time to 12 h, however, resulted in 100 % spermiation in males, regardless of hormonal preparation used for stimulation. The stimulation of spermiation in barbel is best performed using Ovopel 12 h upon application. Extending the latency period to 24 h following the application of this preparation results in a significant decrease in the volume of milt obtained, sperm count and motility parameters, including MOT and VCL, which may influence sperm fertilization ability.  相似文献

In this study, the effect of gonadotropin‐releasing hormone agonist (GnRHa) injection on milt production in spent rainbow trout was investigated. On day 0, 25 newly matured male rainbow trout (Oncorhynchus mykiss) were stripped manually, and sperm quantity (vol: mL fish?1) and quality, spermatocrit (%), sperm count (cell mL?1), motile sperm percentage and motility duration (s) were evaluated. After stripping, fish were randomly divided into five groups: intact; sham (injected with propylene glycol as a hormone vehicle); and groups receiving 4, 8 or 16 μg kg?1 BW of [d ‐Ala6 Des‐Gly10] mGnRHa. On day 7, the fish were stripped again and the same sperm characteristics as on day 0 were measured. At the beginning of the experiment, there were no significant differences in any of the sperm quantity characteristics between groups. On day 7, expressible milt volume was significantly reduced compared with day 0 (P<0.05, t‐test) in the intact and sham groups but milt quality remained the same (P>0.05, t‐test). The present study shows that GnRHa injection with a concentration as low as 4 μg kg?1 BW after first stripping could prevent a significant reduction in milt quantity collected 7 days later without any adverse effects on sperm quality.  相似文献

Kutum Rutilus frisii kutum (Kamenskii, 1901), Cyprinidae is an endemic fish of the Caspian Sea. Iranian Fisheries Organization (Shilat) produce up to 200 million fry (1–2 g body weight (b.w.)) to restock the Caspian Sea population annually. Some of these fry are produced by spawning induction in broodfish by carp pituitary extract (CPE). The objective of this study was to assay the effectiveness of the gonadotropin releasing hormone analogue (d ‐Ala6, Pro9‐Net GnRH) alone or in combination with metoclopramide (MET), a dopamine antagonist, on the percentage of ovulated females, latency period, ovulation index and fertilization success. The following hormone treatments were tested: single injection of 2 mg kg?1 b.w. of CPE as a positive control, GnRHa alone 20 and 40 μg kg?1 b.w. and combination of GnRHa and MET as follows: 5 μg+2.5 mg, 10 μg+ 5 mg and 20 μg+10 mg kg?1 b.w. Negative control group was injected with 0.7% saline. The percentage of ovulated females, ovulation index and fertilization success were 90%, 71.3±1.24%, 68.4±2.3%, respectively, in the group treated with GnRHa+MET at a dose of 20 μg+10 mg kg?1 b.w. and were significantly higher than those in the positive control (60%, 64.5±0.23%, 69.1±4.5%) (P<0.05). However, the latency period in this group was longer than that in the positive control (P<0.05). Only 20% and 40% fish ovulated in groups that received 20 or 40 μg kg?1 b.w. GnRHa. No fish ovulated in the negative control.  相似文献

For the first time was characterized the semen of Pimelodus britskii, hormonally induced and non‐induced induced, during the reproductive period. The experiment 1 was conducted with 12 fish per month, divided into: (i) induced with Carp Pituitary Extract (CPE), and (ii) without hormonal induction (testes macerated). The experiment 2 was conducted, with 30 fish, divided into groups for comparison of different doses of CPE and human Chorionic Gonadotrophin (hCG; T1: control; T2: 3.0 mg kg?1 CPE; T3: 0.5 mg kg + 3.0 mg kg?1 CPE; T4: 0.5 mg kg?1 + 7.0 mg kg?1 CPE; T5: 200 UI kg?1 hCG), the same fish were used every month, and the semen was collected by abdominal massage. In both, experiments were assessed the sperm motility and velocity by means of the CASA software. In experiment 1, the concentration of spermatozoids was significantly increase by application of CPE compared untreated males. The volume and motility decreased gradually until the end of the experiment, the highest values were recorded for treatment induced (0.49 ± 0.25 mL and 62.18, respectively). The same occurred to Gonadosomatic Index, showing the smallest value at the end of the reproductive period. The fish from experiment 2 released reduced volume of watery semen (0.1 mL). The values of sperm concentration, motility velocity decreased gradually throughout the months. For P. britskii, the reproductive period influenced the production and sperm quality. Despite small seminal volume released the dose of 7.5 mg kg?1 of CPE proved effective.  相似文献

To aid in artificial spawning of sciaenid fishes, the present authors developed techniques to collect, handle and cryopreserve sperm from red drum, Sciaenops ocellatus L. Sperm were collected by removing and slicing the testis, and adding Hanks' balanced salt solution (HBSS) or NaCl solution (each at 200-400 mOsm kg?1) as an extender. Sperm were activated with 800 mOsm kg?1 artificial sea water (ASW) to characterize motility. Sperm reached maximum motility (highest percentage motility observed for that sample) within 8 ± 1 s (mean ± SD) and remained at maximum motility for 33 ± 4 s. Sperm were exposed to graded osmotic pressures of ASW (8-800 mOsm kg?1) to determine the range of osmolalities that elicited motility. Threshold activation (defined as ~10% motility) occurred at 351 ± 4 mOsm kg?1 and complete activation occurred at 539 ± 2 mOsm kg?1. Sperm stored at 200 mOsm kg?1 retained motility for up to 13 days. Dimethyl sulphoxide (DMSO) was used as a cryoprotectant at concentrations ranging from 7.5% to 15% (v:v) in HBSS (200 mOsm kg?1). There were no significant differences among post-thaw motilities of sperm cryopreserved at any concentration of DMSO. Sperm thawed on the benchtop at 21°C had lower post-thaw motility than did sperm thawed at 10, 20, 30, 40, 50 or 60°C in a water bath.  相似文献

The effects of an intraperitoneal hormone injection of gonadotropin‐releasing hormone agonist (D‐Ala6, Pro9‐NEt GnRHa) alone or in combination with a dopamine antagonist, domperidone (DOM), on ovulation induction in yellow catfish Pelteobagrus fulvidraco were tested. The hormone treatments were as follows: 6 mg kg−1 body weight (BW) of carp pituitary extract as a positive control, GnRHa 10, 20, 40 and 80 μg kg−1 BW and a combination of GnRHa and DOM as follows: 10 μg+5 mg, 20 μg+10 mg, 40 μg+20 mg and 80 μg+40 mg kg−1 BW. Physiological saline (0.7% NaCl) was used as a negative control. Significant differences in the ovulation ratio, latency period and ovulation index (OI) were observed among treatments (P<0.05). The combination of GnRHa and DOM at doses of 40 μg+20 mg kg−1 BW had higher values of the ovulation ratio and OI, and a shorter latency period compared with other treatments. The highest OI in GnRHa treatments was only 56.67%, suggesting a dopaminergic tone on gonadotropin secretion in this fish at the pre‐ovulatory stage. Therefore, ovulation can be successfully induced in yellow catfish with 40 μg kg−1 GnRHa+20 mg kg−1 DOM without affecting the egg quality.  相似文献

The present study examined the possibility of long‐term storage, by cryopreservation in liquid nitrogen, of the sperm of filefish (Thamnaconus septentrionalis). Changes in motility, survival rate, ultrastructure and fertilization rate of the sperm after freezing and thawing were tested. For selection of the immobilizing solution, artificial seawater (ASW) of 250, 350 and 450 mOsmol kg?1 were tested. Sperm motility was significantly inhibited in 350 mOsmol kg?1 ASW, and restored entirely after 100% ASW (1200 mOsmol kg?1) was added. Two cryoprotectants, dimethyl sulphoxide and glycerol, were employed. The sperm was diluted at the ratio of 1:6 with the extenders, and frozen at a freezing rate of ?40°C min?1 to ?100°C after equilibration for 10 min at room temperature, followed by plunging into liquid nitrogen. The highest post‐thawed sperm motility and survival rate were obtained with 5% glycerol. Afterwards, the effect of different freezing rates was examined using 5% glycerol as a cryoprotectant, and the rate of ?30°C min?1 to ?100°C showed the best result.  相似文献

The effects of dopamine on the secretion of two sturgeon gonadotropins (stGTH I and stGTH II) in sexually mature male white sturgeon (Acipenser transmontanus) were evaluated. In Experiment I, sturgeon were given intraperitoneal injections of physiological saline (PS), dopamine (100 mg kg−1), the gonadotropin releasing hormone analog D-Ala6-des-Gly10-GnRH ethylamide (GnRHA) (10 μg/kg−1), and a combination of GnRHa and dopamine. Fish receiving only GnRHa had significantly higher (p < 0.05) concentrations of plasma stGTH I and stGTH II compared to fish receiving PS, dopamine, or a combination of GnRHa and dopamine. Two hours following its administration, dopamine was effective in decreasing plasma concentrations of both stGTHs that were previously elevated by GnRHa. Dopamine or PS administered by themselves did not alter plasma concentrations of either stGTH. In Experiment II, sturgeon injected intraperitoneally with a combination of GnRHa and pimozide had significantly higher (p < 0.05) concentrations of plasma stGTH I and stGTH II compared to males receiving GnRHa or pimozide alone. While this effect of GnRHAa + pimozide was observed in the spring, no such potentiation was seen in these fish during the summer (Experiment III). These results represent the first evidence of dopaminergic inhibition of GnRH-induced pituitary gonadotropin secretion in Chondrostean fish.  相似文献

Sperm were collected in Florida from wild common snook, Centropomus undecimalis (Bloch), and were shipped to Louisiana State University for analysis and cryopreservation. Threshold activation of sperm (10% motility) occurred at 370 mOsmol kg?1, and complete activation occurred at 680 mOsmol kg?1. These values were significantly different. Sperm samples stored at 1°C in Hanks' balanced salt solution (HBSS) or in 0.6% NaCl solution at 200 mOsmol kg?1 retained motility for as long as 22 days. Mean motility remained above 50% for 9 days for sperm stored in HBSS and for 7 days for sperm stored in NaCl solution. Sperm exposed to 5% dimethyl acetamide (62±10%; mean±SD), 10% dimethyl sulphoxide (DMSO) (39±16%), 5% glycerol (26±5%) or 10% glycerol (6±2%) for 30 min had significantly lower motility than did unexposed sperm (89±9%). When used as a cryoprotectant, samples frozen with 5% or 10% DMSO or 5% methanol had significantly higher post‐thaw motility than did samples frozen with other cryoprotectants. Sperm cryopreserved with 10% DMSO (38±12%) had significantly higher post‐thaw motility than did sperm cryopreserved with 15% DMSO (19±10%) or 20% DMSO (4±4%). There were no significant differences in hatch rates of eggs fertilized with fresh sperm (54±29%) or cryopreserved sperm (41±35%). Survival to first feeding was not different between fish produced with fresh sperm (37±30%; range, 0–86%) or with thawed sperm (24±29%; 0–77%). Transport of sperm to a cryopreservation laboratory and back to a hatchery for thawing and use enabled collaboration between groups with specific expertise and provides a model for the application of cryopreservation by transport of fresh and frozen samples.  相似文献

The effect of two commercial preparations containing different GnRH analogues with dopamine antagonists on quantitative and qualitative parameters of semen from chub Leuciscus cephalus L. collected in artificial conditions were examined. Semen was collected after the application of [(D‐Ala6, Pro9 NEt)‐mGnRH + metoclopramide] (Ovopel, n = 9), [(D‐Arg6, Pro9 Net)‐sGnR + domperidone] (Ovaprim, n = 9) and from the control group (0.9% NaCl, n = 9). Afterwards, semen volume, sperm concentration, total sperm production and semen pH were determined. Osmolality and pH of seminal plasma were also determined. Using the Computer‐assisted sperm analysis system (CASA), selected sperm parameters such as sperm motility (MOT %), progressively motile sperm (PRG, %), curvilinear velocity (VCL, μm s?1), straight‐line velocity (VSL, μm s?1), movement linearity (LIN, %), wobbling index (WOB, %), amplitude of lateral head displacement (ALH, μm) and beat cross frequency (BCF, Hz) were analysed. While Ovopel can also be used to stimulate chub spermation, the application of Ovaprim was much more effective for obtaining higher amounts of semen.  相似文献

The effectiveness of common carp pituitary extract (CPE), luteinizing hormone releasing hormone analogue (LHRH‐A2) injections and LHRH‐A2 implants for spawning induction in female sturgeon, Huso huso was examined. In the first trial, fish were injected with 7% physiological saline (control), 50 mg kg?1 CPE or LHRH‐A2 at 3.5, 7, 8 or 10 μg kg?1. In the second trial, fish were treated with LHRH‐A2 cholesterol pellet implants containing 0, 3.5, 7, 8 and 10 μg kg?1 LHRH‐A2. Ovulated eggs were removed using a minimally invasive surgical technique and were artificially fertilized. Injection of CPE and LHRH‐A2 at doses of 3.5, 7, 8 and 10 μg kg?1 resulted in the number of ovulated fish more than LHRH‐A2 implants (similar doses) or controls, although there was no significant difference at doses of 8 and 10 μg kg?1 (P ≥ 0.05). The latency period of fish receiving CPE and LHRH‐A2 injections was approximately 20 h, which was significantly lower than in fish receiving LHRH‐A2 implants (P ≤ 0.05). Furthermore, there were no significant differences in rates of fertilization or hatching among the progeny produced in any of the treatment groups (P ≥ 0.05). In conclusion, the data from this study could be useful for artificial propagation of not‐fully‐matured females of H. huso at sturgeon hatcheries.  相似文献

The effect of sodium and potassium concentrations as well as optimal pH on the motility of common carp Cyprinus carpio L. sperm during short-term storage in artificial seminal plasma (ASP) was investigated. Sperm was collected from individual males (n?=?5) and each sample diluted tenfold (1:9) in ASP (sperm:extender) containing 2 mM CaCl2, 1 mM Mg2SO4 and 20 mM Tris at pH 8.0 and supplemented by the following concentrations of sodium and potassium (mM/mM): 0/150, 20/130, 40/110, 75/75, 110/40, 130/20 and 150/0. The osmolality of all ASP variants was set at 310 mOsm kg?1. Sperm motility was measured using a CASA system during 72 h of storage. Immediately after dilution, sperm motility was high (90%) both in each variant and in the control group (fresh sperm). After 72-h storage, the highest sperm motility was noted in ASP containing 110 mM NaCl and 40 mM KCl. No differences were found in the motility of samples preserved within the pH range of 7.0–9.0. Our data suggest that for the short-term storage of common carp sperm, whereas the pH of the solution does not play a crucial role, a specific potassium concentration of around 40 mM is required.  相似文献

Milt of the Leuciscus idus L. was collected from five experimental groups, and selected parameters of its quality were analysed for 36 h (group II), 60 h (group III), 84 h (group IV) and 108 h (group V), respectively, after hormonal stimulation with Ovopel (1 granule kg?1 of body weight). The control (group I) fish were not subjected to hormonal stimulation. The highest milt volume was obtained from the fish in group IV (0.70 ± 0.55 mL), where the largest volume of milt expressed per kilogram was also obtained (3.03 ± 1.94 mL kg?1). Significant differences were also found in milt volumes obtained between group I and groups III (P<0.01) and IV (P<0.05). The highest percentage of motile spermatozoa was found in the milt of group IV (59%); significant differences were found between group I and groups II (P<0.01) and III (P<0.001). The value of osmotic pressure of seminal plasma was the highest in group IV (203.19 ± 37.63 mOsm kg?1), and the lowest in group I (118.31 ± 41.13 mOsm kg?1). Parameters determining milt quality and quantity indicate that the period of 60–84 h after hormonal stimulation with Ovopel is optimal for obtaining milt from ide.  相似文献

Changes in semen quality and selected biochemical markers were analyzed during a week of spawning season of common carp Cyprinus carpio L. Semen was obtained twice, on May 30 and on June 7, and each time it was collected 24 h after hormonal stimulation using Ovopel [(d-Ala6, Pro9 NEt)-mGnRH + metoclopramide] in 1 pellet kg?1. The total volume of semen (ml), volume of semen per kg of body weight (ml kg?1 b.w.), sperm concentration (×109 ml?1), total number of sperm per kg of body weight (×109 kg?1 b.w.), pH of semen, pH of seminal plasma, seminal plasma osmotic pressure (mOsm kg?1) and the total protein content in seminal plasma (mg ml?1) were determined. A 10 mM Tris buffer containing 100 mM NaCl with 0.5 % BSA (pH 9.0, osmolality 200 mOsm kg?1) was used to activate sperm. The following computer-assisted sperm analysis (CASA) parameters were determined: percentage of motile sperm (MOT, %), progressively motile sperm (PRG, %), curvilinear velocity (VCL, μm s?1), straight-line velocity (VSL, μm s?1), movement linearity (LIN, %), wobbling index (WOB, %), amplitude of lateral head displacement (ALH, μm) and beat cross-frequency (BCF, Hz). The volume of semen per kg of BW, total number of sperm per kg of BW and semen pH were significantly lower at the second semen sampling compared to the first semen sampling. Volume of semen at the second sampling correlated positively with CASA parameters. A lack of differences among CASA parameters between both collection periods indicates good quality of carp sperm hormonally stimulated with Ovopel twice at a 1-week interval.  相似文献

In spite of interest in the cultivation of the pejerrey fish Odontesthes bonariensis (Cuvier & Valenciennes 1835), there are few studies on subjects required to advance this activity. One of the problems is the synchronization of female and male maturation to provide eggs and sperm for larval production. The low volume of expressible milt, either in wild or culture fish, is a major problem. The aim of this work was to study the effectiveness of the administration of different hormones on sperm production in pejerrey. Milt production was enhanced by the injection of human chorionic gonadotropin (hCG) (16.7‐fold increase, 625 IU kg?1), carp pituitary extracts (13.5‐fold increase, 30 mg kg?1), salmon pituitary extracts (12.8‐fold increase, 30 mg kg?1), salmon‐type gonadotropin‐releasing hormone analogue (GnRH) (16.7‐fold increase, 10 μg kg?1) and mammalian‐type GnRH analogue (10.8‐fold increase, 20 μg kg?1). Sperm concentration, motility and the fertilization rate were not statistically different compared with control groups. It was also demonstrated that sperm could be obtained off‐season. Taken together, hCG is recommended to stimulate pejerrey spermiation because it is effective in low doses is inexpensive and is widely available.  相似文献

This study reports on the spermatological properties, and on the development of a protocol for refrigerator storage (4°C) of Labeo calbasu (Hamilton, 1822) sperm for artificial breeding. Volume, motility, concentration and pH of the freshly collected sperm were 2.21 ± 0.53 (μL g?1 of fish weight) (mean ± SD), 95 ± 1 (%), 1.93 ± 0.44 × 109 (cells mL?1) and 7.56 ± 0.17 respectively. Sperm activation was evaluated at different osmolalities of NaCl solution, and motility ceased completely when osmolality of the extender was ≥287 mOsmol kg?1. Sperm retained motility for 24, 72 and 108 h, after refrigerator storage when sperm were undiluted, suspended in Alsever's solution and suspended in Alsever's solution containing 5% methanol respectively. Fertilization rate of fresh eggs with sperm stored at 4°C in Alsever's solution and Alsever's solution containing 5% methanol was 77% and 60% with a hatching rate of 60% and 43% respectively. The fertilization and hatching success of the stored sperm suggests potential to use refrigeration for transporting genetic material to hatcheries for artificial breeding of L. calbasu in Bangladesh.  相似文献

Sperm motility, pH and osmolality of seminal plasma varied throughout the reproductive season spanning the period from June to September. Initially, sperm motility was low, peaked in July and August and then fell again towards the end of the spawning season. While the pH of seminal plasma increased from pH 7.4 to 7.9 during the period of spermiation, the average seasonal pH (7.78 ± 0.03) remained close to an experimentally determined optimum pH range for ocean pout sperm motility (pH 8–9). Likewise, although the values for seminal plasma osmolality fell during the reproductive season, from 416–339 mmol kg-1, the average osmolality value 356 ± 3 was within the optimum for sperm motility (300–400 mmol kg-1). In comparing fluctuations in sperm motility with the biochemical composition of ocean pout seminal plasma during the spawning season, this analysis showed that increased Mg++ levels were correlated with the summer period of maximum sperm motility. A seasonal decline in Na+ and Cl ion levels was reflected in lower seminal plasma osmolality values.  相似文献

Aquaculture of Spinibarbus denticulatus (Oshima, 1926), a fish species indigenous to North Vietnam and Eastern China, is constrained by lack of fingerlings for stocking ponds and cages. As these fish do not naturally breed in captivity, carp pituitary extract (CPE), luteinizing hormone-releasing hormone analogue (LHRHa) with domperidone (DOM) and human chorionic gonadotropin (HCG) were administered at various doses to induce ovulation. A first set of experiments evaluated the response to LHRHa (30, 40 and 50 μg kg−1) with or without DOM (10 mg kg−1), CPE (20, 30 and 40 mg kg−1) and HCG (3000, 4000 and 5000 IU kg−1). A second set of experiments evaluated the dose response to LHRHa (30, 35, 40 and 50 μg kg−1) primed with 6 mg kg−1 of CPE, and HCG (3000, 3500, 4000, 5000 IU kg−1) primed with 6 mg kg−1 of CPE. The treatment groups were compared with each other and the control (injected with 0.9% saline solution). Only 25% and 50% ovulation resulted when treated with LHRHa at 40 and 50 μg kg−1, whereas 100% ovulation was achieved with an addition of DOM to LHRHa. Both 30 and 40 mg kg−1 CPE induced 100% ovulation. However, HCG (4000 and 5000 IU kg−1) induced ovulation in only 33% of females. When primed with CPE, the minimum dose of LHRHa required was 35 μg kg−1 to achieve 70% ovulation. Priming HCG with CPE also resulted in 100% ovulation, and the minimum effective dose of HCG to induce ovulation was 3500 IU kg−1 with 60% ovulation. Fertilization and hatch rates observed in this study with different hormonal stimulation were high (80–93%). The results indicate that while the use of combined hormone strategy has no apparent advantage over a single hormone strategy, LHRHa+DOM (40 μg kg−1+10.0 mg kg−1) and CPE (30 mg kg−1) are most effective in consistently inducing ovulation and thus can be used for commercial hatchery production of S. denticulatus larvae.  相似文献

Black sea bass Centropristis striata L. are protogynous hermaphrodites that develop and spawn as females before changing sex to male. Since all fish eventually become males, determining the relationship between sperm production, sperm quality and seasonal changes in plasma levels of testosterone (T) and 11‐ketotestosterone (11‐KT) could be useful for identifying appropriate males to maintain as broodstock. Milt and blood samples were collected three times during an 8‐week spawning season. Milt volume (3.5±0.76 mL kg?1), sperm density (3.2 × 108± 0.31 cells mL?1), sperm production [11 × 108±3.4 cells kg?1 body weight (BW)] and sperm motility (80±0.6%) were at their highest during the first sampling interval and coincided with the highest 11‐KT levels (1.0± 0.11 ng mL?1). All of the sperm indices decreased to their lowest levels during the final 3 weeks of the study. Sperm viability was highly correlated (adjusted R2=0.84) with sperm motility. Sperm cryopreserved in modified Mounib's extender (MME) had the highest post‐thaw motility compared with two other extenders. Post‐thaw motility of sperm cryopreserved in MME was not different from fresh after 90 days of storage. There was no difference in fertilization rates between fresh (69±2.4%) and post‐thaw (67±4.1%) sperm samples taken from the same male or among males. These results demonstrate that the quality of black sea bass spermatozoa is higher earlier in the spawning season and that acceptable post‐thaw fertilization rates can be obtained from cryopreserved sperm.  相似文献
