Viral encephalopathy and retinopathy (VER) is one of the most devastating and economically relevant diseases for marine aquaculture. The presence of betanodavirus in freshwater fish is recorded, but very little is known about VER outbreaks in marine species reared in freshwater. Our study investigated the ability of betanodavirus to cause disease in European sea bass, Dicentrarchus labrax, reared at different salinity levels. Fish were challenged with RGNNV or mock infected by bath at different salinity levels (freshwater, 25‰ and 33‰). Fish were checked twice a day and the dead ones were examined by standard virological techniques, by rRT‐PCR and by histochemical and immunohistochemical analyses. All the infected groups showed a significant higher mortality rate than the one of the mock‐infected group. VERv presence was confirmed by rRT‐PCR. Histochemical and immunohistochemical analyses highlighted the typical lesions associated with VER. Our results highlight that salinity does not affect the ability of betanodavirus to induce clinical signs and mortality in European sea bass infected under experimental conditions. These results underline the great adaptation potential of VERv, which in combination with its already known high environmental resistance and broad host range, may explain the diffusion of this disease and the threat posed to aquaculture worldwide.  相似文献

Aquaflor® [50% w w?1 florfenicol (FFC)], is approved for use in freshwater‐reared warmwater finfish which include tilapia Oreochromis spp. in the United States to control mortality from Streptococcus iniae. The depletion of florfenicol amine (FFA), the marker residue of FFC, was evaluated after feeding FFC‐medicated feed to deliver a nominal 20 mg FFC kg?1 BW d?1 dose (1.33× the label use of 15 mg FFC kg?1 BW d?1) to Nile tilapia O. niloticus and hybrid tilapia O. niloticus × O. aureus held in a recirculating aquaculture system (RAS) at production‐scale holding densities. Florfenicol amine concentrations were determined in fillets taken from 10 fish before dosing and from 20 fish at nine time points after dosing (from 1 to 240 h post‐dosing). Water samples were assayed for FFC before, during and after the dosing period. Parameters monitored included daily feed consumption and biofilter function (levels of ammonia, nitrite and nitrate). Mean fillet FFA concentration decreased from 13.77 μg g?1 at 1‐h post dosing to 0.39 μg g?1 at 240‐h post dosing. Water FFC concentration decreased from a maximum of 1400 ng mL?1 at 1 day post‐dosing to 847 ng mL?1 at 240 h post‐dosing. There were no adverse effects noted on fish, feed consumption or biofilter function associated with FFC‐medicated feed administration to tilapia.  相似文献

Phytotherapy can replace antibiotic administration as an alternative to control Aeromonas hydrophila, one of the main bacteria involved in the aetiology of farmed fish diseases. Given that plants of the Lippia spp. genus show biological potential for antimicrobial activity, this study evaluated the chemical composition of essential oils extracted from Lippia alba, Lippia origanoides and Lippia sidoides and their activity against A. hydrophila. The oils were obtained by steam distillation in a Clevenger‐type apparatus and their composition determined by gas chromatography and mass spectrometry (CG/MS). Antibacterial activity was assessed by calculating the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) using broth microdilution method. The main compounds identified were geranial (25.4%) and neral (16.6%) in L. alba oil, carvacrol (40.4%) and p‐cymene (11.4%) in L. origanoides oil and thymol (76.6%) and ortho‐cymene (6.3%) in L. sidoides oil. The three Lippia species showed bacteriostatic and bactericidal action against A. hydrophila, with MICs and MBCs ranging from 1250 to 5000 μg mL?1. Of the species tested, the best performance was obtained with essential oil of L. sidoides.  相似文献

An 8‐week feeding trial was conducted to investigate the optimum dietary protein and lipid levels for growth, feed utilization and body composition of Pseudobagrus ussuriensis fingerlings (initial weight: 3.40 ± 0.01 g). Twelve diets containing four protein levels (350, 400, 450 and 500 g kg?1 crude protein) and three lipid levels (50, 100 and 150 g kg?1 crude lipid) were formulated. Fish were randomly allotted to 36 aquaria (1.0 × 0.5 × 0.8 m) with 25 fish to each glass aquarium. Fish were fed twice daily (08:00 and 16:00) to apparent satiation. The results showed that weight gain and specific growth rate (SGR) decreased with increasing dietary lipid level from 50 to 150 g kg?1 at the same dietary protein level. Fish fed the diets containing 150 g kg?1 lipid exhibited higher feed conversion ratio (< 0.05), lower protein efficiency ratio (PER) and nitrogen retention efficiency (NRE) relative to fish fed the diet containing 50 and 100 g kg?1 lipid. Weight gain and SGR significantly increased with increasing dietary protein from 350 to 450 g kg?1 at the same dietary lipid level, and even a little decline in growth with the further increase in dietary protein to 500 g kg?1. Daily feed intake, NRE and PER were significantly affected by both dietary protein and lipid levels (P < 0.05) and tended to decrease with increasing dietary protein and lipid levels. Whole‐body protein content increased as protein levels increased and lipid levels decreased. Whole‐body lipid and muscle lipid content increased with increasing dietary lipid level, and decreased with increasing dietary protein at each lipid level. There was no significant difference in condition factor and viscerosomatic index among fish fed the diets. Hepatosomatic index was affected by dietary lipid level (P < 0.05), and increased with increasing dietary lipid level at the same protein level. These results suggest that the diet containing 450 g kg?1 protein and 50 g kg?1 lipid with a P/E ratio of 29.1 mg protein kJ?1 is optimal for growth and feed utilization of P. ussuriensis fingerlings under the experimental conditions used in the study.  相似文献

The effect of Bio‐Mos incorporated into feed formulation on the growth, feed utilization, body composition and histological response of sea bass (Dicentrarchus labrax) was investigated. Final fish weight, feed conversion ratio (FCR), specific growth rate (SGR), protein efficiency ratio (PER), protein productive value (PPV) and energy utilization (EU) of sea bass feed Bio‐Mos supplemented diet and the control diet were determined at the end of experiment. Data present mean ± SD from triplicate determination (n = 3) for a 75‐day feeding trial. Different concentrations (0.5, 1.0, 2.0. 3.0 and 4.0 g kg diet?1) of Bio‐Mos supplementation and a control without Bio‐Mos incorporation were fed to the fish, feed in the form of dry diets. Average final weight was significantly higher at 1.0 g kg diet?1 of Bio‐Mos and lowest in control fish group; SGR significantly higher at the 1.0 g kg diet?1 of Bio‐Mos. In addition,, PER, PPV and EU showed the same trend. The chemical body composition, crude protein, dry matter, crude fat and ash were significant (P ≤ 0.01) compared to control groups. We concluded that 1.0 g kg diet?1 of Bio‐Mos improved the growth, feed efficiency, histology intestine and survival of sea bass.  相似文献

Invertebrate meals (e.g. polychaetes and insects) present novel and sustainable high‐quality nutrient sources for use in fish feed formulations. To test this innovative source, an eleven‐week feeding trial was conducted evaluating the effects of replacing the fishmeal (FM) component as an example of a superior protein source (FM CTRL) with ragworm meal (RW, Nereis virens) and/or silkworm pupae (SWP, Bombyx mori) in mirror carp (Cyprinus carpio) diets. Three experimental diets with partial replacement of FM (diets: RW + FM, SWP + FM and RW + SWP + FM) were formulated. All diets were formulated to be iso‐nitrogenous, iso‐lipidic and iso‐energetic. Growth performance and feed utilization indices were assessed, and the feeding trial concluded with the analysis of haematological parameters to provide an indication of carp physiological and health status. Mean weight gain was greatest in mirror carp fed RW + FM (60.83 fish?1 day?1; P < 0.05 vs. all other diets) followed by SWP + FM (40.62 g fish?1 day?1; P < 0.05 vs. all other diets). The least weight gain was achieved in fish fed FM + SWP + RW+ and FM CTRL (34.34 and 33.96 g fish?1 day?1, respectively; not significantly different from each other). Fish fed on RW + FM diet had significantly lower plasma ammonia concentrations than any other dietary groups (= 0.04). Mirror carp fed on SWP + FM diet (111.52 units mL?1) were observed to have a marked enhancement in alternative complement activity than FM CTRL (79.21 units mL?1, = 0.041). Both ragworm and silkworm pupae meal present attractive sustainable functional feed component in carp diets, with benefits on enhancing growth performance and specific physiological parameters.  相似文献

Fusarium oxysporum species complex (FOSC) is a highly diverse fungus. Recently, F. oxysporum infection was identified from zebrafish (Danio rerio) culturing system in Korea. Initially, a rapid whitish smudge was appeared in the water with the fungal blooming on walls of fish tanks. Microscopic studies were conducted on fungal hyphae, colony pigmentation and chlamydospore formation and the presence of macro‐ and microspores confirmed that the isolated fungus as F. oxysporum. Furthermore, isolated F. oxysporum was confirmed by internal transcribed spacer sequencing which matched (100%) to nine F. oxysporum sequences available in GenBank. Experimental hypodermic injection of F. oxysporum into adult zebrafish showed the development of fungal mycelium and pathogenicity similar to signs observed. Histopathologic results revealed a presence of F. oxysporum hyphae in zebrafish muscle. Fusarium oxysporum growth was increased with sea salt in a concentration‐dependent manner. Antifungal susceptibility results revealed that F. oxysporum is resistant to copper sulphate (up to 200 μg mL?1) and sensitive to nystatin (up to 40 μg mL?1). This is the first report of FOSC from zebrafish culture system, suggesting it appears as an emerging pathogen, thus posing a significant risk on zebrafish facilities in the world.  相似文献

The antibacterial effects of organic salts, chemical disinfectants and antibiotics were evaluated on cultures of Aeromonas hydrophila C03, Aeromonas sobria C26, A. sobria C29, Aeromonas caviae C24 and Acinetobacter sp. SH‐94B, the pathogens that cause black disease found in fairy shrimps (Streptocephalus sirindhornae Sanoamuang et al. (2000) and Branchinella thailandensis Sanoamuang, Saengphan & Murugan) of Thailand. The minimal inhibitory concentrations (MICs) of organic salts (sodium chloride and potassium chloride) and antibiotics (oxytetracycline dihydrate, streptomycin sulphate, kanamycin monosulphate, chloramphenicol and ampicillin) were determined using the agar‐dilution method. The effect of chemical disinfectants (sodium hypochlorite and chlorine dioxide) was evaluated by exposing bacteria to different concentrations of these chemicals for different periods of time. Interestingly, all strains were intrinsically resistant to 0.25–3% sodium chloride and potassium chloride. The effect of sodium hypochlorite was greater than that of chlorine dioxide, and 5–20 μg mL?1 of sodium hypochlorite was sufficient to inhibit the growth of these bacteria, but the exposure time varied, depending on the bacterial species. Of the antibiotics tested, chloramphenicol and oxytetracycline dihydrate completely inhibited the selected strains. Chloramphenicol showed the highest antibacterial effect against all pathogenic species – the MIC and minimal bactericidal concentration (MBC) ranged from 0.8 to 3.1 μg mL?1 from 3.1 to 6.25 μg mL?1, respectively. To achieve control of black disease during cultivation of fairy shrimp, data derived from this study can be used as a basis for further toxicity tests in vivo.  相似文献

Juvenile Litopenaeus vannamei farmed at 3.0 psu were fed five diets containing glucose, sucrose, wheat starch, corn starch or potato starch as the carbohydrate (CBH) source. Shrimp were fed for 50 days to explore the effect of dietary CBH source on growth, body composition and ammonia tolerance. The specific growth rate of body length of shrimp fed glucose was the highest and significantly higher than those fed potato starch. The survival rate of shrimp fed glucose was 89.44%, and it was the highest and significantly higher than those fed wheat starch. Whole shrimp body crude protein and lipid of the corn starch group were 140.2 g kg?1 and 10.1 g kg?1 respectively. And they were significantly higher than those fed wheat starch. Shrimp fed potato starch had higher hepatopancreas and muscle glycogen. Shrimp fed sucrose had higher glucose‐6‐phosphate dehydrogenase and lower pyruvate kinase activities (P < 0.05). Besides, shrimp fed starch produced more B cells in hepatopancreas tubules than those fed glucose or sucrose. Shrimp fed different sources of CBH differed in the number of R cells. After 96‐h of ammonia nitrogen challenge, the survival rate of the treatments from high to low in turn was glucose, wheat starch, corn starch, sucrose and potato starch, and no significant differences were observed among all treatments. Based on shrimp growth and the economic problems of practical production, we recommend wheat starch as CBH source in practical diets for L. vannamei farmed at low salinities.  相似文献

To assess the effects of A3α‐peptidoglycan (A3α‐PG) extracted from Bifidobacterium sp. on the immune response and disease resistance of sea cucumber, different concentrations (0, 0.5, 5 and 50 mg mL?1) of A3α‐PG suspensions were used to perform hypodermic injection on Apostichopus japonicus, followed by a Vibrio splendidus challenge. Total coelomocyte count (TCC), phagocytosis activity and activities of four immunological enzymes in both cell‐free coelomic fluid (extra‐cellular, EC) and coelomocyte lysate supernatant (intracellular, IC), including acid phosphatase (ACP), alkaline phosphatase (ALP), superoxide dismutase (SOD) and peroxidase (POD), were measured at 2, 6, 14 and 24 h post injection (hpi). The TCC was not significantly affected (> 0.05) by A3α‐PG, ranging from 1.84 × 106 to 3.53 × 106 cells mL?1. The coelomocyte phagocytosis activity was significantly activated (< 0.05) in all the A3α‐PG treatments, whereas no significant difference was observed between them except 24 hpi (> 0.05). The EC‐ACP activity in the 5.0 mg mL?1 treatment increased significantly (< 0.05) at all sampling times, while the IC‐ACP activity in the 50 mg mL?1 treatment increased significantly (< 0.05) at 2 hpi. Also, the 5.0 mg mL?1 treatment had significant (< 0.05) increase in the EC‐ALP activity within 14 hpi and the EC‐POD activity at 2 hpi, respectively, while significantly (< 0.05) enhanced IC‐ALP and IC‐POD activities were observed in the 50 mg mL?1 treatment within 6 hpi and at 2 hpi, respectively. Only the 5.0 mg mL?1 treatment showed significant (< 0.05) increase in the EC‐SOD activity at 2 hpi and IC‐SOD activity within 14 hpi, respectively. The challenge test showed that the animals treated with 50 mg mL?1 of A3α‐PG had notably lower cumulative mortality after 14 days following V. splendidus exposure. All together, these results suggest that A3α‐PG could positively enhance immune response that effectively promotes the health status of A. japonicus against V. splendidus infection.  相似文献

A 12‐week feeding trial was conducted to estimate the dietary copper requirement of fingerling Channa punctatus. Six casein?gelatin‐based test diets (450 g kg?1 crude protein; 18.81 kJ g?1 gross energy) with graded levels of copper as copper sulphate (3.7, 4.7, 5.7, 6.7, 7.7 and 8.7 mg copper equivalent kg?1 diet) were formulated and fed to triplicate groups of fish (7.25 ± 0.81 cm; 5.21 ± 0.27 g) near to satiation. Fish fed diet with 6.7 mg kg?1 copper had highest absolute weight gain (AWG; 51.63 g fish?1), protein efficiency ratio (PER; 1.42 g fish?1), protein gain (PG; 8.34 g fish?1), haemoglobin (Hb; 9.68 g dL?1), haematocrit (Hct; 31.18%) and RBCs (3.24 × 106 × mm?3). Feed conversion ratio (FCR) was found to be best (1.57) at above level of dietary copper. Whole body copper concentration was found to increase with the increasing levels of dietary copper. Hepatic thiobarbituric acid‐reactive substances concentration was found to decrease with increasing dietary concentrations of copper up to 6.7 mg kg?1 beyond which a reverse trend in this parameter was noted. Broken‐line regression analysis of AWG, FCR and PG concentrations against varying levels of dietary copper yielded the requirement in the range of 6.66–6.78 mg kg?1. Data generated during this study would be useful in formulating copper‐balanced commercial feeds for the intensive culture of this fish.  相似文献

This study presents a simple urease method for analysis of ammonia and urea in freshwater aquaculture systems. Urea is hydrolysed into ammonia using urease followed by analysis of released ammonia using the salicylate‐hypochlorite method. The hydrolysis of urea is performed at room temperature and without addition of a buffer. A number of tests were performed on water samples obtained from a commercial rainbow trout farm to determine the optimal urease concentration and time for complete hydrolysis. One mL of water sample was spiked with 1.3 mL urea at three different concentrations: 50 μg L?1, 100 μg L?1 and 200 μg L?1 urea‐N. In addition, five concentrations of urease were tested, ranging from 0.1 U mL?1 to 4 U mL?1. Samples were hydrolysed for various time periods ranging from 5 to 120 min. A urease concentration of 0.4 UmL?1 and a hydrolysis period of 120 min gave the best results, with 99.6–101% recovery of urea‐N in samples spiked with 100 or 200 μg L?1 urea‐N. The level of accurate quantification of ammonia using the method is 50 μg L?1 NH4+‐N, and the detection level is 5–10 μg L?1 NH4+‐N.  相似文献

The aim of the present work was to obtain the lipid utilization of Octopus vulgaris supplying formulated semi‐moist diets with different contents in cod oil (reduced from water content): 0 g kg?1 (A0, 138 g kg?1 lipids DW; N = 4), 100 g kg?1 (A100, 286 g kg?1 lipids DW; N = 6) and 200 g kg?1 (A200, 388 g kg?1 lipids DW; N = 6). The rest of the ingredients were constant in the three diets: 200 g kg?1 gelatin, 100 g kg?1 egg yolk powder, 150 g kg?1 freeze‐dried Todarodes sagittatus and 50 g kg?1 freeze‐dried Sardinella aurita). Survival was 100% with the three diets. The highest absolute feeding (15.8 ± 1.2 g day?1), growth (9.6 ± 1.4 g day?1; 0.91% BW day?1) and feed efficiency rates (60.3%) were obtained with diet A0. This diet also showed greater retention of lipid and protein than A100 and A200. Protein digestibility was above 95% in all of the diets. Only diet A0 led to a high lipid digestibility coefficient (81.25%), which fell drastically to 12.3% in A200. It was notable the high polar lipid digestibility rates (83–89%) respect to neutral lipids (2–87%) in all diets. The best results were obtained with lipid feeding rates of around 1 g day?1 and a suitable lipid content on 130–140 g kg?1 DW in formulated diets for O. vulgaris.  相似文献

In this study, we examined the effects of 500 IU mL?1 penicillin + 500 μg mL?1 streptomycin sulphate on semen quality indices of endangered caspian brown during 12 days short‐term storage at 4°C. Twenty‐four millilitre semen samples with good quality were considered for the experiment. The semen samples were then stored in the presence and absence of 500 IU mL?1 penicillin + 500 μg mL?1 streptomycin sulphate. The semen quality parameters including percentage and duration of sperm motility were measured 0, 3, 6, 9 and 12 days after storage. In the antibiotic receiving group, the values of percentage and duration of sperm motility reduced 3 and 6 days after storage respectively and reduced to lowest levels at day 12. In the antibiotic‐free group, the duration and percentage of sperm motility decreased significantly after 3 days of storage and reached to lowest values at day 12. Also, percentage and duration of sperm motility in each storage time were significantly higher in the antibiotic receiving group than in the antibiotic‐free group. The overall values of percentage and duration of sperm motility for all storage periods were higher in the antibiotic receiving group than in the antibiotic‐free group. In conclusion, our results demonstrated that 500 IU mL?1 penicillin + 500 μg mL?1 streptomycin sulphate improves the viability of caspian brown trout during short‐term storage.  相似文献

To investigate the effect of dietary sodium chloride (NaCl) on meat quality of white shrimp (Litopenaeus vannamei) reared in low‐salinity (2 g L?1) water, shrimp were distributed into four groups (treatments T‐1, T‐2, T‐3 and control) with three replicates. All shrimps were completely randomised stocked into 12 tanks at an initial density of 40 shrimps per tank. Diets for the control, T‐1, T‐2 and T‐3 groups consisted of the basal diet supplemented with 0 g kg?1, 10 g kg?1, 20 g kg?1 and 40 g kg?1 of NaCl respectively. After 50 days, shrimps in T‐3 showed significantly better (< 0.05) moisture, crude protein and ash than those of the control and T‐1. Higher muscle Na content was observed (< 0.05) in T‐3 than that of the control. Significant increases (< 0.05) in contents of inosinic acid, total free amino acid (TFAA) and essential free amino acid (EFAA) were also found in T‐3. Texture assays showed significant differences (< 0.05) in hardness, adhesiveness and springiness between group T‐3 as compared with those of T‐1 and control. It indicated that dietary supplementation of NaCl appeared to be a promising practice to improve meat quality of white shrimp reared in low‐salinity waters.  相似文献

A 35 days feeding trial was conducted to assess the haemato‐immunological response of Labeo rohita fingerlings fed ethanolic leaf extracts of Psidium guajava and Mangifera indica, and infected with Aeromonas hydrophila. Six iso‐nitrogenous (354.6–361.6 g kg?1) purified diets were prepared with graded level of leaf extracts viz., control (basal feed without any extract); TG‐5 (5 g kg?1 guava extract); TG‐10 (10 g kg?1 guava extract); TM‐5 (5 g kg?1 mango extract); TM‐10 (10 g kg?1 mango extract); and TGM (5 g kg?1 guava extract +5 g kg?1 mango extract). Haematological, immunological, biochemical, along with antioxidant enzyme activities were examined after a 35 day‐feeding trial and following a 7 day challenge with A. hydrophila. The haemoglobin, total leucocyte and erythrocyte counts, respiratory burst activity, lysozyme, total protein, albumin and globulin contents increased significantly (P < 0.05) in leaf extracts fed groups compared with the control in pre‐ and post‐challenge conditions. A significant (P < 0.05) decrease was observed in SOD (superoxide dismutase) and catalase activities of the treatment groups compared with the higher value in control. The trends in mortality indicated that groups of fish showing significantly elevated haemato‐immunological responses had the lowest mortality following challenge with A. hydrophila. The results showed that extracts of P. guajava and M. indica appear to be potential immunostimulant at an inclusion level of 5 g kg?1 in the diet of rohu. But, mixing of both the extract at similar level did not show any synergistic effect, which needs to be tested at its lower level of inclusion.  相似文献

The aim of the present work was to test the capacity of Octopus vulgaris to use carbohydrates supplied in three diets: a diet without added carbohydrates (diet C0: 500 g kg?1 water, 200 g kg?1 gelatine, 100 g kg?1 egg yolk powder, 50 g kg?1 freeze‐dried Sardinella aurita and 150 g kg?1 freeze‐dried Todarodes sagittatus) and two obtained by substituting 50 g kg?1 of T. sagittatus by glucose (diet GLU50) or by starch (diet STA50). The most stable and best‐accepted diet was STA50 (SFR 1.26%BW day?1) although there were no significant differences in the growth rates obtained with the three diets: 10.12 g day?1, 9.37 g day?1 and 11.22 g day?1 for C0, GLU50 and STA50, respectively (P > 0.05). The feed efficiency indices were better for GLU50, of particular note being the protein productive value of 71.88% and a feed conversion ratio lower than 1. Protein and lipid digestibility were similar in all the three diets (96–98% for proteins and 85–94% for lipids), whereas carbohydrate digestibility was higher in GLU50 (98%) than in C0 (84%) and STA50 (0.33%). The content of carbohydrates increased in muscle and the digestive gland as a consequence of the increased carbohydrates intake.  相似文献

This study was designed to evaluate the efficacy of eight sources (designated A–H) of soybean meal (SBM) which included six new non‐genetically modified soya varieties in practical feed formulation for Pacific white shrimp, Litopenaeus vannamei, using both growth and digestibility trials. A soybean meal‐based reference diet was formulated using conventional soybean meal (527 g kg?1 diet), which was then replaced on an isonitrogenous basis with various other experimental soybean meals. In a 6‐week growth trial, shrimp in four replicate tanks per dietary treatment (10 shrimp per tank, initial weight 0.52 ± 0.04 g) were cultured in a recirculating system. There were no significant differences with respects to per cent weight gain and survival across all dietary treatments; however, final weights and feed conversion ratio (FCR) were lower in shrimp offered diet 3. Apparent digestibility coefficients for the eight (A–H) different soybean meals were determined in L. vannamei for dry matter (ADMD), gross energy (ADE) and crude protein (ADP) using 10 g kg?1 chromic oxide as inert marker with 70 : 30 replacement techniques. Coefficients ranged from 71.3% to 88.3%, from 76.6% to 91.3% and from 93.6% to 99.8%, for ADMD, ADE and ADP, respectively. Improved digestibility values were observed in soybean C which was characterized by crude protein (471 g kg?1), crude fat (97 g kg?1), low cooking temperature (180 °C), higher nitrogen solubility index (689 g kg?1) and protein dispersibility index (619 g kg?1). This indicates that new lines of soybean meal can be used to improve digestibility coefficients in shrimp feeds.  相似文献

A 12‐week feeding trial was conducted in eighteen 70 L indoor polyvinyl circular troughs provided with a water flow‐through system (1–1.5 L min?1) at 28 ± 1 °C to evaluate the dietary tryptophan requirement of fingerling Catla catla (3.45 ± 0.24 cm; 0.60 ± 0.13 g). Six casein‐gelatin‐based amino acid test diets (330 g kg?1 crude protein; 13.6 kJ g?1 digestible energy) containing graded levels of L‐tryptophan (1.0, 1.4, 1.9, 2.3, 2.8, 3.4 g kg?1 dry diet) were fed to triplicate groups of fish near to satiation at 08:00, 12:30 and 17:30 h. Absolute weight gain, feed conversion ratio, protein gain, RNA/DNA ratio, hepatosomatic index, viscerosomatic index, condition factor and haematological indices improved with the increasing levels of tryptophan from 1.0 to 2.3 g kg?1 of dry diet. Significantly higher carcass protein was obtained at 2.3 g tryptophan per kilogram of the dry diet. Exponential analysis of absolute weight gain, feed conversion ratio, protein gain and RNA/DNA ratio against dietary tryptophan levels at 95% maximum and minimum responses displayed the tryptophan requirement at 2.5, 2.3, 2.5 and 2.1 g kg?1 dry diet, respectively. Inclusion of dietary tryptophan in the range of 2.1–2.5 g kg?1 dry diet, equivalent to 6.4–7.6 g kg?1 dietary protein, is recommended in formulating tryptophan‐balanced feed for the culture of this fish species.  相似文献

Vibrio harveyi is a causative agent of the Vibriosis or luminescent bacterial disease in worldwide aquaculture industry. A reliable assay for identification of V. harveyi infection is important to prevent the bacterial spread. In this study, biotinylated loop‐mediated isothermal amplification (LAMP) amplicons were produced by a set of four designed primers that recognized specifically the V. harveyi vhhP2 gene, encoding a putative outer membrane protein with unknown function, followed by hybridization with an fluorescein isothiocyanate (FITC)‐labelled probe and lateral flow dipstick (LFD) detection. A novel set of PCR primer was also designed specifically to vhhP2 gene and appear to be a species‐specific tool for V. harveyi detection. The optimized time and temperature conditions for the LAMP assay were 90 min at 65°C. The LAMP‐LFD and PCR methods accurately identified 22 isolates of V. harveyi but did not detect 16 non‐harveyi Vibrio isolates, and 34 non‐Vibrio bacterial isolates. The sensitivity of LAMP‐LFD for V. harveyi detection in pure culture was 1.1 × 102 CFU mL?1 or equivalent to 0.6 CFU per reaction, while that of PCR was 6 CFU per reaction. For spiked shrimp sample, the sensitivity of LAMP was 1.8 × 103 CFU g?1 or equivalent to 5 CFU per reaction, while that of PCR was 50 CFU per reaction. In conclusion, the established LAMP‐LFD methods provided a valuable tool for rapid identification of V. harveyi and can be used to distinguish V. harveyi from V. campbellii.  相似文献
