O‐GlcNAc transferase gene (OGT) was considered as the sole rate‐limiting enzyme in the O‐GlcNAc modification. In the present study, the OGT gene of hybrid grouper (Epinephelus fuscoguttatus ♀ × E. lanceolatus ♂) was cloned and characterized, and its expression in response to dietary carbohydrate level and acute glucose treatment was investigated. The full‐length of OGT (GenBank accession no. KY656469 ) was 4,063 bp, including a 302 bp 5′untranslated terminal region (UTR), a 3,165 bp coding region that encoded 1,054 amino acids residues and a 596 bp 3′ UTR. The highly conservation of OGT gene between fish and mammals was also observed through multiple sequences alignment and phylogenetic analysis. O‐GlcNAc transferase gene was ubiquitously expressed in all detected tissues with highest expressions in brain and liver, to a lesser degree, in eye, heart, kidney and intestine. The increasing dietary carbohydrate from 8.02% to 16.08% had no significant effect on the mRNA expression of OGT. However, the expression of OGT was slightly elevated at 6 hr post‐glucose injection, and the elevation became significant at 24 hr time‐point. These data may enhance our understanding on the nutritional regulation of OGT and O‐GlcNAc modification in fish species.  相似文献

A 12‐week feeding trial was conducted to determine the dietary threonine requirement of fingerling Indian major carp, Catla catla (3.35 ± 0.11 cm; 0.59 ± 0.06 g). Six casein‐gelatin based (33% crude protein; 3.23 kcal g?1 digestible energy) amino acid test diets with graded levels of analysed threonine (0.74%, 0.96%, 1.21%, 1.48%, 1.72% and 1.93% dry diet) were fed to satiation to triplicate groups of fish. Absolute weight gain (g per fish), feed conversion ratio, protein retention efficiency, threonine deposition, RNA/DNA ratio and carcass protein significantly improved with the increase in dietary threonine and peaked at 1.48% of the dry diet. Haematological indices were also found to be best in fish fed at 1.48% threonine diet. Quadratic regression analysis of absolute weight gain, feed conversion ratio, protein retention efficiency, threonine deposition, RNA/DNA ratio, carcass protein, haemoglobin (g dL?1), haematocrit (%) and RBCs (106 × mm?3) at 95% of maximum and minimum response exhibited the threonine requirement of fingerling C. catla between 1.35% and 1.48% dry diet, corresponding to 4.09–4.48% dietary protein. Present finding would be useful in formulating threonine‐balanced feeds for the intensive culture of C. catla.  相似文献

Reproduction allows organisms to produce offspring. Animals shift from immature juveniles into mature adults and become capable of sexual reproduction during puberty, which culminates in the first spermiation and sperm hydration or ovulation. Reproduction is closely related to the precise control of the hypothalamic–pituitary–gonadal (HPG) axis. Kisspeptin peptides are considered as the important regulator of HPG axis in mammalian. However, the current understanding of kisspeptin in flatfish is not comprehensive. In this study, we cloned and analyzed the kiss2 and kissr2 genes in Cynoglossus semilaevis. Interesting alternative splicing in the 5′-untranslated regions (UTR) of the Cskissr2 gene was found. The expression profiles of Cskiss2 and Cskissr2 showed relative high messenger RNA (mRNA) levels at the late gastrula stage during embryonic development, at total length = 40 mm during early gonadal differentiation, and in the brains and gonads of all investigated tissues. These results suggested that the kisspeptin system participated in embryogenesis and in the regulation of gonadal differentiation and development. Considering that the control and regulatory mechanisms of kisspeptin in the central reproductive axis are still unclear, we documented that the intramuscular injection of kisspeptin caused different sGnRH and cGnRH mRNA levels in a dose- and tissue-dependent manner. The mRNA expressions of FSH and LH were stimulated in the ovary and were inhibited in the testis under the kisspeptin treatments. These results provided foundations for understanding the roles of kisspeptin in the neuroendocrine system in fish. The manipulation of the kisspeptin system may provide new opportunities to control the gonadal development and even reproduction in fish.  相似文献

In recent years, kisspeptin peptides, encoded by kiss genes have been used to manipulate reproductive processes in farmed animals, including fish. Our previous studies demonstrated that the chub mackerel brain expresses kiss1 and kiss2 and intramuscular injection of synthetic Kiss1 pentadecapeptide (Kiss1‐15) but not Kiss2 dodecapeptide (Kiss2‐12) accelerates spermatogenesis in prepubertal male chub mackerel (Scomber japonicus). In the present study, we evaluated their effects in prepubertal female chub mackerel. The gonadosomatic index (GSI) values of experimental fish did not show any significant changes. Condition factor (CF) values increased significantly in Kiss1‐15 treated fish, in comparison with control and GnRH analogue (GnRHa) injected fish. Histologically, only perinucleolar oocytes were found in all experimental fish. However, Kiss and GnRHa treated fish showed a significant increase in the perinucleolar oocyte diameter, in comparison with the control fish. Gene expression analyses revealed decreased expression of gnrh1 in the telencephalon‐preoptic region of the brain of Kiss2‐12 and GnRHa injected fish, in comparison with control fish. In contrast, GnRHa injected fish exhibited higher levels of fshβ in the pituitary, with no changes in the levels of lhβ among different treatments. Levels of circulating sex steroids, testosterone, and estradiol‐17β were significantly higher in Kiss1‐15 injected fish, in comparison with control fish. These results indicate that synthetic kisspeptin peptides and GnRHa can induce oocyte growth in prepubertal female chub mackerel.  相似文献

Interleukin‐10 (IL‐10) is a multifunctional cytokine and plays an important role in diseases. In this study, IL‐10 gene was cloned and characterized from catla (Catla catla), which is a highly commercially important fish species in the Indian subcontinent. The result indicated that the full‐length catla IL‐10 (cIL‐10) gene had five exons and four introns with an open reading frame of 540 nucleotides encoding a polypeptide of 179 amino acids. A phylogenetic analysis of cIL‐10 gene sequence showed that cIL‐10 clustered with freshwater carps group as expected. Quantitative real‐time polymerase chain reaction analysis showed that cIL‐10 was expressed in gill, liver, kidney, intestine, skin and heart and its expression profile was up‐regulated in bacterial infection and LPS treatment. A close relationship of high cIL‐10 expression and low pro‐inflammatory cytokine IL‐1β expression was observed in the treated group of fish, which might reveal the role of cIL‐10 as an anti‐inflammatory cytokine. Mechanism of cIL‐10 induction was investigated by blocking nuclear factor (NF)‐κB ‐signalling with BAY 11‐7082 in catla kidney cell culture. Blocking NF‐κB suppressed IL‐10 induction by LPS, and thus it revealed that cIL‐10 was induced through NF‐κB signalling. These data could be helpful to understand the function of IL‐10 in fish in response to vaccinations, probiotics and various diseases.  相似文献

The transformer‐2 gene (tra‐2) plays a key role in the sexual differentiation regulatory hierarchy. In this study, tra‐2 gene homologs designated as Mntra‐2 was cloned and characterized from Macrobrachium nipponense. The full‐length cDNA of Mntra‐2 consists of 1724 bp with an open reading frame (ORF) encoding 192 amino acids, an 827 bp 5′‐untranslated region (UTR) and a 318 bp 3′‐UTR. The predicated molecular mass of Mntra‐2 was 20.805 kDa with an estimated theoretical isoelectric point of 10.36. The deduced amino acid sequence shares high homology with Penaeus monodon. Real‐time quantitative polymerase chain reaction (RT‐qPCR) analyses demonstrated that the expression levels of Mntra‐2 varied significantly during different developmental stages of embryogenesis, larvae, and post‐larvae and in various adult tissues. During embryogenesis, the expression level of Mntra‐2 was slightly higher at the cleavage stage than at the blastula stage, and reached the highest level at the nauplius stage. During the larvae, the Mntra‐2 expression gradually increased from 1 d larvae post hatch (L1) to L10 and decreased to a lowest level at the end of metamorphosis. During the post‐larvae, the Mntra‐2 expression was higher level at the 5 d after the metamorphosis (P5). RT‐qPCR showed the Mntra‐2 mRNA was expressed in ovary, testis, muscle, heart, abdominal ganglion, brain, and intestine with the highest level of expression in muscle and intestine. The results indicate that Mntra‐2 is an arthropods tra‐2 homolog and probably plays important roles in embryonic development and sex differentiation of M. nipponense .  相似文献

Facilitative glucose transporter 1 (GLUT1) is a transporter protein for glucose transport via the plasma membrane of the cells to provide energy through carbohydrate metabolism. GLUT1 cDNA from Litopenaeus vannamei was obtained and analysed in this study. Full‐length GLUT1 cDNA is 2062 bp long and contained a 1506‐bp ORF encoding a 502 amino acid protein, a 270‐bp 5′UTR and a 284‐bp 3′UTR. When shrimp were under acute low salinity stress, the expression in hepatopancreas, muscle, gill and eyestalk was all up‐regulated at 12 h (P < 0.05) and 96 h (P < 0.05), while the expression in the four tissues was all down‐regulated at 6 h (P < 0.05) and 48 h (P < 0.05) . The expression in the muscle of shrimp at water salinity of 3 was lower than that at water salinity of 30 independent of dietary carbohydrate levels, while expression in hepatopancreas, gill and eyestalk was up‐regulated at 200 and 300 g kg?1 carbohydrate levels. The expression in all tissues fed glucose was up‐regulated when compared to the expression in shrimp held at a water salinity of 30. This study suggests that GLUT1 is a conserved protein in L. vannamei, and changes in expression due to environmental salinity and dietary carbohydrate level and source.  相似文献

Long‐day photoperiods are considered as an effective managerial tool in manipulating of reproduction and somatic growth in a number of fish species. In this study, the effects of three different artificial photoperiods on the gonadal development and somatic growth of rainbow trout (Oncorhynchus mykiss L.) were investigated. Two years‐old immature female rainbow trout (279.94 ±2.25 g) were exposed to three artificial photoperiod regimes of 24L:0D, 18L:6D and 6L:18D and natural light (NL) regime for 5 months. The highest gonadosomatic indices were recorded in NL and 6L:18D groups while the rates were significantly lower in fish maintained under 18L:6D and 24L:0D photoperiods (< 0.05). Mean oocyte diameters in fish exposed to 24L:0D and also to 18L:6D were significantly lower than the 6L:18D and NL groups. Photoperiods with 24L:0D and 18L:6D regimes resulted in significantly higher mean final weights and specific growth rates (SGR) than NL regime. The highest mean final weight (635.45 ± 16.19 g) and SGR (1.03 ± 0.04% day?1) were obtained under 24L:0D photoperiod. Fish exposed to 24L:0D and 18L:6D showed the highest condition factor as 1.44 ± 0.01 and 1.44 ± 0.02 respectively, when compared with the NL (1.27 ± 0.01) and 6L:18D (1.34 ± 0.02) groups. Basically, the results suggested that continuous artificial lightning can be used as an influential factor in delaying gonadal development and enhancing somatic growth in rainbow trout during gonadal growth phase.  相似文献

The aim of this study was to evaluate the anaesthetic effect of MS‐222 and propofol and determine their optimal concentrations for safe handling of the tetra Astyanax altiparanae in the laboratory. The fish were separated by length into three classes: I (1.5–5.0 cm), II (5.1–8.0 cm) and III (greater than 8.1 cm). Pilot tests were performed to evaluate the appropriate anaesthetic concentrations for inducing the five possible anaesthetic stages: I – sedation; II – light anaesthesia; III – deep anaesthesia; IV – surgical anaesthesia; and V – spinal collapse. After defining the maximum and minimum concentrations required to induce stage IV anaesthesia, the animals were exposed to five intermediate concentrations (n = 10 fish/concentration) of each anaesthetic for 15 min. The animals were then transferred to clean water to evaluate the time required for recovery. In addition, blood glucose levels were measured for class II and class III fish subjected to the previously defined ideal concentrations for each of the tested anaesthetics (n = 10 fish/treatment). Both evaluated substances are suitable to anaesthetize A. altiparanae. The optimal MS‐222 concentration was 90 mg L?1, and this result was similar for all three size classes. The optimal propofol concentrations for inducing surgical anaesthesia in the size classes I, II and III were 0.22, 0.23 and 0.27 respectively.  相似文献

The γ‐aminobutyrate type A receptor‐associated protein (GABARAP) is a ubiquitin‐like modifier implicated in membrane trafficking and fusion events involving the γ‐aminobutyrate type A receptor, autophagy and apoptosis. In this study, the gene encoding GABARAP was cloned from swimming crab Portunus trituberculatus (PtGABARAP) based on the expression sequence tag (EST). The full‐length cDNA of 664 bp includes a 5′ untranslated region (UTR) of 87 bp, a 3′ UTR of 223 bp with a poly(A) tail, and an open reading frame (ORF) of 354 bp encoding a polypeptide of 117 amino acids with a predicted molecular weight of 13.96 kDa. The deduced amino acid sequence shares high similarity (93%–100%) with GABARAPs from other species and includes a conserved Atg8 domain. In a phylogenetic analysis PtGABARAP clustered with GABARAPs from other species, and more widely with other GABARAP family proteins. The impact of elevated ocean acidification (OA) on P. trituberculatus behaviours was investigated, and real‐time RT‐PCR revealed that PtGABARAP expression was up‐regulated after OA exposure. Ocean acidification also caused crabs anxiety‐like behaviours, like the shoal average speed increase, preference for dark environment (scototaxis) and fast exploration. The results indicated that GABARAP might be involved in the interactions of GABAA receptors and elevated‐CO2 seawater.  相似文献

Systematic research was performed to assess the survival, growth performance, gonadal structure and sex ratio during and after sex reversal induced by estradiol‐17β in tiger puffers. The fish of 20 days after hatching (dah) were immersed in different doses (0.2, 2, 20, 100 ppb) for 60 days. There were significant differences in survival rates among groups (< .05). With the increase in the estradiol‐17β dose, the survival rates of tiger puffers decreased, revealing a linear regression relationship with log10 (estradiol + 1; < .05). The growth tendency of fish in the low‐dose group was similar to those in the control group over the same period. The growth in the 20 ppb dose group was significantly slower than in the low‐dose and control groups in fish of same age (< .05), but was significantly faster than in the 100 ppb dose group (< .05) after 60 days. Intersex fish were categorized into intersex I, II and III types, according to the feminization degree from low to high. Phenotypic intersex puffer fish were found in the 2, 20 and 100 ppb dose groups. Intersex ratios in 2, 20 and 100 ppb were 10%, 43.3% and 46.7%, respectively, the ratios except the ones in 2 ppb group being significantly higher than those in the control group (< .05). Moreover, 100.0% male induction for intersex II and III types occurred in 20 and 100 ppb groups. Together, these results indicated that estradiol‐17β at a dose of 20 ppb was most effective for pseudo‐female production.  相似文献

This study aimed to develop the consistent ovulation induction method in a pelagic egg spawning marine teleost, nibe croaker Nibea mitsukurii. Attempts to induce oocyte maturation and ovulation in nibe croaker using human chorionic gonadotropin (hCG; 0.5 IU g?1) resulted in the normal progression of oocyte maturation and hydration, but a failure to induce ovulation in many individuals. This ovulation disorder was similarly observed even when the dose of hCG was increased 10 times (5 IU g?1) or decreased to one tenth (0.05 IU g?1), indicating that it cannot be completely overcome solely by hCG administration. However, this ovulation disorder could be completely overcome by subsequent administration of 17,20β‐dihydroxy‐4‐pregnen‐3‐one (DHP) at the appropriate dose (0.5 μg g?1) and time (20 h after hCG administration). An increase in the number of individuals that ovulated due to DHP administration led to an increase in individuals producing larvae, resulting in an approximately threefold increase in the estimated number of larvae produced compared with the group of fish administered hCG alone. Thus, this ovulation induction method using DHP administration after hCG was demonstrated to overcome the ovulation disorder in nibe croaker and could be applicable to commercially important species with similar ovulation problems.  相似文献

To assess the compensatory growth, a 10‐month pond experiment was conducted in which three species of Indian major carps, viz. Catla catla, Labeo rohita and Cirrhinus mrigala (1 : 1 : 1), were stocked at 7500 ha?1 and the fish were subjected to different restricted feeding and refeeding protocols, viz. Control (C): Continuously fed for 10 months; Treatment‐1 (T‐1): Initial supplementary feeding for 2 months+ No supplementary feeding for 1 month + Refeeding for 7 months; Treatment‐2 (T‐2): Initial supplementary feeding for 2 months + No supplementary feeding for 2 months + Refeeding for 6 months; and Treatment‐3 (T‐3): Initial supplementary feeding for 2 months + No supplementary feeding for 3 months + Refeeding for 5 months in triplicate ponds. At the end of experiment, 100% growth compensation was achieved in T‐2 for all the three carp species. Among all treatments, higher weight gain, PER and PPV and lower AFCR were also recorded in T‐2, resulting in maximum fish production. The restricted feeding had significant effect (P < 0.05) on whole body chemical composition of fish. From this study, it is concluded that in 10‐month pond culture of carp in fertilized pond, the T‐2 would be the best and most economic feeding strategy.  相似文献

Formulation of nutritionally complete and cost efficient diets for yellow perch (Perca flavescens) is a prerequisite for successful intensive culture of this species. One of the objectives of this study was to determine the optimum diet for the grow‐out phase of juvenile yellow perch. Fish at the size of 12.9 ± 4 g were individually marked with passive integrated transponders (PIT)‐tags and randomly distributed into six 400 L tanks, 45 fish per tank. This experiment included lysine‐deficient [(?) Lys] and lysine‐supplemented [(+) Lys] wheat‐gluten‐based diets in triplicate groups. Our experiment showed that the mean weight of fish fed (+) Lys diet (83.9 ± 1.5 g) was significantly larger than fish fed (?) Lys diet (68.6 ± 5.2 g) (< 0.05). This experiment also showed that the blood plasma concentration of free lysine in (+) Lys group was significantly higher than in (?) Lys group (< 0.05) and the same trend appeared also in methionine concentrations. The concentration of Lys in deficient group of fish, 3 h after a meal was lower compared with levels of Lys 24 h post‐feeding (< 0.05). Lysine deficiency in diet resulted in significantly higher level of serine, and a similar trend occurred in small and large fish.  相似文献
