尼罗罗非鱼六个性别相关标记的FISH分析   总被引:2,自引:0,他引:2       下载免费PDF全文
从尼罗罗非鱼(Oreochromis niloticus)BAC基因文库5个克隆中提取和纯化含有6个性别连锁或相关标记(CLC5,GM204,GM271,GM354,UNH995和UNH104)的重组质粒DNA作为模板,以简并核苷酸为引物,通过PCR制备原位杂交探针。探针用荧光素进行标记,并与尼罗罗非鱼中期相染色体进行荧光原位杂交以确定这些标记在尼罗罗非鱼染色体上的位置和分布。结果显示,这些性别连锁或相关标记都位于尼罗罗非鱼第一对染色体长臂近末端,从分子细胞学角度验证了第一对染色体是尼罗罗非鱼的性染色体。另外由于这些标记的荧光信号在XY个体的2条性染色体上都有,一方面说明这些标记在罗非鱼上还不是性别特异的;另一方面也验证了尼罗罗非鱼的性染色体还处于分化的早期阶段。【中国水产科学,2006,13(4):525—529】  相似文献

为探究黑素皮质素受体1基因(MC1R)在橘色双冠丽鱼胚胎发育和体色形成过程中的表达定位及功能,本研究首先制备了MC1R基因的RNA正反义探针,T7方向转录的正义RNA探针质量浓度为447.529 ng/μL,SP6方向转录的反义RNA探针质量浓度为342.698 ng/μL。经10~20倍稀释后的探针用于原位杂交,MC1R基因探针表达定位显示,随橘色双冠丽鱼胚胎的发育杂交信号总体呈逐渐减弱趋势,原肠期和视泡期其卵黄和胚体侧卧部分有杂交信号分布;出膜期其脊柱、卵黄囊及其内容物和色素细胞内出现杂交信号;出膜期第1天,卵黄表面有信号分布。总体来看,杂交信号在脊索、卵黄囊、卵黄囊内容物质及色素细胞有特异表达,而以正义探针作为阴性对照组在5个胚胎阶段均无任何信号,杂交信号显现的MC1R表达定位说明其在橘色双冠丽鱼色素细胞的分化、迁移中起到重要调控作用,也与神经、营养、免疫功能相关,初步建立了鱼类体色相关基因功能和定位研究的整胚原位杂交方法。  相似文献

奥利亚罗非鱼、尼罗罗非鱼MyoD1和MyoD2基因特征及差异   总被引:1,自引:0,他引:1  
采用RT-PCR和RACE法,分离了奥利亚罗非鱼(Oreochromis aureus)、尼罗罗非鱼(O.niloticus)MyoD1和MyoD2基因全长cDNA。结果显示,2种罗非鱼MyoD1全长均为1090bp,包括5′非翻译区(UTR)137bp,3′UTR50bp,开放阅读框(ORF)903bp,编码300个氨基酸,其中第110~161个氨基酸为bHLH结构,第233~249个氨基酸为helixIII结构;MyoD2全长均为1478bp,包括5′UTR215bp,3′UTR471bp,ORF792bp,编码263个氨基酸,其中第91~142个氨基酸为bHLH结构,第212~228个氨基酸为helixIII结构。2种罗非鱼MyoD1与其他鱼类MyoD1的相似性为73%~92%;MyoD2与其他鱼类MyoD2的相似性为74%~79%。系统发育树显示,MyoD1和MyoD2分属两支,MyoD1所反映的不同鱼类间的亲缘关系符合传统分类。2种罗非鱼的MyoD1、MyoD2cDNA序列之间只存在个别碱基的差别,而氨基酸序列一致;奥利亚罗非鱼MyoD1的2个内含子均比尼罗罗非鱼的长。根据MyoD1内含子2的差异构建鉴别奥利亚罗非鱼和尼罗罗非鱼基因混杂的标记,对形态上典型的15尾奥利亚罗非鱼、18尾尼罗罗非鱼及15尾奥尼罗非鱼[Oreochromis aureus(♂)×Oreochromis niloticus(♀)]进行鉴定。结果其中1尾奥利亚罗非鱼中在MyoD1位点混杂了尼罗罗非鱼的基因,尼罗罗非鱼和奥尼罗非鱼则与预期的一致。该研究为选择基因纯合的奥利亚罗非鱼和尼罗罗非鱼提供了新的分子手段。  相似文献

高碳酸盐碱胁迫对尼罗罗非鱼氨代谢基因表达变化的影响   总被引:1,自引:0,他引:1  
为了解尼罗罗非鱼(Oreochromis niloticus)在碱环境适应过程中氨代谢调节途径,本研究选取了5个氨代谢相关酶:谷氨酰胺合成酶(glutamine synthetase,GS)、碳酸酐酶5A(carbonic anhydrase 5A,CA-5A)、谷氨酰胺酶(glutaminase 2,GLS2)、氨甲酰磷酸合成酶(carbamyl phosphate synthetase 1,CPS1)、氨转运蛋白(ammonium transporter Rh type C-2 like,Rhcgl2),研究了急性碳酸盐碱度胁迫条件下,尼罗罗非鱼血氨浓度变化、氨代谢相关酶基因表达水平及其酶活性变化。结果表明,随碳酸盐碱胁迫浓度升高,尼罗罗非鱼血氨浓度上升,随时间推移呈先上升后下降的变化趋势,在胁迫后12 h达到峰值。氨代谢相关基因在不同碱度下、不同组织中均有不同程度的上调表达,随着胁迫时间推移呈先上升后下降的变化趋势,胁迫后12~24 h各基因表达水平显著升高,随后逐渐恢复到稳定水平;氨代谢相关基因具有一定的组织表达差异:氨转运蛋白基因(Rhcgl2)主要在鳃中表达,碳酸酐酶5A基因(CA-5A)、谷氨酰胺合成酶基因(GS)、氨甲酰磷酸合成酶基因(CPS1)主要在肝中表达,谷氨酰胺酶基因(GLS2)主要在肾和鳃中表达。碳酸酐酶和谷氨酰胺合成酶活性随胁迫碱度的升高而上升,碳酸酐酶、谷氨酰胺合成酶活性变化分别在鳃、肝中最为显著。研究结果表明,碳酸盐碱度胁迫会引起尼罗罗非鱼血氨水平升高,随着时间推移血氨水平下降,推测鳃、肝、肾中不同氨代谢基因共同参与调节氨代谢,在鳃中通过直接排氨,在肝中通过合成谷氨酰胺、尿素途径,共同调节降低血氨水平。  相似文献

罗非鱼是重要的经济鱼类,而嗜水气单胞菌(Aeromonashydrophila)是其主要病原菌之一,只有充分的掌握该病原菌的感染机制才能提供更好的预防和治疗手段。本研究首先通过转录组筛查尼罗罗非鱼(Oreochromisniloticus)脾脏中与细菌感染相关的基因,然后通过荧光定量PCR证实上述基因的表达变化,最后使用秋水仙素试验和交叉免疫试验进行验证。研究结果显示,尼罗罗非鱼在感染嗜水气单胞菌之后,吞噬作用发生显著增强。秋水仙素试验发现,微管蛋白的功能受到抑制可以显著降低罗非鱼对嗜水气单胞菌的防御能力。交叉免疫试验证实,通过迟缓爱德华菌(Edwardsiella tarda)激活吞噬作用也可以提高罗非鱼对嗜水气单胞菌的防御能力。本研究旨在了解吞噬作用在罗非鱼抵御嗜水气单胞菌感染过程中发挥重要作用,并试图通过交叉免疫试验为细菌疫苗的制备提供新思路。  相似文献

为获得尼罗罗非鱼(Oreochromis niloticus)抗链球菌病相关的单核苷酸多态性(single nucleotide polymorphism,SNP)标记,本研究通过染色体步移法克隆了尼罗罗非鱼Ikaros基因5′调控区序列,长度为4178 bp,并使用生物信息学软件对Ikaros基因的启动子和转录调控元件进行预测和分析。利用PCR产物直接测序法从亲本(P0)尼罗罗非鱼Ikaros基因5′调控区中筛查到5个SNPs,分别为SNP1(g.562,GA)、SNP2(g.217,GT)、SNP3(g.–53,CT)、SNP4(g.–220,TC)和SNP5(g.–579,TC)。利用Snapshot技术对子一代(F1)尼罗罗非鱼易感群体和抗病群体进行相应SNPs的基因分型,分析两个群体遗传多样性参数,结果显示多态信息含量(polymorphism information content,PIC)值在0.0872~0.3747之间,表明Ikaros基因5′调控区中所有SNPs的多态性均较低。Ikaros基因5′调控区SNPs与抗链球菌病性状关联分析结果表明,SNP2、SNP3、SNP4和SNP5的基因型频率和等位基因频率在易感群体和抗病群体中存在显著差异(P0.05)。连锁不平衡分析结果显示Ikaros基因5′调控区SNPs可形成1个单倍块和5种单倍型,其中GGCTT单倍型与抗病性显著相关(P0.05),GGTCT和GTCCC单倍型与易感性显著相关(P0.05)。此外,还发现SNP2和SNP5处于完全连锁状态(r~2=1,LOD=57.25,D′=1),可作为罗非鱼抗链球菌病遗传育种的标签SNP。综上所述,在Ikaros基因5?调控区筛选到4个抗链球菌病相关SNPs和1个单倍型(GGCTT),均可作为尼罗罗非鱼分子育种的候选分子标记。  相似文献

本研究采用RT-PCR方法对生长快、耐盐性弱的尼罗罗非鱼(Oreochromis niloticus),耐盐性强、生长慢的萨罗罗非鱼(Sarotherodon melanotheron)以及生长与耐盐均较优的杂交子代(O.niloticus ♀早×S. melanotheron ♂)的催乳素PRLI基因cDNA序列片段进行了克隆与序列分析,以探讨罗非鱼耐盐性的分子机制,分析和筛选与耐盐性相关的基因.主要结果:(1)序列克隆及ClustalX1.83分析表明克隆所得cDNA序列长度为339 bp,编码112个氨基酸.(2)3种罗非鱼PRLI的cDNA具有高度的保守性,核苷酸序列同源性介于97.94%~99.71%之间,氨基酸序列同源性均在99.11%以上,表明罗非鱼的PRLI基因具有高度保守性.(3)杂交子代同尼罗罗非鱼的cDNA序列相比有5个碱基的变异,变异比例为1.47%;同萨罗罗非鱼相比有1个碱基的变异,变异比例为0.30%.(4)尼罗罗非鱼推导氨基酸序列中第80位氨基酸残基为脯氨酸残基(P),而杂交子代和萨罗罗非鱼在该位点均为丝氨酸残基(S).(5)MEGA4系统进化树分析显示,杂交子代同萨罗罗非鱼聚为1支.结果(3)、(4)、(5)表明,在PRLI基因的表达方面,杂交子代同父本萨罗罗非鱼的关系较近.  相似文献

加州鲈肌肉生长抑制素(MSTN)cDNA的克隆和序列分析   总被引:1,自引:2,他引:1  
肌肉生长抑制素是抑制肌肉生长和发育的生长调控因子。对运用RT-PCR和RACE技术从加州鲈成鱼肌肉总RNA中扩增得到的MSTN cDNA全序列进行了序列分析。结果表明,加州鲈MSTN cDNA全长为1626bp,其开放阅读框为1 134bp,共编码377个氨基酸,前面的22个氨基酸为信号肽,中间有四个氨基酸(RARR)为蛋白水解加工位点;该基因总共有13个半胱氨酸残基,后面9个在蛋白水解加工位点之后的C端生物活性区,与其它脊椎动物比较,它们的位置完全一致,对该基因的结构和功能非常重要。与GenBank中已知的条纹狼鲈、金鲷、斑马鱼、虹鳟、斑点叉尾鮰、人、猪、鸡、鸽MSTN的ORF相比较,核苷酸序列同源性为63%~94.4%,氨基酸同源性为61.4%~96%,特别是在C端生物活性区氨基酸同源性为88.1%~100%,高度的保守性反映了该基因受到了高度的进化限制以及功能的重要性。加州鲈MSTN基因的克隆为研究该基因打靶和鱼类肌肉发育调控机理奠定了基础。  相似文献

An experiment using differently marked larval striped bass, Morone saxatilis (Walbaum). and its hybrid (M. saxatilis x M. chrysops (Rafinesque)) was performed in two grow-out ponds. Larval striped bass were immersed in oxytetracycline solution to mark their otoliths; hybrids received no treatment. Larvae from both groups were mixed and stocked at 8 days post hatch into the ponds. Striped bass and hybrid larvae grew to a significantly larger size at 35 days in one pond. In both ponds, hybrid lengths at 35 days after hatch were significantly greater than striped bass lengths. However, the magnitude of the size difference between hybrids and striped bass was twofold greater (10%) in one pond than in the other (5%). The proportion of striped bass juveniles at 35 days differed from the initial stocking proportion (0.53) only in one pond, where hybrids showed 12% greater survival than striped bass. Results suggested that the relative survival of hybrids was influenced by growth conditions in the ponds. Based on the ease of protocol and analysis of the marking experiment, we recommend its use to (1) investigate relative performance between genetic groups of young fish in common environments; and (2) predict the effects of introductions of genetically altered fishes.  相似文献

Kisspeptin(简称Kiss或者Kp)是由KISS1/Kiss1基因编码的一种下丘脑神经肽,通过其受体KissR(也称作GPR54)的介导参与了多种生理过程,如抑制肿瘤转移和参与生殖调控。目前,尽管在鲤形目(Cypriniformes)、鲈形目(Perciforms)、鲽形目(Pleuronectiforms)、鲀形目(Tetraodontiforms)、颌针目(Beloniforms)、鲉形目(Scorpaeniformes)、鲑形目(Salmoniformes)及鳕形目(Gadiformes)等多种鱼类中均鉴定出了kiss/kissr基因,但Kiss/KissR系统在鱼类生殖调控中的精确作用及其分子机制尚未完全阐明。尤其是在鱼类中存在2种kiss及3种kissr基因,Kiss/KissR系统对鱼类生殖调控的作用方式更加复杂。本文简要总结鱼类Kiss及其受体的研究进展,并对Kiss的生理学功能、信号转导机制以及kiss/kissr表达调控研究进行概括讨论,旨在加深对鱼类Kiss/KissR系统的认识和了解,为后续研究指明方向。  相似文献

In recent years, infectious disease caused by Streptococcus agalactiae has increased in aquaculture, threatening the healthy development of tilapia and resulting in substantial economic losses. Cultivating disease‐resistant tilapia by transgenic technology is one strategy that has been developed to address this issue. Here, we used a transposon system to investigate gene expression and tissue lytic activity in transgenic zebrafish expressing the tilapia lysozyme. The transpose vector contained the Tgf2 transposon skeleton, tilapia heat shock protein 70 promoter (Hsp70) and two target genes (tilapia C‐type lysozyme 3 [lysozyme‐C3] and green fluorescent protein [GFP]). The transgenic zebrafish F0 generation was obtained by microinjecting the donor vector and transposase mRNA into fertilized zebrafish eggs. There was 45% green fluorescence in the zebrafish F0 generation, which was spread over the entire body based on microscope observation. The F0 generation was reared to sexual maturity, at which point individuals were mated with wild‐type zebrafish to produce the F1 generation. Tilapia lysozyme‐C3 expression was detected in the liver of F1 generation by RT‐PCR and western blotting, but was not detected in the skin, intestine, or muscle. Significantly higher liver bacteriolysis activity was detected in transgenic zebrafish compared with wild‐type zebrafish. Therefore, transgenic zebrafish may have an increased potential for bacterial resistance.  相似文献

Francisella orientalis is a highly virulent, emerging bacterium that causes mass mortalities in tilapia. This pathogen also affects numerous other warm-water fish species, including three-line grunt, hybrid striped bass and various ornamental fish. This study sheds light on two new species of fish that are susceptible to F. orientalis. Asian seabass and largemouth bass showed variable levels of susceptibility in a bacterial challenge experiment. After intraperitoneally injected with a dose of 106 CFU/fish, a total of 64.28% and 21.42% mortalities were obtained in Asian seabass and largemouth bass, respectively. Meanwhile, Nile tilapia showed acute mortality of 100%. All fish showed typical lesions of francisellosis, including multifocal granulomas in the spleen and head kidney. Immunohistochemical analysis revealed strong positive signals inside the granulomas of all fish. The bacterial recovery in solid media from infected fish was highest in Nile tilapia (85.71%), followed by Asian seabass (35.71%) and largemouth bass (21.42%). PCR results tested 100% positive for Nile tilapia, and 78.57% and 21.42% for Asian seabass and largemouth bass, respectively. In conclusion, Asian seabass and largemouth bass are susceptible to this pathogen, which warrants new management strategies when employing predation polyculture systems of these species with tilapia.  相似文献

Abstract.— Meiotic gynogens were produced using hybrid striped bass (♀ white bass, Morone chrysops , ×♂ striped bass, M. saxatilis ) eggs and white perch M. americana UV-irradiated sperm. Diploidy of the fertilized eggs was restored by application of hydrostatic pressure, which induced retention of the second polar body. Use of white perch sperm provided an unmistakable marker for detection of a paternal genetic contribution. Two assays were developed using the polymerase chain reaction (PCR) to amplify specific regions of the Morone genome . Primers for gene amplification were developed based on the DNA sequence of the striped bass growth hormone gene (SB-GH) or an anonymous striped bass locus (SBI-10). Control experiments using DNA from the three Morone species demonstrated that gene amplification yielded species-specific patterns of DNA fragments for both of these loci. Therefore, any progeny with a paternal contribution of a set of white perch chromosomes could be identified. Using these assays, we demonstrated that greater than 75% of the progeny obtained from successful experiments were true gynogens. These striped bass hybrid gynogens will provide the basis for future efforts to calculate gene-centromere distances and to identify markers linked to specific traits of interest to aquaculture.  相似文献

Abstract. Fifty each of F2-generation gold ( gg ), bronze ( Gg ), and black ( GG ) Oreochromis mossambicus (Peters) and either zero, four or eight largemouth bass, Micropterus salmoides , (Lacepède) were stocked in 20-m2 concrete tanks. After 7 days, water in all tanks was drained, and fish in each tank were censused. Largemouth bass ate 18% of the tilapia in the four-bass treatment (1 tilapia/bass/day) and 60% of the tilapia in the eight-bass treatment (1·6 tilapia/bass/day); the difference was significant (P=0–05). In the four-bass treatment there was a greater observed mean predation rate on gold than that on black or on bronze tilapia, but the difference was not significant: largemouth bass ate 25% of the gold tilapia, 16% of the bronze tilapia, and 13% of the black tilapia. In the eight-bass treatment, predation on the gold tilapia was significantly greater than that on both bronze (P=0·05) and on black (P=0·06) tilapia; predation on bronze and black tilapia was similar: largemouth bass ate 80% of the gold tilapia, 48% of the bronze tilapia, and 51% of the black tilapia. Overall average total predation (both treatments combined) on gold tilapia was significantly (P=0·06) greater than that on both bronze and on black tilapia, which did not differ: largemouth bass ate 52% of the gold tilapia, 32% of the bronze tilapia, and 32% of the black tilapia. The increased vulnerability of gold tilapia to predation was a negative pleiotropic effect of the gg genotype.  相似文献

甘露糖受体是先天免疫系统中重要的模式识别受体和内吞受体,主要存在于巨噬细胞和树突状细胞的细胞膜表面,在维持内稳态、识别病原、诱导细胞因子、抗原递呈等过程中发挥重要作用。有关甘露糖受体靶向药物与分子疫苗的研究不断深入,显示出良好的应用前景。斑马鱼(Danio rerio)、尼罗罗非鱼(Oreochromis niloti-cus)、舌齿鲈(Dicentrarchus labrax)、斑点叉尾鮰(Ictalurus punctatus)的基因组中都预测有甘露糖受体基因。笔者首次克隆到团头鲂(Megalobrama amblycephala)甘露糖受体基因全长序列(JQ345719)和草鱼(Ctenopharyn-godon idellus)甘露糖受体基因部分序列(JX391027)。开展鱼类甘露糖受体的研究对深入了解养殖鱼类生理机能与抗感染免疫机制,开发鱼类甘露糖受体靶向新药和分子疫苗具有重要意义。  相似文献

Previous reports have indicated there are significant differences in both the dietary lysine requirement and the metabolic rate of striped bass and its hybrids. However, there is very little directly comparative data to confirm these suggestions. A series of experiments was conducted to comparatively assess efficiency of protein and energy retention between striped bass Morone saxatilis and sunshine bass M. chrysops♀×M. saxatilis♂ grown under identical culture conditions. In experiment one, a dose response study was conducted using digestible energy (DE) levels of 3,200 kcal/kg and 3,600 kcal/kg. At each level of DE, six levels of dietary lysine were fed to striped bass and sunshine bass. Ten sunshine bass weighing 3.48 ± 0.08 g or six striped bass weighing 3.23 ± 0.14 g were stocked into 15-L tanks supplied with single-pass flow-through fresh water. After 12 wk on the experimental regime, feed conversion ratios (FCR), weight gain, and % nitrogen (N) retention were determined. Nonlinear regression analysis indicated that the dietary lysine requirement of both bass groups was similar. The dietary lysine requirement was determined to be 2.0 ± 0.08% of dry diet (or 6.0 ± 0.26 g lysine/1,000 kcal DE), and 1.7 ± 0.08% of dry diet (or 4.7 ± 0.22 g lysine/1,000 kcal DE), for the 3,200 and 3,600 kcal DE/kg diets, respectively. A second experiment comparatively assessed growth, metabolism, and energy partitioning between striped bass and sunshine bass. Two diets were formulated to be isonitrogenous and isocaloric with a calculated DE level of 3462 kcal/kg and contained lysine concentrations approximating the published dietary requirements of sunshine bass (low lysine = LL diet) and striped bass (high lysine = HL diet). The diets were fed at an average rate of 2% of body weight/d to 25 juvenile striped bass or hybrid bass held separately in eight 2000-L single-pass flow-through tanks supplied with freshwater. In general, growth performance of sunshine bass was superior to striped bass. Both bass groups performed better when fed the HL diet. Mean FCRs were 1.19 ± 0.12 and 1.94 ± 0.29, respectively, for hybrids and striped bass fed the LL diet (P= 0.066); and 1.17 ± 0.07, and 1-58 ± 0.08, respectively, for hybrids and striped bass fed the HL diet (P = 0.011). Mean % gain/d values were significantly higher (P= 0.001) for hybrids (2.77 ± 0.85) than for striped bass (1-30 ± 0.27) when fed the LL diet. When fed the HL diet, mean % gain/d was significantly higher (P = 0.003) for sunshine bass (2.72 ± 0.83) compared to striped bass (1.51 ± 0.25). Additionally, when fed the HL diet, sunshine bass had significantly higher percent nitrogen (P= 0.006) and energy retention (P= 0.014) when compared to striped bass. These results further document that sunshine bass are more efficient at protein and energy retention compared to striped bass in freshwater.  相似文献
