为确诊新疆北屯市某牛场致犊牛肺炎死亡的病原,试验无菌采集2头病死牛肺脏组织分离培养牛支原体,并进行特异性PCR鉴定及oppF基因的序列比对。结果表明:分离获得2株牛支原体,分别命名为M.bovis BT-1和M.bovis BT-2;2株分离株的菌落形态呈典型的"煎蛋样";PCR扩增出牛支原体oppF基因片段,与预期大小一致;2株分离株的oppF基因与国际牛支原体标准株PG45的同源性分别为97.3%和97.8%;与HB0801、CQ-70W在同一进化分支上,亲缘关系较近。  相似文献

国内首次从患肺炎的犊牛肺脏中分离到牛支原体   总被引:23,自引:2,他引:21  
牛支原体(Mycoplasma boris)是引起犊牛肺炎、关节炎和成牛乳房炎的主要病原之一.本研究利用选择性培养基从2份患肺炎的犊牛肺脏病料中得到2株支原体,分别命名为Hubei-1和Hubei-2,并在固体培养基进行了3次克隆纯化.通过生化实验、特异性PCR鉴定和序列比较证明为牛支原体.其16S rRNA 核酸序列与牛支原体国际标准株PG45同源性为99.3%.以该培养物作为包被抗原的ELISA 检测方法对13份发病犊牛血清进行了检测,其中11份血清抗体OD值为阴性血清对照值的2倍以上.该结果首次证实,国内部分地区存在牛支原体的流行.  相似文献

致犊牛肺炎和关节炎牛支原体新疆株的分离与鉴定   总被引:6,自引:5,他引:1  
自2010年7月以来,新疆北疆部分地区某些奶牛场犊牛陆续出现严重的肺炎和关节炎,为确定其病原,无菌采集8个牛场病死犊牛的肺脏和关节液,患病犊牛的血清和部分鼻拭子,病料接种筛选培养基,通过菌落形态观察、特异性PCR鉴定、生长抑制试验和血清学检测,确定牛支原体15株,其中7株牛支原体克隆测序,与PG45 OPP F基因同源性为97.32%~100.00%。牛支原体ELISA试剂盒检测92份血清显示,阳性率82.61%。结果表明,该地区奶牛场发病犊牛以牛支原体感染为主。  相似文献

为调查新疆规模化奶牛场病牛死亡原因并确定病原,本研究无菌采集7份肺炎病死牛病变肺组织样,通过牛支原体液体培养基和固体培养基分离到1株支原体,采用形态学观察和生化试验鉴定该分离株,采用支原体特异性引物和牛支原体16S rRNA通用引物扩增基因序列并测序,使用DNAStar软件将分离菌株测序结果与GenBank中的标准株序列进行同源性比对,采用Mega 6.0软件中的邻接法(Neighbor-Joining,NJ)依据16S rRNA序列构建分离株系统进化树。结果显示,分离株菌落呈典型的"煎蛋样",菌落中心凹陷深入培养基,周边菲薄而透明,经Dienes染液染色后,菌落中心呈深蓝色。该分离株不分解葡萄糖、尿素、不水解精氨酸,血细胞吸附试验和溶血试验均呈阴性,氯化三苯基四氮唑还原反应呈阳性,产生膜和斑。PCR反应扩增出大小为1 911 bp的牛支原体特异性目的片段;分离株16S rRNA基因序列与牛支原体标准株PG45的序列同源性为99.8%,与牛支原体地方株(Mb NM2012、Mb HB0801、Mb Hubei-1、Mb Ningxia-1、Mb CQ-W70和Mb 08M)的同源性为99.3%~99.7%。系统进化树显示,分离株16S rRNA基因与Mb Ningxia-1株和Mb 08M株亲缘关系较近,处于同一分支。本研究结果证实了引起病牛死亡的病原为牛支原体,为新疆牛支原体病的防治提供了科学依据。  相似文献

为研发用于预防犊牛支原体肺炎及关节炎灭活油佐剂疫苗并评价其免疫效果,本研究采用牛支原体(Mycoplasma bovis,M.bovis)新疆分离株做为制苗菌株,经牛支原体专用液体扩大培养基培养后进行膜分离浓缩、甲醛灭活,加入油佐剂乳化制备牛支原体灭活疫苗,经无菌检验、实验动物安全性检验及乳化效果检测后进行犊牛免疫效果测定。结果显示,免疫组犊牛在二免后14 d,血清中M.bovis抗体达到0.465(OD_(450)),3头攻毒犊牛均保持良好的精神状态且体温变化不明显,临床症状综合评分为3,肺脏解剖学及病理组织学观察无异常,肺脏病变指数为2且肺脏中未分离回收到M.bovis;未免疫对照组犊牛同期血清M.bovis抗体为0.142(OD_(450)),3头攻毒犊牛均先后出现体温升高、轻度咳喘、脓性鼻液、明显消瘦等症状,临床症状综合评分为11,肺脏病变指数为17,肺脏尖叶、心叶及部分膈叶均表现明显肝变,2头犊牛表现胸膜与肺脏黏连,1头犊牛整个尖叶广泛分布黄白色蚕豆状大小坏死性结节,与自然感染病例相同,病理组织学观察显示肺泡腔和支气管中有大量中性粒细胞浸润,支气管管壁充血、水肿,肺脏部分坏死灶呈均质红染,为典型的化脓性及坏死性肺炎变化,从病变肺组织中均分离回收到M.bovis。结果表明,本研究制备的牛支原体灭活疫苗给犊牛免疫后能产生良好的免疫应答反应,并能抵抗M.bovis感染所致的肺脏病变作用。  相似文献

牛支原体(Mycoplasma bovis)是引起犊牛肺炎、关节炎和成牛乳房炎的主要病原之一。从湖北恩施暴发牛支原体的肉牛场(70头引种肉牛出现20%的死亡)分离得到一株牛支原体,命名为HB2015,并对其进行了保藏(保藏号:CCTCC M 2016559),进化分析结果显示该菌株与M.bovis NM2012内蒙株亲缘关系较近,为牛支原体的防控提供了理论依据。  相似文献

牛呼吸道疾病综合征病例的病原分析   总被引:1,自引:0,他引:1  
本试验旨在查清广西某牛场1起牛呼吸道疾病综合征(BRDC)病例的病原,指导牛场进行疾病防控。采取现场调查、临床症状与病理变化、病原分离鉴定等方法对病例病原进行分析,根据病原药敏试验结果进行治疗。从病例的肺脏组织中分离到1株支原体和1株革兰氏阴性致病杆菌。支原体分离株在PPLO固体培养基上可见典型的"煎蛋样"菌落,PCR扩增出牛支原体oppF基因特异性的448 bp目的片段,其oppF基因序列与美国分离的牛支原体国际标准株PG45的核苷酸序列同源性为98.4%。革兰氏阴性细菌分离株生化特性符合黏质沙雷氏菌特性,其16S rRNA基因PCR扩增出1 400 bp的目的片段,测序结果与GenBank上登录的黏质沙雷氏菌的核苷酸序列同源性达到99.0%,对小鼠具有致病性。牛支原体和黏质沙雷氏菌分离株均对壮观霉素、阿奇霉素、阿米卡星、庆大霉素和新霉素高度敏感,用高敏药物壮观霉素联合地塞米松等相关措施进行治疗,收到良好效果。结果表明,引起这次牛呼吸道疾病综合征的病原为牛支原体和黏质沙雷氏菌。  相似文献

对采集到的疑似牛支原体肺炎肺组织病料进行病原的分离,并对分离株进行形态学、生化和分子生物学鉴定,结果显示成功分离获得1株牛支原体,命名为NM001。该分离株的菌落形态呈典型的“荷包蛋状”,不能发酵葡萄糖,不能水解精氨酸,不分解尿素。PCR能够扩增出牛支原体特异的P48基因条带,16S rRNA基因序列与Ningxia-1序列同源性为99.03%。将该分离株接种2头6月龄犊牛均出现明显的临床症状,剖检后胸腔中少量淡黄色渗出液,肺脏出现肉样实变。试验结果表明,分离到的牛支原体NM001株对牛具有较强的致病性,为牛支原体攻毒模型的建立奠定了基础。  相似文献

新疆南疆地区某奶牛场3月龄犊牛陆续出现咳嗽、气喘、黏性鼻液等临床症状,发病死亡数只,怀疑为支原体感染引起。为确定病原,本研究采集发病犊牛鼻拭子30份、病死犊牛肺脏1份,提取基因组DNA进行PCR检测。同时,使用支原体培养基进行支原体分离,对纯化后单菌落进行分子生物学鉴定。结果显示,病牛鼻拭子中牛鼻支原体PCR阳性率为70%,从病死牛肺脏组织中分离到1株牛鼻支原体,未检测、分离到其他支原体。  相似文献

为确诊银川市某奶牛场成年奶牛病因,本研究对两头表现明显临床症状的犊牛进行解剖,无菌采集肺脏、肺积液、关节液及患临床型乳房炎的奶牛乳样1份进行病原分离,分离到2株巴士杆菌、2株链球菌、1株大肠杆菌和3株支原体,通过革兰氏染色、菌落形态观察、特异性PCR鉴定和16S r RNA序列比对,结果显示引起此次流行的主要病原为牛支原体。采用比利时进口牛支原体ELISA诊断试剂盒对银川地区12个规模化奶牛场的230份奶牛血清进行牛支原体血清学调查,结果显示牛群平均阳性率26.52%,牛场阳性率高达100%,表明牛支原体病在银川地区呈高感染率。  相似文献

Nasal swabs of 293 calves were examined for Mycoplasma. The samples were collected from calves affected with respiratory diseases on 71 farms in various parts of Japan between 1996 and 1997. Mycoplasma bovirhinis was isolated from 47 of 293 calves (16.0%). Mycoplasma alkalescens, M. bovis, M. arginini, M. bovigenitalium and Acholeplasma spp. were isolated from 19 (6.5%), seven (2.4%), four (1.4%), four (1.4%) and 18 (6.1%) calves, respectively. Pasteurella multocida and P. haemolytica were isolated from 60% of Mycoplasma-positive calves. However, other bacteria were not isolated from calves. To evaluate the antimicrobial susceptibility of their isolates, 68 M. bovirhinis, 21 M. alkalescens and 10 M. bovis strains were examined for 12 antimicrobial agents. All isolates showed higher susceptibility to tiamulin than to the other drugs used in the study. However, erythromycin had no effect on any of the Mycoplasma strains studied. The field isolates were less susceptible than the type strains to some drugs, such as spiramycin, oxytetracycline and tylosin.  相似文献

Mycoplasma bovis is perceived as an emerging cause of mortality in feedlot beef cattle. This study examined the lesions and infectious agents in naturally occurring M. bovis-associated bronchopneumonia and arthritis and the relationship of this condition with bovine viral diarrhea virus (BVDV) infection. Standardized pathologic, immunohistochemical, and microbiologic investigations were conducted on 99 calves that died or were euthanized within 60 days after arrival in 72 feedlots. Cranioventral bronchopneumonia with multiple foci of caseous necrosis was identified in 54 of 99 calves, including 30 with concurrent fibrinosuppurative bronchopneumonia typical of pneumonic pasteurellosis. Mycoplasma bovis was consistently identified in these lesions by culture and immunohistochemistry, but also commonly in healthy lungs and those with pneumonia of other causes. Focal lesions of coagulation necrosis, typical of pneumonic pasteurellosis, were often infected with both Mannheimia haemolytica and M. bovis. Arthritis was present in 25 of 54 (46%) calves with M. bovis pneumonia, and all calves with arthritis had pneumonia. BVDV infection was more common in calves with lesions of bacterial pneumonia than in those dying of other causes, but BVDV infection was not more common in calves with caseonecrotic bronchopneumonia than those with fibrinosuppurative bronchopneumonia. Retrospective analysis identified cases of M. bovis pneumonia in the early 1980s that had milder lesions than the current cases. The findings suggest that, in at least some calves, M. bovis induces caseonecrotic bronchopneumonia within the lesions of pneumonic pasteurellosis.  相似文献

The prevalence of mycoplasmas in the respiratory tracts of 148 pneumonic calves originating from 25 herds in the Netherlands is reported. Four types of culture media were used to isolate mycoplasmas: solid modified Edward medium, 2 types of Friis media, and A7B differential agar medium. Mycoplasmas were isolated both from nasal swab specimens and lung lavage fluids collected from live calves and from nasal mucosa and lung tissue specimens collected post mortem. All of the mycoplasma strains isolated could be identified as either Ureaplasma diversum (isolated from 80% of 25 herds), Mycoplasma dispar (92%), M. bovirhinis (88%), M. bovis (20%), M. bovigenitalium (4%), M. arginini (16%), or M. canis (12%). Isolation rates of M. dispar and U. diversum were considerably higher from lung lavage fluids than from nasal swab specimens. M. bovis was detected only in fattening herds and not in dairy herds. The respiratory tracts of 75% of the calves examined contained at least 2 mycoplasma species. In total, 25 different combinations of mycoplasma species were detected in specimens collected from noses and lungs. The pathogenic species U. diversum and M. dispar had not been isolated before in the Netherlands.  相似文献

A pure culture of Mycoplasma (M.) bovis was isolated from calves with respiratory disease, exhibiting the picture of lymphohistiocytic proliferative pneumonia with presence of eosinophil plasmatic cells. A mixed infection of M. bovis and Pasteurella (P.) multocida was demonstrated in calves with exudative pneumonia. Both M. bovis and Haemophilus (H.) somnus were recovered from calves with necrotic pneumonia. All 3 organisms--M. bovis, P. multocida, and H. somnus--were present in cases of exudative-necrotic pneumonia. It was also shown that M. bovis played a primary role in the aetiopathogenesis of respiratory diseases caused by mixed infections.  相似文献

Multiple intranasal experimental Mycoplasma (M.) bovis infection was induced to 22 calves and resulted in clinical manifestations. M. bovis was re-isolated from 17 of 24 lungs which had been altered by pneumonia. Pasteurella haemolytica was the only pathogen isolated from the pathologically affected lung of one of the experimental animals. The haematogenic proliferation phase was successfully identified in 5 of 8 examined calves. Arthritis was not recordable from any of the experimental animals. Interstitial and catarrhal to catarrhal-purulent bronchopneumonias were the most common histological findings. Peribronchial lymphoid cell accumulations occurred frequently, as time passed from onset of infection. Antibodies against M. bovis were detected in blood serum of infected animals by means of film inhibition, as of the 3. week from infection, and by means of indirect haemagglutination, as of the 4. week from infection.  相似文献

The aim of this study was to identify morphometric histological features of pneumonia caused by Mycoplasma bovis in calves. Eight three-month-old calves were infected with M. bovis and samples of their lung tissue, three weeks after exposure, compared to samples from four uninfected calves. In the uninfected animals the goblet cells were clustered in the crypt area of the epithelial folds, while in the infected calves they had migrated towards the tips of the folds and were distributed evenly throughout the folds. In infected lung tissue there was goblet cell hyperplasia and metaplasia in the bronchioles and an increased epithelial height. Goblet cell mucin in uninfected calves was acidic, but in infected calves most goblet cells contained neutral mucins. These morphometric and histochemical bronco-epithelial changes may be able to be used as markers of the severity of bovine respiratory mycoplasmosis.  相似文献
