Cryopreservation of silver barb Puntius gonionotus (Bleeker) spermatozoa: effect of extender composition,cryoprotective agents and freezing rate on their postthawing fertilization ability

The effects of extender composition, cryoprotectant concentration and freezing and thawing on the fertilization efficiency of cryopreserved spermatozoa of Puntius gonionotus were evaluated. Computer‐aided motility analysis of semen was conducted to check the suitability of spermatozoa for cryopreservation after mixing with different extenders and cryoprotective agents (CPAs). Extender‐4 with an osmolality 260 mOsmol kg⁻¹and pH 7.6 was used for the cryopreservation study. Among the CPAs, dimethyl sulphoxide (DMSO) was least toxic and more than 60% fertilization was achieved when used at 1.4 M at 0 °C for 10 and 30 min, whereas the toxicity of all CPAs to spermatozoa was evident when tested at 30 °C. Semen frozen at −16 °C min⁻¹ with 1.4 M DMSO showed 70% fertilization, which was significantly higher (P<0.05) than other freezing rates. Samples thawed at 35 °C water showed a fertilization rate comparable with that of fresh semen. Computer‐assisted semen analysis of fresh and frozen semen after thawing showed variations in different types of motility in spermatozoa and in their class. There was no significant difference in motility before or after cryopreservation; however, significant differences could be observed in the average path velocity (VAP), straight line velocity (VSL) and curve linear velocity (VCL). Semen of silver barb could be cryopreserved with extender‐4 by addition of 1.4 M DMSO to a final cryopreservation medium (MED 2) cooled at a rate of −16 °C min⁻¹, stored in liquid nitrogen (−196 °C) and utilized after thawing at 35±2 °C.  相似文献

Effect of Lentil Residues on the Yield and Response of Succeeding Rice to Nitrogen

A field experiment was conducted at the Division of Agronomy, Indian Agricultural Research Institute, New Delhi during 1983-84 and 1984-85 to study the effect of lentil residues on the yield and response of succeeding rice to nitrogen. Lentil residues benefitted the succeeding rice and gave longer and heavier panicles, more grains per panicle, higher 1000-grain weight and higher grain and straw yield of rice as compared to fallow. Rice responded well to applied nitrogen both in the absence and presence of lentil residues. The effect of lentil root residues and root residues plus incorporation of lentil straw was equivalent to 11 and 33 kg N ha⁻¹ when 60 kg N ha⁻¹ was applied to rice; the corresponding values being 54 and 60 kg N ha⁻¹ when 120 kg N ha⁻¹ was applied to rice.  相似文献

The contribution of the maternal inbreeding to the depression of characters was more pronounced on 1-d-old chicks, but declined as the chicks grew. 3. The body weight taken near sexual maturity showed a low and non-significant decline. 4. Glyceryl guaiacolate was used with thiopentone sodium for general anaesthesia in 10 clinically healthy male buffalo calves. When the animals were in the stage of surgical anaesthesia, there was marked hypotension and tachycardia along with a decrease in central venous pressure, tidal volume, partial pressure of oxygen in arterial blood and venous plasma potassium. Hyperglycaemia was another consistent feature. The changes in the acid base status were mild and inconsistent.  相似文献

Mouse neuroblastoma tumors have only the fifth isozyme band (A(4)) of lactate dehydrogenase, whereas this band is missing in the brain which contained four other bands of lactate dehydrogenase. The alpha-esterase isozyme patterns of tumors, kidney, and brain are similar except that there is an additional slowest-moving form of esterase in all tumor tissues. A field experiment conducted at the Indian Agricultural Research Institute, New Delhi brought out that summer maize green fodder yield was 31.8 t/ha to 34.1 t/ha, while that of cowpea fodder was only 15.5 to 16 t/ ha.
