Cryopreservation of semen collected from dead fishes showed that it is possible to use it for fish production. We humanely killed individuals of the Indian major carp rohu, Labeo rohita and stored at different temperature regimes of 31 °C, 0 °C, − 10 °C and − 30 °C till 8 h. At every one hour interval semen from these fishes were collected by Pasteur pipette and evaluated for sperm yield/kg body weight, motility, pH, spermatocrit (%) and sperm count. The semen having suitable characteristics such as; 70% spermatocrit or above and motility index of 4 or above was cryopreserved by following a specific protocol. The cryopreserved semen of the dead fishes was stored for 7 days and then thawed in a water bath at 37 °C for 50 s. It was found that up to 8 h, spermatozoa of rohu were viable when stored at 0 °C or − 10 °C. Sperm collected after 8 h of fish death and maintained at 0 °C was the best stored condition that showed 30% larval survival. The spermatozoa collected 8 h after fish death was mostly normal as observed under scanning electron microscope and the total length of rohu spermatozoa was 25-30 μm. The hatchlings produced with this cryopreserved semen grew normally and juvenile fishes of rohu could be produced. This study suggests that germ cells such as spermatozoa of dead fishes can be cryopreserved and utilized for restoration of a species. It has the potential use in cryo-conservation of endangered fishes, restoration of animals through fertilization and genetic manipulation studies.  相似文献

The effects of extender composition, cryoprotectant concentration and freezing and thawing on the fertilization efficiency of cryopreserved spermatozoa of Puntius gonionotus were evaluated. Computer‐aided motility analysis of semen was conducted to check the suitability of spermatozoa for cryopreservation after mixing with different extenders and cryoprotective agents (CPAs). Extender‐4 with an osmolality 260 mOsmol kg−1and pH 7.6 was used for the cryopreservation study. Among the CPAs, dimethyl sulphoxide (DMSO) was least toxic and more than 60% fertilization was achieved when used at 1.4 M at 0 °C for 10 and 30 min, whereas the toxicity of all CPAs to spermatozoa was evident when tested at 30 °C. Semen frozen at −16 °C min−1 with 1.4 M DMSO showed 70% fertilization, which was significantly higher (P<0.05) than other freezing rates. Samples thawed at 35 °C water showed a fertilization rate comparable with that of fresh semen. Computer‐assisted semen analysis of fresh and frozen semen after thawing showed variations in different types of motility in spermatozoa and in their class. There was no significant difference in motility before or after cryopreservation; however, significant differences could be observed in the average path velocity (VAP), straight line velocity (VSL) and curve linear velocity (VCL). Semen of silver barb could be cryopreserved with extender‐4 by addition of 1.4 M DMSO to a final cryopreservation medium (MED 2) cooled at a rate of −16 °C min−1, stored in liquid nitrogen (−196 °C) and utilized after thawing at 35±2 °C.  相似文献

The fish-seed production industry in India has recorded remarkable growth over the last three decades. The hypophysation technique was successfully introduced into India in 1957 and steady progress towards the refinement of the technique has been registered, which has revolutionized carp seed production in the Indian subcontinent. Advancement of carp maturity through brood stock management and multiple breeding has enabled spawn production well ahead of the monsoon and even beyond, ensuring seed availability throughout the year. The quality of seed is an important consideration for commercial aquaculturists. So, partial stock replenishment in carp hatcheries is practised to overcome the problem of inbreeding, which otherwise leads to poor growth of carps. Similarly, gametes of improved stock are cryopreserved and utilized for quality seed production as well as upgrading the brood stock of carps. The gamete cryopreservation protocol for carps is the focus of this review. An attempt has also been made to incorporate information on carp brood stock management, inducing agents, and seed production, including hatchery management.  相似文献

The viability of matured oocytes stored in vitro were assessed using carp ovarian fluid (OF) and artificial carp ovarian fluid (ACOF) under different temperature regimes (4, 24, 26, 28 and 30°C) for different storage durations (0, 60, 120, 150 and 180 min). Significantly higher fertilization (74%) was achieved when the oocytes were stored using ACOF and 65% in OF after 180 min at 28°C. Similarly the hatching rates of larvae were significantly higher in the ACOF and OF, that is, 64% and 47%, respectively, after 180 min of storage. The oocytes kept in the storage containers with ACOF having 65% moisture level showed a significantly higher fertilization rate than the 59% moisture level. This study demonstrated that unfertilized matured oocytes (eggs) of rohu can be stored in vitro for 180 min without compromising the viability (fertilization and hatching) to a great extent in OF and ACOF.  相似文献

Seed samples of 27 landraces of wheat were collected from farmers’ fields of hilly areas of Himalaya in Uttaranchal state of India during April 2004. Genetic diversity among 41 genotypes (cultivars and landraces of wheat) was studied using morphological traits, microsatellite markers and SDS-PAGE of HMW-GS. The dendrogram and PCA (Principal Component Analysis) based on morphological data clearly separated landraces of wheat from cultivars. In the dendrogram based on microsatellite markers data all the wheat cultivars released after the introduction of high yielding dwarf wheat varieties from CIMMYT, used in this study, were grouped separately with the exception of NP4. The pre-green revolution indigenous varieties grouped with landraces suggesting that the same had been probably developed through selection among landraces in India. The landraces had higher diversity for HMW-glutenin subunits coded by Glu-B1, with distinct subunit combinations 6 + 8, 7 + 9, 13 + 16, than within the wheat cultivars analyzed. Most of the landraces except IITR10 and IITR14 are clearly distinct from the indigenous and modern wheat cultivars released in India in the 20th century. More than half of the landraces were heterogeneous mixture of plants with different glume color, awnness, grain color and HMW-GS profile and hence need purification through single plant selection. Some of the landraces with resistance to yellow rust and powdery mildew and distinct HMW-GS subunits can be used in appropriate breeding programs. It will be desirable to conserve and protect the landraces as geographical indications of Uttaranchal.  相似文献

Soil erosion is one form of land degradation, which is caused by the interacting effects of numerous factors such as biophysical characteristics and socio‐economic condition of a particular watershed. Previous erosion studies focused on the use of soil erosion models (e.g. USLE, EUROSEM, SLEMSA etc.), which have been developed under local conditions (e.g. United States, Europe, Africa, etc) and mostly use only biophysical factors as inputs to the models. In this study, a methodology that integrates both biophysical and socio‐economic aspects into a framework for soil erosion hazard assessment using principal component analysis (PCA) is described. The analysis is done at the land unit level. With the particular conditions of the study area that is characterized by Inceptisols and Alfisols soil types, nine different land uses with mixed vegetation and forest area dominant in the steep slope, high annual rainfall (>2500 mm), high population with mostly low income and low education, were considered. These were used in formulating a soil erosion hazard index (EHI) equation which relates a number of key factors consisting of biophysical and socio‐economic variables, namely soil texture, slope steepness, land cover, soil conservation practices, income and farmers' knowledge. Weighting and scoring of these key factors were used to develop the EHI equation and to calculate an index value of erosion hazard for every land unit. Results indicate that more than 60% of the area has erosion hazard ranging from moderate to very severe, and most of the land units with high erosion hazard were found at the mountain areas. It was also found that erosion hazard was severe in areas with high silt content, followed by high rainfall and steep slope, low crop cover without any soil conservation practices coupled with lack of awareness on soil erosion and low income. The key factors identified and level of erosion hazard obtained can be used to formulate conservation measures in critical areas which are prone to soil erosion. This study aimed at developing a suitable cryopreservation protocol for embryonic stem (ES)‐like cells of a tiny freshwater fish Leopard danio (Brachydanio frankei). Embryonic stem (ES)‐like cells derived from blastomeres of the early blastulae stage of the developing embryo were cultured in vitro in a medium containing Leibowitz‐15 supplemented with 10% foetal bovine serum, leopard danio embryo extract, sodium bicarbonate, sodium selenite, basic fibroblast growth factor, epidermal growth factor and leukaemia inhibitory factor. The ES‐like cells showed properties similar to ES cells in other species. They were morphologically small, round to polygonal and present in patches and extensively expressed alkaline phosphatase and stage‐specific embryonic antigen. The toxicity and chilling sensitivity of these cells were determined using ethylene glycol (EG), propylene glycol (PG) and glycerol as cryoprotective agents at molar concentrations of 0.6, 1.0, 1.4, 1.8 and 2.0. Among them, 1.8 M EG showed 70% significant viable ES‐like cells (P<0.05). The post‐thawed cells retained similar properties of non‐cryopreserved ES‐like cells with a viability rate of 65%. Similarly, blastomeres cryopreserved following the slow cooling rate with EG and PG yielded a viability of more than 70%.  相似文献
