The formation of glutaminyl transfer RNA (Gln-tRNA(Gln)) differs among the three domains of life. Most bacteria employ an indirect pathway to produce Gln-tRNA(Gln) by a heterotrimeric glutamine amidotransferase CAB (GatCAB) that acts on the misacylated Glu-tRNA(Gln). Here, we describe a series of crystal structures of intact GatCAB from Staphylococcus aureus in the apo form and in the complexes with glutamine, asparagine, Mn2+, and adenosine triphosphate analog. Two identified catalytic centers for the glutaminase and transamidase reactions are markedly distant but connected by a hydrophilic ammonia channel 30 A in length. Further, we show that the first U-A base pair in the acceptor stem and the D loop of tRNA(Gln) serve as identity elements essential for discrimination by GatCAB and propose a complete model for the overall concerted reactions to synthesize Gln-tRNA(Gln).  相似文献

Bovine lactoferrin was administered orally, in feed, to six bottlenosed dolphins (Tursiops truncatus) before they were transported for approximately six hours; their stress responses were compared with those of five untreated dolphins. During the journey the dolphins had an increased plasma concentration of cortisol, and lymphopenia, eosinopenia and mild neutrophilia, indicating a stress response. The administration of lactoferrin did not affect the function of the dolphins' polymorphonuclear leucocytes, but affected their leucogram by maintaining the number of circulating eosinophils.  相似文献

OBJECTIVE: To evaluate the efficacy of percutaneous administration of iohexol into the popliteal lymph node as a non-invasive technique for thoracic duct lymphangiography in dogs. STUDY DESIGN: Experimental study and clinical report. ANIMALS: Normal adult dogs (n=4) and 1 dog with recurrent chylothorax. METHODS: For the experimental study, 4 dogs (weight, 8.4-12.3 kg) had 5-10 mL iohexol injected percutaneously into 1 popliteal lymph node and then thoracic radiographs were taken. Popliteal lymph nodes were examined by histopathology 8 days later. One 25-kg dog with recurrent chylothorax had 25 mL iohexol injected into the right popliteal lymph node followed by thoracic radiography. RESULTS: In experimental dogs, the thoracic duct was best visualized on thoracic radiographs after administration of 10 mL iohexol. Clinically, no abnormalities were identified in the injected limb and except for 1 dog that had large numbers of siderocytes and erythrophagocytic macrophages in the injected lymph node, the histopathologic findings in the other injected popliteal lymph nodes were not different from contralateral nodes. In the clinical case, the thoracic duct was visualized, but there was leakage of iohexol around the node. CONCLUSION: The thoracic duct in dogs can be visualized by lymphography after percutaneous injection of iohexol (1 mL/kg at 2 mL/min) into the popliteal lymph node. CLINICAL RELEVANCE: Percutaneous popliteal lymph node administration of iohexol should be considered as an alternative to mesenteric lymph node injection for radiographic identification of the thoracic duct in dogs.  相似文献

Neoplasms of skeletal muscle origin are very rare in the rat. Recently, we experienced a case of rhabdomyosarcoma as a white mass involving the junction of the esophagus and stomach in the abdominal cavity of a 12-month-old female Donryu rat. Histopathologically, the neoplastic cells composing the mass invasively spreaded from the lamina propia to the tunica serosa in the stomach as well as the esophagus. Although the neoplastic cells varied in appearance, pleomorphic atypical cells with abundant eosinophilic cytoplasm were prominent. Some tumor cells were stained blue with phosphotungstic acid hematoxylin. The nuclei of spindle-shaped neoplastic cells were arranged longitudinally like beads. Multinucleate giant cells and mitotic figures were also frequently observed. Immunohistochemically, these neoplastic cells were positive for desmin and myoglobin, whereas they were negative for alpha-smooth muscle actin. Taken together these findings, this tumor was diagnosed as a pleomorphic rhabdomyosarcoma, probably derived from the muscle layer of the lower part of the esophagus. This is the first report of rhabdomyosarcoma in a Donryu rat.  相似文献

The objective was to evaluate effects of feeding level of milk replacer on body growth, plasma metabolite and insulin concentrations, and allometric growth of visceral organs in suckling calves. Holstein bull calves (n = 8; 3–4 days of age) were fed either a low amount (average 0.63 kgDM/day, LM) or high amount (average 1.15 kgDM/day, HM) of high protein milk replacer until they were slaughtered at 6 weeks of age. Body weight (BW) at 4, 5, and 6 weeks of age, feed intake, average daily gain, and feed efficiency were higher in the HM than LM calves. The HM group had higher plasma glucose at 3 and 4 weeks of age and insulin levels after the age of 4 weeks compared with LM calves whereas no effect was detected on plasma nonesterified fatty acid or urea nitrogen concentrations. The HM calves had greater empty body weight (EBW), viscera‐free BW and most of the organs dissected than LM calves. Relative weights (% of EBW) of liver, spleen, kidneys, and internal fat were higher, whereas head and large intestine was lower in HM than LM calves. The results suggest that increased milk feeding levels would accelerate the growth of the body and specific organs.  相似文献

OBJECTIVE: To determine whether cross-reactivity exists between canine chromogranin A (CgA) and anti-human CgA antibody and investigate the usefulness of plasma CgA concentration measurements as an index of acute stress responses in dogs. ANIMALS: 12 healthy Beagles. PROCEDURE: Canine CgA was extracted and purified from canine adrenal glands of cadaver dogs for studying cross-reactivity with anti-human CgA antibody. Western blotting with anti-human CgA antibody was performed. Blood samples were collected from dogs at 0, 10, 20, 30, 40, 60, 120, and 180 minutes after IV administration of saline (0.9% NaCl) solution or insulin. Canine plasma CgA concentrations were determined by use of a CgA ELISA kit with rabbit antiserum against the carboxy-terminal fragment of human CgA. Plasma cortisol and catecholamine (ie, norepinephrine and epinephrine) concentrations were measured by use of an ELISA and a high-performance liquid chromatography method, respectively. RESULTS: Purified canine CgA was specifically detected by use of western blot analysis and an ELISA with anti-human CgA antibody. An increase in plasma CgA concentrations was observed in insulin-induced hypoglycemic dogs. Changes in plasma CgA concentration were correlated with changes in plasma cortisol or catecholamine concentrations of hypoglycemic dogs. CONCLUSIONS AND CLINICAL RELEVANCE: Use of the CgA ELISA kit for determination of human plasma CgA concentrations is applicable to the measurement of canine plasma CgA concentrations. Canine plasma CgA concentrations, along with measurements of plasma cortisol and catecholamine concentrations, correctly reflect insulin-induced hypoglycemic stressed conditions in dogs. Measurement of canine plasma CgA concentrations may provide a useful index for evaluation of an acute stress response.  相似文献

The immunomodulatory effects of phagocytic cells in common carp Cyprinus carpio L., by alpha melanocyte stimulating hormone (alpha-MSH) was analyzed in vitro. Carp head kidney leucocytes were cultured in RPMI 1640 medium containing 0.05, 0.1, 1 and 10 nM alpha-MSH and the production of superoxide anion was measured via the reduction of nitroblue tetrazolium (NBT) in vitro. Macrophages incubated with alpha-MSH showed an increase in the production of superoxide anion in comparison to control macrophages cultured without hormone. Phagocytic cells treated with alpha-MSH also displayed increased phagocytosis. Furthermore, carp lymphocytes treated with alpha-MSH increased the mitogenic responses to phytohaemoagglutinin (PHA-P). These results show that alpha-MSH directly influences fish immune responses.  相似文献

The effect of dietary energy levels on the urinary excretion of purine derivatives (PD) was investigated in sheep fed hay and concentrate diets. In addition, the possibility of using PD concentrations in spot samples of urine as a useful tool for determining daily PD excretion was also examined. Three female lambs (mean bodyweight (BW) ± standard error, 31.4 ± 1.0 kg) were fed three diets consisting of timothy hay (20 g dry matter/kg BW per day) and rolled barley at three different energy levels (low‐energy (LE), medium‐energy (ME) and high‐energy (HE) at 1.0‐, 1.2‐ and 1.5‐fold greater than the maintenance level of energy (450 KJ/BW0.75/day), respectively) in a 3 × 3 Latin square design. The dietary protein level was almost constant (380 mg nitrogen/BW0.75/day as digestible crude protein base) and was adjusted using soybean protein. Digestible organic matter intake (DOMI) increased (P < 0.05) with an increase in the dietary energy level and there was a difference (P < 0.05) in the organic matter digestibility between the LE and HE diets. For urinary PD excretion, the differences between the LE and ME diets and the HE diet were significant (P < 0.05), although there was no clear difference between the LE and ME diets. There was a positive correlation (r = 0.47, P < 0.05) between urinary PD excretion and DOMI. There was also a correlation (r = 0.59, P < 0.05) between the urinary PD level at 7–8 h after the morning feed and the daily total PD excretion, suggesting that the PD level in spot urine samples could be useful for estimating microbial protein synthesized in the rumen and/or digested in the lower gut.  相似文献

Porphyra (Bangiales, Rhodophyta), which includes several valuable marine crops, has recently received great interest as a model plant for fundamental and applied studies in marine sciences. Amplified fragment length polymorphisms (AFLPs) are a robust and efficient means for genetic mapping, linkage analysis of genetic characters for breeding and population studies in land plant genomes. To examine whether AFLPs are applicable as genetic markers in the present study, we detected AFLP markers with three pure lines in order to promote genetic analysis in Porphyra yezoensis . The following five sets of AFLP primer pairs (E-AA, M-CAA) (E-AA, M-CAC) (E-AA, M-CAG) (E-AA, M-CAT) (E-AA, M-CTA) were tested with template DNAs from three pure lines and they showed a total of 227 bands. This suggests that AFLP markers are promising tools for genetic analysis in Porphyra .  相似文献
