The aim of this study was to test the influence of post ‐ thaw storage time on the duration of sperm motility, percentage of motile sperm, and fertilization and hatching rates of fresh sperm and sperm stored for 0, 30 and 60 min at 4°C post‐thawing. After being frozen in liquid nitrogen and then thawed, the percentage of motile sperm and duration of motility were not affected by 30 min of storage at 4°C, whereas a significant decline in these parameters was observed after 60 min of storage. Similarly, fertilization and hatching rates were significantly affected within 60 min of storage at 4°C, and the fertility of frozen‐thawed sperm was significantly lower than that of fresh sperm. We conclude that cryopreserved sperm of beluga sturgeon could be stored for 30 min without the loss of sperm quality. This described procedure for beluga sturgeon cryopreservation is reliable and efficient and therefore can be recommended for hatchery practice after scaling up this technique. 相似文献

The objectives were (i) to evaluate the effect of temperament, determined by modified 2‐point chute exit and gait score, on artificial insemination (AI) pregnancy rates in beef heifers following fixed time AI and (ii) to determine the effect of temperament on cortisol, substance‐P, prolactin and progesterone at initiation of synchronization and at the time of AI. Angus beef heifers (n = 967) at eight locations were included in this study. At the initiation of synchronization (Day 0 = initiation of synchronization), all heifers received a body condition score (BCS), and temperament score (0 = calm; slow exit and walk or 1 = excitable; fast exit or jump or trot or run). Blood samples were collected from a sub‐population of heifers (n = 86) at both synchronization initiation and the time of AI to determine the differences in serum progesterone, cortisol, prolactin and substance‐P concentrations between temperament groups. Heifers were synchronized with 5‐day CO‐Synch+ controlled internal drug release (CIDR) protocol and were inseminated at 56 h after CIDR removal. Heifers were examined for pregnancy by ultrasound 70 days after AI to determine AI pregnancy. Controlling for synchronization treatment (p = 0.03), facility design (p = 0.05), and cattle handling facility design by temperament score interaction (p = 0.02), the AI pregnancy differed between heifers with excitable and calm temperament (51.9% vs 60.3%; p = 0.01). The alley‐way with acute bends and turns, and long straight alley‐way had lower AI pregnancy rate than did the semicircular alley‐way (53.5%, 56.3% and 67.0% respectively; p = 0.05). The serum hormone concentrations differed significantly between different types of cattle handling facility (p < 0.05). The cattle handling facility design by temperament group interactions significantly influenced progesterone (p = 0.01), cortisol (p = 0.01), prolactin (p = 0.02) and substance‐P (p = 0.04) both at the initiation of synchronization and at the time of AI. Inter‐ and intra‐rater agreement for temperament scoring were moderate and good (Kappa = 0.596 ± 0.07 and 0.797 ± 0.11) respectively. The predictive value for calm and pregnant to AI was 0.87, and excited and non‐pregnant to AI was 0.76. In conclusion, the modified 2‐point temperament scoring method can be used to identify heifers with excitable temperament. Heifers with excitable temperament had lower AI pregnancy. Exposure to environmental levels of the sheep-dip insecticides cypermethrin and diazinon inhibited embryo development in Atlantic salmon. Brief exposure to eggs and milt during fertilisation affected both the number of fry that emerged and the time of emergence. Following insecticide exposure there was a higher incidence of fry deformities and a reduction in the survival of the fry. First-feeding chinook salmon were fed either live adult Artemia franciscana or commercial feed over a 15-week period. Unexplained mortality began occurring in the Artemia-fed fish after 35 days on the diets, with cumulative mortality reaching 34.5% in the Artemia-fed fish compared with 4.3% in feed-fed fish. Necropsy examinations revealed systemic fungal yeast infections and the causative agent was cultured from kidneys. Fungal cells were observed in the transport water of Artemia shipments and within the Artemia. The Artemia were purchased from a retail supplier, and originated from salt ponds in San Francisco Bay (SFB), California. Artemia infection rates ranged from 0.5% to 37.5% (mean 16.5±3.59%) in shipments received over a 5-week period. The fungus was characterized by morphological and physiological properties and was identified as Metschnikowia sp., a pathogenic yeast of aquatic invertebrates. The yeast grew at 9–27 °C, and 0–180 ppt NaCl, and could tolerate salinity of at least 270 ppt. Sequence analysis of the divergent D1/D2 domain of ascomycetous yeast 26S ribosomal DNA identified the organism as Metschnikowia bicuspidata var. bicuspidata. The organism was found in water from the salt ponds and probably entered from the bay. Mortality due to the fungal infection stopped after changes were made in the handling of incoming Artemia shipments. 相似文献

Previous research has generally not found a strong relationship between participation in outdoor recreation and environmental concern. Research has suggested, however, that the motivation underlying recreation can mediate this relationship for some nature-based recreation groups. We hypothesized that the motivation underlying participation in birding mediates the participation–environmental concern relationship. We collected data via mail-back surveys from 529 American Birding Association members. Using structural modeling, we compared a direct effects model with partial and full mediation models with motivation variables as the mediators. The partial mediation model was weakly supported for two out of three motivation variables. Fit indices showed all models to have less than adequate fit. The results suggested that participation may not lead to environmental concern and that other variables, including motivation, may be more influential. 相似文献
