Azostix-reagent-tests(R) strips (Ames, Miles, Inc., Diagnostic Division, Elkhart, IN) were used to measure blood urea nitrogen values in blood samples from 125 dogs and cats at the North Carolina State University, College of Veterinary Medicine. Results of the tests were compared with standard serum urea nitrogen results. Sensitivity, specificity, and negative predictive values were high (86.4, 90.3, and 96.5%, respectively). Positive predictive value was low, 65.5% of the dogs and cats with elevated blood urea nitrogen values were correctly classified as abnormal The test performs well when the prevalence of abnormal values is near 50%.  相似文献   

Identification of feline blood B and T lymphocytes by cell electrophoresis.

The electrokinetic properties of feline blood lymphocytes isolated by centrifugation over Ficoll-Isopaque gradient were investigated. A biphasic electrophoretic mobility (EPM) distribution was regularly observed with a low-mobility (LM) population (mean EPM: 0.82) accounting for 32% of blood lymphocytes and a high-mobility (HM) population (mean EPM: 1.09) representing 68% of blood lymphocytes. Following fractionation on nylon-wool columns, lymphocytes with B-cell properties (64% sIg⁺; 9% guinea pig erythrocytes (GPE)-rosette⁺, PHA and Con A unresponsive) were enriched in the adherent fraction and belonged mainly (78%) to the LM population. In contrast, lymphocytes with T properties (5% sIg⁺, 42% GPE-rosette⁺, PHA and Con A responsive) were recovered in the effluent fraction and comprised 84% of HM elements.Thus, in cat blood, LM lymphocytes are likely to represent in majority B cells and HM lymphocytes T cells. This indicates that cell electrophoresis provides an interesting mean for differentiating B and T cells in the cat.  相似文献   

Permeability of the endothelium and partitioning of the pulmonary blood flow resistance in isolated perfused pig lungs: Effects of breed and age

The right and left lungs of 5 healthy Minipigs and of 13 healthy Landrace piglets were isolated, perfused at constant pressure and maintained in an isogravimetric state under zone III conditions (pulmonary venous pressure>alveolar pressure). By applying the double, arterial and venous, occlusion technique, the total blood flow resistance (R _t) was partitioned into four components: arterial (R _a), pre-(R _a) and post-capillary (R _v) and venous (R _v). The capillary filtration coefficient (K _f,c) was evaluated by measuring the weight gained by the lungs when the arterial and venous pressures were suddenly increased. In the youngest Landrace piglets (5 weeks old), there was an uncontrolled vasoconstriction which sometimes prevented perfusion of the lungs and induced a large increase inR _t. These high values ofR _t were decreased by tolazoline administration. The values ofR _t recorded in older pigs (12–13 weeks old) were lower in Minipigs (33.66±3.77 cmH₂O min L^–1 per 100 g of lungs;n=5) than in Landrace piglets (55.20±6.18 cmH₂O min L^–1 per 100 g;n=5). This breed difference was due to the differences inR _a andR _v. The mean values ofK _f,c were 0.193±0.015 and 0.202±0.029 ml min (cmH₂O)^–1 per 100 g of the lungs in Minipigs and Landrace piglets respectively. All these parameters were stable for the 3 hours following the equilibrium period. It was concluded that: (1) There is an age-related maturation of the control of the vasomotor tone in porcine lungs. (2) Pulmonary microvascular haemodynamics are influenced by the breed of the pigs. (3) There was no difference in theK _f,c values between both the breeds. (4) A comparison of the values reported for dogs and rabbits with our data shows that the pre- and post-capillary resistances and, to a lesser extent, the arterial and venous resistances are relatively high in pigs.  相似文献   

大豆黄酮对奶牛血清钙、磷及葡萄糖水平的影响

本研究选用40头胎次、产奶量、泌乳期相近的中国荷斯坦奶牛随机分成4组(n=10),试验组奶牛日粮中分别添加45、60和75mg/kg大豆黄酮,在试验前及试验开始后第10天、第20天和第30天颈静脉采血,测定4组奶牛血清钙、磷和葡萄糖水平。结果表明:与对照组相比较,试验1组中血清钙、磷和葡萄糖变化均不显著(P>0.05);试验2组仅血清钙在第30天时显著高于对照组(P<0.05);试验3组第10天和20天时血清钙水平显著高于对照组(P<0.05),第30天时血清钙水平极显著高于对照组(P<0.01),而血糖水平仅第30天显著高于对照组(P<0.05)。  相似文献   

Study on adherent precursors from human cord blood

AIM: To confirm the existence of the endothelial progenitor cells in human cord blood and to study its differentiation and development process. METHODS: The mononuclear cells in human cord blood were isolated using lymphocyte separation solution. Then the mononuclear cells were cltured in MCDB131 containing 20% fetal bovine serum. The effects of 5 μmol/L dexamethasone, the extract from bovine brain, insulin and hypoxanine on the proliferation and differentiation of the adherent cells were observed. The morphology of the adherent cells were examined twice daily by inverted phase contrast microscope. CD34 and CD14 expression were determined by FACS. Immunohistochemistry was used to confirm the expression of factor Ⅷ. RESULTS: The proliferative endothelial progenitor cells existed within the CD34^-adherent mononuclear cells of human cord blood. Dexamethasone and hypoxanine decreased the number of spindle-shaped cells and caudated cells. Bovine brain extract, insulin and FCS enhanced the number of spindle-shaped cells and caudated cells. CONCLUSION: The existence of endothelial progenitor cells within the CD34 - adherent monouclear cells of the human cord blood was observed and these cells were able to differentiate into endothelial-like cells in vitro.  相似文献   

Analysis of the distribution and clonality of TCR Vβ T cells in cord blood

AIM:To investigate the distribution and clonality of TCR Vβ subfamily T cells in cord blood. METHODS:The CDR3 of TCR Vβ 24 subfamily genes were amplified in mononuclear cells from 13 cases of cord blood. To observe the usage of TCR Vβ repertoire, the PCR products were further labeled with fluorescent and analyzed by genescan technique for the CDR3 size, to evaluate clonality of the detectable TCR Vβ T cells. Peripheral bloods from 10 cases of normal individuals and T cell line Molt-4 and Jurkat served as controls. RESULTS:Only 38.78%±16.26% of 24 Vβ subfamily T cell were selectively expressed in cord blood, predominantly in Vβ 3, 5, 8, 9 and 13, whereas all 24 Vβ subfamilies could be detected in T cells from peripheral blood of normal individuals. Genescan analysis showed that all PCR products of TCR Vβ subfamilies from cord blood or normal individual peripheral blood displayed multi-peaks. CONCLUSION:Some TCR Vβ subfamily T cells were absent in cord blood. All TCR Vβ subfamily T cells in cord blood displayed polyclonality.  相似文献

富铁酵母对家兔红细胞免疫功能的影响

为探讨富铁酵母对家兔红细胞免疫功能的影响,将32只家免随机分成富铁酵母高、低剂量组,FeSO4组及空白对照组,分别灌服富铁酵母(Fe元素8 mg/kg、4 mg/kg)、FeSO4(Fe元素4 mg/kg)及空白生理盐水;连续5 d后,检测红细胞C3g受体花环率和复合物(IC)花环率.结果:富铁酵母较FeSO4及空白组能显著提高红细胞C3h受体花环和红细胞复合物(IC)花环的百分率,但高、低剂量间无显著性差异.结论:富铁酵母能显著提高红细胞免疫功能.  相似文献

依据淋巴细胞脉络丛脑膜炎病毒(LCMV)主要保护性抗原CD8 T细胞表位VRRPQASGVYMGNLTAQ和卵清白蛋白(OVA)CD8 T细胞表位SIINFEKL,设计、合成两条编码LCMV和OVA CD8 T细胞表位的寡核苷酸片段,经退火后,克隆入绿色荧光蛋白表达质粒pYAGFP,经PCR扩增和序列测定分析,证实成功构建T细胞表位与绿色荧光蛋白融合表达的重组质粒pYAGFPL-O,将此重组质粒pYAGFPL-O转化减毒鼠伤寒沙门氏菌X4550,获得重组沙门氏菌X4550(pYAGFPL-O).用SDS-PAGE电泳测得重组菌表达的融合蛋白约为30kD.同时,荧光显微镜下观察到X4550(pYAGFPL-O)发出黄绿色荧光.这些结果表明LCMV和OVA T细胞表位已成功表达.重组菌X4550(pYAGFPL-O)的获得为研究沙门氏菌载体携带外源抗原的T细胞应答规律及调控机理打下了重要基础.  相似文献

应用饲养试验与氮平衡试验,研究了不同尿素水平的日粮对青山羊血清总蛋白(STP)、血清清蛋白(ALB)、血清球蛋白(GLB)、血清清球蛋白比值(A/G)、血清尿素氮(SUN)及血清谷丙转氨酶的含量变化的影响。试验采用4×4拉丁方试验设计,试验分4个阶段,在基础日粮一致的情况下,采用4、9、14、0(对照)g/kg·DM4种不同的处理,试验结果表明:不同尿素水平极显著影响ALB、SUN浓度和GPT活性浓度(P<0.01),对血清清球比(A/G)无显著差异(P>0.05),尿素水平与STP、GLB浓度存在负相关;血清含氮生化指标STP、ALB、GLB、A/G、GPT、SUN随时间呈动态变化,出现高峰的时间比瘤胃氨浓度出现的时间平均推迟1～2h。进食尿素后,血液生化指标由平稳逐渐缓慢升高到高峰,随后又下降并趋于平衡。  相似文献
