从日本中央实验动物研究所购买的2对IQI无菌小鼠饲养在无菌隔离器中,饲养温度为22~24℃,相对湿度为58~60%,并成功地繁殖了214只无菌小鼠。无菌检验每2月进行一次,共检验4次,结果均为无菌生长。对鼠肝炎病毒和仙台病毒进行监测,结果为阴性。无菌小鼠剖解肉眼观察其盲肠巨大。  相似文献

Fundamental processes influencing human growth can be revealed by studying extreme short stature. Using genetic linkage analysis, we find that biallelic loss-of-function mutations in the centrosomal pericentrin (PCNT) gene on chromosome 21q22.3 cause microcephalic osteodysplastic primordial dwarfism type II (MOPD II) in 25 patients. Adults with this rare inherited condition have an average height of 100 centimeters and a brain size comparable to that of a 3-month-old baby, but are of near-normal intelligence. Absence of PCNT results in disorganized mitotic spindles and missegregation of chromosomes. Mutations in related genes are known to cause primary microcephaly (MCPH1, CDK5RAP2, ASPM, and CENPJ).  相似文献

Twelve pairs of SSR markers were used to evaluate the genetic diversity of 138 accessions of potato cultivars from the Western Highlands region of Cameroon. The average Polymorphism information content (PIC) value (0.74) and number of alleles (7.08) indicated a high genetic diversity of the potato cultivars tested. Analysis of molecular variance revealed that most of the genetic variations are found within geographic region (≥ 91%), resulting in high gene flow (Nm > 4 individuals). Local varieties had significantly more alleles than exotic varieties. Genetic diversity estimates for accessions from low elevations were significantly lower than those from medium and high elevations. Cluster analysis showed three clusters; the model-based approach inferred two gene pools. Genotypes revealed a high level of admixture between gene pools within locations and elevations. A core collection identified in this study composed of 27 individuals representing 19.57% of the whole collection and captured 99.15% of the total alleles found.  相似文献

Consumer demand for fish and shellfish has led to an increase in aquatic species movement for aquaculture purposes. One potential drawback to the successful transplantation of animals for aquaculture is unpredictable performance due to local adaptation effects. This study used a common environment experiment to examine the potential for local adaptation in Mytilus trossulus (Lamark) on the east coast of Vancouver Island, British Columbia (BC). Newly settled mussels were collected from two sites, Chemainus and Quadra Island (approximately 150 km apart), reared in cages at the Quadra collection site, and mortality and shell length were monitored. Mussels were cage‐reared for 384 days with an overall survival of 59% (Quadra) and 39% (Chemainus). The Quadra mussels were initially smaller (5.07 mm) than the Chemainus mussels (5.90 mm), but the two were similar at the end of the experiment (23.5 and 23.6 mm). Transplanted mussels had a significantly higher mortality over the course of the experiment, primarily due to a severe episode in the early summer, and had significantly lower relative growth rates at three of 11 measurement dates. Overall, the local mussels performed better than the transplanted mussels. This study demonstrates the potential for local adaptation effects between populations of mussels separated by only a relatively small geographic distance.  相似文献

The nitrate threshold concentration in rearing water of African catfish (Clarias gariepinus) was assessed. Female African catfish with an initial mean (SD) weight of 154.3 (7.5) g were exposed to 0.4 (Control), 1.5, 4.2, 9.7 and 27.0 mM nitrate for 42 days. Mean (SD) plasma concentrations of nitrate increased from 71 (29) to 6623 (921) μM at the highest ambient nitrate level. Mean (SD) plasma nitrite concentration ranged from 1.2 (0.5) to 7.9 (9.0) μM. Haematocrit, plasma concentrations of non‐esterified fatty acids (NEFA), cortisol, glucose, lactate, osmolality, gill morphology and branchial Na+/K+‐ATPase activity were not affected. Feed intake and specific growth rate were significantly reduced at the highest nitrate concentration. We advise not to exceed a water nitrate concentration of 10 mM (140 mg L?1 NO3‐N) to prevent the risk of reduced growth and feed intake in African catfish aquaculture.  相似文献
