A single dose of 5 x 10(8) bacilli of Pseudomonas pseudomallei by intratracheal injection resulted in acute (21 cases) or chronic (19 cases) melioidosis in 40 of 48 pigs. Fifteen (10 acute and 5 chronic) had been immunosuppressed by cyclophosphamide before inoculation. The major clinical signs were initial fever, marked neutrophilia and, in the acute cases, respiratory distress. There were no signs of the nasal and ocular discharge, paresis or diarrhoea seen in acute cases in south-east Asia. The cyclophosphamide treatment caused a significant decrease in the neutrophil count by 7 d after inoculation in all 15 immunosuppressed pigs, and all were culture positive at necropsy. Eight of the 33 non-treated pigs were culture negative at necropsy. Pigs overcoming the initial phase of infection had more abscess-like nodules that were bacteriologically sterile at necropsy than the pigs with acute cases of melioidosis. P. pseudomallei was isolated predominantly from the spleen, lungs and the injection site. Although only one strain was used in this study, it is likely that Australian strains of P. pseudomallei are not as virulent as the south-east Asian isolates.  相似文献

Three experiments were conducted to evaluate methods of immunization against GnRH on antibody titer, luteal activity, and pregnancy in beef heifers. Experiment 1 evaluated the efficacy of adjuvants with 30 heifers. Control heifers were immunized against human serum albumin (HSA) emulsified in Freund's complete adjuvant (FCA). The other 4 treatments contained GnRH conjugated to HSA (HSA-GnRH) emulsified in FCA, Freund's incomplete adjuvant (FIA), DEAE dextran (DD) + mineral oil (MO), or DD+FIA. Treatment was in the mammary gland for all experiments. Titers against GnRH for heifers immunized against HSA-GnRH with FCA, DD+MO, or DD+FIA were greater than titers for HSA-GnRH with FIA or control heifers (P < 0.01). Body weight was reduced (P < 0.05) in control and FCA heifers compared with FIA, DD+MO, and DD+FIA heifers. Heifers immunized with DD+MO and DD+FIA had fewer granulomas in mammary glands than heifers treated with FCA (P < 0.01). In Exp. 2, 36 heifers were used to determine the effect of the protein conjugated to GnRH on titers against GnRH. Heifers (6/treatment) received a primary immunization against GnRH conjugated to HSA (HSA-GnRH), ovalbumin (OA-GnRH), or keyhole limpet hemocyanin (KL-GnRH), or heifers were immunized against each carrier protein. Antigens were emulsified in DD+FIA. Immunization of heifers against OA-GnRH, KL-GnRH, or HSA-GnRH suppressed luteal activity (P < 0.01) for 23, 16, and 12 wk, respectively, and antibody titers against GnRH were greater (P < 0.01) for 19, 5, and 7 wk, respectively, compared with heifers immunized against the carrier proteins. In Exp. 3, 90 heifers were used to determine the effect of immunization against GnRH on ovarian activity and pregnancy rate. Heifers (30/treatment) received a primary and 2 or 3 booster immunizations against GnRH conjugated to OA, and controls received a primary and 2 booster immunizations against OA. All antigens were emulsified in DD+FIA. At 8 wk after primary immunization, heifers were exposed to fertile bulls for 24 wk. Pregnancy rate was less (P < 0.01) for 3-booster heifers (13%) compared with control (83%) and 2-booster (62%) heifers. We conclude that immunization against GnRH, conjugated to OA and emulsified in DD+FIA, does not influence ADG and produces sufficient titers against GnRH to prevent estrous cycles with few mammary granulomas. Immunization against GnRH with 3 booster immunizations prevented luteal activity and pregnancy in most beef heifers for more than 4 mo.  相似文献

Effects of small population size and reduced genetic variation on the viability of wild animal populations remain controversial. During a 35-year study of a remnant population of greater prairie chickens, population size decreased from 2000 individuals in 1962 to fewer than 50 by 1994. Concurrently, both fitness, as measured by fertility and hatching rates of eggs, and genetic diversity declined significantly. Conservation measures initiated in 1992 with translocations of birds from large, genetically diverse populations restored egg viability. Thus, sufficient genetic resources appear to be critical for maintaining populations of greater prairie chickens.  相似文献

Twelve ruminally cannulated crossbred Angus steers were used to evaluate ruminal fermentation characteristics and diet digestibility when 30% (DM) corn dried distillers grains with solubles (DDGS) containing 0.42 or 0.65% (DM) of dietary S was incorporated into finishing diets based on steam-flaked corn (SFC) or dry-rolled corn (DRC). The study was a replicated, balanced randomized incomplete block design with a 2 × 2 factorial arrangement of treatments. Factors consisted of dietary S concentration (0.42 and 0.65% of DM; 0.42S and 0.65S, respectively) and grain processing method (SFC or DRC). The 0.65S concentration was achieved by adding H(2)SO(4) to DDGS before mixing rations. Steers were assigned randomly to diets and individual, slatted-floor pens, and fed once daily for ad libitum intake. Two 15-d experimental periods were used, each consisting of a 12-d diet adaptation phase and a 3-d sample collection phase. Samples were collected at 2-h intervals postfeeding during the collection phase. Ruminal pH was measured immediately after sampling, and concentrations of ruminal ammonia and VFA were determined. Fecal samples were composited by steer within period and used to determine apparent total tract digestibilities of DM, OM, NDF, CP, starch, and ether extract. Feeding 0.65S tended (P = 0.08) to decrease DMI but resulted in greater apparent total tract digestibilities of DM (P = 0.04) and ether extract (P = 0.03). Ruminal pH increased (P < 0.05) in steers fed 0.65S diets, which may be attributable, in part, to decreased (P = 0.05) VFA concentrations and greater (P < 0.01) ruminal ammonia concentrations when 0.65S was fed, compared with feeding 0.42S. These effects were more exaggerated in steers fed DRC (interaction, P < 0.01), compared with steers fed SFC. Steers fed DRC-0.65S had greater (P < 0.01) acetate concentration than steers fed DRC-0.42S, but acetate concentration was not affected by S concentration when SFC was fed. Propionate concentration was decreased (P < 0.01) in steers fed SFC-0.65S compared with steers fed SFC-0.42S, but dietary S concentration had no effect on propionate concentration when DRC was fed. Butyrate concentration was less (P < 0.01) in steers fed 0.65S diets than in steers fed 0.42S. Lactate concentrations tended (P = 0.06) to decrease in steers fed 0.65S diets. Feeding DDGS with increased S concentration may decrease feed intake and ruminal VFA concentration but increase ruminal ammonia concentration.  相似文献

The objectives of this study were to determine effects of cyfluthrin and pyrethrin spray products, used in combination with cyfluthrin topical and ear tag applications, on bull reproductive parameters over 18 weeks. Angus or Angus x Simmental bulls were randomly assigned to one of three treatment groups: (i) no exposure to pyrethrins/cyfluthrin (CONT; n = 10), (ii) cyfluthrin ear tag and topical applications (ET; n = 10), or (iii) cyfluthrin ear tag, topical, premise spray and pyrethrin fog spray applications (ET+S; n = 8). Bull body weight was measured every 3 week, and body condition score and scrotal circumference were recorded on weeks 0, 9 and 18. Semen and serum were collected every 3 weeks for sperm evaluation and testosterone measurement, respectively. There was a treatment × week interaction (p < 0.01) for sperm with primary defects; bulls in CONT group had a greater (p = 0.01) percentage of sperm with primary defects than bulls treated with insecticides at week 18. Overall and progressive sperm motility, normal sperm morphology, secondary sperm defects and serum testosterone concentrations changed (p < 0.01) over time in all bulls; however, treatment did not affect (p ≥ 0.13) any of these parameters. There were also no treatment effects (p ≥ 0.08) on bull body weight, body condition score or scrotal circumference. The use of pyrethrin‐ and cyfluthrin‐based insecticides, regardless of application, did not negatively affect reproductive parameters in beef bulls when administered over 18 weeks.  相似文献

Six hundred ten crossbred-yearling heifers (347 +/- 5 kg of initial BW) were obtained and used in a randomized complete-block design finishing study. Finishing diets were based on steam-flaked corn and ground alfalfa hay. The control (CONT) treatment contained no distillers grains with solubles (DGS), the second diet was formulated to contained 13% (DM basis) dried corn DGS derived from a traditional dry-grind ethanol process (TRAD), and the third diet was formulated to contained 13% (DM basis) dried corn DGS derived from a partial fractionation dry-grind process (FRAC). Dry matter intake, ADG, and gain efficiency were not different (P >/= 0.48) for yearling heifers fed CONT when compared with heifers fed DGS. Heifers fed TRAD consumed more (P = 0.01) feed than heifers fed FRAC. However, ADG and feed efficiency were not different (P >/= 0.07) for heifers fed DGS. Moderate inclusion levels of DGS in finishing flaked corn diets yielded satisfactory performance. Growth performance was not different for heifers fed DGS originating from either ethanol processing method.  相似文献

Local control of the domain orientation in diblock copolymer thin films can be obtained by the application of electric fields on micrometer-length scales. Thin films of an asymmetric polystyrene-polymethylmethacrylate diblock copolymer, with cylindrical polymethylmethacrylate microdomains, were spin-coated onto substrates previously patterned with planar electrodes. The substrates, 100-nanometer-thick silicon nitride membranes, allow direct observation of the electrodes and the copolymer domain structure by transmission electron microscopy. The cylinders aligned parallel to the electric field lines for fields exceeding 30 kilovolts per centimeter, after annealing at 250°C in an inert atmosphere for 24 hours. This technique could find application in nanostructure fabrication.  相似文献

One experiment was conducted during 1989 to determine whether a deficiency exists for either ruminally degradable or escape protein in steers grazing summer native range. Increasing levels of ruminally degradable (.15, .27, and .37 kg/d) and escape protein (.07, .14, and .21 kg/d) replaced a cornstarch and molasses (energy control) supplement. Supplements were isoenergetic and fed individually to steers (.88 kg/d). No response to the degradable protein supplement (P = .15) was observed; however, a linear gain response (P less than .01) was observed in steers fed escape protein in addition to ruminally degradable protein. An in vitro study indicated that more (P less than .01) microbial protein was synthesized from the energy supplement than from the degradable protein; this finding presumably relates to the numerical decrease in weight gains observed for steers fed degradable protein supplements. Analyses of esophageal extrusa samples indicated that CP was relatively constant for the 1989 growing season compared with the 1988 growing season (P less than .05). Escape protein values differed (P less than .01) between years and among months within year. Forages that were apparently grazed in 1989 were never deficient in degradable protein. Additional gain observed from feeding escape protein would indicate that microbial protein synthesis may be insufficient to satisfy the metabolizable protein requirement, which probably limited gains by steers grazing summer native range.  相似文献

Research was conducted to determine the effects of supplemental dietary vitamin D3 on DMI, carcass traits, Warner Bratzler shear (WBS) force, calpastatin activity, plasma minerals, pH (0, 3, 12, and 24 h after slaughter), water-holding capacity (WHC), and sensory characteristics of three muscles. Pre-slaughter vitamin D3 treatments included no supplemental vitamin D3, 6 x 106 IU (MIU) of vitamin D3 for 4 d, or 6 MIU of vitamin D3 for 6 d. Cattle were slaughtered and carcasses were chilled for 48 h before removal of steaks from the longissimus, gluteus medius, and biceps femoris muscles. Steaks were aged at 2 degrees C for 7, 14, or 21 d before cooking to a final internal temperature of 70 degrees C for WBS and sensory panel analysis. Dry matter intake was lower for steers supplemented with vitamin D3 for 4 or 6 d. Live and carcass weights were lower (P < 0.05) in steers supplemented with vitamin D3. Supplementing 6 MIU/6 d of vitamin D3 decreased (P < 0.05) WBS values of gluteus steaks (pooled over aging times). Longissimus steaks from steers supplemented with vitamin D3 for 6 d had lower (P < 0.05) WBS force values than these steaks from control steers or steers fed vitamin D3 for 4 d at 7 d postmortem. Biceps femoris steaks from steers receiving vitamin D3 for 4 d had higher WBS values than steaks from control steers at 14 and 21 d postmortem. Feeding vitamin D3 at 6 MIU for 6 d decreased (P < 0.05) the percentage of steaks that had WBS values > or = 3.86 kg for all steaks. Feeding vitamin D3 had no effect on palatability traits evaluated by trained panelists. Blood Ca concentrations were greater (P < 0.05) when vitamin D3 was fed and with increased vitamin D3 feeding time. Feeding vitamin D3 for 6 d (vs 4 d) delayed pH decline for all muscle types after 0, 3, and 12 h postmortem. Water-holding capacity was increased (P > 0.02) after 0 h, 24 h, and 21 d postmortem when vitamin D3 was fed and was greater at 0 and 24 h if vitamin D3 was fed for 6 d rather than 4 d. These data suggest that supplementing 6 MIU of vitamin D3 will decrease DMI and improve beef tenderness through increased blood plasma Ca concentrations and WHC.  相似文献
