  1. The ocean crisis is urgent and central to human wellbeing and life on Earth; past and current activities are damaging the planet's main life support system for future generations. We are witnessing an increase in ocean heat, disturbance, acidification, bio‐invasions and nutrients, and reducing oxygen levels. Several of these act like ratchets: once detrimental or negative changes have occurred, they may lock in place and may not be reversible, especially at gross ecological and ocean process scales.
  2. Each change may represent a loss to humanity of resources, ecosystem function, oxygen production and species. The longer we pursue unsuitable actions, the more we close the path to recovery and better ocean health and greater benefits for humanity in the future.
  3. We stand at a critical juncture and have identified eight priority issues that need to be addressed in unison to help avert a potential ecological disaster in the global ocean. They form a purposely ambitious agenda for global governance and are aimed at informing decision‐makers at a high level. They should also be of interest to the general public.
  4. Of all the themes, the highest priority is to rigorously address global warming and limit surface temperature rise to 1.5°C by 2100, as warming is the pre‐eminent factor driving change in the ocean. The other themes are establishing a robust and comprehensive High Seas Treaty, enforcing existing standards for Marine Protected Areas and expanding their coverage, especially in terms of high levels of protection, adopting a precautionary pause on deep‐sea mining, ending overfishing and destructive fishing practices, radically reducing marine pollution, putting in place a financing mechanism for ocean management and protection, and lastly, scaling up science/data gathering and facilitating data sharing.
  5. By implementing all eight measures in unison, as a coordinated strategy, we can build resilience to climate change, help sustain fisheries productivity, particularly for low‐income countries dependent on fisheries, protect coasts (e.g. via soft‐engineering/habitat‐based approaches), promote mitigation (e.g. carbon storage) and enable improved adaptation to rapid global change.

This study was undertaken to enable the determination of hydrolysis and functionality of proteins in situ during fermentation of wheat doughs. Wheat proteins were fractionated and labeled with fluorescein-isothiocyanate (FITC). Fluorescent proteins were incorporated into wheat sourdoughs inoculated with lactobacilli and into neutral and acid control doughs. Doughs containing fungal protease were furthermore evaluated. Doughs were analyzed by extraction and size exclusion chromatography analysis of sodium dodecyl sulfate soluble proteins. Labeled proteins exhibited characteristics comparable to native proteins, with respect to proteolytic degradation and polymerization. Proteolytic breakdown of proteins was enhanced at low pH. Glutenin subunits were incorporated into the gluten macropolymer at neutral pH. Polymerization of FITC proteins was not observed at low pH. Sourdoughs were comparable to acid control doughs, major effects were attributed to changes of pH, rather than microbial metabolism. A synergistic effect with respect to proteolytic activity was observed between fungal protease and L. pontis.  相似文献

Sulfonamide antibiotics reach soil via manure and adversely affect microbial diversity. Clear effects of these bacteriostatic, growth‐inhibiting antibiotics occur in the presence of a parallel input of microbial activity stimulating manure. Natural hot spots with already increased soil microbial activity are located in the rhizosphere, comprising microorganism such as Pseudomonas with plant growth promoting and pathogenic strains. The hypothesis was therefore that the antibiotic activity of sulfonamides is promoted in the rhizosphere even in the absence of manure, followed by shifts of the natural plant‐specific microbial community structure. This was evaluated by a laboratory experiment with Salix fragilis L. and Zea mays L. After 40 d of incubation, sub‐areas such as non‐rhizosphere soil, rhizosphere soil and plant roots were sampled. Effects on microbial community structure were analyzed using 16S rRNA gene fragment patterns of total bacteria community and Pseudomonas. Selected exoenzymes of N‐, P‐, and C‐cycling were used to test effects on microbial functions. Compared to the factors soil sub‐area and sulfadiazine (SDZ) content, plant species had the largest influence on the bacterial community structure and soil exoenzyme activity pattern. This was also reflected by an up to 1.5‐fold higher acid phosphatase activity in samples from maize‐ compared to willow‐planted soil. We conclude that antibiotic effects on the bacterial community structures are influenced by the antibiotic concentration and root influence.  相似文献

Sulfonamides are the second most used antibiotic class in veterinary medicine and applied to livestock to treat bacterial infections. Subsequently, they are spread onto agricultural soils together with the contaminated manure used as fertilizer. Both manure and antibiotics affect the soil microbial community. However, the influence of different liquid manure loads on effects of antibiotics to soil microorganisms is not well understood. Therefore, we performed a microcosm experiment for up to 32 d to clarify whether the function and structure of the soil microbial community is differently affected by interactions of manure and the antibiotic sulfadiazine (SDZ). To this end selected concentrations of pig liquid manure (0, 20, 40, 80 g kg–1) and SDZ (0, 10, 100 mg kg–1) were combined. We hypothesized that incremental manure amendment might reduce the effect of SDZ in soils, due to an increasing sorption capacity of SDZ to organic compounds. Clear dose‐dependent effects of SDZ on microbial biomass and PLFA pattern were determined, and SDZ effects interacted with the liquid manure application rate. Soil microbial biomass increased with incremental liquid manure addition, whereas this effect was absent in the presence of additional SDZ. However, activities of enzymes such as urease and protease were only slightly affected and basal respiration was not affected by SDZ application, while differences mostly depended on the concentration of liquid manure. These results illustrated that the microbial biomass and structural composition react more sensitive to SDZ contamination than functional processes. Furthermore, effects disproportionally increased with incremental liquid manure addition, although extractable amounts of SDZ declined with increasing liquid manure application.  相似文献

PAH decrease in soil samples from different polluted industrial sites by activating the autochthonous microflora In pot experiments under field conditions the PAH decrease in ten different soil samples from former industrial sites with differing PAH load were investigated over a period of 74 and 168 weeks. 15 out of 16 PAH according to US EPA (without acenaphthylene) were determined. Easily degradable organic matter, mineral fertilizers, synthetic surfactants and in some experiments also lime were added to the soil samples in varying amounts. Depending on the nature and quantity of the amendments, the biological activity and (co-)metabolic decomposition of PAH by soil microorganisms could considerably be increased. In the different soil samples a decrease of the initial PAH contents between 12 and 90% was achieved within 74 weeks. Even from 74 up to 168 weeks for some soil samples a remarkable further decrease of the PAH contents could be observed. The decrease of the extractable PAH with time is mainly caused by microbial decomposition and formation of nonextractable residues. This behavior can be fitted by two coupled exponential functions, one for an initial phase of rapid decomposition and the other for a subsequent phase of slow decomposition. Therefore, two different processes (I, II) determine the decrease of PAH. In the first week of the experiment the decomposition rates for process I amount to 4.2–88.3 and for process II to 0.06–5.3 mg PAH ? kg?1 soil ? week?1; in the 168th week they are no longer determinable for process I and amount to 0.05–2.3 mg ? kg?1 ? week' for process II. Higher initial PAH contents often led to higher relative PAH decreases, but also to absolute higher contents of residues. The persistence of PAH is mainly determined by their bioavailability. PAH degradation is increased by the soil treatments. The addition of easily degradable organic matter (C/N ratio < 20) in a quantity of 30g ? kg?1 (w/w) combined with the addition of nitrate and a surfactant most effectively increased the degradation of PAH. Raising the pH of a very strongly acidic soil sample revealed a maximum PAH decomposition at a pH of 6.  相似文献

During the 1980s, when a flexibleapproach to research, known asfarmer-back-to-farmer, was developed, theInternational Potato Center (CIP) became famousfor participatory research. Subsequently itappeared to have lost leadership in this field.This article documents participatory researchactivities in CIP over the past thirty years tofind out what happened. Even in the 1980s,implementation of participatory research wasactually limited. Participatory research in thecenter grew unevenly, with little clearencouragement from the CGIAR. Decentralizationof social scientists in the 1990s led to thefragmentation of participatory research and, inthe absence of any clear champion, it seemedthat it might wither away. Recently, increasinginterest in IPM, donor support, and therecruitment of international staff who havebeen exposed to other currents of participatoryresearch and training has led to a revival ofinterest. This needs to be carefully nurturedto ensure that the flexibility for whichparticipatory research in CIP earlier gainedworld renown is retained.  相似文献
