应用淋巴细胞杂交瘤技术,融合经水稻细菌性条斑病病原菌(Xanthomonas campestris pv.oryzicola)免疫的BALB/C小鼠脾细胞和Sp2/O骨髓瘤细胞,经细条病菌典型菌株R17,R25和GX-1单独及三者混合包被进行ELISA检测筛选,建立了能稳定分泌特异性单克隆抗体的杂交瘤细胞株18G 9,19D8,21F2,21G3,25E6和26F9细胞株。体外连续传代培养三个月,液氮冻存四个月后复苏培养,分泌抗体稳定。六株杂交瘤细胞的腹水单克隆抗体ELISA效价在1:2000左右。各株细胞的培养上清液ELISA效价为1:8左右。18G9,19D8和25E6细胞株所分泌的抗体为小鼠IgM类,21F2和21G3分泌的抗体为小鼠IgG_3亚类,26F9分泌的抗体为小鼠IgG_(2b)亚类。根据与植物病原细菌4个属6个种的20个不同菌株的特异性反应,将六株单克隆抗体分为A,B,C三组,20个菌株分为3个血清型和1个菌株群。  相似文献

为了探明华中地区种鸡场沙门菌(Salmonella)的优势血清型和耐药情况,本研究从湖北、河南、湖南等省市22个规模化鸡场采集病鸡、死胚及弱雏组织样品3 724份,通过分离培养、生化试验、PCR鉴定及血清型试验确定分离菌种属及其血清型,并采用Kirby-Bauer法对分离菌株进行了耐药性分析。结果显示,本试验从3 724份病料中共分离鉴定出124株沙门菌,其中79株为D群肠炎沙门菌(63.71%,79/124),34株为D群鸡白痢沙门菌(27.42%,34/124),8株为B群鼠伤寒沙门菌(6.45%,8/124),有3株沙门菌未能确定血清型。O抗原鉴定79株肠炎沙门菌和34株鸡白痢沙门菌为O9,8株鼠伤寒沙门菌为O4。H抗原鉴定79株肠炎沙门菌为Hg,m,8株鼠伤寒沙门菌为Hi。药敏试验结果显示,124株分离菌株对萘啶酸、氨苄青霉素、四环素和多西环素耐药率分别为95.97%(119/124)、91.94%(114/124)、57.26%(71/124)和70.16%(87/124);对复方新诺明和红霉素耐药率分别为25.81%(32/124)和12.10%(15/124);对氯霉素、庆大霉素、头孢噻肟和卡那霉素耐药率分别为6.45%(8/124)、1.61%(2/124)、1.61%(2/124)和0.81%(1/124);对左氧氟沙星、阿米卡星和多黏菌素B完全敏感。99.19%(123/124)的分离株至少对一种药物耐药,87.10%(108/124)的分离株表现多重耐药。本研究为华中地区养鸡场沙门菌的诊断及防控提供了数据支撑。  相似文献

Salmonella enterica subsp. enterica serovar Eppendorf, with antigenic formula 1,4,12,[27]:d:1,5, is an infrequent serovar. However, 14% (20 of 142) of the isolates recovered during June–July 2012 in chicken farms in Tunisia belonged to S. Eppendorf. These isolates were analysed for resistance and virulence profiles. None of them were susceptible to all antimicrobials tested, while 70%, 60%, 50%, 50%, 20% and 5% were resistant to sulphonamides (sul1, sul2 and sul3), streptomycin (aadA1‐like), trimethoprim (dfrA1‐like), nalidixic acid (GyrA Asp87→Asn and not identified), gentamicin (not identified) and ampicillin (blaTEM‐1‐like). About 30% of the isolates showed decreased susceptibility to ciprofloxacin and carried the qnrB gene; 65% of the isolates were multidrug resistant and contained class 1 integrons with sul1 or sul3 in the 3′ conserved segment. The orgA, ssaQ, mgtC, siiD and sopB virulence genes located on SPI1 to SPI5 and the fimbrial bcfC gene were present in all isolates; the sopE1 and sodC1 carried by prophages were variably detected; however, the prophage gipA gene and the spvC gene of serovar‐specific virulence plasmids were absent. Altogether, ten resistance and three virulence profiles were identified. Typing of the isolates with XbaI‐ and BlnI‐PFGE supports a close relationship, although they appear to be evolving under selective pressure probably caused by antimicrobial use in chicken husbandry. As far as we know, this is the first study investigating the molecular bases of antimicrobial drug resistance, the virulence gene content and the PFGE profiles of S. Eppendorf. The epidemiological surveillance of this serovar would be necessary to evaluate its possible impact on human health, particularly in Tunisia and other African countries where it was already reported.  相似文献

为了探索 cpxR 对鼠伤寒沙门菌的黏菌素耐药相关基因 pmrB 和 phoQ 的调控作用,用overlapping PCR扩增得到 cpxR 、 pmrB 和 cpxR 、 phoQ 共表达基因 cpxR - pmrB 、 cpxR - phoQ ,用2倍微量肉汤稀释法对构建的 pmrB 、 phoQ 单基因过表达菌株JSΔ acrB Δ cpxR :: kan /p pmrB (JSΔΔ/p B )、JSΔ acrB Δ cpxR :: kan /p phoQ (JSΔΔ/p Q )和 cpxR - pmrB 、 cpxR - phoQ 共表达菌株JSΔ acrB Δ cpxR :: kan /p cpxR - pmrB (JSΔΔ/p RB )、JSΔ acrB Δ cpxR :: kan /p cpxR - phoQ (JSΔΔ/p RQ )进行黏菌素最小抑菌浓度(MICs)的测定,同时以各菌株在LB中的OD 600 nm 值绘制生长曲线,以各菌株在不同浓度的黏菌素中的存活率绘制杀菌曲线。结果:成功构建的鼠伤寒沙门菌 pmrB 、 phoQ 的单基因过表达菌株JSΔΔ/p B 、JSΔΔ/p Q 和 cpxR - pmrB 、 cpxR - phoQ 共表达菌株JSΔΔ/p RB 、JSΔΔ/p RQ 的MIC结果显示,JSΔ acrB Δ cpxR :: kan /p cpxR (JSΔΔ/p R )的MIC值较JS下降93.75%,与本实验室前期研究结果一致。JSΔΔ/p B 、JSΔΔ/p Q 的MIC值较JS均上升3倍,而JSΔΔ/p RB 、JSΔΔ/p RQ 的MIC值较JS分别降低50%和75%。生长曲线结果显示JSΔΔ/p R 的生长活性最低,JSΔΔ/p B 、JSΔΔ/p Q 、JSΔΔ/p RB 和JSΔΔ/p RQ 的生长活性均低于JSΔ acrB Δ cpxR :: kan /pHisA(JSΔΔ/pHisA)。黏菌素的杀菌曲线结果显示JSΔΔ/p B 和JSΔΔ/p Q 在不同浓度的黏菌素中的存活率显著高于黏菌素较敏感的JSΔΔ/p RB 和JSΔΔ/p RQ 。上述结果表明:CpxR能够通过PmrB和PhoQ调控鼠伤寒沙门菌对黏菌素的敏感性。  相似文献

Sera obtained from cattle in the state of Yucatan, Mexico, were screened using the microscopic agglutination test against 13 serovars of Leptospira interrogans. A total of 62.8% (461/734) cows were positive for one or more serovars. This seroprevalence probably reflects infection because vaccination against leptospirosis has not been practised in Yucatan. The most common antibodies detected were those against antigens of serovars hardjo (54.1%) and tarassovi (53.3%). Region was the only risk factor associated with the seroprevalence of leptospirosis (p < 0.05). This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

副猪嗜血杆菌(Haemophilus parasuis,Hps)的血清型及基因型具有复杂多样性。本研究用间接血凝试验对某一猪场分离的Hps进行血清分型鉴定,并对部分菌株的外膜蛋白P5基因进行克隆测序和进化发育分析。试验结果显示,在21头病死猪不同部位共分离了42株Hps,其中 Hps血清5型17株(40.7%)、血清14型12株(28.5%)、血清4型1株(2.3%)和未定型12株(28.5%)。本试验中有5头猪同时感染2种不同血清型的Hps。21株Hps临床分离株分属5个不同基因序列型(STA~STE),其中STA有12株(12/21)为优势基因型;相同血清型的Hps分离株具有不同的STs型,来自同一头猪的Hps血清型和STs型也不相同。以上结果表明,同一猪群感染的Hps血清型和基因型存在明显的多样性,同一头猪感染Hps的血清型和基因型也存在多样性。本研究为进一步揭示猪群感染Hps的复杂性,研究该病的感染与流行机制提供了有价值的参考。  相似文献

The spread of the emerging pathogen Vibrio vulnificus biotype 2 serovar A in Danish anguilliculture is reported. Serovar A was originally isolated in a Spanish eel farm in 2000 and occurred in Denmark in the summer of 2004, affecting eels of 5-10 g body weight cultured in fresh water. The Danish eels showed clinical signs different from those reported for Spanish eels, such as severe haemorrhages in the head and gill region with necrosis of the soft tissues. Danish isolates were biochemically and serologically identical to Spanish serovar A strains and also highly virulent for eels by both intraperitoneal injection and immersion challenges. Vaccination with Vulnivaccine, a vaccine against V. vulnificus serovar E, cross-protected eels against serovar A. The LD(50) for experimentally infected vaccinated animals was significantly higher than for non-vaccinated animals.  相似文献

Vibrio vulnificus biotype 2 is subdivided into two main serovars, serovar E, able to infect fish and humans, and serovar A, only virulent for fish. Serovar E emerged in 1976 as the causative agent of a haemorrhagic septicaemia (warm‐water vibriosis) affecting eels cultured in brackish water. Serovar A emerged in 2000 in freshwater‐cultured eels vaccinated against serovar E, causing warm‐water vibriosis with fish showing a haemorrhagic intestine as the main differential sign. The aim of the present work was to compare the disease caused by both serovars in terms of transmission routes, portals of entry and host range. Results of bath, patch‐contact and oral‐anal challenges demonstrated that both serovars spread through water and infect healthy eels, serovar A entering mainly by the anus and serovar E by the gills. The course of the disease under laboratory conditions was similar for both serovars in terms of transmission and dependence of degree of virulence on water parameters (temperature and salinity). However, the decrease in degree of virulence in fresh water was significantly greater in serovar E than in serovar A. Finally, both serovars proved pathogenic for tilapia, sea bass and rainbow trout, but not for sea bream, with significant differences in degree of virulence only in rainbow trout. In conclusion, serovar A seems to represent a new antigenic form of V. vulnificus biotype 2 with an unusual portal of entry and is better adapted to fresh water than serovar E.  相似文献

Salmonella enterica serovar Choleraesuis (Salmonella Choleraesuis) causes a lethal systemic infection (salmonellosis) in swine. Live attenuated Salmonella Choleraesuis vaccines are effective in preventing the disease, and isolates of Salmonella Choleraesuis with mutations in the cAMP-receptor protein (CRP) gene (Salmonella Choleraesuis ∆crp) are the most widely used, although the basis of the attenuation remains unclear. The objective of this study was to determine if the attenuated phenotype of Salmonella Choleraesuis ∆crp was due to alterations in susceptibility to gastrointestinal factors such as pH and bile salts, ability to colonize or invade the intestine, or cytotoxicity for macrophages. Compared with the parental strain, the survival rate of Salmonella Choleraesuis ∆crp at low pH or in the presence of bile salts was higher, while the ability of the mutant to invade intestinal epithelia was significantly decreased. In examining the role of CRP on the secretory function of the Salmonella pathogenicity island 1 (SPI-1) encoded type III secretion system (T3SS), it was shown that Salmonella Choleraesuis ∆crp was unable to secrete the SPI-1 T3SS effector proteins, SopB and SipB, which play a role in Salmonella intestinal invasiveness and macrophage cytotoxicity, respectively. In addition, caspase-1 dependent cytotoxicity for macrophages was significantly reduced in Salmonella Choleraesuis ∆crp. Collectively, this study demonstrates that the CRP affects the secretory function of SPI-1 T3SS and the resulting ability to invade the host intestinal epithelium, which is a critical element in the pathogenesis of Salmonella Choleraesuis.  相似文献
