从H9N2亚型禽流感病毒感染的鸡胚尿囊液中提取病毒RNA,RT-PCR克隆全长血凝素(Hemagglutinin,HA)基因,并进一步克隆HA的主要抗原区HA1基因.将HA1克隆到原核表达载体pGEXKG,与谷胱苷肽(GST)融合表达,表达的融合蛋白GST-HA1以包涵体形式存在.包涵体经变性、复性处理,Western blot分析表明,表达蛋白具有良好的免疫学活性.ELISA检测发现,GST-HA1只能与H9亚型禽流感病毒抗体发生反应,而与H5和H7亚型禽流感病毒抗体无交叉反应.进一步将纯化的融合蛋白与佐剂混合后免疫Balb/c小鼠,免疫小鼠体内产生了较高滴度的特异性抗体.制备的HA1蛋白特异性强,具有良好的免疫原性,为禽流感病毒的鉴别诊断和禽流感疫苗开发奠定了基础.  相似文献

血凝素(hemagglutinin,HA)是禽流感病毒的主要抗原之一,原核表达HA是该抗原特性研究和疫苗制备的关键。利用分子生物学方法构建H5N1亚型的禽流感病毒HA基因原核表达载体,转化至大肠埃希菌Rosetta菌株,用不同IPTG浓度和时间诱导HA表达,检测蛋白表达量,分析不同诱导条件对HA表达效率的影响,以确定最佳诱导条件。重组表达载体pET32a-HA经BamHⅠ和EcoRⅠ双酶切鉴定,获得预期大小的DNA片段,说明HA基因成功插入质粒pET32a。Western blot检测结果表明,IPTG诱导重组菌株成功表达HA蛋白,当诱导浓度为0.1mmol/L,诱导时间为6h时,HA蛋白表达量最高。研究结果为HA抗原制备及禽流感疫苗的研究奠定基础。  相似文献

根据NCBI中紫花苜蓿1型金属硫蛋白基因(MET1, 登录号:AF189766.1)cDNA序列设计一对特异性引物,以紫花苜蓿品种“农菁1号”的cDNA为模板,利用PCR技术克隆出的一个基因,命名为MsMT1,测序发现该基因全长228 bp,编码75个氨基酸。通过碱基序列比对发现MsMT1与MET1的相似度为99%。通过氨基酸序列分析和进化树分析,发现与其他植物的1型金属硫蛋白基因具有较高的同源性。利用qRT-PCR对MsMT1在苜蓿不同器官中的表达进行分析,发现该基因在根和子叶中表达量较高。将苜蓿幼苗进行不同浓度NaCl、Na2CO3、NaHCO3及不同pH值胁迫处理后, 观察MsMT1基因的表达,发现MsMT1的表达量随盐碱处理液浓度和pH值变化发生改变,说明MsMT1与植物的抗逆性相关。采用农杆菌介导法将MsMT1导入苜蓿植株体内,卡那抗性的筛选和Northern blot结果显示MsMT1基因能够在转基因苜蓿中高效表达。用不同浓度NaCl、NaHCO3处理野生型和转化MsMT1基因的苜蓿幼苗,观察幼苗受胁迫后的表型,发现转基因的苜蓿幼苗比未转基因的苜蓿幼苗抵抗力高。本研究表明MsMT1基因能够增加苜蓿对胁迫的抗性。  相似文献

盐胁迫应答基因GmWRKY6的克隆及转基因百脉根的抗盐分析   总被引:1,自引:0,他引:1  
根据大豆盐胁迫RNA-Seq数据,筛选并克隆得到1个大豆WRKY类转录因子GmWRKY6基因。序列分析表明,该基因含有1个1674bp的开放阅读框,编码557个AA,编码蛋白属于IIb类WRKY家族成员,含有1个WRKY保守结构域和1个C2H2锌指结构基序。基因表达模式分析表明,GmWRKY6基因在大豆根中表达量较高,并且该基因受盐胁迫诱导表达。构建GmWRKY6基因的植物超表达载体,获得7株T0代阳性转GmWRKY6基因百脉根。进一步对T1代转基因株系进行抗盐分析发现,在200mmol·L-1 NaCl处理14d后转基因株系生长状态良好,而野生型对照生长矮小,叶片干枯。此外,对相关生理指标的测定发现,转基因株系叶片中脯氨酸和叶绿素含量显著高于野生型对照,而丙二醛含量和相对质膜透性显著低于野生型对照。此外,盐胁迫下转基因百脉根叶片中钠离子含量显著低于对照,而钾离子含量显著高于对照。以上结果表明大豆GmWRKY6基因的超表达能够显著增强百脉根的抗盐能力。  相似文献

本研究以多年生黑麦草‘高帽2号’叶片为试材,根据同源基因的CDS序列设计简并引物,采用RT-PCR和RACE技术,克隆了多年生黑麦草γ-谷氨酰基半胱氨酸合成酶基因LpGCS。该基因全长为1674 bp(GenBank登录号:KJ551844),具有完整的开放阅读框(ORF),共1125 bp;对其氨基酸序列特性、结构以及与其他8种同源基因氨基酸序列间的同源性进行了研究,预测该蛋白分子量为42.86 kDa,属于不稳定类蛋白质,其蛋白质二级结构以α-螺旋为主;蛋白质氨基酸序列与其他8种同源基因氨基酸序列间同源性都比较高;此外,成功构建了该基因的正、反义植物表达载体pCAMBIA-Ubi-LpGCS+和pCAMBIA-Ubi-LpGCS-,并通过农杆菌介导法获得转正、反义基因烟草。对转基因植株和野生型烟草进行镉离子胁迫试验,生理生化指标测试结果表明镉离子胁迫处理10 d后,转LpGCS+植株中MDA含量低于野生型,光合色素含量与POD、SOD、CAT活性均高于野生型;而转LpGCS-植株中MDA含量高于野生型,光合色素含量与POD、SOD、CAT活性均低于野生型。综上所述,LpGCS基因在烟草中的过量表达可以提高植株的耐镉胁迫能力,为进一步利用该基因转化多年生黑麦草培育抗重金属植株奠定基础。  相似文献

利用PCR技术,从A型产气英膜梭菌染色体和pEWD299中分别扩增出a毒素(CPA)基因和LTB基因,通过分离、纯化、内切酶酶切、连接和转化,构建了含CPA—LTB融合基因表达质粒的重组菌株BL21(DE3)(pCPA—LTB)。经酶切鉴定和核苷酸序列测定证实,构建的重组质粒pET—CPA含有CPA—LTB融合基因,其基因序列和阅读框架均正确。经EISA检测和SDS—PAGE分析,重组菌株表达的CPA—LTB融合蛋白能够被α、LT毒素抗体所识别。重组菌株BL21(DE3)(pCPA—LTB)经IPTG诱导后,融合蛋白CPA—LTB的表达量占菌体总蛋白相对含量的12.5%。  相似文献

本研究选择禽流感病毒H9N2株的HA的两个保守T细胞表位基因和M2基因胞外保守序列(M2e),经人工合成并通过PCR扩增获得HA-M2融合表位基因,将其与含有增强型绿色荧光蛋白的载体pEGFP-N1连接,构建了重组真核表达质粒HA-M2-pEGFP-N1。采用脂质体法转染体外培养的鸭胚成纤维细胞(DEF)后,通过RT-PCR、直接荧光观察和间接免疫荧光检测,结果表明,成功构建了重组真核表达质粒HA-M2-pEGFP-N1,且以HA抗原表位和保守的M2e序列为基础构建的重组蛋白能够在鸭胚成纤维细胞中成功表达。本试验为研制新型的禽流感通用疫苗,进一步探索HA基因与靶细胞相互作用及AIV的感染机理等奠定了基础。  相似文献

为提高豆科牧草百脉根抗旱方面的能力,从沙生植物胡杨和铃铛刺中克隆了4个抗逆转录因子基因并进行功能分析,从中筛选了2个具有抗旱特性的基因PeDREB2aHhERF2,构建了以磷酸甘露糖异构酶为筛选标记的单、双价植物表达载体,转化百脉根Leo品种,结果表明,转录因子基因明显改善了百脉根的抗旱特性,经过抗旱复水试验证明转基因百脉根成活率达到100%。  相似文献

金属硫蛋白对热应激下体外培养奶牛淋巴细胞的影响   总被引:1,自引:0,他引:1  
本研究的目的在于探讨外源性金属硫蛋白(MT)对热应激下体外培养奶牛淋巴细胞凋亡的影响。将屠宰获取的奶牛脾脏淋巴细胞随机分为4组,每组细胞培养液中MT的终浓度分别为0 μg/mL(对照组)、70 μg/mL(第Ⅰ组)、140 μg/mL(第Ⅱ组)、210 μg/mL(第Ⅲ组),同时置于细胞培养箱(37℃、5%CO2)中培养20 h,之后同时置于43℃水浴锅中进行热应激处理1 h,再回细胞培养箱中培养3 h,然后利用形态学观察、RT-PCR等方法进行各项指标的检测。结果如下,1)通过形态学观察和AO/EB染色观察,表明添加MT可以有效缓解热应激对奶牛淋巴细胞的损伤,但缓解效果随MT浓度的增加而下降; 2)第Ⅰ~Ⅲ组的CAT活性、SOD活性和GSH-Px活性均高于对照组(P<0.01),MDA含量均低于对照组(P<0.01),表明MT能够有效改善淋巴细胞的抗氧化能力;3)试验Ⅰ~Ⅲ组的Bax基因相对表达量均显著低于对照组(P<0.01),但试验Ⅰ~Ⅲ组Bcl-2基因的表达量和p50基因的表达量均显著低于对照组(P<0.01),表明MT可以调节与细胞凋亡相关基因的表达。综上所述,添加MT可以降低奶牛淋巴细胞死亡率,改善奶牛淋巴细胞抗氧化性能,抑制奶牛淋巴细胞凋亡,以添加终浓度为70 μg/mL的MT效果最好。  相似文献

赖氨酸脱羧酶(LDC)基因是苦豆子中氧化苦参碱(OMA)生物合成的第一个关键酶基因。在已有苦豆子赖氨酸脱羧酶基因(SaLDC)基础上克隆得到该基因上游1260 bp的启动子序列,GenBank登录号为KY038928,前期在苦豆子愈伤组织中的瞬时表达研究显示该启动子具有启动活性。生物信息学分析发现该启动子区域除了拥有启动子区的基本顺式作用元件TATA-box和CAAT-box外,还具有多个与光信号、逆境应答等相关的顺式作用元件。为进一步研究SaLDC启动子的功能,构建了该启动子与β-葡萄糖苷酸酶(GUS)报告基因融合的植物表达载体并通过农杆菌介导遗传转化拟南芥,同时对光诱导和聚乙二醇(PEG)胁迫的转基因拟南芥进行GUS活性染色和定量分析。结果显示,在T2代转基因拟南芥幼苗的不同生长阶段和成株的各组织器官中均可检测到GUS酶活性,且随幼苗生长时间的延长,叶片中的表达活性下降;在成株叶片和花萼中的表达活性强于根、茎、花瓣和角果。光和PEG胁迫均能诱导转基因拟南芥中GUS的表达;GUS酶活性定量测定显示,短时间的PEG胁迫(1~2 h)GUS酶活性显著上调(P<0.05),而连续胁迫8 h时GUS酶活性下调至最低(P<0.01),比胁迫前下降了28.2%。以上结果表明SaLDC启动子既有时空表达特异性又有组织表达特异性,光诱导和干旱胁迫对结构基因的表达有重要的调控作用。  相似文献

Escherichia coli heat-labile enterotoxin B subunit (LTB) can be used as an adjuvant for co-administered antigens. Our previous study showed that the expression of neutralizing epitope GP5 of porcine reproductive and respiratory syndrome virus (PRRSV) in transgenic tobacco plant (GP5-T) could induce PRRSV-specific immune responses in pigs. A transgenic tobacco plant co-expressing LTB and PRRSV GP5 as a fusion protein (LTB-GP5-T) was further constructed and its immunogenicity was evaluated. Pigs were given orally three consecutive doses of equal concentration of recombinant GP5 protein expressed in leaves of LTB-GP5-T or GP5-T at a 2-week interval and challenged with PRRSV at 7 weeks post-initial immunization. Pigs receiving LTB-GP5-T or GP5-T developed PRRSV-specific antibody- and cell-mediated immunity and showed significantly lower viremia and tissue viral load and milder lung lesions than wild type tobacco plant (W-T). The LTB-GP5-T-treated group had relatively higher immune responses than the GP5-T-treated group, although the differences were not statistically significant.  相似文献

Background

Gentamicin is an aminoglycoside antimicrobial commonly used in horses at 6.6 mg/kg IV once daily. Therapeutic drug monitoring (TDM) can confirm desired peak concentration is reached for common bacterial isolates, and detect toxicosis associated with high trough values.

Objectives

Determine the relationship between gentamicin dose and plasma concentration in hospitalized horses, and identify a starting dose range to achieve peaks > 32 μg/mL.

Animals

Sixty‐five horses (2002–2010) receiving once‐daily gentamicin with TDM performed (N = 99 sets).

Methods

Retrospective study. Data from hospitalized horses including weight, dose, plasma peak, and trough gentamicin concentration, creatinine concentrations and presence of focal or systemic disease were collected from medical records. Peak concentrations measured 25–35 minutes after administration were included (N = 77). Data were divided into low (<7.7 mg/kg), medium (7.7–9.7 mg/kg) and high (>9.7 mg/kg) dose groups, and were grouped by the horse having focal or systemic disease.

Results

Peak concentrations resulting from doses ≥7.7 mg/kg were 5.74 μg/mL (SE 2.1 μg/mL) greater than peaks from doses <7.7 mg/kg (P = .007). Peak concentrations was 3.6 times more likely to be >32 μg/mL if dose was ≥7.7 mg/kg (P = .04). There were no significant effects of dose on trough or creatinine concentration. At a given dose, horses with focal disease had higher peaks than those with systemic disease (P = .039).

Conclusions and Clinical Importance

These data suggest gentamicin dosage should be individually determined in horses using TDM, but support an initial once‐daily dose of 7.7–9.7 mg/kg IV to achieve peaks >32 μg/mL and trough concentrations <2 μg/mL. Further studies evaluating the safety of doses >6.6 mg/kg are required.  相似文献

以新疆大叶苜蓿及其为母本的4种转基因株系(SN:转AtNDPK2基因;SC:转codA基因;SOR:转IbOr基因;SAF:转AtABF3基因)为研究材料,通过盆栽控制试验,比较研究了不同土壤水分条件下始花期各株系苜蓿的光合和荧光参数特征。结果表明,随土壤含水量从80% FC(田间持水量)降至40% FC,各株系叶片净光合速率(Pn)、蒸腾速率(Tr)和气孔导度(Gs)均呈下降趋势,叶片瞬时水分利用效率(WUEi)呈上升趋势。在40% FC时,各转基因株系Pn值(11.19~21.58 μmol·m-2·s-1)均显著(P<0.05)大于非转基因株系(6.06 μmol·m-2·s-1),各株系间WUEi无差异但以转AtABF3基因株系最大(3.22 μmol·mmol-1)。土壤水分降低过程中,各株系叶片初始荧光(Fo),最大光化学效率(Fv/Fm)和光化学淬灭系数(qP)值总体呈下降趋势,实际光化学效率(ΦPSⅡ)和表观电子传递效率(ETR)值呈先上升后下降,而非光化学淬灭系数(NPQ)值整体呈上升趋势。转基因株系qP值出现显著下降时的土壤水分含量(50% FC)低于非转基因(70% FC)苜蓿。在40% FC时,转基因株系Fv/Fm(0.78~0.82)值显著高于非转基因苜蓿(0.58)。总体表明,转多抗基因均不同程度地提高水分胁迫条件下新疆大叶苜蓿的光合性能,其中转codA基因株系可维持较高光合速率,转IbOr基因株系能维持较高光能利用能力,转AtABF3基因株系能够高效利用水分,转AtNDPK2基因株系总生物量最高。  相似文献

The dosenresponse relationship for a synthetic gonadotrophin-releasing hormone (GnRH) was studied in normally cycling heifers using the area under the luteinizing hormone (LH) curve as a response parameter. Oestrus was synchronized by an injection of 0.5 mg cloprostenol before the experiment started and after the 3rd treatment with GnRH. Treatment with GnRH as assigned in a Latin square included 5 dose levels (0, 10, 50, 100, 250 μg) and 5 treatment days over a period of 22 days. GnRH was capable of inducing an increase of plasma LH within 30 min after injection. Plasma LH response increased with increasing doses of GnRH, the largest increase being observed when the dose was raised from 50 μg to 100 μg. One heifer did not respond to any of the doses applied. The existence of an individual treshold dose of GnRH is suggested.  相似文献

The aim of this study was to determine the viability and cardiorespiratory effects of the association of epidural alpha-2 adrenergic agonists and lidocaine for ovariohysterectomy (OH) in bitches. Forty-two bitches were spayed under epidural anesthesia with 2.5 mg/kg body weight (BW) of 1% lidocaine with adrenaline (CON) or in association with 0.25 mg/kg BW of xylazine (XYL), 10 μg/kg BW of romifidine (ROM), 30 μg/kg BW of detomidine (DET), 2 μg/kg BW of dexmedetomidine (DEX), or 5 μg/kg BW of clonidine (CLO). Heart rate (HR), respiratory rate (fR) and arterial pressures were monitored immediately before and every 10 min after the epidural procedure. Blood gas and pH analysis were done before, and at 30 and 60 min after the epidural procedure. Animals were submitted to isoflurane anesthesia if they presented a slightest sign of discomfort during the procedure. Time of sensory epidural block and postoperative analgesia were evaluated. All animals in CON and DEX, 5 animals in ROM and CLO, 4 animals in XYL, and 3 in DET required supplementary isoflurane. All groups, except CLO, showed a decrease in HR. There was an increase in arterial pressures in all groups. Postoperative analgesia lasted the longest in XYL. None of the protocols were totally efficient to perform the complete procedure of OH; however, xylazine provided longer postoperative analgesia than the others.  相似文献

Blood selenium levels were found to fluctuate throughout the year, being highest during the indoor season when the greatest amounts of compound concentrates were fed. From October to January the average blood selenium levels increased from 0.10 μg/ml to 0.18 μg/ml (15 cows). Subcutaneous injections of barium selenate (500 mg selenium) increased the blood selenium levels significantly. The treated group (15 cows) reached average levels of about 0.21 μg/ml blood during the indoor season.The effect of oral supplementation of sodium selenite (for 2 months) on the levels of plasma selenium, blood selenium and glutathione peroxidase activity was investigated. Plasma selenium was found to give an immediate reflection of the daily selenium intake. Maximum activity of glutathione peroxidase was reached 1 month after the end of the supplementation period.It is concluded that if selenium enriched concentrates are used in a normal feeding regimen, further supplementation with selenium does not seem to be necessary.  相似文献

[目的] 探明内蒙古4种家畜乳的硒含量水平。[方法] 从内蒙古东北部到西部8个旗县采集双峰驼、蒙古马、萨能山羊和荷斯坦牛4种家畜代表性混合乳样48份,涉及188头(只/匹)动物个体乳样。利用原子荧光光谱法测定乳硒含量,并对乳硒含量的地区和畜种差异进行统计学分析和比较。[结果] 4种家畜乳硒含量存在地区差异:阿拉善左旗双峰驼乳硒含量[(5.96±0.41)μg/kg]约为鄂温克族自治旗双峰驼乳硒含量[(2.89±0.12)μg/kg]的2倍;呼和浩特市荷斯坦牛乳硒含量[(8.74±0.22)μg/kg]约为鄂温克族自治旗荷斯坦牛乳硒含量[(2.26±0.41)μg/kg]的3.8倍;正蓝旗蒙古马乳硒含量[(6.65±0.57)μg/kg]约为鄂温克族自治旗蒙古马乳硒含量[(1.02±0.42)μg/kg]的6.5倍;同在西部区,杭锦旗萨能山羊乳硒含量为[(8.95±1.32)μg/kg],略高于五原县萨能山羊乳硒含量[(6.28±0.80)μg/kg],但二者差异不显著(P>0.05)。家畜种类不同乳中硒含量也存在差异:双峰驼、萨能山羊和荷斯坦牛的乳硒含量整体高于蒙古马;萨能山羊乳硒含量显著(P<0.05)高于蒙古马乳。4种家畜乳硒含量数据都较为离散,提示乳中硒含量不仅存在跨东西部地区的差异,还可能存在局部地质丰度效应,也可能与外来饲草料和舔砖等有关。[结论] 内蒙古家畜乳硒含量整体较低,并存在东北部显著低于中西部地区的现象;富硒饲养是内蒙古地区养殖业和乳品加工业高品质发展的一个有效途径。  相似文献

叶绿素含量直接影响植物光合效率,从而决定其生物量。叶绿素酸酯a加氧酶(CAO)催化叶绿素酸酯a转化成叶绿素酸酯b,是叶绿素合成的限速酶之一。本研究克隆了蒺藜苜蓿MtCAO,该基因编码531个氨基酸。进化分析表明,MtCAO与双子叶植物的CAO亲缘关系较近。高等植物CAO基因的结构保守,均含有9个外显子。MtCAO主要在绿色组织尤其是叶片中高丰度表达。瞬时表达拟南芥原生质体试验显示,MtCAO-GFP融合蛋白定位于叶绿体。表达模式分析表明,茉莉酸甲酯(MeJA,100 μmol·L-1)及黑暗处理抑制MtCAO的表达,而24 h内NaCl (100 mmol·L-1)及聚乙二醇(PEG,5%) 处理先诱导后抑制其表达,且该基因受昼夜节律调控。这与对MtCAO启动子区顺式作用元件的预测结果一致。类似于超表达AtCAO的拟南芥,超表达MtCAO的拟南芥株系叶色正常,叶绿素含量及鲜重与野生型差异不显著,推测是由于MtCAO高度保守的A结构域中的降解子导致的。  相似文献

通过分离青海高寒草地针茅根际土壤细菌,以番茄早疫病菌、马铃薯枯萎病菌、马铃薯坏疽病菌和马铃薯炭疽病菌为指示菌,采用平板对峙法对其拮抗功能进行评价,并利用16SrDNA序列分析法进行鉴定。结果表明,从针茅根际土壤中共分离获得23株土壤细菌,17株可转代培养,根际土壤细菌数量介于6.4×10^6~36×10^6cfu·g^-1,其中4T1、5T1、10T1和10T3至少对1种病原真菌有抑菌作用,占细菌总数的17.39%,抑菌率介于46.68%~75.71%;菌株10T1的抑菌效果最好,对番茄早疫病菌、马铃薯枯萎病菌、马铃薯坏疽病菌和马铃薯炭疽病菌的抑菌率分别为67.14%、68.57%、61.43%和75.71%;23株土壤细菌均呈革兰氏阳性,杆状,大小介于3.79μm×0.85μm^10.50μm×2.37μm。结合培养形状、形态特征和16SrDNA序列分析将17株土壤细菌鉴定为7属12种,其中芽孢杆菌属8株,占鉴定总数的47.05%,为优势属。通过对青海高寒极端生境针茅根际土壤细菌的拮抗功能评价,为生防菌剂的开发提供了菌株资源。  相似文献
