The production of muramidase-released protein (MRP), extracellular protein factor (EF) and hemolysin (suilysin) by 101 Canadian field strains of Streptococcus suis capsular type 2 is described. Most strains (72%) isolated from diseased pigs were MRP-EF- and only 1 strain was MRP+EF+. This strain was also the only 1 to produce the hemolysin. Thirteen strains (15%) were MRP+ EF- and only 3 strains were MRP* EF-. All the strains isolated from clinically healthy pigs as well as a bovine and 2 human isolates had a MRP-EF- phenotype. In addition, 7 strains (8%) had a MRPS phenotype, which had so far been described for S. suis capsular type 1. In conclusion, most Canadian field isolates of S. suis capsular type 2 tested in this study do not produce the virulence-related proteins described so far for this bacterial pathogen.  相似文献

A total of 142 strains from different serotypes of Streptococcus suis isolated in Spain from diseased pigs (88 strains) and healthy carrier pigs (54 strains) were studied for the presence of a muramidase released protein (MRP) and an extracellular factor (EF). The following five phenotypes: MRP+EF+, MRP+EF-, MRP-EF+, MRP+EF* and MRP*EF- were detected. A high percentage of S. suis serotype 2 strains isolated from diseased pigs (84 per cent) belonged to phenotype MRP+EF+, but this phenotype has also been noticed in other serotypes (serotypes 1, 1/2 and 14). Both proteins were detected in S. suis serotype 2 strains (26%) isolated from healthy carrier pigs and one of both proteins in serotypes 1 and 14 (phenotype MRP+EF*). The isolation of S. suis strains from healthy pigs which have shown both proteins may support the epidemiological significance of these carriers in the maintenance, transmission and distribution of virulent strains within and between swine farms.  相似文献

Isolation and characterization of Streptococcus suis capsular types 9-22   总被引:7,自引:0,他引:7  
The incidence and biochemical patterns of Streptococcus suis capsular types 9-22 are presented. Of 148 untypeable (with types 1-8 antisera) isolates of S. suis recovered from diseased pigs, 10% were not capsulated. Of the remaining 134 isolates, only 53% belonged to capsular types 9-22; capsular types 22 and 9 were the most prevalent, representing 19% and 13%, respectively. Capsular type 15 (de Moor's group T Streptococcus) is reported here for the first time in North America since it was described in 1963 in Europe. Of 188 untypeable isolates recovered from clinically healthy pigs, 25% were noncapsulated. Of the remaining 141 isolates, 90% belonged to the new capsular types, and 87% were identified as 1 of 4 types: 17, 18, 19, and 21. Capsular types 12 and 20 were not detected among the Canadian isolates. Almost half of strains were arginine dihydrolase-negative, and 45% fermented mannitol, which is seldom a positive test with capsular types 1-8. Although some strains were negative with salicin or trehalose, none were negative for both sugars. Only 54% of isolates tested with 1 rapid multitest system were correctly identified as S. suis. A tentative biochemical profile that might be used with a microplate identification system is also presented. Biochemical identification using the conventional system instead of the rapid multitest system is preferable.  相似文献

The present study was undertaken to evaluate the ability of 33 Streptococcus suis capsular type 2 isolates to adhere to frozen sections of porcine lung. Twenty isolates originated from diseased pigs and 13 from the nasal cavities of clinically healthy pigs. All isolates from diseased animals adhered to lung sections; isolates from pneumonia adhered, in general, in greater numbers than isolates from meningitis. Only four isolates from clinically healthy animals showed a weak adherence to lung sections. Hydrophobic surface properties were also evaluated. All isolates tested appeared to possess a hydrophilic cell surface. The thickness of the capsular material correlated well with the degree of adherence. However, when the adherence capacity of a noncapsulated mutant was compared with that of the parent strain, it was found that the mutant strain had at least the same adherence capacity as the capsulated parent strain. The data suggest that S. suis capsular type 2 isolates involved in pathological conditions can adhere to porcine lung tissue. The adherence activity does not seem to involve hydrophobic interactions. The amount of capsular material seems to influence the adherence activity, but is probably not the only mechanism involved.  相似文献

This study was undertaken to determine the prevalence, capsular serotype, and antimicrobial susceptibility of Streptococcus suis isolated from slaughter pigs. Capsular serotype and antimicrobial susceptibility were determined by coagglutination test and agar dilution minimum inhibitory concentration, respectively. Streptococcus suis was isolated from 55 of the 406 palatine tonsillar samples tested (13.8%) and 14 of the 29 sampled herds (48.3%). Of the 55 isolates recovered from slaughter pigs, 26 (47.3%) were untypeable. Of the remaining 29 isolates, capsular serotypes 9 (9 isolates) and 16 (4 isolates) were the most common, followed by capsular serotypes 4 (3 isolates) and 7 (3 isolates). Every capsulated isolate was typeable and no palatine tonsillar sample yielded more than one serotype. Most of isolates were susceptible to low concentrations (MIC90) of amoxicillin (2 microg/mL), ceftiofur (1 microg/mL), and penicillin (1 microg/mL). No correlation was found between antimicrobial susceptibility and capsular serotype.  相似文献

A double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) was developed for the detection and the identification of Streptococcus suis capsular types 1, 2, 1/2, 3 and 22. The specificity of this test was first evaluated using reference strains of S. suis capsular types 1 to 28 and 1/2 as well as 15 different bacterial species susceptible to be isolated from swine. The ELISA developed was very specific for capsular types 1, 3 and 22 but it could not discriminate between capsular types 2 and 1/2. In a second study, S. suis isolates from 328, 493, 368 and 76 diseased pigs were used to detect capsular types 1, 2 or 1/2, 3 and 22 respectively. The relative specificity and sensitivity varied between 98% and 100%. The ELISA results were in excellent agreement with the standard techniques (biochemical tests, coagglutination and capsular reaction tests) in detecting both positive and negative strains. Kappa values were 0.80, 0.99, 0.97 and 1.00 for detecting S. suis capsular types 1, 2 or 1/2, 3, and 22 respectively. To evaluate the relative-sensitivity of the test, primary cultures from 73 diseased pigs and tissue samples from 67 diseased pigs were used directly for detecting these capsular types. With primary cultures, the relative specificity and sensitivity (95.9% and 91.6% respectively) remained high and the test was very suitable (Kappa = 0.87). 猪链球茵(SS)病是一种严重危害养猪业的人畜共患病,呈世界性分布.根据荚膜抗原特性的不同,猪链球茵被分为35个血清型(1/2型,1型~34型),其中以猪链球菌2型(SS2)流行最广,毒力最强.SS2主要的毒力因子有溶菌酶释放蛋白、胞外蛋白因子、荚膜抗原、溶血素等.然而,新近发现的一些与SS2致病性相关的基因序列和蛋白片段已经成为当前研究的重点和热点.在基因水平上的新发现主要有orf2毒力因子、89 kb毒力岛、转录调节因子、氨基酸通透酶、ABC转运子及表面锚定蛋白基因、反应调节因子RevS基因、纤连蛋白结合蛋白基因、编码噬茵体的基因序列Ssl和分泌性核酸酶SsnA基因等.蛋白水平上的新发现主要有脂磷壁酸的D-丙氨酰位点、自溶素、精氨酸脱亚氨酸酶系统、浑浊因子,38,45,39,44 ku蛋白,以及一些蛋白酶和纤维蛋白溶血酶原受体等.  相似文献

根据猪链球菌2型的mrp基因自设计和合成了一对可扩增长度为885bp目的片段的引物,成功地建立了检测溶菌酶释放蛋白(MPR)的PCR方法,用XbaⅠ内切酶进行酶切,获得了与预期一致的578bp和307bp2的2个片段,并进行了PCR的特异性试验和敏感性试验。对马链球菌兽疫亚种、猪丹毒杆菌、猪肺疫巴氏杆菌和猪肺炎支原体的PCR检测结果均呈阴性;检测的敏感度可达100个细菌。另外,对9株从病猪体内分离的猪链球菌2型菌株进行检测了8个呈阳性;对15份正常屠宰猪扁桃体分离物的检测结果是1份为阳性。结果表明此法特异性敏感性均很高,可作为MRP快速诊和流行病学调查的手段。  相似文献

从临床表现为脑膜炎、关节炎及败血症的发病猪分离到2株细菌,分别命名为JX02和JX03,经鉴定其形态、染色及培养特性均符合链球菌的特点,具α或β溶血,生化试验结果不稳定。用猪链球菌2型、1型和9型抗血清进行凝集试验,结果2株菌均能与猪链球2型抗血清凝集,其中1株菌对小鼠和家兔均有致病性,另1株菌只能致死家兔。PCR均能扩增2株菌的主要毒力因子基因。表明发病猪的病原不同于以往的C群链球菌,不是马链球菌兽疫亚种,而是属于R群的猪链球菌。  相似文献

为研究携带不同毒力因子的猪链球菌2型对细胞侵袭作用的差异,本实验室从全国各地分离100多株猪链球菌,PCR鉴定出10株链球菌2型,其中携带毒力因子菌株为7-4[MRP+EF+]、8-3[MRP+EF-]、ZH-1[MRP-EF+],另外选择1株LN-2 [MRP-EF-]与PK 15细胞进行黏附动力学试验,结果表明MPR毒力因子与菌株对PK-15细胞黏附具有密切相关性。  相似文献

Of a total of 561 isolates of Streptococcus suis recovered from various tissues of diseased pigs, 464 were from veterinary laboratories in Quebec and 97 were from western Canada, particularly Alberta (84). Almost 83% of all these isolates belonged to the 23 known S. suis capsular types. There was no marked difference between the two groups of isolates. Capsular type 2 was the most prevalent and represented 32% of all isolates. The other important capsular types were, in decreasing order, 3, 1/2, 8 and 4. Lungs, brain, and meninges were the source of the majority of isolates. More than 40% of all S. suis isolates were found in pure culture. The number of isolations of this microorganism was higher in piglets aged five to eight weeks than in any other age group.  相似文献

Streptococcus suis was recovered from 9 outbreaks of septicaemia and meningitis in weaned pigs between 1979 and 1983. Fifteen isolates from 7 outbreaks were identified as S. suis type 9, and 3 isolates from 2 outbreaks as S. suis type 2. Three further isolates of S. suis type 2 and an isolate of S. suis type 3 were recovered from cases of bronchopneumonia in weaned pigs from 4 other piggeries.  相似文献

Five inbred strains of mice were tested for their susceptibility to Streptococcus suis type 2 including the type strain, two isolates from meningitis in pigs and two isolates from tonsils of clinically healthy pigs. C57BL/6, ICR and ddY strain mice showed lower susceptibility to all strains of S. suis type 2 than BALB/c and SS strain mice. The type strain and the isolates from diseased pigs produced septicaemia and meningitis in BALB/c and SS mice inoculated with 10(8) colony forming unit of the bacteria and 60 to 100% of these infected mice died. On the other hand, mice inoculated with the isolates from healthy pigs showed mild clinical signs but none of them died. In BALB/c mice which died or developed nervous signs, the purulent meningo-encephalitis, myocarditis, ophthalmitis, labyrinthitis and otitis media were observed. S. suis type 2 antigen was demonstrated in these lesions by immunoperoxidase staining using rabbit S. suis type 2 antiserum. These results were similar to those in the experimentally infected pigs with these virulent and avirulent strains against mice. These results indicate that BALB/c and SS strains of mice are useful as an experimental model of S. suis type 2 infections in pigs, and that there are virulent and avirulent strains against mice and pigs among the strains of S. suis type 2.  相似文献

Streptococcus suis is an important pig pathogen which is associated with respiratory problems, meningitis and less fre-quently with a variety of other conditions(Hommez et al. 1986). S. suis type 1 causes disease mainly in 1–2 week old pigs while serotype 2 is found commoaily in 2–22 week old pigs, S. suis type 2 is a zoonosis. It can cause meningitis and septicaemia in man (Christensen & Kronvall 1985). Several other serotypes of S. suis have also been identified on the basis of the capsular poly-saccharide (Perch et al. 1983, Hommez et al. 1986). We present a case where we isolated S. suis types 1 and 2 from the brain and lungs respectively of the same diseased suckling piglet. This i/s the first reported case of S. suis types 1 and 2 in Finland.  相似文献

Streptococcus suis strains (n=411), isolated from diseased pigs in seven European countries were serotyped using specific antisera against serotype 1 to 28, and were phenotyped on the basis of their muramidase-released-protein (MRP) and extracellular-factor protein (EF) production. Overall, S. suis serotype 2 appeared to be most prevalent (32%), followed by serotype 9 (20%) and serotype 1 (12%). Serotype 2 was most frequently isolated in France, Italy and Spain, whereas serotype 9 was most frequently isolated in Belgium, The Netherlands and Germany. In the United Kingdom serotypes 1 and 14 were most frequently isolated. High percentages of S. suis serotype 1, 2, 1/2 and 14 strains, isolated from tissues associated with S. suis infections such as brain, serosa, joint, heart and organs expressed the EF-protein, indicating that in these serotypes expression of EF is likely to be associated with virulence. In contrast, strains belonging to serotype 7 and 9, isolated from tissues associated with S. suis infections did not produce EF. These results strongly suggest that in the serotypes 7 and 9 EF expression is not related to virulence. More than 80% of the S. suis serotype 9 strains produced an MRP* protein, a high molecular variant of the 136kDa MRP. Expression of MRP* in serotype 9 strains is possibly associated with virulence.  相似文献

The present study describes the prevalence of muraminidase-released protein (MRP) and extracellular factor (EF) proteins associated with virulence of Streptococcus suis serotype 2 from a collection of USA strains. Sixty-six strains belonging to serotypes 1, 2, 3, 4, 7, and 10, were analyzed with a set of double antibody sandwich ELISAs and Western blots. Nineteen of 34 serotype 2 strains from cases of swine meningitis had the MRP+EF+ phenotype. Five of 7 serotype 2 strains isolated from lungs had an MRP*EF- phenotype. An MRP-EF+ phenotype was found in 4/34 strains isolated from swine meningitis. The MRP*EF- and MRP-EF+ phenotypes have not been reported previously. All strains of serotypes other than 2, including isolates from cases of meningitis, had the MRP-EF- phenotype, suggesting that these strains must have other, as yet undetected, virulence factors.  相似文献

A total of 58 Streptococcus suis strains were isolated from deceased pigs submitted to the National Veterinary Institute, Regional Laboratory in Kuopio, Finland, over a 3 1/2 year period, most frequently from cases of pneumonia. The bacteria were isolated from cases of meningitis, sepsis, rhinitis, endocarditis and abortion. S. suis was also isolated from nasal cavity, lung and brain of some sick piglets without signs of inflammation. Further S. suis was detected in 12 out of 107 tonsils of healthy fatteners tested. S. suis strains were identified by biochemical methods followed by typing. The most common capsular types were 7, 3 and 2, respectively. Only one type 1 strain and no types 6 and 9 strains were found. All S. suis strains tested were sensitive to penicillin and ampicillin.S. suis is not uncommon in Finnish pig herds. S. suis may be regarded as a potentially pathogenic organism which under certain predisposing conditions may cause serious disease.  相似文献

The objective of this study was to determine the capsular serotypes and potential virulence factors of Streptococcus suis isolated from pigs with polyserositis. Among the 24 isolates evaluated, serotype 3 [7 (29%) of the isolates] and serotype 4 [5 (21%)] were the most common. The isolates were also studied for the presence of the genes mrp, epf, and sly, which encode muramidase-released protein (MRP), extracellular factor (EF), and suilysin (SLY), respectively. Of the 24 isolates, 8 carried mrp: 4 of serotype 3, 2 of serotype 2, and 2 of serotype 4. One mrp(+) isolate (serotype 2) also carried the epf gene. All 24 isolates carried the sly gene. The serotype and genotype distribution greatly differed from that reported for isolates from pigs with other clinical manifestations of S. suis infection in other countries.  相似文献

Virulence-associated gene profiling of Streptococcus suis isolates by PCR   总被引:11,自引:0,他引:11  
Definition of virulent Streptococcus suis strains is controversial. One successful approach for identification of virulent European strains is differentiation of capsular serotypes (or the corresponding cps types) and subsequent detection of virulence-associated factors, namely the extracellular factor (EF, epf), the muramidase-released protein (MRP, mrp) and the hemolysin suilysin (SLY, sly). In this work we present a novel multiplex PCR (MP-PCR) and an mrp variant PCR for identification and characterization of virulent S. suis strains. These new methods were used to identify association of disease with particular profiles of virulence-associated genes. The MP-PCR allowed identification of S. suis through detection of the housekeeping gene gdh, differentiation of four cps types (1, 2, 7 and 9), and detection of epf, mrp, sly and arcA (arginine deiminase from S. suis). Furthermore, this study describes the first PCR assay for differentiation of at least six mrp variants. Expression of the corresponding size variants of MRP was shown for four of the six mrp variants, but was undetectable for the two larger mrp variants in the particular strains investigated. The results of this study suggest that cps7 strains are associated with pneumonia and that variation of mrp is very pronounced among these strains. Gene profiles of invasive, pneumonia and carrier S. suis isolates by combination of PCR assays allowed differentiation of 24 different genotypes among cps1, 2, 7 and 9 strains. Forty-five percent of the invasive S. suis diseases investigated in this study were caused by only two of these genotypes, namely cps2/mrp+/epf+/sly+ and cps9/mrp(*)/epf-/sly+. Thus, this study demonstrates for the first time a uniform profile of the particular virulence-associated genes for the vast majority of the investigated invasive cps9 strains.  相似文献

A selective medium containing tryptic soy agar, 5% defibrinated bovine blood, crystal violet, nalidixic acid and gentamicin significantly improved the isolation rate of Streptococcus suis from tonsilar tissue of slaughtered pigs. Ninety-five percent of the S. suis isolates identified in Guelph were confirmed as S. suis in Copenhagen, but only six out of 21 isolates typed as capsular serotype 2 in Guelph were confirmed to possess serotype 2 antigen in Copenhagen. Sixty-four percent of the S. suis isolates were not typable within the current scheme of capsular serotypes from 1 to 13 and type 1/2.  相似文献
