用血凝/血凝抑制试验(HA/HI)检测自制的猪血凝性脑脊髓炎病毒(HEV)氢氧化铝胶灭活苗和蜂胶佐剂灭活苗免疫BALB/c小鼠后的抗体效价,依据小鼠抗体效价比较不同免疫佐剂的效果。结果表明,用氢氧化铝胶佐剂配制的疫苗可刺激接种动物产生快速免疫应答反应,抗体产生效价高、维持时间长,其效果优于其它佐剂,有望成为HEV灭活疫苗的候选佐剂。  相似文献

The immunogenicity of a sonicated extract (SE) and of outer membrane proteins (OMP) of Salmonella enteritidis was tested in birds of about 8 weeks of age. The dose, route of vaccination and the adjuvant used varied in different groups of birds. Two vaccine doses with or without adjuvant were given parenterally or orally 3 weeks apart. OMP vaccines gave significantly higher antibody titres than SE vaccines, as indicated by ELISA. The vaccines adjuvanted with oil produced higher antibody titres than those without any adjuvant. A dose of 1 mg of vaccine produced higher antibody titres than 0.5 mg of vaccine. Adjuvanted vaccine given subcutaneously elicited higher antibody responses than oral vaccines given without adjuvant. The birds were challenged with virulent S. enteritidis organisms at the end of the second week after a booster dose. None of the birds given 1 mg of OMP vaccine subcutaneously shed the organisms when tested by culturing cloacal swabs, although a few birds vaccinated with 0.5 mg of OMP vaccine did so. In general, adjuvanted OMP vaccines gave better protection than SE vaccines.  相似文献

蜂胶极其复杂的化学成分,决定了它具有广泛的生物学作用。其中,蜂胶具有抗菌、抗病毒及增强机体免疫功能等作用;蜂胶颗粒能与抗原相互交联成类免疫刺激复合物(ISCOM)结构,具有抗原“仓库”和免疫刺激复合物作用;蜂胶本身没有抗原性,但可起到免疫佐剂的作用,促进抗体的生成;这些作用及其间的协同作用决定了蜂胶是一种天然的免疫增强剂。自20世纪60年代以来,蜂胶的免疫增强作用及其在疫苗佐剂中的研究不断深入,并取得了满意效果,这为新型而有效疫苗的研制和开发提供一些理论指导。  相似文献

为筛选出理想的鸭TMUV-AIV(H9亚型)二联灭活疫苗免疫佐剂,将灭活的坦布苏病毒液和禽流感病毒(H9亚型)尿囊液分别与白油佐剂、卡波姆佐剂、铝胶佐剂、蜂胶佐剂制成抗原含量相同的TMUV-AIV二联灭活疫苗。应用阻断ELISA方法和血凝抑制方法测定免疫鸭血清中坦布苏病毒和禽流感病毒抗体,比较不同佐剂疫苗对试验鸭的免疫效力。结果表明,以白油作为佐剂的灭活疫苗组免疫效果最好,抗体产生快、抗体水平优于其他各组,免疫2周后60%(6/10)血清样品HI抗体可达1∶8以上,80%血清样品呈TMUV抗体阳性,是制备鸭TMUV-AIV(H9亚型)二联灭活疫苗的理想佐剂。  相似文献

采用鸽新城疫ND-GS 01株毒为种毒,分别研制出了尿囊液毒蜂胶佐剂灭活疫苗和四元材料(鸡胚尿囊液、羊水、尿囊膜、羊膜)混合病毒蜂胶佐剂灭活苗。对研制的两种灭活疫苗经安全性检验、急性毒性试验,免疫抗体消长水平与免疫持续期对比试验,结果表明,四元材料的疫苗与尿囊液疫苗性能一致。  相似文献

佐剂是疫苗的重要组成部分,不同佐剂对体液免疫会产生不同的作用.选择合适的佐剂不仅可以增强疫苗的免疫效果,而且可以提高机体的抵抗力.本试验选用白油佐剂、弗氏佐剂和蜂胶佐剂分别与灭活的新城疫病毒制成疫苗,免疫雏鸡,探讨不同佐剂对鸡体液免疫的影响.结果表明,试验组鸡抗体水平高,饲养至61日龄均未发生新城疫,其中弗氏佐剂的效果最好,蜂胶佐剂的效果次之,油佐剂的效果最次.  相似文献

作者制备了3批牦牛大肠埃希氏菌病蜂胶疫苗。制备了蜂胶佐剂和培养基,进行了牦牛大肠埃希氏菌的菌液培养、纯粹检验、活菌计数、灭活及无菌检验,配苗、分装及成品检验。3批牦牛大肠埃希氏菌病蜂胶疫苗无菌检验为阴性;物理性状检验为:静置后上层是乳黄色液体,下层为土黄色沉淀,振荡后呈均匀混浊液;经家兔安全检验结果为安全;经家兔效力试验,免疫后家兔抗体效价≥12.709lg2,对家兔具有免疫保护作用;蜂胶疫苗经防冻试验证明,随着蜂胶含量的增高,防冻效果越好。本试验为西藏牦牛大肠埃希氏菌病的预防提供依据。  相似文献

为研究比较5种塞内卡病毒疫苗佐剂,分别用4种自主研发佐剂和进口ISA206佐剂配制塞内卡病毒灭活疫苗,检测疫苗的理化性质、安全性及免疫效力,并对检测结果进行对比分析。结果显示,进口ISA206佐剂疫苗黏度为43.02 cP,自主研发佐剂疫苗黏度均在27.88 cP以下,其他理化性质无差异;5批疫苗安全性试验均未见明显异常;5批疫苗免疫动物后均可诱导机体产生中和抗体,二免后14 d抗体滴度在211以上,4批自主研发佐剂疫苗免疫组抗体滴度水平高于进口ISA206佐剂疫苗约1个滴度,攻毒保护结果均在4/5以上,2批自主研发佐剂疫苗免疫组保护率可达5/5。结果表明,用4种自主研发佐剂制备的塞内卡病毒灭活疫苗质量不低于用进口ISA206佐剂制备的塞内卡病毒灭活疫苗。  相似文献

为了探讨不同佐剂的奶牛乳房炎多联苗对小鼠免疫机能的影响,笔者将小白鼠分为3个组,每组30只,分别为蜂胶苗组、铝胶苗组和空白对照组。用制备的氢氧化铝胶和蜂胶2种乳房炎佐剂多联苗分别免疫小鼠,分别于免疫后的7、14、21、30和45 d采血,进行2种多联苗免疫后E-玫瑰花环率和淋巴细胞转化率测定。结果表明,蜂胶和铝胶2种佐剂苗与空白对照组相比均能显著提高小鼠的细胞免疫水平,2种苗的细胞免疫水平均在免疫后30 d左右达到最高峰(P<0.01),但在免疫后第30和45天时E-玫瑰花环率和淋巴细胞转化率,蜂胶苗组均极显著高于铝胶苗组(P<0.01),说明蜂胶佐剂比铝胶佐剂能更强地增强多联苗抗原对小鼠T细胞的致敏程度。该研究为进一步明确乳房炎多联苗免疫机制及筛选更好的免疫佐剂提供了科学数据。  相似文献

以蜂胶为佐剂制备小鹅瘟蜂胶佐剂灭活苗,将病毒液灭活后按一定比例加入蜂胶佐剂制备疫苗,并进行疫苗安全性、免疫持续期、效力对比、保存期及区域应用试验,结果表明小鹅瘟蜂胶灭活疫苗安全性好,第5天即产生抗体且抗体持续时间长(90d),免疫效果远好于油乳剂灭活苗,推广试验也取得了令人满意的结果。  相似文献

Enzyme-linked immunosorbent assay (ELISA) and crossed immunoelectrophoresis (IEP) were used to investigate antibody responses of sheep vaccinated with a double adjuvanted or single adjuvanted commercial foot rot vaccine. ELISA detected an antibody response of greater magnitude to the double adjuvant vaccine compared with the single adjuvant vaccine. Sera from sheep vaccinated with double adjuvant vaccine recognised at least six antigens of Bacteroides nodosus in crossed IEP while sera from the single adjuvant vaccinated sheep recognised one antigen. The use of non-denatured antigens of B nodosus in ELISA and crossed IEP enabled quantitative comparisons of antibody responses to the different foot rot vaccines to be made.  相似文献

Avridine, a lipoidal amine with interferon-inducing and adjuvant properties, was an effective adjuvant for Newcastle disease antigen (NDA) in chickens. Eleven vaccine lots were evaluated: 2 commercial water-in-oil vaccines, 4 experimental oil emulsion vaccines, 4 avridine-containing vaccines, and a control lot of nonadjuvanted antigen. Avridine significantly enhanced the immunologic responses of chickens against NDA. Chickens vaccinated with the avridine-containing vaccines had significantly higher antibody titers (hemagglutination inhibition) than did chickens vaccinated with the commercial vaccines. Experimental oil emulsion vaccines prepared from the same antigens as avridine-adjuvanted vaccines induced higher hemagglutination inhibition antibody titers after primary but not after booster vaccination. Use of avridine as an adjuvant for NDA in vaccines for chickens induced immunologic protection rates similar to those induced by oil emulsion vaccines, without causing the reactogenic and tissue residue problems associated with the use of oil vaccines in chickens.  相似文献

BackgroundNew-generation adjuvants for foot-and-mouth disease virus (FMDV) vaccines can improve the efficacy of existing vaccines. Chinese medicinal herb polysaccharide possesses better promoting effects.ObjectivesIn this study, the aqueous extract from Artemisia rupestris L. (AEAR), an immunoregulatory crude polysaccharide, was utilized as the adjuvant of inactivated FMDV vaccine to explore their immune regulation roles.MethodsThe mice in each group were subcutaneously injected with different vaccine formulations containing inactivated FMDV antigen adjuvanted with three doses (low, medium, and high) of AEAR or AEAR with ISA-206 adjuvant for 2 times respectively in 1 and 14 days. The variations of antibody level, lymphocyte count, and cytokine secretion in 14 to 42 days after first vaccination were monitored. Then cytotoxic T lymphocyte (CTL) response and antibody duration were measured after the second vaccination.ResultsAEAR significantly induced FMDV-specific antibody titers and lymphocyte activation. AEAR at a medium dose stimulated Th1/Th2-type response through interleukin-4 and interferon-γ secreted by CD4+ T cells. Effective T lymphocyte counts were significantly elevated by AEAR. Importantly, the efficient CTL response was remarkably provoked by AEAR. Furthermore, AEAR at a low dose and ISA-206 adjuvant also synergistically promoted immune responses more significantly in immunized mice than those injected with only ISA-206 adjuvant and the stable antibody duration without body weight loss was 6 months.ConclusionsThese findings suggested that AEAR had potential utility as a polysaccharide adjuvant for FMDV vaccines.  相似文献

IgG antibody response in chickens immunized with F4 fimbriae extracted from local enterotoxigenic Escherichia coli (ETEC) strain was studied during a 98-day immunization period for comparing the efficacy of four adjuvants: Freund' adjuvant, Quil A (QA), propolis and extract from Cochinchina momordica seed (ECMS). For this purpose, chickens were immunized with F4 fimbriae alone or combined with one of the above adjuvants on days 1 and 21. IgG antibody levels in serum and egg yolk (by ELISA) were measured on days 0, 7, 14, 21, 28, 35, 42, 49, 56, 70, 84 and 98. The egg production of each group was also determined during days 1-7 and the following four weeks. The results showed that QA could enhance antibody titre, as good or almost as good as Freund's adjuvant, whereas the titres of ECMS and propolis groups were relatively lower, with the overall order: Freund's adjuvant>QA>ECMS>propolis both in serum and egg yolk. However, the significant decrease of egg production was merely observed in the Freund's adjuvant group. It is concluded that the four adjuvants tested can stimulate immune response to F4 fimbriae in chickens, with Freund's adjuvant giving the best results, followed by QA.  相似文献

采用热抽提法提取4种肠毒素性大肠杆菌菌毛蛋白。K88、K99、F41和987p菌毛蛋白分别制成弗氏佐剂苗;K88还制成白油佐剂苗,氢氧化铝胶苗和蜂胶佐剂苗;另将4种菌毛等比例混合制成弗氏佐剂苗。分别对产蛋鸡进行免疫,用微量凝集反应和血凝抑制试验检测卵黄抗体效价。结果表明,K88菌毛较其他3种菌毛免疫性好,诱导抗体效价最高而且能长时间维持;987p菌毛能快速诱导抗体的产生,但整体效价低。K88不同佐剂苗中,铝胶佐剂能较快地诱导抗体的产生,蜂胶佐剂苗抗体持续时间短,弗氏佐剂能诱导高效价的抗体产生而且能长时间持续。  相似文献

选择4种中药成分(黄芪多糖、淫羊藿多糖、峰胶黄酮、人参皂甙)组成2个复方,各按3个剂量水平与新城疫疫苗混合后免疫雏鸡,以无佐剂苗和油乳苗为对照,分别于免疫后7、14、21、28、35、42d采血,用MTT法和微量血凝法测定T淋巴细胞转化和血清抗体效价的动态变化。结果表明。中药佐剂能显著促进淋巴细胞转化。提高血清抗体效价,多数时间点与油佐剂的效果相当,部分时间点显著强于油佐剂。综合判定方1高剂量和方2中剂量的效果最好。  相似文献

旨在筛选适宜于猪丹毒丝菌灭活疫苗的佐剂。以分离鉴定出的猪丹毒丝菌1a型HG-1株灭活菌体为抗原分别配制矿物油佐剂疫苗(简称矿物油疫苗)、氢氧化铝胶佐剂疫苗(简称铝胶疫苗)、ISA201双相油乳佐剂疫苗(简称ISA201疫苗)、GEL02水溶性聚合物佐剂疫苗(简称GEL疫苗)、IMS1313水溶性纳米佐剂疫苗(简称IMS1313疫苗)共5种佐剂的灭活疫苗。小鼠免疫保护试验结果表明,二免14 d后使用约为4 LD50的HG-1株对小鼠进行腹腔攻毒,矿物油疫苗和GEL疫苗的保护率分别为100%(7/7)和71%(5/7),其他三种佐剂疫苗的保护率均为14%(1/7)。本研究进一步选择铝胶疫苗和GEL疫苗进行猪体对比试验;仔猪安全性试验结果表明,两种佐剂疫苗的副反应均较小;免疫保护试验结果表明,两次免疫后使用约为16 LD100的HG-1株对免疫仔猪进行耳缘静脉攻毒,两种佐剂疫苗的保护率分别为60%(3/5)和100%(5/5)。本研究最终选择GEL佐剂作为开发猪丹毒灭活疫苗的最适佐剂。  相似文献

Flagellin, a bioactive Toll-like receptor (TLR) 5 ligand, may trigger the innate immunity that in turn is important for subsequent adaptive immune responses. In the present study, the adjuvant effects of the monomeric and polymeric forms of Salmonella flagellin (mFliC and pFliC, respectively) were examined in specific-pathogen free (SPF) chickens immunized intramuscularly (i.m.) or intranasally (i.n.) with formalin-inactivated avian influenza virus (AIV) H5N2 vaccines. Results showed that mFliC cooperating with the 64CpG adjuvant significantly induced influenza-specific antibody titers of plasma IgA in the i.m.-vaccinated animals. The nasal IgA levels in the i.n.-mFliC-coadministrated AIV vaccinated chickens were significantly elevated compared to levels observed in the control group (H5N2 vaccine alone). The pFliC cooperating with the 64CpG adjuvant significantly enhanced cell proliferation of splenocytes in the i.m.-vaccinated animals. TLR3 and TLR5 expressions were activated by flagellin stimulation in vitro and in vivo. These results suggest that flagellin can be used as an adjuvant in an AIV H5N2 vaccine, especially for mucosal immunity.  相似文献
