为探讨水牛睾丸生精上皮细胞冷冻保存的适宜冷冻保护剂及其浓度,在DMEM中分别添加不同浓度的冷冻保护剂二甲基亚砜(DMSO)、甘油(G)、丙二醇(PG)和乙二醇(EG),以及10%的胎牛血清(FBS),对3~5月龄水牛生精上皮细胞进行冷冻保存,解冻后台盼蓝染色测定细胞活率.结果显示,当以0%,5%.10%,15%,20%DMSO添加时,10%DMSO组的细胞解冻后活率均显著高于其他各组(P<0.05);当以0%,20%,25%,30%,35%,40%G添加时,35%G组的细胞解冻后活率均显著高于其他各组(P<0.05);当以0%,5%,10%,15%,20%,25%PG添加时,15%~25%PG各组的细胞解冻后活率均显著高于其他各组(P<0.05),但以20%PG组为最高;当以0%,5%,10%,15%,20%EG添加时,5%~20%EG各组的细胞解冻后活率均显著高于0%EG组,但以10%EG组为最高;而对4种冷冻保护剂各最优浓度组的细胞解冻后活率比较,10%DMSO、35%G组的细胞活率均显著高于20%PG、10%EG组(P<0.05).结果表明,以添加10%DMSO或35%G于含10%FBS的DMEM冷冻液中,采用两步慢速冷冻,液氯保存,37℃水浴1 min解冻,是一种具有较高复苏率的冷冻保存水牛生精上皮细胞的方法.  相似文献

采用两步酶解和差异贴壁法分离、纯化鸡精原干细胞(SSCs),比较了6种冷冻保护液对其冷冻保存效果,结果表明:FBS浓度为10%时,10%DMSO与10%甘油解冻后细胞存活率分别为54.05%和37.49%,差异显著(P<0.05);以10%DMSO为冷冻保护剂时,FBS浓度从10%增加到20%,解冻后细胞存活率分别为54.05%和69.06%,差异显著(P<0.05);在冷冻液Ⅰ基础上,添加3种不同浓度的蔗糖溶液,解冻后细胞存活率分别为52.49%、47.65%和51.94%,三者之间差异不显著(P>0.05),且与冷冻液Ⅰ组差异也不显著(P>0.05);将解冻后细胞接种饲养层上培养,除甘油组外,SSCs均能增殖并形成AKP阳性集落,但10%DMSO加上20%FBS组细胞增殖速度和集落数优于其他组合,表明10%DMSO加上20%FBS为鸡SSCs最佳冷冻保护剂。  相似文献

通过比较不同冷冻保存方法和冷冻保护剂对小鼠耳皮肤成纤维细胞冷冻-解冻复苏后细胞存活率、48h贴壁率和细胞生长曲线的影响,筛选适宜的小鼠耳皮肤成纤维细胞冷冻保存方法和冷冻保护剂。结果表明,以100mL/L二甲基亚砜(DMSO)作为冷冻保护剂进行小鼠耳皮肤成纤维细胞冷冻保存时,方法3处理组解冻复苏后细胞存活率和培养48h细胞贴壁率均高于方法2处理组(P>0.05),并分别极显著高于方法1和方法4。采用方法3进行冷冻保存时,以100mL/L DMSO作为冷冻剂的细胞贴壁率显著高于100mL/L甘油(GL)组(P<0.05),且冷冻解冻后的细胞呈现正常的分裂增殖生长模式。因此,宜选择100mL/L胎牛血清(FBS)+100mL/L DMSO+DMEM作为冷冻保护液,采用方法3进行小鼠耳皮肤成纤维细胞的冷冻保存。  相似文献

以甘油与DMSO为冷冻保护剂,添加5%-20%胎牛血清的DMEM为基础冻存液,对鲁西黄牛皮肤成纤维细胞进行冷冻保存,结果表明:①同浓度DMSO为冷冻保护剂效果优于同浓度的甘油(P<0.05),但添加50%血清时并不明显(P>0.05),可能与较高血清浓度有关,血清对冻存细胞的影响高于冷冻保护剂的作用。②血清浓度一定时,DMSO和甘油浓度在10%-15%时,冻存效果较好。③当甘油与DMSO浓度一定时,血清浓度越高冷冻保存效果越好。④甘油与DMSO浓度一定时(DMSO浓度5%除外),添加10%和20%血清效果无差异(P>0.05)。因此,在实际应用中,考虑成本等因素,血清浓度达到10%时就已经能达到一般试验要求。  相似文献

采用PBS作为冷冻基础液,分别用甘油和二甲基亚砜(DMSO)作为冷冻保护液,在程序化冷冻保存和玻璃化冷冻保存条件下,研究小鼠生发泡期(GV期)卵母细胞的抗冻能力。结果表明,2种冷冻方法对小鼠GV期卵母细胞解冻后形态正常率和存活率无显著影响(P>0.05)。冷冻保护剂种类对小鼠GV期卵母细胞解冻后形态正常率无显著影响(P>0.05);但对存活率有显著影响,玻璃化冷冻采用二甲基亚砜作为冷冻保护液效果极显著优于甘油(P<0.01)。以冷冻效果较好的二甲基亚砜作为冷冻保护液,采用玻璃化冷冻不同发育阶段(GV期和MⅡ期)的小鼠卵母细胞,解冻后形态正常率无显著差异(P>0.05),但存活率GV期要显著优于MⅡ期卵母细胞(P<0.05)。  相似文献

本试验采用不同的稀释液对兔精液进行冷冻保存,并比较了在稀释液中添加不同防冻剂以及平衡时间对兔精液冷冻保存效果的影响.结果表明:稀释液Ⅱ稀释的精液,解冻后,精子活率明显高于稀释液Ⅰ稀释的(P<0.01),其复苏率显著高于后者(P<0.05).以稀释液Ⅱ为基础液,添加不同浓度(27.3%,15%,12%,9%)的DMSO对兔精液冷冻保存,结果解冻后精子活率以15%及12%两组较好,其中添加15%的优于12%的,但差异不显著(P>0.05).选择不同种类的防冻剂(7%甘油与15%DMSO)比较时,发现7%甘油组解冻后精子活率明显低于15%DMSO组,二者差异极显著(P<0.01),其复苏率显著低于后者(P<0.05).以15%DMSO作为防冻剂,采用不同的平衡时间在5 ℃平衡,结果发现平衡30 min和60 min的解冻后精子活率较好,但差异不显著(P>0.05),而90 min和120 min的冷冻效果较差.  相似文献

为了探讨新西兰兔精液超低温冷冻保存的最佳方法,试验采集新西兰成年公兔精液,采用0.25 mL细管冷冻法进行超低温冷冻,以解冻后精液活率、质膜完整性、顶体完整率为指标,研究距离液氮面不同高度和冷冻保护剂种类对冷冻保存兔精液品质的影响。结果表明:冷冻精液距离液氮面3 cm,其精液活率、质膜完整性和顶体完整率明显优于1 cm和5 cm;在冷冻稀释液中添加甘油相对于其他冷冻保护剂对精液品质的保护效果好,甘油浓度为3%时相对其他浓度效果较好。说明了距液氮面3 cm冷冻,冷冻稀释液中添加3%甘油对兔精液的保存效果较好。  相似文献

采用假阴道法采集成年种公牛精液,经精液品质检查后,合格精液用四种不同冷冻稀释液进行稀释、冷冻、以筛选出适宜的冷冻稀释液和最佳冷冻保护剂.结果表明:Ⅱ号稀释液优于其它3种稀释液(P<0.05);甘油为最佳冷冻保护剂.优于乙二醇和DMSO冷冻效果(P<0.05),其最佳浓度为7%,精子冷冻后活率平均可达0.45±0.575,畸形率15.44%,项体完整率57.82%;奶牛细管冻精的解冻效果以75℃水浴,2 S到6 S的解冻时间最佳.  相似文献

二甲基亚砜(DMSO)、丙二醇(PROH)、乙二醇(EG)和甘油(GL)4种冷冻保护剂程序化冷冻牛GV期卵母细胞的结果表明,EG和PROH的保护效果比GL和DMSO好。4种不同冷冻方法冷冻保存牛GV期卵母细胞,比较解冻后卵母细胞的体外成熟率、受精后卵裂率。结果表明,在程序化冷冻法与细管玻璃化法(Straw)之间的差异不显著(P>0.05),在开放式拉管法(OPS)与毛细玻管法(GMP)之间的差异不显著(P>0.05);但OPS和GMP与程序化冷冻法和Straw之间的差异极显著(P<0.01)。玻璃化冷冻效果优于程序化冷冻。说明GMP和OPS玻璃化冷冻优于Straw玻璃化冷冻。说明可以采用GMP方法冷冻保存牛GV期卵母细胞。  相似文献

在BF5稀释液中分别添加不同水平的牛血清白蛋白(BSA)(0.5,1,1.5 g/L)、二甲基乙酰胺(DMA)(0.5%,1%,1.5%)、甲基-β-环糊精载胆固醇(CLC)(1,2.5,5 g/L),对成年健康猪精液进行冷冻保存,于不同温度(37,50,70℃)解冻后分别检测冷冻后精子的活率、活力、质膜完整性、项体完整率、线粒体活性.结果显示,0.5 g/L BSA组冷冻后精子活力、质膜完整率、顶体完整率和线粒体活性均高于其他组,但差异不显著(P0.05).1% DMA组冷冻后精子活率和活力显著优于1.5%DMA组(P<0.05),同时也高于对照组(3%甘油)冻后精子活率和活力,但差异不显著.不同质量浓度CLC组冷冻后精子的活率、活力、质膜完整性和线粒体活性与对照组相比差异不显著.50℃和70℃解冻组精子的活率和线粒体活性显著高于37℃解冻组的精子;50℃解冻组精子的活力和质膜完整率显著高于其他2组.因此,在猪精液冷冻稀释液中,用DMA可以代替甘油作为渗透性保护剂,且以1%DMA,50℃解冻最佳.  相似文献

1-(2-Chloroethyl)3-cyclohexyl-1-nitrosourea (CCNU) is an alkylating agent in the nitrosourea subclass. A prospective evaluation of CCNU was done to determine the maximally tolerated dosage of CCNU in tumor-bearing cats. Response data were obtained when available. Twenty-five cats were treated with CCNU at a dosage of 50-60 mg/m3 body surface area. Complete hematologic data were available for 13 cats. Neutropenia was the acute dose-limiting toxicity. The median neutrophil count at the nadir was 1,000 cells/microL (mean, 2,433 cells/microL; range, 0-9,694 cells/microL). The time of neutrophil nadir was variable, occurring 7-28 days after treatment, and counts sometimes did not return to normal for up to 14 days after the nadir. Based on these findings, a 6-week dosing interval and weekly hematologic monitoring after the 1st treatment with CCNU are recommended. The nadir of the platelet count may occur 14-21 days after treatment. The median platelet count at the nadir was 43,500 cells/microL. No gastrointestinal, renal, or hepatic toxicities were observed after a single CCNU treatment, and additional studies to evaluate the potential for cumulative toxicity should be performed. Five cats with lymphoma and 1 cat with mast cell tumor had measurable responses to CCNU. Phase II studies to evaluate antitumor activity should be completed with a dosing regimen of 50-60 mg/m3 every 6 weeks.  相似文献

Human and equine athletes are reported to have a high prevalence of gastric disease, and anecdotal evidence suggests a similar phenomenon applies to racing sled dogs. To investigate the prevalence of gastric disease in racing sled dogs, we conducted 2 gastroscopy studies on dogs competing in the annual Iditarod Sled Dog Race. A pilot study of dogs that were either dropped from the 2000 Iditarod Sled Dog Race because of illness or that finished the race indicated that, approximately 5 days after competing, 10 of 28 dogs (35%) had endoscopic evidence of gastric ulceration, erosion, or hemorrhage. The next year, an endoscopic study of 73 dogs participating in the 2001 Iditarod race was performed in order to evaluate a larger population of dogs. Data from 70 of these dogs could be used; 34 (48.5%) had ulceration, erosion, gastric hemorrhage, or some combination of these findings. When this group of 70 dogs was compared retrospectively to a control group of 87 dogs presented to the Texas A&M University (TAMU) Veterinary Medical Teaching Hospital, the Iditarod sled dogs had a significantly higher prevalence (P = .049) of gastric lesions. These findings suggest that, similar to athletes of other species, elite canine athletes have an increased prevalence of gastric disease compared to the canine population at large.  相似文献

Continuous rate infusion (CRI) of furosemide in humans is considered superior to intermittent administration (IA). This study examined whether furosemide CRI, compared with IA, would increase diuretic efficacy with decreased fluid and electrolyte fluctuations and activation of the renin-angiotensin-aldosterone system (RAAS) in the horse. Five mares were used in a crossover-design study. During a 24-hour period, each horse received a total of 3 mg/kg furosemide by either CRI (0.12 mg/kg/h preceded by a loading dose of 0.12 mg/kg IV) or IA (1 mg/kg IV q8h). There was not a statistically significant difference in urine volume over 24 hours between methods; however, urine volume was significantly greater after CRI compared with IA during the first 8 hours ([median 25th percentile, 75th percentile]: 9.6 L [8.9, 14.4] for CRI versus 5.9 L [5.3, 6.0] for IA). CRI produced a more uniform urine flow, decreased fluctuations in plasma volume, and suppressed renal concentrating ability throughout the infusion period. Potassium, Ca, and Cl excretion was greater during CRI than IA (1,133 mmol [1.110, 1,229] versus 764 mmol [709, 904], 102.7 mmol [96.0, 117.2] versus 73.3 mmol [65.0, 73.5], and 1,776 mmol [1,657, 2.378] versus 1,596 mmol [1,457, 1,767], respectively). Elimination half-lives of furosemide were 1.35 and 0.47 hours for CRI and IA, respectively. The area under the excretion rate curve was 1,285.7 and 184.2 mL x mg/mL for CRI and IA, respectively. Furosemide CRI (0.12 mg/kg/h) for 8 hours, preceded by a loading dose (0.12 mg/kg), is recommended when profound diuresis is needed acutely in horses.  相似文献

The epidemiology of H. placei and of other gastrointestinal nematodes in yearling dairy cattle was examined on two farms in Kiambu District, central Kenya during each of 13 one-month periods from April 1993 to April 1994. On each farm, 32 newly weaned dairy calves were given a single dose of albendazole and then placed on experimental pastures. Twelve of the animals were designated for bi-monthly slaughter (n = 2) and analysis of worm population characteristics and 20 were designated for blood and faecal collection and for weighing. Two parasite-free tracer calves were grazed alongside the weaner calves each month throughout the study period and were also slaughtered for analysis of worm populations. Faecal egg counts, haematological and serum pepsinogen determinations, herbage larval counts, and animal live weight changes were recorded monthly. The study revealed that Haemonchus placei, Trichostrongylus axei, Cooperia spp. and Oesophagostomum radiatum were responsible for parasitic gastroenteritis and that H. placei was the predominant nematode present in the young cattle on both farms. Faecal egg counts from resident cattle and necropsy worm counts revealed that pasture larval levels were directly related to the amount of rainfall. The total worm burdens in the animals were highest during the rainy season (March–June and October–December) and lowest during the dry seasons (July–September and January–February). The very low recovery of immature larvae of H. placei from the tracer calves indicated that arrested development is not a feature of the life cycle of this parasite in central Kenya. The maintenance of the parasite population depended on continuous cycling of infection between the host and the pasture. The agroclimatic conditions of the study area were such that, in general, favourable weather conditions for the development and survival of the free-living stages of gastrointestinal nematodes existed all year round.  相似文献

The occurrence of ectoparasites in sheep flocks is frequently reported but seldom quantified. Sheep production used to be a predominantly family activity in the state of Sa~o Paulo (Brazil), but it began to become a commercial activity in the past decade. Thus, information about the ectoparasites existing in sheep flocks has become necessary. The present data were obtained by means of questionnaires sent to all sheep breeders belonging to the `Associaça~o Paulista de Criadores de Ovinos' (ASPACO; Sa~o Paulo State Association of Sheep Breeders). Response reliability was tested by means of random visits paid to 10.6% of the respondents. Most of the properties (89.5%) reported the presence of one or more ectoparasites. Screw-worm (Cochliomyia hominivorax) was the most frequent ectoparasite (72.5%), followed by bot fly larvae (Dermatobia hominis, 45.0%), ticks (Amblyomma cajennense) and Boophilus microplus, 31.3%) and finally lice (Damalinia ovis, 13.8%). Combined infestations also occurred, the most common one being screw-worm with bot fly larvae (36.0%) followed by bot fly larvae with ticks (13.9%), screw-worm with ticks (9.3%), bot fly larvae with lice (6.9%), and ticks with lice (5.0%). The most common triple combination was screw-worm, bot fly larvae and ticks (12.8%). Breeds raised for meat or wool were attacked by bot fly larvae and ticks more often than other breeds. Lice were only absent from animals of indigenous breeds. The relationships among these ectoparasites are discussed in terms of sheep breeds, flock size, seasonality and the ectoparasitic combinations on the host.  相似文献

Our particular attention in this article was given to natural mediators for macrophages isolated from the sites of tissue injury. A number of chemotactic factors, which may satisfy many criteria making them acceptable as inflammatory leucocyte chemotactic factors, has been separated. An experiment was conducted to study disappearance of C14 to C18 fatty acids, lag times and biohydrogenation (BH) rates of C18 fatty acids of ground soya bean and canola seeds in situ. Three ruminally fistulated Dallagh sheep were used to determine ruminal BH of unsaturated fatty acids (UFAs). Differences in the disappearance of fatty acids through the bags and lag times were observed between the oilseeds. We saw that the longer the incubation time of the oilseeds in the rumen, the lower the content of C18:2 and C18:3. Significantly higher lag times for both C18:2 and C18:3 were observed in ground canola compared to ground soya bean. BH rates of C18:2 and C18:3 fatty acids in soya bean were three times higher than those of canola. These results suggest that the fatty acid profile of fat source can affect the BH of UFAs by rumen micro‐organisms. So that UFAs of canola had higher ability to escape from ruminal BH. It seems that fatty acid profile of ruminant products is more affected by canola seed compared to soya bean seed.  相似文献

Serum biochemical values and the wet weight to dry weight ratios of tissues were determined in yaks in Shandan county of Gansu province. The liver, kidney, heart and muscle contents of seven elements in yaks were also analysed. Most serum biochemical values were similar to those of cattle, camels and sheep, but the calcium concentration was considerably above the normal range for other ruminants.
