This study was conducted to estimate different levels of protein supplementary diet on gene expressions related to intramuscular deposition in early‐weaned yaks. Results showed that supplementary dietary protein significantly increased final weight, average daily gain (ADG), intramuscular fat (IMF), serum free fatty acid (FFA), total triglycerides, total cholesterol (Ch), low‐density lipoprotein cholesterol (LDL) and high‐density lipoprotein cholesterol (HDL) content. There was a quadratic response of ADG, IMF, FFA, Ch, HDL and LDL to dietary crude protein (CP) level. Lipoprotein lipase (LPL), fatty acid synthase (FAS) and acetyl‐CoA carboxylase (ACC) enzyme activities were significantly increased by supplementary dietary CP, while hormone‐sensitive lipase (HSL) and carnitine palmitoyltransferase‐1 (CPT‐1) activities were significantly decreased. LPL, ACC and FAS enzyme activities showed quadratic increase as dietary CP increased. Peroxisome proliferator‐activated receptor γ (PPARγ), LPL, FAS, sterol regulatory element binding protein 1 (SREBP‐1), ACC, stearoyl‐CoA desaturase (SCD) and heart fatty‐acid binding protein (H‐FABP) gene expression were significantly increased by supplementary dietary CP, while HSL and CPT‐1 gene expression were significantly decreased. PPARγ, LPL, SREBP‐1, ACC and H‐FABP gene expression showed quadratic increase as dietary CP increased. These results indicated that supplementary dietary protein increased IMF accumulation mainly to increased intramuscular lipogenic gene expression and decreased lipolytic gene expression.  相似文献

This study was conducted to estimate the effect of dietary energy level on lipid metabolism‐related gene expression of subcutaneous adipose tissue in Yellow breed × Simmental cattle. The experiment was conducted for 60 days. The results showed that final weight, average daily gain, average backfat thickness, (testicles + kidney + pelvic) fat percentage and subcutaneous fat percentage in the high and medium energy groups were significantly higher than in the low‐energy group but that the feed conversion ratio was significantly lower. The glucose, triglycerides, cholesterol, high‐density lipoprotein and low‐density lipoprotein in the high‐energy group were significantly higher than in the low‐energy group. With dietary energy increasing the activities of lipoprotein lipase (LPL), fatty acid synthase (FAS) and acetyl‐CoA carboxylase (ACC) significantly increased, whereas hormone‐sensitive lipase (HSL) and carnitine palmitoyltransferase‐1 (CPT‐1) significantly diminished. Peroxisome proliferator‐activated receptor γ (PPARγ), LPL, FAS, sterol regulatory element binding protein 1 (SREBP‐1), ACC, stearoyl‐CoA desaturase (SCD) and adipocyte‐fatty acid binding proteins (A‐FABP) gene expression were significantly increased by dietary energy increasing, and HSL and CPT‐1 gene expression were significantly decreased. These results indicated that with dietary energy increasing, the subcutaneous fat accumulation mainly increased due to adipose tissue lipogenic gene expression and decreased lipolytic gene expression.  相似文献

This study was designed to estimate dietary energy level on intramuscular fat (IMF) deposition in Simmental × Yellow breed cattle. Results showed that ultimate weight and average daily gain in high and medium energy groups were significantly higher than low‐energy group, yet feed conversion ratio was significantly lower. IMF content was significantly increased by dietary energy increasing, whereas longissimus muscle shear force significantly decreased. Serum‐free fatty acids, triglycerides and glucose significantly increased by dietary energy increasing, whereas growth hormone (GH) significantly decreased. Enzyme activities of lipoprotein lipase (LPL), fatty acid synthase (FAS), and acetyl‐CoA carboxylase (ACC) significantly increased by dietary energy increasing, whereas hormone‐sensitive lipase (HSL) and carnitine palmitoyltransferase‐1 (CPT‐1) significantly diminished. Peroxisome proliferator‐activated receptor γ, sterol regulatory element‐binding protein 1, stearoyl‐CoA desaturase, adipocyte‐fatty acid‐binding proteins, ACC, LPL, and FAS gene or protein expression significantly increased by dietary energy increasing, whereas HSL, CPT‐1, and GH gene or protein expression significantly decreased. These results indicated that high dietary energy promoting IMF deposition is mainly by downregulating pituitary GH gene expression, decreasing serum GH concentration, increasing lipogenic genes levels of mRNA, enzyme activities and protein expression, and decreasing lipolytic genes levels of mRNA, enzyme activities, and protein expression.  相似文献

Antibiotics stimulate the growth of animals but result in drug residues and bacterial resistance. In this study, the negative effect of antibiotics on abdominal fat deposition was evaluated in broilers. The results showed that adding both chlortetracycline (50 g/1,000 kg) and tylosin (50 g/1,000 kg) significantly increased abdominal fat weight, abdominal fat percentage (p < .05), and triglyceride and cholesterol levels (p < .05) in blood. Also, both products synchronously stimulated intestinal absorption and synthesis of liver fat. The expression levels of the peroxisome proliferator‐activated receptor γ (PPARγ), diacylgycerol acyltransferase 2 (DGAT2), lipoprotein lipase (LPL), and fatty acid‐binding protein (FABP4) genes in abdominal fat tissue significantly increased (p < .05 or 0.01) when antibiotics were added to the feed. However, no significant difference was found in expression of the fatty acid synthesis (FAS) or acetyl CoA carboxylase (ACC) genes. Further in vitro study results revealed that antibiotics had no effect on fat content or the related gene expression levels in preadipocytes. In summary, the antibiotics induced fat deposition in adipose tissues by activating extracellular absorption of fatty acids from intestinal absorption and synthesis of liver fat. However, it shows no direct regulation by adipose tissue.  相似文献

为系统筛选对贵州地方猪脂肪沉积具有影响的相关酶基因,本实验以雄性野猪和雌性柯乐猪的F1代杂交猪作为研究对象,采用qRT-PCR方法检测猪背最长肌和皮下脂肪组织PPARγ、LPL、ACC、ATGL基因的mRNA表达水平,并通过Pearson方法分析背最长肌和皮下脂肪组织4个基因mRNA表达水平与胴体及肉质性状间的相关性。结果表明:在背最长肌和皮下脂肪组织中相对表达量最高的为ACC和PPARγ基因。关联分析得出,背最长肌组织中LPL与PPARγ、ACC基因表达量呈显著正相关,皮下脂肪组织中PPARγ与ACC基因间呈显著正相关;在野柯F1代猪背最长肌组织中PPARγ、LPL基因表达量均与肌肉水分呈显著负相关;在皮下脂肪组织中LPL基因表达量与皮厚间呈显著负相关。本研究结果可进一步印证脂肪沉积相关酶基因LPL在柯乐猪×野猪F1代脂肪沉积过程中发挥着重要作用。  相似文献

本试验旨在研究饲粮添加锰对生长獭兔脂肪组织脂质代谢的影响。选用初始体重相近、健康状况良好的3月龄獭兔200只,随机分为5组(每组40个重复,每个重复1只),分别饲喂在基础饲粮中添加0(对照)、5、10、20、40 mg/kg锰(以硫酸锰的形式)的试验饲粮。预试期7 d,正试期29 d。结果表明:1)与对照组相比,饲粮中添加10~20 mg/kg锰显著降低了肩胛脂肪沉积率(P<0.05),饲粮中添加5~40 mg/kg锰显著降低了胃周脂肪沉积率(P<0.05)。饲粮中锰添加水平对肾周脂肪沉积率没有显著影响(P>0.05)。2)饲粮中锰添加水平对肉碱脂酰转移酶2(CPT2)、激素敏感脂酶(HSL)和过氧化物酶体增殖物激活受体β(PPARβ)基因的表达量没有显著影响(P>0.05)。与对照组相比,饲粮中添加20~40 mg/kg锰显著降低了脂肪酸合成酶(FAS)基因的表达量(P<0.05);饲粮中添加20~40 mg/kg锰显著降低了乙酰辅酶A羧化酶(ACC)基因的表达量(P<0.05);饲粮中添加10~40 mg/kg锰显著提高了肉碱脂酰转移酶1(CPT1)基因的表达量(P<0.05);饲粮中添加10~20 mg/kg锰显著提高过氧化物酶体增殖物激活受体α(PPARα)基因的表达量(P<0.05)。综上所述,饲粮中添加锰影响了3~4月龄獭兔的脂肪沉积和脂肪组织脂质代谢,锰添加水平为20 mg/kg时,可以有效促进脂肪组织的氧化分解,抑制脂肪组织的合成,并能降低体脂肪的沉积率。  相似文献

The objective of this study was to discover the expression of key genes for fatty deposition in Inner Mongolia cashmere goat,so that deeply analysize the change features of intramuscular fat deposition.Real-time PCR was used to detect the expression pattern of seven different genes (PPARγ,FTO,Lipin,LPL,HSL,A-FABP and Perilipin) in ten different muscle tissues,and in intramuscular adipocyte for three periods as early (D3),middle (D7),and end (D11),as well as the changes in key points of induced differentiation time (24,48,72 and 96 h).The correlation between the seven genes and the intramuscular fat content was researched from the prospective of analyzing the tissues and cells.The results showed that FTO gene mRNA had a higher expression in different tissues than that of other genes.While the Lipin gene mRNA expression was the lowest than others,LPL and HSL were the key enzymes in the fat synthesis and splitting,the expression trends of which were basically similar,they only showed the opposite expression pattern in the psoas major muscle,and trapezius A-FABP and Perilipin gene expression patterns were more special,the overall expression level was lower,the former's expression level was higher in next moment chest muscle than other tissues,while the latter's expression level was higher in biceps femoris muscle than other tissues.Analyzing from the cell level,the expression patterns of Lipin,PPARγ,A-FABP and Perilipin were similar,with the increase of adipocytes cultivating day age and the triglyceride accumulation,the corresponding expression level increased gradually;The expression abundance of Lipin gene in each time point was extremely low;The expression level of HSL gene reduced gradually as the differentiation degree increases;The expression level of Perilipin gene was higher only in the late period of induction and differentiation.Through statistical analysis of the correlation,the PPARγ gene mRNA of cashmere goat formed negative correlation with intramuscular fat;While other adipogenic genes like HSL,LPL,FTO,Lipin,Perilipin and A-FABP mRNA formed positive correlation with intramuscular fat content.Therefore,the key gene expression of fat deposition had the specificity,generally,the amount of gene expression by quantitative analysis was FTO> PPARγ> LPL >HSL> A-FABP >Perilipin >Lipin,such results lay foundation for further researching the fat deposition mechanism and improving the quality of cashmere goat.  相似文献

试验旨在研究脂肪沉积关键基因在内蒙古白绒山羊的表达规律,以期深入分析绒山羊肌内脂肪沉积特征.利用实时荧光定量PCR技术检测PPARγ、FTOLipinLPLHSLA-FABPPerilipin7个基因在绒山羊10个不同肌肉组织及肌内前体脂肪细胞分化的3个时期(早期(D3)、中期(D7)、末期(D11))、诱导分化关键时间点(24、48、72和96 h)的变化,分别从组织和细胞角度对其进行分析,并研究其与肌内脂肪含量的关系.结果表明,FTO基因的表达水平高于其他基因;Lipin基因的表达量最低;LPL和HSL是脂肪合成和分解的关键酶类,其表达趋势大致相同,仅仅在腰大肌和斜方肌这两个部位的表达模式相反.A-FABPPerilipin基因表达模式也较为特殊,整体表达量较低,前者仅仅在胸下矩肌中有较高表达,后者在股二头肌中有较高表达.从细胞水平分析发现Lipin、PPARγ、A-FABP和Perilipin的表达模式相似,随着前体脂肪细胞培养日龄的增加和甘油三酯的积聚,其表达量逐渐升高;Lipin基因在各个时间点的表达丰度极低;HSL基因的表达量随分化程度的升高而逐渐降低;Perilipin基因仅仅在诱导分化的后期有较高表达.相关性统计分析发现,PPARγ基因mRNA与肌内脂肪多呈负相关;而其他成脂基因HSLLPLFTOLipinPerilipinA-FABP mRNA与肌内脂肪含量多呈正相关.因此,脂肪沉积关键基因的表达具有特异性,总体上看基因表达量为FTO> PPARγ> LPL >HSL> A-FABP >Perilipin >Lipin,该结果为进一步研究脂肪沉积机制及优化山羊肉品质奠定基础.  相似文献

Intramuscular fat (IMF) in cattle is an important component of traits that influence meat quality. We measured carcass characteristics and gene expression in Korean steers to clarify the molecular mechanism(s) underlying IMF deposition in LM tissue by determining the correlation between IMF content and gene expression abundance and by developing models to predict IMF content using gene expression abundance. The deposition of IMF is determined by a balance between fat deposition and fat removal in the LM. We measured mRNA abundance of lipid metabolic genes including lipogenesis [acetyl CoA carboxylase (ACC), fatty acid synthase (FASN)], fatty lipid uptake [lipoprotein lipase (LPL), fatty acid translocase (CD36), fatty acid transport protein 1 (FATP1)], fatty acid esterification [glycerol-3-phosphate acyltransferase 1 (GPAT1), acylglycerol phosphate acyltransferase 1 (AGPAT1), diacylglycerol acyltransferase 1 (DGAT1), DGAT2], lipolysis [adipose triglyceride lipase (ATGL), hormone-sensitive lipase (HSL), monoglyceride lipase (MGL)], and fatty acid oxidation [carnitine palmitoyl transferase 1B, very long-chain acyl-CoA dehydrogenase (VLCAD), medium-chain acyl-CoA dehydrogenase (MCAD)] in the LM. The mRNA abundance of the GPAT1 gene showed the greatest correlation (r = 0.74; P < 0.001) with IMF content among 9 fat deposition genes. The gene expression abundance of other fat deposition genes including ACC, FASN, LPL, CD36, FATP1, AGPAT1, DGAT1, and DGAT2 also exhibited significant positive correlations (P < 0.05) with IMF content in the LM. Conversely, ATGL mRNA abundance showed the greatest negative correlation (r = -0.68; P < 0.001) with IMF content in the LM among 6 fat removal genes. The expression of other fat removal genes including MGL, VLCAD, and MCAD showed significant negative correlations (P < 0.05) with IMF content. Our findings show that the combined effects of increases in lipogenesis, fatty acid uptake, fatty acid esterification, and of decreases in lipolysis and fatty acid oxidation contribute to increasing IMF deposition in Korean steers. The multiple regression analysis revealed that the mRNA abundance of the GPAT1 gene in the LM was the first major variable predicting IMF content (54%) among 15 lipid metabolic genes. The second was mRNA abundance of ATGL (11%). In conclusion, these results suggest that GPAT1 and ATGL genes could be used as genetic markers to predict IMF deposition in the LM.  相似文献

本试验旨在研究饲粮添加不同水平的过瘤胃保护性精氨酸(rumen protectedarginine,RP Arg)和大豆油对细毛羊肌内脂肪和脂肪酸组成的影响。试验选用健康、体重相近的 15只细毛羊,随机分为5组,对照组饲喂基础饲粮,试验组分别饲喂在基础饲粮中添加1.5g/dRP Arg、2.0g/dRP Arg、1.5g/dRP Arg+3%大豆油和 2.0g/dRP Arg+3%大豆油的饲粮。预试期为5d,正试期为 45d。试验结束后每组选 2只羊进行屠宰,取背最长肌样用于测定肉质指标和脂肪酸组成。结果表明:试验组肌肉 pH、系水率和熟肉率与对照组差异不显著(P>0.05),而肌内脂肪含量显著高于对照组(P<0.05);试验组肌肉多不饱和脂肪酸含量与对照组相比显著提高(P<0.05);添加 RP Arg和大豆油有提高肌肉过氧化物酶体增生物激活受体 γ(PPARγ)、脂蛋白脂酶(LPL)mRNA表达量,降低激素敏感性甘油三酯脂肪酶(HSL)mRNA表达量的趋势。综合指标,以 1.5g/dRP Arg组与 1.5g/dRP Arg+3%大豆油组添加效果较好。  相似文献

The purpose of this article was to investigate the effects of dietary resveratrol supplementation during gestation and lactation of sows on the milk composition of sows and the fat metabolism of sucking piglets. Forty sows were allotted to two experimental treatment groups that included the following: (a) control sows (CON treatment, n = 20) fed with a corn–soybean meal control diet and (b) treatment sows (RES treatment, n = 20) fed with a control diet with addition of 300 mg/kg resveratrol. The results showed that the content of lactose in the colostrum was increased (p < 0.05) and the content of fat in 21‐day milk was increased (p < 0.05) by dietary resveratrol supplementation. In the RES treatment group, the concentrations of high‐density lipoprotein‐cholesterol (HDL‐C), low‐density lipoprotein‐cholesterol (LDL‐C), lipase activity and insulin (INS) in plasma of sucking piglets were increased (p < 0.05). In the adipose tissue, the enzyme activities of hormone‐sensitive lipase (HSL), acetyl‐CoA carboxylase (ACC) and lipoprotein lipase (LPL) increased significantly by RES treatment (p < 0.05), and the mRNA levels of acetyl coenzyme A‐alpha (ACCα), LPL, fatty acid transport protein (FATP1) and CCAAT–enhancer‐binding protein gene (C/EBPα) were higher in the RES treatment group (p < 0.05). In conclusion, resveratrol supplementation on gestational and lactating sows improved the content of lactose in the colostrum and the content of fat in milk at day 21 of lactation. In addition, resveratrol supplementation on sows increased HDL and LDL in the plasma of piglets. In piglet adipose tissue, the enzyme activity and mRNA level related to lipolysis, fatty acid uptake from circulating triacylglycerols and lipogenesis are partially improved by resveratrol supplementation on sows. These aspects affect fat metabolism in piglets.  相似文献

试验旨在研究染料木黄酮对3T3-L1细胞增殖与分化的影响以及对分化过程中相关基因表达的影响。以浓度为0、10、50、100、200μmol/L的染料木黄酮处理细胞,测定不同天数3T3-L1细胞相对增殖量、细胞合成脂肪量以及第8天与脂肪合成有关的基因PPARγ、FASN、LPL、ACC、HSL的相对表达量。结果表明:在增殖方面,染料木黄酮能显著抑制3T3-L1前脂肪细胞的增殖(P〈0.05)。随着染料木黄酮浓度的增加,细胞增殖的抑制作用越强。当浓度为200μmol/L时,细胞停止增殖。在分化方面,染料木黄酮能显著减少3T3-L1前脂肪细胞分化合成脂肪的量(P〈0.05)。随着染料木黄酮浓度的增加,细胞分化合成脂肪量减少。染料木黄酮能显著减少3T3-L1细胞与脂肪合成有关的基因PPARγ、FASN、LPL、ACC、HSL的相对表达量(P〈0.05)。在浓度为50μmol/L时,PPARγ基因相对表达量最低;在浓度为200μmol/L时,FASN基因相对表达量最低;在浓度为10μmol/L时,LPL基因相对表达量最低;在浓度为200μmol/L时,ACC基因相对表达量最低;在浓度为10μmol/L时,HSL基因相对表达量最低。分析推测,染料木黄酮通过抑制与脂肪合成相关基因的表达来减少脂肪合成量。  相似文献

The aim of this study was to explore the hepatic lipolysis in broiler chickens with different fat deposition during embryonic development. The mRNA expression of CPT-1 (carmitine palmtoyltransferase-1), PPARα (peroxisome proliferator-activated receptor alpha) and LPL (lipoprotein lipase) genes were determined using Real time RT-PCR. The start of incubation was called day 1 (E1) and after hatching called day 1 (H1). On incubation days 9 (E9), 14 (E14) and 19 (E19) as well as at hatching (H1), samples of liver were collected. Blood samples were obtained during days 14 (E14) and 19 (E19) of embryonic development and at hatching. This study showed that serum TG (triglycerol) decreased and TC (total cholesterol) and NEFA (non-estered fatty acid) increased during embryonic development. The expression of CPT-1, PPARα and LPL genes exhibited different developmental changes. For example, little LPL gene was expressed at hatching and PPARα gene expression peaked before hatching. However, CPT-1 gene exhibited no significance during the embryonic development. Our results showed that expression of these genes in Arbor Acres (AA) broilers was significantly higher than that in San Huang (SH) broilers. Therefore, this study suggested that hepatic lipolysis in broiler chickens exhibited developmental changes during embryogenesis and breed difference which may be one of the factors in the fat deposition difference between fat line and lean line broilers during embryonic development.  相似文献

为了研究氨氮胁迫下饥饿与复投喂对黄颡鱼脂质代谢的影响,将360尾黄颡鱼幼鱼[初始体重为(14.95±0.03)g]随机分配到12个养殖桶中,每桶30尾。对照组人工饱食投喂42 d,试验组饥饿14 d后恢复投喂28 d。对照组和试验组分别被暴露到总氨氮浓度为0(低氨氮处理,正常养殖环境)和5.70 mg/L(高氨氮处理,氨氮胁迫)的水体中,每组分配3桶试验鱼。结果显示:饥饿14 d后,高氨氮处理黄颡鱼肝脏中6-磷酸葡萄糖酸脱氢酶(6PGD)、脂肪酸合酶(FAS)活性以及6PGD、葡萄糖-6-磷酸脱氢酶(G6PD)、FAS、胆固醇调节元件结合蛋白-1(SREBP-1)、过氧化物酶体增殖物激活受体α(PPARα)和过氧化物酶体增殖物激活受体γ(PPARγ)基因的mRNA相对表达量显著低于低氨氮处理(P<0.05),而肝脏中肉碱棕榈酰转移酶(CPT)、脂蛋白脂酶(LPL)活性以及肉碱棕榈酰转移酶1(CPT1)和LPL基因的mRNA相对表达量则显著高于低氨氮处理(P<0.05);在氨氮胁迫或正常养殖环境下,饥饿14 d后,试验组黄颡鱼肝脏中6PGD、FAS活性以及6PGD、G6PD、FAS、SREBP-1和PPARγ基因的mRNA相对表达量显著低于对照组(P<0.05),而CPT、LPL活性以及PPARα、CPT1和LPL基因的mRNA相对表达量显著高于对照组(P<0.05);恢复投喂28 d后,高氨氮处理黄颡鱼肝脏中6PGD、FAS活性以及6PGD、G6PD、FAS和PPARα基因的mRNA相对表达量显著低于低氨氮处理(P<0.05),而肝脏中CPT、LPL活性以及CPT1和LPL基因的mRNA相对表达量则显著高于低氨氮组(P<0.05);在氨氮胁迫或正常养殖环境下,恢复投喂28 d后,试验组黄颡鱼肝脏中6PGD活性以及6PGD和G6PD基因的mRNA相对表达量显著高于对照组(P<0.05),而肝脏中LPL活性显著低于对照组(P<0.05);此外,在氨氮胁迫下,试验组黄颡鱼肝脏中FAS、SREBP-1和PPARγ基因的mRNA相对表达量显著高于对照组(P<0.05),而LPL基因的mRNA相对表达量则显著低于对照组(P<0.05)。综上可知,氨氮胁迫和饥饿胁迫均会促进黄颡鱼的脂质分解代谢,并抑制脂质合成代谢;氨氮胁迫下饥饿后复投喂能够恢复黄颡鱼的脂质代谢稳态。  相似文献

选择初始体重约20kg的长白×荣昌杂交猪72头,随机分为3个处理,研究日粮能量水平对生长育肥猪脂肪酸合成酶(FAS)和激素敏感酯酶(HSL)活性及基因表达量的影响。日粮消化能分别为11.75MJ/kg(低能量组)、13.05MJ/kg(中能量组)和14.36MJ/kg(高能量组)。试验猪体重达到100kg时屠宰。结果表明:与低能量组相比,中能量组和高能量组的脂肪组织、肝脏组织和肌肉组织的FAS活性显著提高(P<0.05),而HSL活性明显降低,并且FAS和HSL活性具有组织差异性,脂肪组织中活性最高,肝脏组织中的活性其次,肌肉组织中的活性最低;与低能量组相比,中能量组和高能量组的脂肪组织和肌肉组织的FASmRNA表达量均明显提高,HSLmRNA表达量均显著降低(P<0.05)。本研究结果初步表明,能量水平调控脂肪沉积可能是通过调节FAS和HSL的活性及基因表达量来实现的。  相似文献

Adiponectin is a protein hormone secreted exclusively by adipocytes that plays an important role in the modulation of glucose and lipid metabolism. To investigate the effect of adiponectin on lipid metabolism in chicken, rosiglitazone (agonist of adiponectin) and dexamethasone (inhibitor of adiponectin) were used to treat 23‐day‐old broilers in vivo. To verify the functionality of adiponectin on fat deposition, chicken pre‐adipocytes were cultured in the medium containing 10 μg/ml adiponectin. Serum adiponectin and lipids and fat distribution were analysed. Oil Red O staining was used to determine lipid deposition in adipocytes. The expression levels of adiponectin, adiponectin receptors (AdipoR) and lipid metabolism–related genes in different tissues and pre‐adipocytes were measured using real‐time PCR, and the abundance of lipid metabolism–related proteins was measured by Western blot. Rosiglitazone increased serum adiponectin concentration and the expression levels of adiponectin and adiponectin receptor 1 (AdipoR1) in tissues and significantly decreased levels of serum lipids and fat deposition. Rosiglitazone significantly increased the expression levels of adipose triglyceride lipase (ATGL) and AdipoR1 and decreased the expression levels of fatty acid synthase (FAS). Dexamethasone had the converse effects compared with rosiglitazone. Oil red O staining results showed a marked decrease in fat deposition in cells treated with adiponectin. In adipocytes, adiponectin could decrease the expression levels of CCAAT/enhancer‐binding protein α (C/EBPα) and FAS and increased the expression levels of ATGL and AdipoR1. These results indicate that adiponectin has a remarkable effect on impairment of adipocyte differentiation, which contributes to the negative regulation of fat deposition in chicken.  相似文献

本试验旨在研究长期饲喂不同蛋白质水平饲粮对猪脂肪代谢相关基因表达的影响。试验选用18头三元(杜×长×大)杂交28日龄断奶仔猪,随机分为3组,每组6个重复,每个重复1头。对照组[高粗蛋白质(HCP)组]采用符合NRC(2012)推荐营养需要的饲粮,试验组是根据NRC(2012)标准,在添加赖氨酸(Lys)、蛋氨酸(Met)、苏氨酸(Thr)、色氨酸(Try)4种必需氨基酸基础上,将饲粮蛋白质水平分别降低3%[中粗蛋白质(MCP)组]和6%[低粗蛋白质(LCP)组]。试验期125 d。结果表明:1)在肝脏中,与HCP组相比,LCP组显著降低脂肪酸合成相关基因乙酰辅酶A羧化酶(ACC)、脂肪酸合成酶(FAS)、苹果酸酶1(ME1)及锚蛋白1(ANK1)的基因表达量(P0.05);同时显著提高脂肪酸转运相关基因过氧化物酶体增殖物激活受体γ(PPARγ)及脂肪酸结合蛋白(FABP)的基因表达量(P0.05);而MCP组与HCP组相比,脂肪酸合成、转运相关基因表达量均无显著性差异(P0.05);与HCP组相比,MCP及LCP组均显著降低脂肪酸氧化分解相关基因、过氧化物酶体增殖物激活受体α(PPARα)、肉毒碱棕榈酰转移酶(CPT)的基因表达量(P0.05)。2)在背最长肌(LDM)中,MCP组中脂肪酸合成相关基因胆固醇调节元件结合蛋白(SREBP)、FAS、硬脂酰辅酶A去饱和酶(SCD)的基因表达量显著高于其他2组(P0.05);ACC及FABP在LCP组的基因表达量显著低于其他2组(P0.05);CPT基因表达量及LDM的肌内脂肪(IM F)含量在LCP及M CP组显著低于HCP组(P0.05)。3)肝脏和LDM中,各组甘油三酯脂酶(ATG L)及二烯酰辅酶A还原酶(DECR)基因表达量均无显著性差异(P0.05)。由此可见,在NRC(2012)基础上,适当的降低饲粮蛋白质水平(3%)可促进LDM中脂肪酸合成、转运相关基因的表达,但对肝脏中脂肪酸合成相关基因的表达无显著影响;适当的降低饲粮蛋白质水平(3%)可降低LDM及肝脏中脂肪酸氧化分解相关基因的表达,但并不增加LDM肌内脂肪含量。  相似文献
