开菲尔粒是一个复杂的微生物共生体系,包含很多有益生作用的微生物,其主要功能菌的分离鉴定,对进一步制备复合发酵剂具有重要的意义。因此本文研究了一种开菲尔粒的主要组成菌。通过形态学特征、生理生化分析,初步分离纯化得出:该开菲尔粒样品主要由两株酵母菌、三株乳酸菌以及两株醋酸菌构成。经16SrDNA序列分析进一步确定其种属,得出其分别为Kluyverom ycesm arxianus和Pichiakudriavzevii、Lactobacilluspontis和Lactobacilluskefiri、Acetobacterlovaniensis和Acetobactercibinongensis。  相似文献

以采集自内蒙古锡林郭勒盟正蓝旗的2份发酵酸乳样品为研究对象,采用传统纯化培养技术、16S rDNA基因序列测定并构建系统发育树的方法,对样品中的乳酸菌进行了分离鉴定,以期解析发酵乳中乳酸菌的多样性,积累食品微生物资源。研究结果显示,共分离15株乳酸菌,被鉴定为3个属9个种,3个属分别为乳酸乳球菌属、明串珠菌属和乳杆菌属,9个种分别为乳酸乳球菌叶蝉亚种(Lactococcus lactis subsp. hordniae,3株)、乳酸乳球菌(Lactococcus lactis,1株)、乳酸乳球菌乳脂亚种(Lactococcus lactis subsp. cremoris,1株)、乳明串珠菌(Leuconostoc lactis,3株)、肠膜明串珠菌肠膜亚种(Leuconostoc mesenteroides subsp. mesenteroides,1株)、肠膜明串珠菌右旋葡聚糖亚种(Leuconostoc mesenteroides subsp. dextranicum,1株)、植物乳杆菌(Lactobacillus plantarum,1株)、副干酪乳杆菌(Lactobacillus paracasei,3株)和干酪乳杆菌(Lactobacillus casei,1株)。综上表明,该研究采集的酸乳样品中乳酸菌资源丰富,是内蒙古传统乳制品优良发酵剂的研发和产业化生产的潜在微生物资源。  相似文献

通过革兰染色、菌落形态观察、生化分析鉴定、牛津杯抑菌试验、Caco-2细胞黏附试验对分离到的6株乳酸产生菌进行研究。结果显示:经16SrDNA序列测定,比对分析6株乳酸产生菌与鼠李糖乳杆菌、乳酸乳球菌乳亚种、唾液乳杆菌、乳链球菌、哥伦比亚肠球菌和鼠乳杆菌高度同源。6株乳酸菌抑菌试验检测显示:对大肠杆菌、沙门菌、蜡样芽胞杆菌和金黄色葡萄球菌具有很强的抑菌效应,以CM3菌株的抑菌效果最强。同时,这6株乳酸菌对Caco-2细胞均有一定的黏附能力。结果表明:6株番鸭肠道乳酸产生菌能够黏附于肠道模型Caco-2细胞上并具有较强的抑菌活性,但其黏附性能及保护机制还有待进一步研究。  相似文献

开菲尔是通过开菲尔粒中的细菌和酵母发酵形成的乳制品,具有特殊的风味特征。体外和动物实验显示,开菲尔中的共生菌群具有抗癌、抗突变以及抗菌等功效。本实验采用开菲尔商用发酵剂制备开菲尔,分析了不同保藏温度对开菲尔中微生物菌相的影响,发现贮藏温度控制在4℃-15℃内,20 d保质期结束后开菲尔中菌株衰减量少,稳定性较高,可保证产品最终质量。  相似文献

试验以生牛奶、自制泡菜水、青贮料、市售奶酪为样品,进行乳酸乳球菌和魏斯氏菌的筛选与鉴定。通过培养基中菌落形态观察和镜检细胞形态观察,共筛得6株疑似乳酸球菌(分别命名为ST1、ST2、ST6、ST7、ST8、ST9)。经生理生化、耐盐性、耐热性试验以及16S rD-NA序列分析鉴定,这6株菌分属两个属:ST1、ST2、ST7、ST9为乳酸乳球菌(Lactococcus lac-tis),其中ST2为乳酸乳球菌乳脂亚种(Lactococcus lactis subsp. Cremoris),ST7为乳酸乳球菌乳酸亚种(Lactococcus lactis subsp. Lactis );ST6、ST8属于魏斯氏菌属(Weissella),其中ST6为食窦魏斯氏菌(Weissella cibaria)。研究表明,生牛奶和泡菜水分别是乳酸乳球菌和魏斯氏菌的优良生活环境,传统方法与分子生物技术相结合可更准确快速地分离及鉴定菌株。  相似文献

开菲尔是一种由开菲尔粒发酵的功能性酒精性的乳制品.在开菲尔粒发酵培养期间,对外加营养元素进行单因素与正交试验,以研究其对开菲尔多糖产量的影响.结果表明:在无菌灭菌乳中添加3%麦芽糖、4%蔗糖、4%葡萄糖时,开菲尔多糖的产量最高,达到1.92 g/L;在无菌灭菌乳中添加酵母膏0.2%、蛋白胨1%、L-酪氨酸0.004%时,开菲尔多糖的产量最高,达到2.07 g/L;在无菌灭菌乳中添加0.4% KH2PO4、0.08% MgSO4·7H2O、0.02%CaCO3、0.25%NaC1时,开菲尔多糖的产量最高,达到2.01 g/L.  相似文献

从海南热带地区的香蕉茎叶中分离出3株高产酸菌,利用经典分类法即在形态学、生理生化特性等方面对其进行了细菌学鉴定,结果表明,3株菌分别为植物乳杆菌(Lactobacillus planetarium)、短乳杆菌(Lactobacillus brevis)和乳酸片球菌(P.pentosaceus)。对这3株菌产酸性能的测试结果表明,植物乳杆菌HN1和短乳杆菌HN2产酸速率大于乳酸片球菌HN3。  相似文献

从山竹岩黄芪(Hedysarum fruticosum Pall.)青贮饲料中分离到3株乳酸菌(HF84,HF69,HF49)。经传统鉴定方法及16SrRNA序列分析,菌株HF84为蒙氏肠球菌(Enterococcus mundtii),菌株HF69为乳酸乳球菌乳酸亚种(Lactococcus lactis subsp.Lactis),菌株HF49为植物乳杆菌(Lactobacillus plantarum)。3株乳酸菌均为同型发酵乳酸菌,蒙氏肠球菌在pH 3.0条件下不能生长,乳酸乳球菌乳酸亚种在pH 3.0、温度5℃时及6.5%NaCl溶液中不能生长,植物乳杆菌均可在试验设定条件下生长。通过测定3株乳酸菌的生长曲线和产酸速率,菌株HF49生长速度快、产酸性能好,可作为制备乳酸菌青贮饲料添加剂的优良菌株。  相似文献

The aim of this study was to identify lactic acid bacteria (LAB) using polymerase chain reaction (PCR) amplification of variable regions of the 16S rRNA gene. Thirteen LAB strains were isolated from the intestinal microbiota of healthy salmonids. A approximately 500-bp region of the highly conserved 16S rRNA gene was PCR-amplified and following this, a portion of the amplicon (272-bp) including the V1 and V2 variable regions was sequenced. The sequence containing both the V1 and V2 region provided strong evidence for the identification of LAB. The LAB strains were identified as Carnobacterium maltaromaticum, Lactobacillus curvatus, Lactobacillus sakei, Lactobacillus plantarum, Lactococcus lactis subsp. cremoris, Lactococcus lactis subsp. lactis, and Leuconostoc mesenteroides. The method described was found to be a very simple, rapid, specific, and low-cost tool for the identification of unknown strains of LAB.  相似文献

The present study describes the screening of five lactic acid bacteria (LAB) for use as probiotics based on their competitive adhesion and production of antagonistic substances against some fish pathogens. A reduction of adhesion of all pathogenic strains tested was obtained with three of the LAB strains (Lactococcus lactis subsp. lactis CLFP100, Lactococcus lactis subsp. cremoris CLFP102 and Lactobacillus curvatus CLFP150). With the exception of fish pathogens Flavobacterium psychrophilum and Renibacterium salmoninarum that were not inhibited by LAB strains, production of antagonistic compounds by all tested LAB was observed against at least one of the indicator strains. Based on mucus adhesion, competitive exclusion, and suppression of fish pathogen growth, the selected LAB strains can be considered for future challenge experiments in fish as a very promising alternative to the use of chemotherapeutic agents.  相似文献

采用发酵乳粘丝法初筛与发酵乳综合品质对比法复筛相结合的方法,从我国西北部三种不同来源的发酵制品中筛选了三株产粘性能良好的乳酸菌。经鉴定其中两株为嗜热链球菌,一株为乳酸乳球菌乳脂亚种。选取两株嗜热链球菌与保加利亚乳杆菌进行复配发酵试验,以市售酸奶做对照,筛选出使发酵乳粘性好、乳清析出少、感官评价好的菌株,命名为Streptococcus thermophilus ST-1。  相似文献

This study aimed to verify the occurrence of Listeria monocytogenes and Salmonella spp. in raw milk produced in Brazil. On account of the poor microbiological quality of this product, possible interference from the indigenous microbiota in these pathogens was also evaluated. Two-hundred and ten raw milk samples were collected in four important milk-producing areas in Brazil, tested for L. monocytogenes and Salmonella spp. presence, and for enumeration of indicator microorganisms: mesophilic aerobes, total coliforms and Escherichia coli. The interference of the indigenous microbiota in the isolation procedures was also tested, as well the frequency of naturally occurring raw milk strains with antagonistic activity against both pathogens. The pathogens were not isolated in any raw milk sample, but poor microbiological quality was confirmed by the high levels of indicator microorganisms. When present at high levels, the indigenous microbiota generated an evident interference in the methodologies of L. monocytogenes and Salmonella spp. isolation, mainly when the pathogens appeared at low levels. Three-hundred and sixty raw milk strains were tested for antagonistic activity against both pathogens, and 91 (25.3%) showed inhibitory activity against L. monocytogenes and 33 (9.2%) against Salmonella spp. The majority of the antagonistic strains were identified as Lactic Acid Bacteria species, mainly Lactococcus lactis subsp. lactis and Enterococcus faecium, known by antimicrobial substance production.  相似文献

采用实时荧光定量聚合酶链反应(real-time quantitive polymerase chain reaction,q-PCR)技术对从内蒙古鄂尔多斯地区采集的28份传统发酵乳制品(7份酸绵羊乳、8份酸山羊乳以及13份酸牛乳)样品中分离到的乳酸菌优势菌群L.helveticus、Leu.mesenteroides及ac.lactis subsp.lactis的数量进行比较分析.结果表明:酸山羊乳和酸牛乳中,3种优势菌属的数量关系为Leu.mesenteroides>L.helveticus> Lac.lactis subsp.lactis;酸绵羊乳中,3种优势菌属数量关系为L.helveticus> Leu.mesenteroides> Lac.lactis subsp.lactis.  相似文献

以分离自西藏那曲县罗玛镇传统发酵酸牦牛奶中的乳酸乳球菌乳脂亚种(Lactococcus lactis subsp.Cremoris)IMAU60064为试验菌株,研究其最佳的培养条件。对碳源、氮源、缓冲盐等培养基成分及培养条件进行优化,并采用响应面法对优选的碳源、氮源和缓冲盐类的组成含量进行优化,得到IMAU60064的增殖培养基为:葡萄糖23g/L、大豆蛋白胨11g/L、牛肉膏11g/L、胰蛋白胨5g/L、NaAC1.8g/L、K2HP041.2g/L、柠檬酸钠1.2g/L、MgSO4·7H200.4g/L、MnSO4·5H2O54mg/L、L-半胱氨酸盐酸盐0.5g/L、吐温80为1g/L。Lactococcus lactis subsp.cremorisIMAU60064在此增殖培养基中经30℃,14h培养活菌数可达到3.26×10^8cFu/mL,比在MRS中(6.54×10^7CFU/mL)提高近5倍。  相似文献
