Qualitative tests are used to monitor antimicrobial resistance in bacteria of animal origin in the Netherlands. Quantitative information on trends in resistance is thus not obtained. Moreover, in general a limited panel of antibiotics is tested. The present study describes resistance in zoonotic food-borne pathogens Salmonella, Campylobacter, and Escherichia coli O157 isolated from human clinical cases and from faeces of healthy food animals in 1998 and 1999, as determined with quantitative susceptibility tests. The resistance of the indicator organisms E. coli and Enterococcus faecium isolated from faecal samples of broilers and pigs randomly sampled at slaughterhouses was also determined. For this end, faecal samples from veal calves were sampled in 1996 and 1997 at the three main Dutch veal calf slaughterhouses. In 1998 only a limited number of faecal samples of veal calves were taken at farms. For E. coli and Salmonella the following antibiotics were tested: amoxicillin, amoxicillin-clavulanic acid, piperacillin, cefotaxime, ceftazidime, imipenem, gentamicin, doxycycline, trimethoprim, trimethoprim/sulphamethoxazole, ciprofloxacin, chloramphenicol, florfenicol, carbadox, and flumequine. For E. faecium the following antibiotics were tested: amoxicillin, amoxicillin-clavulanic acid, chloramphenicol, doxycycline, erythromycin, vancomycin, teicoplanin, streptomycin ('high level' > 2000 mg/ml), gentamicin ('high level' > 500 mg/ml), ciprofloxacin, bacitracin, flavofosfolipol, salinomycin, quinupristin-dalfopristin, virginiamycin, tilmicosin, avilamycin, and everninomycin. For Campylobacter the following antibiotics were tested: erythromycin, doxycycline, gentamicin, carbadox, flavofosfolipol, ciprofloxacin, trimethoprim/sulphamethoxazole, amoxicillin, and metronidazole.  相似文献

Staphylococcus aureus and coagulase negative staphylococci (CNS) were isolated from ovine and caprine mastitis milk samples originating from more than 40 Swiss farms. CNS dominated as causal microorganisms of mastitis in small ruminants. By restriction fragment length polymorphism (RFLP) analysis of the groEL gene and sequencing of 16S rDNA, various CNS species were identified, albeit certain of them predominated. For susceptibility testing of mastitis pathogens to selected antibiotics, minimal inhibitory concentrations were determined. Of the 67 S. aureus and 208 CNS strains, 31.3 % and 8.2 % were resistant to penicillin, 29.9 % and 1.0 % to ampicillin, 1.5 % and 10.6 % to erythromycin, and 3.0 % and 7.7 % to tetracycline, respectively. Moreover, 10 CNS strains (4.8 %) were resistant to oxacillin and one CNS strain to sulfamethoxazole/trimethoprim. The results obtained describe for the first time the resistance situation of mastitis pathogens from sheep and goats in Switzerland. However, accompanying and preventing measures are also of importance in mastitis control of small ruminants.  相似文献

Faecal samples from 95 healthy pigs and samples of jejunal content from 85 piglets suffering from colienterotoxaemia were tested for the presence of drug resistant E. coli strains. Practically all pigs in both groups harboured E. coli strains resistant to one or more of the 6 antibiotics/chemotherapeutic agents tested (Oxytetracycline, streptomycin, sulphaisodimidin, neomycin, ampicillin, chloramphenicol). Almost 100% of healthy and approx. 90% of diseased pigs harboured strains resistant to Oxytetracycline, streptomycin and sulphaisodimidin. Pigs with strains resistant to neomycin, ampicillin and chloramphenicol were less frequently found. The predominant coliform flora consisted of E. coli strains” resistant to Oxytetracycline, streptomycin and sulphaisodimidin in 71% to 81% of diseased pigs and in 47% to 69% of the healthy pigs. In diseased pigs ¾ of the animals had a coliform flora dominated by neomycinresistant E. coli strains.Of the 721 resistant E. coli strains isolated from healthy pigs, 11% were single resistant while the corresponding figure for the 518 resistant strains isolated from diseased pigs was 6%. Thus 89% and 94% of strains showed simultaneous resistance to 2 or more antibiotics. E. coli strains resistant to 3 or more drugs were found in approx. 60% and 70% of the isolates from healthy and diseased animals, respectively. Oxytetracycline/streptomycin/sulphaisodimidin resistance was most commonly found, approx. 22% and 38% of the strains from healthy and diseased pigs, respectively, showing this resistance pattern.Transmission of drug resistance which was examined in E. coli strains originating from the diseased pigs was demonstrated in approx. 76% of the isolates. The incidence of drug resistance transfer in single, double, triple and quadruple resistant strains was 11%, 68%, 97% and 98%, respectively.  相似文献

Two different groups of bacteria carrying genes encoding for resistance to antibiotics may be transmitted from animals to humans via food products: a.) obligate infectious agents (enteric pathogens, e.g. Salmonella enterica spp., Campylobacter spp., EHEC) and b) facultative pathogenic species (e.g. E. coli, enterococci). Thus far, it is unknown whether genes encoding for resistance to antibiotics from these bacteria may be transferred to bacteria in normal flora of the host. The transfers of genes encoding for resistance to vancomycin from animal sources to the mucosa of humans has been suggested. Thus, there is a threat that these plasmid-encoded resistance genes may also be transferred to other gram-positive organisms present in the human flora. Vancomycin is the antibiotic in reserve for treatment of infections caused by oxacillin (methicillin) resistant strains of S. aureus and by strains of pneumococcus resistant to penicillin.  相似文献

Antimicrobial susceptibility data (n = 1501) and bacterial isolates (n = 258) of important bacterial pathogens from animals were collected in collaboration with eight Swiss laboratories from May 1999 to February 2000. Using these data, the antimicrobial resistance situation could be assessed for the following bacterial species: Escherichia coli, Salmonella, Haemophilus parasuis, Actinobacillus pleuropneumoniae, Pasteurella multocida, Mannheimia haemolytica, Bordetella bronchiseptica, Campylobacter jejuni, Pseudomonas aeruginosa, Staphylococcus aureus, Staphylococcus intermedius, Streptococci, and enterococci. Differences in the distribution of resistance between animal species could be evidenced in E. coli and salmonella. Some resistance frequency data were compared with those obtained in 1980. A significant increase of resistance frequency was observed for several antibiotics. This includes in particular an increase of ampicillin, gentamicin, and cotrimoxazole resistance in E. coli. A similar increase was observed in salmonella for ampicillin, streptomycin, sulfonamides, and nalidixic acid. Staphylococci from dogs (S. intermedius and S. aureus) also presented a clear increase of resistance for penicillin, neomycin, sulfonamides, cotrimoxazole, and erythromycin. Finally, a comparison with data from abroad shows that the antibiotic resistance situation in Switzerland is relatively favorable.  相似文献

Antibiotic susceptibility of bacteria isolated from nasal swabs and lungs of pigs, to 16 commonly used antibiotics, was determined by disc diffusion test. beta-lactams showed the best activity against Streptococcus suis (S. suis) (> 99% of susceptible strains). The lowest sensitivity of S. suis was evidenced to: tylosin, tetracycline and neomycin (50%, 40% and 25%, respectively). Isolates of Escherichia coli (E. coli) demonstrated the highest susceptibility to cephalosporin (85% strains), gentamicin and norfloxacin (over 74%). The lowest susceptibility of E. coli was demonstrated to tiamulin and penicillin (11.3% and 1.9%, respectively). Over 80% of Actinobacillus pleuropneumoniae (App) strains were susceptible to all antibiotics tested. The highest resistance of App, but demonstrated by below 20% of tested isolates only, was evidenced to neomycin and LxS. Isolates of Pasteurella multocida (Pm), Haemophilus parasuis (Hps) and Arcanobacterium pyogenes (A. pyogenes) were highly susceptible to the most antibiotics included in the analysis. The comparison of the in vitro susceptibility of pathogens to the chemotherapeutics used on Polish farms for the therapy of bacterial infection of pigs within the last five years and the last 10 years, showed an increasing percent of E. coli and S. suis strains resistant to commonly used antibiotics. It is also shown that Pm, Hps, App and A. pyogenes isolates were continuously susceptible to the most chemotherapeutics applied.  相似文献

Bacteria with antimicrobial resistance can be transferred from animals to humans and may compromise antimicrobial treatment in case of infection. To determine the antimicrobial resistance situation in bacteria from Swiss veal calves, faecal samples from 500 randomly selected calves originating from 129 farms were collected at four big slaughterhouses. Samples were cultured for Escherichia coli, Enterococcus sp. and Campylobacter sp. and isolated strains were tested for antimicrobial susceptibility to selected antimicrobial agents by the minimal inhibitory concentration technique using the broth microdilution method. From 100 farms, data on farm management, animal husbandry and antimicrobial treatments of the calves were collected by questionnaire. Risk factors associated with antimicrobial resistance were identified by logistic regression. In total, 467 E. coli, 413 Enterococcus sp. and 202 Campylobacter sp. were isolated. Of those, 68.7%, 98.7% and 67.8%, respectively, were resistant to at least one of the tested antimicrobial agents. Resistance was mainly observed to antimicrobials frequently used in farm animals. Prevalence of resistance to antimicrobials important for human treatment was generally low. However, a rather high number of quinupristin/dalfopristin-resistant Enterococcus faecium and ciprofloxacin-resistant Campylobacter sp. were detected. External calf purchase, large finishing groups, feeding of milk by-products and administration of antimicrobials through feed upon arrival of the animals on the farm significantly increased the risk of antimicrobial resistance at farm level. Participation in a quality assurance programme and injection of a macrolide upon arrival of the animals on the farm had a protective effect. The present study showed that veal calves may serve as a reservoir for resistant bacteria. To ensure food safety, veal calves should be included in the national monitoring programme for antimicrobial resistance in farm animals. By improving farm management and calf husbandry the prevalence of resistance may be reduced.  相似文献

The National Resistance Monitoring of the Federal Office of Consumer Protection and Food Safety (BVL) is to determine the prevalence of resistance of bacterial pathogens from animals using a valid database. From 2004 to 2006, a total of 1,472 Pasteurella multocida strains isolated from pigs with acute respiratory tract diseases was submitted to the BVL and examined. Of these, 1,11 (75.5 %) were included in the study and tested using 24 different antimicrobial substances. The results showed that the resistance level is generally low, with the exception of the substances tetracycline, trimethoprim, and the combination trimethoprim/sulfamethoxazole. It also became clear that resistance data need to be evaluated separately for each of the animal production categories, so that a realistic figure of the current resistance level can be presented. This knowledge provides information about the resistance situation in Germany, and helps deduce the necessary management measures that must be taken to minimize resistance to antibiotics. Furthermore, it provides valuable information that can form the basis for empirical therapy, so that the National Resistance Monitoring makes an important contribution to the safety of food derived from animals and consequently aids the improvement of consumer protection.  相似文献

On a farm with therapeutic problems and unsatisfactory management conditions, the occurrence of antibiotic resistance and transferable antibiotic resistance has been studied in E. coli isolated from calves which were 5 and 30 days old. Strains with resistance to up to seven antibiotics as well as transferable resistance against up to five antibiotics were recorded. On an average, 4.2 strains with different patterns and 4.0 different strains with transferable resistance were isolated from each calf. The corresponding figures previously found for healthy control calves were 1.6 and 1.1 strains, respectively. Resistance and transferable resistance were most common against sulphonamide and penicillin.

A comparative study of investigations from 1970 (before the prohibition against therapeutically used antibiotics as food additives for growth promotion) and from 1978 respectively proves that the prevalence of young pigs and calves with resistant E. coli has increased significantly from 61% to 92% and from 69% to 86% (tables I and II). Figures 1 and 2 show the prevalence of young pigs and calves with antibiotic resistant E. coli with resistance determinants towards sulfonamides, tetracycline, streptomycin, ampicillin, chloramphenicol, neomycin and furazolidone. This shows that the prevalence of young pigs with resistance determinants towards sulfonamides, tetracycline, streptomycin, ampicillin, and chloramphenicol has increased significantly from 1970 to 1978. The same facts apply to the calves. However, the increase in tetracycline resistance is not significant. Neomycin resistant E. coli for both young pigs and calves were only found in 1978. Figure 3 shows the distribution of the number of resistance determinants per young pig and calf with resistant E. coli. The figure shows the significant tendency that the pigs and calves investigated contain resistant E. coli with more resistance determinants in 1978 than in 1970. The distribution of R-factors among resistant E. coli seems to be reduced at first sight but a closer examination establishes that this only goes for R-factor mediated sulfonamide resistance (the experimental conditions may account for this). R-factor mediated streptomycin resistance has increased while R-factor mediated tetracycline resistance was at the same level. On the basis of literature the causes and risks of an increased distribution of antibiotic resistant E. coli are discussed.  相似文献

OBJECTIVE: To compare antimicrobial susceptibility patterns of Escherichia coli isolates cultured from fecal samples from cows and calves on dairy farms that used organic (ie, no or severely limited antimicrobial use) versus conventional production methods. DESIGN: Cross-sectional study. SAMPLE POPULATION: Fecal samples from 10 cows and 10 calves on each of 30 organic dairy farms and 30 neighboring conventional dairy farms in Wisconsin. PROCEDURE: E. coli isolates obtained from the fecal samples were tested for susceptibility to 17 antimicrobials by means of a microbroth dilution test. Prevalence of antimicrobial resistance was compared between organic and conventional dairy farms. RESULTS: E. coli was isolated from 1,121 (94%) fecal samples. Farm type (organic vs conventional) and animal age (cow vs calf) were significantly associated with odds that E. coli isolates would be resistant to various antimicrobials. After controlling for age, logistic regression analyses indicated that isolates from conventional dairy farms had significantly higher rates of resistance to ampicillin, streptomycin, kanamycin, gentamicin, chloramphenicol, tetracycline, and sulfamethoxazole than did isolates from organic dairy farms. However, no significant differences were detected for the 10 other antimicrobials that were tested. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated that compared with isolates from conventional dairy farms, E. coli isolates from organic dairy herds have significantly lower prevalences of resistance to 7 antimicrobials; however, prevalence of resistance was not significantly different for 10 other antimicrobials. Resistance was more common for isolates from calves than for isolates from adult dairy cows.  相似文献

A total of 417 isolates of Escherichia coli collected from five animal species/organ system combinations from swine [urinary/genital tract (UGT) incl. mastitis metritis agalactia syndrome], horses [genital tract (GT)] and dogs/cats [respiratory tract (RT), UGT and gastrointestinal tract (GIT)] were analysed quantitatively for their susceptibility against different antimicrobial agents by determination of minimum inhibitory concentrations. Regardless of which animal species the strains originated from, resistance appeared most frequently against sulfamethoxazole (18-59%), tetracycline (14-54 %), and ampicillin (14-39%). High percentages of intermediate isolates were observed for cephalothin (39-46 %). In general, low prevalences of resistance were detected for amoxicillin/clavulanic acid (1-4%), gentamicin (1-9%), and cefazolin (0-11%). Generally speaking, the antimicrobial resistance situation among E. coli isolates from horses and small animals is relatively good.  相似文献

In the study 2004/2005, the current quantitative resistance level of Escherichia coli, Pasteurella multocida, Pseudomonas aeruginosa, Salmonella sp. and Staphylococcus aureus from chickens and turkeys was determined for the first time within the framework of the National Resistance Monitoring of the Federal Office of Consumer Protection and Food Safety (BVL).The objective was to implement a valid database on the basis of which the development and spread of resistance can be evaluated and monitored. During the investigation period from January 2004 to February 2005,927 strains were collected and 857 (92%) bacteria strains which corresponded to the specifications of the study protocol were tested with the broth microdilution method to determine the in vitro susceptibility (minimum inhibitory concentration) to 22 to 28 antimicrobial agents or antibiotic combinations. The results document a prevalence of resistance that exceeds that of bacterial pathogens of other animal species, especially in the case of tetracycline. Apart for S. aureus, clinical resistance to fluoroquinolones can still be considered low in poultry pathogens (E. coli approx. 2%). By applying the MICG of 4 mg/L for enrofloxacin, a susceptibility of approximately 78 % was calculated for S. aureus. A comparison of the prevalence of resistance between chickens and turkeys, showed that a slightly higher prevalence of resistance can be expected in turkeys. Differences between the susceptibility data of chicks and adult animals could only be found in turkeys. In the case of E. coli, the prevalence of resistance of strains isolated from adult turkeys was up to 10% higher than those isolated from chicks for the corresponding antimicrobial agents. It must be pointed out that the number of E. coli strains from adult turkeys was much higher (n = 194) than the number from turkey chicks (n = 21). The results indicate clearly that in a resistance monitoring system it is necessary to categorise poultry by animal species (chicken, turkey) as well as by production stage and type (broiler, laying hen), so that the epidemiology of resistance can be correctly represented and evaluated. This information is the basis for the development of long-term management options for minimizing antibiotic resistance. Furthermore, the knowledge of prevailing resistance levels in Germany is a valuable tool for veterinary practitioners when determining an empirical therapy. The data collected by the BVL make an important contribution to the optimisation of the safety of food from animals, and thus to improving consumer protection.  相似文献

Antimicrobial susceptibility testing was performed on a total of 581 clinical Escherichia coli isolates from diarrhea and edema disease in pigs, from acute mastitis in dairy cattle, from urinary tract infections in dogs and cats, and from septicemia in laying hens collected in Switzerland between 1999 and 2001. Among the 16 antimicrobial agents tested, resistance was most frequent for sulfonamides, tetracycline, and streptomycin. Isolates from swine presented significantly more resistance than those from the other animal species. The distribution of the resistance determinants for sulfonamides, tetracycline, and streptomycin was assessed by hybridization and PCR in resistant isolates. Significant differences in the distribution of resistance determinants for tetracycline (tetA, tetB) and sulfonamides (sulII) were observed between the isolates from swine and those from the other species. Resistance to sulfonamides could not be explained by known resistance mechanisms in more than a quarter of the sulfonamide-resistant and sulfonamide-intermediate isolates from swine, dogs and cats. This finding suggests that one or several new resistance mechanisms for sulfonamides may be widespread among E. coli isolates from these animal species. The integrase gene (intI) from class I integrons was detected in a large proportion of resistant isolates in association with the sulI and aadA genes, thus demonstrating the importance of integrons in the epidemiology of resistance in clinical E. coli isolates from animals.  相似文献

OBJECTIVE: To determine the cause of persistent resistance to chloramphenicol (CP) after the ban on its use in food-producing animals in several countries. SAMPLE POPULATION: 71 CP-resistant and 104 CP-susceptible Escherichia coli strains isolated from sick cattle and pigs in Japan. PROCEDURE: Susceptibility of all bacterial strains to thiamphenicol (TP) and florfenicol (FFC) was tested by use of an agar dilution method. The CP-resistance genes and variable region within class 1 integrons in CP-resistant strains were identified by use of a PCR assay. RESULTS: The CP acetyltransferase gene (ie, cat1) was identified as the predominant CP-resistance gene in strains isolated from cattle, and the cat1and nonenzymatic CP-resistance gene (ie, cmlA) were the predominant CP-resistance genes in strains isolated from pigs. Additionally, strains with cat1 isolated from cattle often were resistant to ampicillin, dihydrostreptomycin (DSM), oxytetracycline, and trimethoprim (TMP), whereas strains with cat1 or cmlA isolated from pigs often were resistant to DSM and TMP. Class 1 integrons were significantly more prevalent in strains with CP-resistance genes, compared with prevalence in strains without CP-resistance genes. All gene cassettes within the integrons were involved in resistance to DSM, TMP, or both. CONCLUSIONS AND CLINICAL RELEVANCE: Coresistance that develops because of the use of DSM and TMP in cattle and pigs apparently contributes to the selection of CP-resistant strains of E coli. Thus, it is possible that bacterial resistance to CP in animals would persist despite a ban on the use of CP in cattle and pigs.  相似文献

The susceptibility of Streptococcus suis strains (n=384) isolated from diseased pigs in seven European countries to 10 antimicrobial agents was determined. For that purpose a microbroth dilution method was used according to CLSI recommendations. The following antimicrobial agents were tested: ceftiofur, cefquinome, enrofloxacin, florfenicol, gentamicin, penicillin, spectinomycin, tetracycline, tilmicosin and trimethoprim/sulphamethoxazole. Using breakpoints established by CLSI for veterinary pathogens, all strains were susceptible to ceftiofur, florfenicol, enrofloxacin and penicillin. MIC-90 values of these antibiotics were < or = 0.03, 0.5, 2 and < or = 0.13 microg/mL, respectively. A low degree of resistance was observed for gentamicin (1.3%), spectinomycin (3.6%) and trimethoprim/sulphamethoxazole (6.0%). MIC-90 values of these antibiotics were 8, 16 and 2 microg/mL, respectively. A high level of resistance was observed for tetracycline (75.1%). A MIC-90 value of 64 microg/mL was found for this antibiotic. Serotype-associated differences in MIC-90 values were observed for tetracycline, tilmicosin and trimethoprim/suphamethoxazole.  相似文献

It is believed that the intensive use of antibiotics in the management of disease in pre-weaned calves contributes to high levels of antibiotic resistance in commensal and pathogenic bacteria. We described the temporal dynamics of antibiotic-susceptibility patterns seen in bovine enteric Escherichia coli in pre-weaned calves on dairy farms and dedicated calf-rearing facilities. Cohorts of 30 calves at each of six farms were sampled at 2-week intervals during the pre-weaning period. Faecal E. coli isolates were analyzed for antibiotic susceptibility to 12 antibiotics with the disk-diffusion method and grouped using cluster analysis of inhibition-zone patterns. The influences of calf age, farm-type, and individual-calf antibiotic therapy on the clusterings were assessed using stratified analyses and cumulative multinomial logistic regression using generalized estimating equation with antibiotic-resistance cluster as an ordinal-dependent variable. The model controlled for farm and cohort by a nested design and included a repeated measure on calf at each sampling occasion. E. coli from calves 2 weeks of age were more likely to be increasingly multiply resistant than E. coli from day-old calves (OR = 53.6), as were 4- and 6-week-old calves (OR = 29.8 and 16.4, respectively). E. coli from calves on dedicated calf-rearing facilities were more likely to be increasingly multiply resistant than E. coli from dairy-reared calves (OR = 2.4). E. coli from calves treated with antibiotics within 5 days prior to sampling were also more likely to be increasingly multiply resistant than E. coli from calves not exposed to individual antibiotic therapy (OR = 2.0).  相似文献

Diagnostic laboratory data on antimicrobial susceptibility of Escherichia coli isolated from feces of spring calves were evaluated retrospectively for the 5-year period from 1999 to 2003. The antimicrobials to which resistance was most prevalent were tetracycline, ampicillin, and trimethoprim/sulphamethoxazole. Resistance to 3 or more antimicrobials was found in 52.5% [95% confidence interval (CI): 47.9 to 56.6] of the E. coli isolates. Incomplete records reduced the usefulness of the diagnostic laboratory data for surveillance. Standardized patient information submitted by veterinary clinics would increase the value of this data for surveillance.  相似文献
