表达鸡传染性法氏囊病病毒VP2基因的重组火鸡疱疹病毒的构建及生物学特性分析

鸡传染性法氏囊病（IBD）是一种严重危害养禽业的高度致死性和免疫抑制性传染病。为研制IBD重组火鸡疱疹病毒（HVT）活载体疫苗,本研究构建了表达鸡传染性法氏囊病病毒（IBDV）保护性抗原VP2基因的重组HVT并对其体外生物学特性进行了分析。通过RT-PCR扩增IBDV超强毒株VP2基因并克隆入pCI载体,获得重组真核表达质粒pCI-VP2。用限制性内切酶将携带CMV启动子的VP2基因表达框架切下,连接于入门质粒pENTR,构建获得重组入门质粒pENTR-VP2。将pENTR-VP2与HVT重组黏粒H3-Kan/ccdB进行LR重组反应,构建重组表达黏粒H3-VP2。用H3-VP2与其他4个相互重叠并覆盖HVT全基因组的黏粒共同转染鸡胚成纤维细胞（CEF）,拯救获得重组病毒rHVT-VP2。将重组病毒在CEF中连续传至20代后用PCR、间接免疫荧光试验和免疫印迹试验进行检测,并绘制重组病毒体外生长曲线,分析其体外复制特性。结果表明,重组病毒rHVT-VP2能够稳定表达VP2蛋白,rHVT-VP2在CEF中的复制能力与亲本病毒无明显差异。重组病毒rHVT-VP2免疫鸡后能够诱导产生IBDV中和抗体,并对IBDV强毒株攻击引起的死亡提供90%免疫保护。重组病毒rHVT-VP2的构建为研制IBD重组HVT活载体疫苗奠定了基础,对IBD的防控具有重要意义。  相似文献

Earning report is a main accounting report that transmits earning information to users. It is the core of financial reports system. Earning information is significant to users' decision making. This paper studies the respond of stock trading volume to earning report and draws the curve of trading volume before and after earning report. This paper further does a regression analysis of accidental earning and abnormal return rate, and draws the relation curve of them on the earning report date to prove stock trading volume does have remarkable respond to earning report. This research is significant to either lessen small investors blindness investment and help them make reasonable forecast to markets and regulate the disclosure of listed companies.  相似文献   

Reference Efficiency of Planetary Gear Train

Reference efficiency of a pair of gear drive without planetary motion is the base to calculate efficiency of the whole planetary gear train. The authors find reference efficiency for speed increase is higher than that of the same drive used for speed reduction. This is different from the earlier viewpoint. New equations are derived for correctly calculating the reference efficiency of spur gear and helical gear with internal conjugate and external conjugate either in the state of speed increase or speed reduction respectively. Analysis on Longitudinal Cracks and Deflection of the Pre-stressed V-shape Slab Roof Structure

In this paper, the finite element method is used to analyze the behavior of the prestressed V-shape Slab Roof structure, especially the effect of temperature change on the stress of slab. The calculated results indicate that when the temperature changes, the tensile stress at the lower extreme fibers of the flange plate near rib exceeds the tensile strength of concrete subjected to the combined action of triaxial stresses and results in the longitudinal cracks of the flange plate. The deflection analysis is based on the mechanical model of the prestressed V shape slab with the longitudinal cracks. The calculated results are in general agreement with the field observations.  相似文献   

Behavior and Design of High-strength Bolt in Tensile Connection

The tensile strength of high strength bolt is affected by many factors, including prying force. The prying force can result in the increment of bolt force, for which the connection failed ahead of time. Several methods are presented for calculating the prying force. In the China's "Code for Design of Steel Structure" (GB50017), there is no formula for calculation of prying force and a factor of 0.8 is considered in formula for tensile strength of bolt for taking the prying force into account. It is proposed that the design value of tensile resistance for high strength bolt should be increased.  相似文献

为了不断完善小流域污染综合治理思路,进一步推进水污染治理工作,对TMDL计划进行了介绍,并在治理思路、总量分配方法及总量分配公平性、合理性等多个方面与小流域污染综合治理思路进行比较,找出差异,吸收其有益的思路和措施。通过比较得出:①小流域污染综合治理和TMDL计划在思路上是一致的,但在总量分配及分配的公平性、水质目标可达性的保证率等方面存在着一定差异;②TMDL计划中的诸多措施可为小流域污染综合治理所吸收借鉴,并以此为基础对完善小流域综合治理提出了诸多合理的建议。  相似文献

为满足现代温室对室内环境越来越高的要求，采用快速、安全、可靠、低成本的气动自动化技术设计气动天窗系统，可以实现天窗系统开闭与开度的自动化调控。为此，讨论了气动天窗系统的原理，得出了驱动气缸负载模型以及各级风速下气缸负载特性，探讨了该系统可行性和特点。与传统技术比较，气动系统完全可以取代机械式开天窗，而且结构简单，操作方便，位置控制精度高，天窗开度和通风面积显著增加，增强了利用自然通风调节温室环境的能力。  相似文献
