In order to understand the infection of Staphylococcus disease in healthy Tibetan pigs in Linzhi city,Tibet.232 samples were collected from the slaughterhouse in Bahe town,Gongbujiangda county,Linzhi city,Tibet,the teaching practice ranch at the Tibet Agriculture and Animal Husbandry University in Bayi district,Linzhi city,and the slaughterhouse in Bayi district,Linzhi city.These samples were isolated,cultured and identified,and one isolated strain was systematically studied for biological characteristics.Epidemiological survey results showed that 73 of 232 samples were positive for Staphylococcus,the positive rate was 31.47%,16 positive samples were detected in the slaughterhouse of Bahe town,Gongbujiangda county,Linzhi,Tibet,with a detection rate of 23.89%(16/67),43 positive samples were detected in the pastures of the teaching practice ranch at the Tibet Agriculture and Animal Husbandry University in Bayi district,Linzhi city,with a detection rate of 34.40% (43/125),14 positive samples were detected in the slanghterhouse in Bayi district,Linzhi city,with a detection rate of 35.00% (14/40).Through PCR identification,gene sequencing and biochemical identification,5 strains of Staphylococcus chromogenes were isolated,and one of them (Sa LZ1) was studied systematically.The biological characteristics of the isolated strains showed that the Sa LZ1 strain formed round white convex colonies on the agar medium,with regular edges,smooth surfaces,and opaque white colonies.The isolates were Gram-positive cocci bacteria.PCR sequencing results showed that the isolate had the highest homology with Staphylococcus chromogenes (GenBank accession No.:MG576253.1),positive for serum inulin,manicol,glucose,saccharose,lactose,negative for arabinose,sodium hippurate,raffinose,xylose,urea,etc,consistent with the biochemical characteristics of Staphylococcus chromogenes.The logarithmic growth period of isolated bacteria was 5 to 10 h.The pathogenicity test of mice showed that the mortality rate of mice reached 100% when the concentration of Sa LZ1 strain reached 3.5×109 CFU/mL.And the results of pathological section showed that Sa LZ1 strain could cause the visceral tissue of mice to develop pathological changes.The above results provided a theoretical basis for the prevention and control of Staphylococcus suis in Linzhi,Tibet.  相似文献

This study was aimed to isolate a mutant strain of porcine epidemic diarrhea virus and prepare a PEDV inactivated vaccine with high valence by suspension culture process for immunizing against PEDV effectively in China.200 small intestines and theirs contents of diarrhea piglets died of diarrhea,collected from many large-scale pig farms in China,were detected by RT-PCR and sequenced,a mutant strain of porcine epidemic diarrhea virus was selected and put on the suspension-cultured Vero cells in a 2 L reactor for virus isolation and continuous cell culture,the harvested virus suspension,which was identified and determined TCID50,was inactivated by formaldehyde and mixed with aluminum hydroxide gel adjuvant to prepare PEDV inactivated vaccine.After its physical behavior,stability viscosity,sterility test were checked out,the safety and immune efficacy were studied by immunizing the pregnant pigs and theirs piglets.The results were as follows:86 samples were detected positive in 200 samples,cytopathy occurred after the mutant strain samples screened were passaged to 5th generation,the virus suspension was harvested in 10th generation and identified as a mutant strain of PEDV,named PEDV-GF10 strain.The virus titer of harvested virus suspension was measured up to 1×108.0 TCID50/mL after concentrated.After the vaccine was checked out,the sows,40 and 25 days before delivery in experimental groups,were injected into Xuehai acupoint with 4 mL vaccine and the pigs in blank group were free of immunifications.The results showed that there were no obvious differences in the production status of the sows in experimental groups and blank group and the temperature of theirs 3-day-old healthy piglets injected different doses of vaccine,and the vaccine was safe to the sows and piglets.Forty 3-day-old piglets producted by pregnant sows in experimental groups and blank group were randomly selected and taken 4 mL 1×108.0TCID50/mL F10 virus culture.The PEDV morbidity of piglets in blank group was 100% after injection and the antibodies were negative;10% piglets in blank group had mild diarrhea symptoms,the protection rate was up to 90%,antibody of passive immunity in piglets lasted for more than 35 days.Virus titer of mutant strain of PEDV-GF10 improved a lot by suspension cell culture,the PEDV-GF10 inactivated vaccine was safe,and could effectively prevent and control the variation strain of PEDV in China.  相似文献

利用细胞培养方法从临床表现神经症状的死亡仔猪脑组织内分离获得1株病毒,同时对该病毒进行电镜负染观察、昆明种小鼠致病性试验、RT-PCR检测和部分基因测序分析。电镜观察可见细胞培养物中有大量的直径约115 nm的冠状病毒样粒子;接种病毒的小鼠均出现典型的神经症状,死亡率100%;血凝性脑脊髓炎病毒(HEV)特异性引物RT-PCR扩增结果阳性;其S基因序列与GenBank中登录的HEV-67 N相应基因序列的同源性高达99.6%。结果表明,分离的病毒为猪血凝性脑脊髓炎病毒,分离株暂命名为HEV-CC-2007。  相似文献

山羊痘病毒的分离鉴定及生物学特性的研究   总被引:6,自引:0,他引:6  
本研究对2003年广西部分地区山羊群发生的疑似山羊痘进行了病毒分离鉴定及生物特性的研究。取疑似山羊痘病羊的皮肤丘疹、水泡或脓泡组织的病毒悬液,接种初生羔羊睾丸细胞观察到明显的细胞病变,免疫荧光试验结果显示,病毒能与山羊痘标准阳性血清反应,在感染的细胞浆内发出特异性的黄绿色荧光。病毒悬液接种乳鼠、小鼠、豚鼠、兔子都未发病,而接种3月龄山羊则出现典型的山羊痘症状和病理变化,接种9~10日龄鸡胚绒毛尿囊膜,未见出现痘斑,连传3代,均无异常变化。通过病理组织学观察可以看到在细胞浆内有大小不一圆形或椭圆形的包涵体,在电子显微镜下可以观察到150nm~300nm大小,卵圆形、砖形,有囊膜的病毒颗粒。利用一对山羊痘病毒P32基因引物进行了PCR扩增,将所得序列与GenBank收录的5株山羊痘病毒P32基因的核苷酸及氨基酸序列比较分析。结果与疫苗株的同源性分别为99.8%和99.4%。与国外其它毒株的同源性为99.6%和98、8%~99.4%。研究结果表明,所分离的病毒为山羊痘病毒,在生物学特性上与资料记载存在一定的差异,P32基因与疫苗毒和国外毒株之间同源性非常高。将该毒株命名为山羊痘病毒LiuJiang/2003株。  相似文献

从徐州地区临床疑似鸭传染性浆膜炎发病鸭中分离到细菌21株,通过细菌形态、培养特怔、生化试验和动物试验,鉴定为鸭疫里默氏杆菌菌株,对不同地区分离到的21株菌进行了24种常用抗菌药物的药敏试验,结果该24个菌株对头孢类药物、红霉素、链霉素、妥布霉素、新霉素、氯霉素、泰乐菌素、痢特灵等几种药物敏感性较高,对喹诺酮类、强力霉素、丁安卡那霉素、林肯霉素、庆大霉素、氟苯尼考、利福平、复方新诺明等均不同程度地产生了耐药性。不同地区分离到的鸭疫里默氏杆菌对不同抗菌药的敏感程度存在较大差异性,研究结果为该地区鸭传染性浆膜炎的药物防治提供了很好的理论依据.  相似文献

本研究采用生化反应,溶血试验,致病力试验,PCR方法对分离的新疆5株绵羊致病株李斯特杆菌90SB1,90SB2,90SB5,90SS1,125SL进行了种型的鉴别测定,并用RAPD技术对这5株李斯特杆菌及标准株李斯特杆菌进行分型。试验结果,五株绵羊致病株(野毒株),在生化反应,培养特性,溶血性,致病性及溶血素基因特异性等方面表现高度的一致性,属于同一种LM。RAPD实验表明此5株LM指纹图谱相同,属于同一个型。此5株致病性LM与标准LM对照株(从食品中分离到)指纹图谱差异显著,属于不同型。  相似文献

从广东省某鸭场的病死番鸭肝脏中分离一株病毒,该病毒对雏鸭、种鸭、蛋鸭均可引起发病。接种鸭胚传至第6代时病毒对鸡红细胞的血凝性稳定,通常在72h开始致死非免疫鸭胚,死亡率为80%。经磷钨酸负染电镜观察,病毒粒子多呈现椭圆形,长轴为167~264nm,短轴为131~139nm,病毒外表有囊膜,囊膜外层有排列整齐的纤突。病毒与禽流感病毒无抗原相关性,能被禽副黏病毒I型阳性血清所中和。根据试验结果,初步将分离的病毒判定为鸭副黏病毒。  相似文献
